OBJECTIVES/SPECIFIC AIMS: We aimed to develop an assay to measure new protein synthesis after Antisense Oligonucleotide treatment, which we hypothesized to be the earliest biochemical identification of RNA-targeting therapy efficacy. METHODS/STUDY POPULATION: We treated 2 transgenic animal models expressing proteins implicated in neurodegenerative disease: human tau protein (hTau) and human superoxide dismutase 1 (hSOD1), with ASO against these mRNA transcripts. Animals received isotope-labeled 13C6-Leucine via drinking water to label newly synthesized proteins. We assayed target protein synthesis and concentration after ASO treatment to determine the earliest identification of ASO target engagement. RESULTS/ANTICIPATED RESULTS: hTau ASO treatment in transgenic mice lowered hTau protein concentration 23 days post-treatment in cortex (95% CI: 0.05%–64.0% reduction). In the same tissue, we observed lowering of hTau protein synthesis as early as 13 days (95% CI: 29.4%–123%). In hSOD1 transgenic rats, we observed lowering of 13C6-leucine-labeled hSOD1 in the cerebrospinal fluid 30 days after ASO treatment compared with inactive ASO control (95% CI: 12.0%–48.4%). DISCUSSION/SIGNIFICANCE OF IMPACT: In progressive neurodegenerative diseases, it is crucial to develop measurements that identify treatment efficacy early to improve patient outcomes. These data support the use of stable isotope labeling of amino acids to measure new protein synthesis as an early pharmacodynamics measurement for therapies that target RNA and inhibit the translation of proteins.
Postemergence Weed Control Systems Without Dinoseb for Peanuts (Arachis hypogaea)

Postemergence treatments utilizing various combinations of fluazifop-P, paraquat, and 2,4-DB were compared to a preplant-incorporated (PPI) application of benefin followed by a ground-cracking application of alachlor and dinoseb plus naptalam and a postemergence application of 2,4-DB for weed control, peanut yield, and net economic return to land, overhead, and management. The greatest peanut yields (3-yr average of 4510 kg/ha) and net returns (3-yr average of $521/ha) were provided by a postemergence system that utilized one ground-cracking and one postemergence application of paraquat and one postemergence application of fluazifop-P and 2,4-DB. Seven postemergence systems provided equivalent or greater yield and net returns than the PPI and dinoseb plus naptalam system. Fresh weight reductions of Texas panicum, sicklepod, Florida beggarweed, and pitted morningglory from postemergence weed control systems were equivalent to reductions obtained from the PPI and dinoseb plus naptalam system. The addition of paraquat and 2,4-DB to the PPI and dinoseb plus naptalam system improved the 3-yr average peanut yield and net economic return by 510 kg/ha and $136/ha, respectively, compared to the same system without paraquat and 2,4-DB.

Absorption of foliar-applied 14C-imazaquin {2-[4,5-dihydro-4-methyl-4-(1-methylethyl)-5-oxo-1H-imidazol-2-yl]-3-quinolinecarboxylic acid} 72 h after treatment was higher than 90% for soybean [Glycine max (L.) Merr. ‘Braxton’], peanut (Arachis hypogaea L. ‘Florunner’), common cocklebur (Xanthium strumarium L. # XANST), sicklepod (Cassia obtusifolia L. # CASOB), and Florida beggarweed [Desmodium tortuosum (SW.) # DEDTO]. Both symplasmic and apoplasmic translocation of the herbicide were evident. Imazaquin half-life was 4.4 days in soybean, 5.3 days in peanut, 9.6 days in Florida beggarweed, 12.7 days in sicklepod, and 39.8 days in cocklebur. Tolerance of these species to foliar-applied imazaquin as determined in greenhouse studies with 21-day-old seedlings was as follows: soybean = peanut > Florida beggarweed > sicklepod > cocklebur. Tolerance was directly correlated to imazaquin halflife within the tissue of the respective species.

Tolerance of species to foliar applications of the ethyl ester of chlorimuron as determined in greenhouse studies with 21-day-old seedlings was: soybean = peanut > prickly sida > sicklepod > Florida beggarweed > common cocklebur. Absorption of foliar-applied 14C-chlorimuron 72 h after application was similar in soybean, peanut, sicklepod, common cocklebur, and prickly sida, but much less in Florida beggarweed. Slight symplasmic and apoplasmic translocation of the herbicide was evident in all species. Metabolism of chlorimuron 72 h after application was greatest in soybean and least in common cocklebur. Species tolerance to chlorimuron was directly correlated (r2 = 0.93) to the amount of unmetabolized chlorimuron (dpm/g dry wt) in the plant. Peanut exhibited increased tolerance to chlorimuron with age; this result was attributed to reduced absorption and translocation and more extensive metabolism of the absorbed herbicide by older plants.

Field studies were conducted from 1985 to 1987 to evaluate postemergence herbicide systems with preemergence systems to control Texas panicum, Florida beggarweed, sicklepod, and pitted morningglory in peanuts. Adding paraquat at 0.14 kg ai/ha to postemergence herbicide systems reduced fresh weight of Florida beggarweed 92% (18% increase over the same systems without paraquat), sicklepod 95% (21% increase), and pitted morningglory 95% (11% increase). Herbicide systems containing paraquat improved peanut yields by 230 kg/ha and net returns by $52/ha over herbicide systems not containing paraquat. Fluazifop-P and sethoxydim systems reduced Texas panicum fresh weight (at least 96%) more than a preemergence system (92% reduction) that used benefin applied preplant incorporated and alachlor plus naptalam and dinoseb applied at cracking (GC) or a postemergence system that used alachlor and naptalam plus dinoseb GC and paraquat applied early postemergence (86% reduction). Systems containing fluazifop-P provided greater yields (4190 kg/ha) and net returns ($383/ha) than systems containing sethoxydim (4010 kg/ha, $305/ha) when averaged across all rates of application.

Imazethapyr was applied at 0.14 kg ae/ha to soybean, peanut, sicklepod. Florida beggarweed, and redroot pigweed as either a soil, foliar, or soil plus foliar application. Soybean and peanut were the most tolerant species; redroot pigweed was the most sensitive, with sicklepod and Florida beggarweed being intermediate. Foliar or foliar plus soil applications were more effective in reducing sicklepod and Florida beggarweed fresh weights than soil application alone. Foliar absorption of 14C-imazethapyr 72 h after treatment was greater than 90% for soybean, peanut, sicklepod, and redroot pigweed, but only 77% in Florida beggarweed. For the species evaluated, the amount translocated from the treated leaf ranged from 5 to 16% after 72 h. Within this same time period, an average of 90% of the root-absorbed imazethapyr had been translocated to the shoot in all species except peanut. The half-life of imazethapyr was 6.6, 6.5, 14.4, 24.0, and 32.1 days in soybean, peanut, Florida beggarweed, sicklepod, and redroot pigweed, respectively. Tolerance was most closely associated with imazethapyr half-life within these species.
Control of Rattail Fescue (Vulpia myuros) in Winter Wheat

Rattail fescue, a winter annual grass weed, has been increasing in Pacific Northwest (PNW) dryland cereal producing areas. Although rattail fescue is not a new weed species in the PNW, its incidence is expanding rapidly in circumstances where soil disturbances are minimized such as in direct seed systems. Options for effective rattail fescue control in winter wheat cropping systems have not been adequately investigated and need to be developed. Rattail fescue control with herbicide treatments was investigated in imidazolinone-resistant winter wheat using imazamox and other herbicides. Across multiple sites and two growing seasons, crop injury from herbicide treatments was minor to negligible with some exceptions. Treatments containing imazamox or mesosulfuron produced minor, transient winter wheat crop injury at some locations in some years. With the exception of flufenacet applied preemergence (PRE), control of rattail fescue in wheat was variable with single herbicide applications, but improved with sequential herbicide treatments. Rattail fescue biomass was greatly reduced by several treatments especially those containing flufenacet or from sequential herbicide application. Crop yield varied among sites due to growing season precipitation, and in some cases from rattail fescue control or herbicide related crop injury.
Design and Synthesis of Nanoparticle Contrast Agents for Spectral (color) X-Ray Imaging

Computed tomography (CT) is an important tool in clinical diagnostic imaging enabling three-dimensional anatomic imaging at high spatial resolution with short scan times. However, X-ray attenuation differences in physiological fluids and soft tissues are relatively small, requiring the use of contrast agents to achieve sufficient imaging contrast. Recent advances in energy-sensitive X-ray detectors have made spectral (color) CT commercially feasible by unmixing the energy-dependent attenuation profile of different materials and will potentially enable molecular imaging in CT. In order to leverage these capabilities for diagnostic imaging, we are developing a spectral library of nanoparticle contrast agents with K-shell absorption edges spaced at least 10 keV apart. The objective of this study was to demonstrate the ability of spectral CT to simultaneously detect up to three different contrast agents and unmixed their signals to create color images. Gadolinium oxide (Gd), hafnium oxide (Hf) and gold (Au) were chosen due to exhibiting K-edges spaced 10-20 keV apart. Core-shell nanoparticles of each composition were synthesized by various methods to have a core diameter of 15-20 nm and were coated with a silica shell at least 2-4 nm in thickness to create a common platform for surface functionalization. The contrast agents were imaged in a soft tissue equivalent phantom using source-side method for spectral CT imaging. The source-side approach utilized monochromatic synchrotron radiation at the Argonne National Laboratory which, while not clinically applicable, served as a gold standard due to providing the highest spectral resolution. The nanoparticles designed for this study have broad applications in biomedical imaging due to their modular assembly, potential for enabling multi-modal detection, and surface functionalization with biomolecules (e.g., antibodies, peptides or enzymes) for active targeting.
Hospital Room Sterilization Using Far-Ultraviolet Radiation: A Pilot Evaluation of the Sterilray Device in an Active Hospital Setting

Environmental contamination of hospital rooms is well recognized as a reservoir for highly resistant nosocomial pathogens such as methicillin-resistant Staphylococcus aureu (MRSA) and vancomycin-resistant Enterococcus (VRE), which can be transferred to patients through contact with healthcare providers and contaminated surfaces. Numerous studies dedicated to environmental cleaning and disinfection have found promising results with several novel technologies, including vaporized hydrogen peroxide and ultraviolet over-head lighting or wands. We conducted a pilot study of one such device, the Sterilray Disinfection Wand (Healthy Environment Innovations), a handheld ultraviolet (UV) room decontamination wand. The Sterilray device claims to generate UV radiation in the far-UV spectrum (185-230 nm), resulting in the rapid killing of contaminant bacteria. The goal of this pilot was to collect preliminary data on the efficacy of this device in reducing surface contamination, particularly of common nosocomial pathogens, in an active hospital setting.
