Examining pathways between genetic liability for schizophrenia and patterns of tobacco and cannabis use in adolescence

Background
It is not clear to what extent associations between schizophrenia, cannabis use and cigarette use are due to a shared genetic etiology. We, therefore, examined whether schizophrenia genetic risk associates with longitudinal patterns of cigarette and cannabis use in adolescence and mediating pathways for any association to inform potential reduction strategies.
MethodsAssociations between schizophrenia polygenic scores and longitudinal latent classes of cigarette and cannabis use from ages 14 to 19 years were investigated in up to 3925 individuals in the Avon Longitudinal Study of Parents and Children. Mediation models were estimated to assess the potential mediating effects of a range of cognitive, emotional, and behavioral phenotypes.
ResultsThe schizophrenia polygenic score, based on single nucleotide polymorphisms meeting a training-set p threshold of 0.05, was associated with late-onset cannabis use (OR = 1.23; 95% CI = 1.08,1.41), but not with cigarette or early-onset cannabis use classes. This association was not mediated through lower IQ, victimization, emotional difficulties, antisocial behavior, impulsivity, or poorer social relationships during childhood. Sensitivity analyses adjusting for genetic liability to cannabis or cigarette use, using polygenic scores excluding the CHRNA5-A3-B4 gene cluster, or basing scores on a 0.5 training-set p threshold, provided results consistent with our main analyses.
ConclusionsOur study provides evidence that genetic risk for schizophrenia is associated with patterns of cannabis use during adolescence. Investigation of pathways other than the cognitive, emotional, and behavioral phenotypes examined here is required to identify modifiable targets to reduce the public health burden of cannabis use in the population.

A new fossil site in a previously unexplored part of western Madagascar (the Beanka Protected Area) has yielded remains of many recently extinct vertebrates, including giant lemurs (Babakotia radofilai, Palaeopropithecus kelyus, Pachylemur sp., and Archaeolemur edwardsi), carnivores (Cryptoprocta spelea), the aardvark-like Plesiorycteropus sp., and giant ground cuckoos (Coua). Many of these represent considerable range extensions. Extant species that were extirpated from the region (e.g., Prolemur simus) are also present. Calibrated radiocarbon ages for 10 bones from extinct primates span the last three millennia. The largely undisturbed taphonomy of bone deposits supports the interpretation that many specimens fell in from a rock ledge above the entrance. Some primates and other mammals may have been prey items of avian predators, but human predation is also evident. Strontium isotope ratios (87Sr/86Sr) suggest that fossils were local to the area. Pottery sherds and bones of extinct and extant vertebrates with cut and chop marks indicate human activity in previous centuries. Scarcity of charcoal and human artifacts suggests only occasional visitation to the site by humans. The fossil assemblage from this site is unusual in that, while it contains many sloth lemurs, it lacks ratites, hippopotami, and crocodiles typical of nearly all other Holocene subfossil sites on Madagascar.

BACKGROUND: IGTS is a rare phenomenon of paradoxical germ cell tumor (GCT) growth during or following treatment despite normalization of tumor markers. We sought to evaluate the frequency, clinical characteristics and outcome of IGTS in patients in 21 North-American and Australian institutions. METHODS: Patients with IGTS diagnosed from 2000-2017 were retrospectively evaluated. RESULTS: Out of 739 GCT diagnoses, IGTS was identified in 33 patients (4.5%). IGTS occurred in 9/191 (4.7%) mixed-malignant GCTs, 4/22 (18.2%) immature teratomas (ITs), 3/472 (0.6%) germinomas/germinomas with mature teratoma, and in 17 secreting non-biopsied tumours. Median age at GCT diagnosis was 10.9 years (range 1.8-19.4). Male gender (84%) and pineal location (88%) predominated. Of 27 patients with elevated markers, median serum AFP and Beta-HCG were 70 ng/mL (range 9.2-932) and 44 IU/L (range 4.2-493), respectively. IGTS occurred at a median time of 2 months (range 0.5-32) from diagnosis, during chemotherapy in 85%, radiation in 3%, and after treatment completion in 12%. Surgical resection was attempted in all, leading to gross total resection in 76%. Most patients (79%) resumed GCT chemotherapy/radiation after surgery. At a median follow-up of 5.3 years (range 0.3-12), all but 2 patients are alive (1 succumbed to progressive disease, 1 to malignant transformation of GCT). CONCLUSION: IGTS occurred in less than 5% of patients with GCT and most commonly after initiation of chemotherapy. IGTS was more common in patients with IT-only on biopsy than with mixed-malignant GCT. Surgical resection is a principal treatment modality. Survival outcomes for patients who developed IGTS are favourable.

Summary
Prediction poses special difficulties, as those of us who were once confidently promised personalized rocket transportation can attest. Some predictions can be left vague, and thus conveniently reinterpretable or indefinitely deferrable. But real predictions are falsifiable, sometimes even within the compass of the predictor's own professional career. Appreciating this awkward possibility, we often succumb to the temptation to limit our predictions to mere extrapolation. Predicting only incremental change along preexisting lines may be less interesting than predicting sharp discontinuities, path dependencies, random walks, catastrophes, chaotic swirls, tipping points, symmetry breaks, cascades, emergent properties, butterfly effects, or exponential acceleration. The payoff for mere extrapolationism is in seeming levelheaded and judicious.
One crucial problem with the strategy of extrapolationism, though, is that current trends often go south surprisingly quickly. And over the truly long haul, even betting on a normally sure thing, such as increasing entropy, will at some point have no future. Most established trends in legal philosophy, it is fair to guess, will unravel long before entropy halts its advance.
Legal philosophers recognize all of this. Asked to depict likely scenarios for legal philosophy over the century hence, most would resist the temptation to limn merely a contest among, say, recognizable descendents of today's inclusive legal positivists, exclusive legal positivists, and a smattering of broadly described natural law theorists. Even if that were indeed the single most likely scenario among many alternatives, it seems too unlikely to place many chips on.

Michael Perry's The Idea of Human Rights raises important and difficult issues. One such issue, reformulated, is whether the latter half of the twentieth century has witnessed both the rise of human rights language in international law, and the erosion, if not the collapse, in the intellectual sphere of the theoretical underpinnings of human rights as traditionally understood. This is part of a broader tension, in which the advance of broadly liberal values has coexisted with increasing skepticism about the objectivity of ethics, freedom of the will and genuine moral responsibility, meaningfulness in a natural order, and the irreducibility of mind.

1. The effect of increasing concentrations of milk solids on the degree of ionisation of electrolytes cannot be measured by determinations of electrical conductivity, owing to the influence of viscosity on ionic mobility.
2. An approximately linear relationship exists between concentration of milk solids and the freezing-point depression. Above a certain concentration (equivalent to 34 g. milk solids per 100 g. of water), however, the solutions are saturated with respect to lactose, and any increase in freezing-point depression beyond this point is limited to that due to the other osmotically active constituents of milk.
3. There is no evidence of the existence of “bound” water in solution of milk solids up to concentrations equivalent to 65 g. milk solids per 100 g. water.
4. An approximately linear relationship exists between concentration of milk solids and hydrogen-ion concentration, increase in the concentration of milk solids resulting in a markedly increased acidity.
5. The applications of these results to the heat coagulation of evaporated and dried milks are briefly discussed.

In the previous paper(1) it was shown that the heat treatment to which a milk powder is subjected during manufacture may result in the production of two distinct types of protein insolubility, namely insolubility due to denaturation of the protein while still in solution, and insolubility caused by over-heating the milk powder while in a dry state. The insolubility due to denaturation was shown to be independent of the temperature of reconstitution of the milk powder. On the other hand the insolubility due to dry heating was found to vary with the temperature of reconstitution, the protein being relatively insoluble when reconstituted in water held at 20° C, but completely soluble when mechanically stirred for 30 min. in water held at 90° C. It was suggested that this latter type of insolubility might be due to the removal of water of imbibition from the protein particles, since such an explanation would account for the effect of hot water in restoring the solubility of the protein. In the previous paper, however, observations on the effect of temperature of reconstitution were limited to the two extreme temperatures, i.e. 20 and 90° C.
