Treatment-resistant depression and peripheral C-reactive protein

Background
C-reactive protein (CRP) is a candidate biomarker for major depressive disorder (MDD), but it is unclear how peripheral CRP levels relate to the heterogeneous clinical phenotypes of the disorder.
AimTo explore CRP in MDD and its phenotypic associations.
MethodWe recruited 102 treatment-resistant patients with MDD currently experiencing depression, 48 treatment-responsive patients with MDD not currently experiencing depression, 48 patients with depression who were not receiving medication and 54 healthy volunteers. High-sensitivity CRP in peripheral venous blood, body mass index (BMI) and questionnaire assessments of depression, anxiety and childhood trauma were measured. Group differences in CRP were estimated, and partial least squares (PLS) analysis explored the relationships between CRP and specific clinical phenotypes.
ResultsCompared with healthy volunteers, BMI-corrected CRP was significantly elevated in the treatment-resistant group (P = 0.007; Cohen's d = 0.47); but not significantly so in the treatment-responsive (d = 0.29) and untreated (d = 0.18) groups. PLS yielded an optimal two-factor solution that accounted for 34.7% of variation in clinical measures and for 36.0% of variation in CRP. Clinical phenotypes most strongly associated with CRP and heavily weighted on the first PLS component were vegetative depressive symptoms, BMI, state anxiety and feeling unloved as a child or wishing for a different childhood.
ConclusionsCRP was elevated in patients with MDD, and more so in treatment-resistant patients. Other phenotypes associated with elevated CRP included childhood adversity and specific depressive and anxious symptoms. We suggest that patients with MDD stratified for proinflammatory biomarkers, like CRP, have a distinctive clinical profile that might be responsive to second-line treatment with anti-inflammatory drugs.

Evidence on adherence to diet-related cancer prevention guidelines and associations with colorectal cancer (CRC) risk is limited and conflicting. The aim of this cohort analysis is to evaluate associations between adherence to the World Cancer Research Fund/American Institute of Cancer Research (WCRF/AICR) 2007 recommendations and incident CRC. The UK Women’s Cohort Study comprises over 35 372 women who filled in a FFQ at baseline in 1995. They were followed up for CRC incidence for a median of 17·4 years, an individual score linking adherence to eight of the WCRF/AICR recommendations was constructed. Cox proportional hazards regression provided hazard ratios (HR) and 95 % CI for the estimation of CRC risk, adjusting for confounders. Following exclusions, 444 CRC cases were identified. In the multivariate-adjusted model, women within the second and third (highest) categories of the WRCF/AICR score had HR of 0·79 (95 % CI 0·62, 1·00) and 0·73 (95 % CI 0·48, 1·10), respectively, for CRC compared with those in the lowest, reference category. The overall linear trend across the categories was not significant (P=0·17). No significant associations were observed between the WCRF/AICR score and proximal colon, distal colon and rectal cancers separately. Of the individual score components, a BMI within the normal weight range was borderline significantly protective only for rectal cancer in the fully adjusted model. In view of the likely different causes of CRC subtypes, further research is needed to identify the optimal dietary patterns associated with reducing colon and rectal cancer risk, respectively.
Quantifying climatic change through the last glacial–interglacial transition based on lake isotope palaeohydrology from central Turkey

Questions remain as to the nature of climatic change through the last glacial–interglacial transition in the eastern Mediterranean region, particularly the relative contribution of evaporation and precipitation to regional water balance. Here changes in oxygen isotope values through this time period from Eski Acıgöl, a crater lake in central Turkey, are investigated using hydrological and isotope mass balance models. These allow changes in evaporation and precipitation to be quantified and their relative importance evaluated. We show that it is the volumetric flux rate of water passing through the lake system and not the precipitation-to-evaporation ratio per se which controlled the stable isotope record in Eski Acıgöl. Early Holocene precipitation is shown to be much greater than that during both the latter part of the last glaciation and the present day. We test these calculated values against other records in the same region, firstly with other lake records in Anatolia, the Konya basin and Lake Van, and secondly with isotope-inferred palaeo-precipitation data from Soreq cave in Israel. This reveals a contrast between pre- and post-LGM precipitation values in Turkey (wetter and drier, respectively) and also suggests that during the last glacial–interglacial transition there was a more marked precipitation gradient than at present between northern/interior and southern/coastal parts of the eastern Mediterranean region.

Six alloys based on Cr-10Ta-7Si (by at.%) with quaternary additions of 0.5Ag, 5Ti, 1Hf, 3Mo, 3Al, or 3Re (by at.%) substituted for Cr were produced by vacuum arc-melting. The microstructures of the alloys were found to predominantly consist of a eutectic mixture of an A2 Cr-based solid solution and a C14 Cr2Ta Laves phase along with proeutectic Cr2Ta dendrites. Microstructural macro- and micro-scale inhomogeneities were observed in all alloy ingots, which were attributed to the non-equilibrium arc-melting process. The measured lattice parameters of the constituent phases and the elemental partitioning behaviour between the phases have been correlated with the respective covalent atomic radii. The bulk hardnesses of the alloys, along with the hardness of individual phases, have also been reported.

The influence of undernutrition during gestation on skeletal muscle cellularity and on the expression of genes that control muscle growth

We examined the effects of two levels of gestational undernutrition (50% and 40% of ad libitum) on postnatal growth rate, skeletal muscle cellularity and the expression of genes that control muscle growth, in the offspring at weaning. The results showed that the rat pups born to mothers fed the 50% diet during gestation and a control diet during lactation had an increased postnatal growth rate compared with the pups fed the more restricted diet (40% of ad libitum). Surprisingly, the growth rate of the control group (ad libitum) was intermediate between the 50% and 40% groups. The restricted diets did not alter the number of muscle fibres in the semitendinosus muscle of the offspring but the number of muscle nuclei was reduced by 16% in the 40% group compared with the control group. In the 50% group, the lightest pups at birth (L) had elevated muscle insulin-like growth factor (IGF)-1, IGF binding protein (BP)-5 and proliferating cell nuclear antigen (PCNA) mRNA compared with the L pups from both the control and 40% groups. The heaviest pups at birth (H) in the 50% group had increased levels of IGFBP-4, PCNA and M-cadherin mRNA compared with both the control and 40% groups. Levels of IGF-1 receptor, myostatin and MyoD mRNA did not correlate with postnatal growth. Both H and L pups from the 40% group had reduced muscle IGF-1 mRNA but all other transcripts examined were similar to control levels. The results suggest that the increased postnatal growth rate, which accompanied milder fetal undernutrition (50%), may be due to a more active local muscle IGF system and increased muscle-cell proliferation.

A range of “single source” Sr-Nb and Sr-Ta heterometal alkoxides precursors are investigated as potential sources for liquid injection MOCVD (metalorganic chemical vapour deposition) and ALD (atomic layer deposition) of SrBi2Ta2O9 (SBT) and SrBi2(TxNb1-x)2O9 (SBTN). These “single source” precursors are designed to alleviate the mis-match between conventional Sr and Ta or Sr and Nb sources. Strontium-tantalate and strontium-niobate thin films were deposited on silicon using the “single source” alkoxide precursors [Sr{Ta(OEt)5(dmae}2] and [Sr{Nb(OEt)5(dmae)}2] (dmae = OCH2CH2NMe2), and the optimum temperatures for deposition of stoichiometric SrTa2O6 and SrNb2O6 were determined. Separate ALD studies of [Sr{Ta(OEt)5(dmae)}2] and [Sr{Ta(OEt)5(mee)}2] (mee = OCH2CH2OMe) for the growth of strontium-tantalate were carried out to assess precursor suitability for this technique. Liquid injection MOCVD of Bi-oxide films using Bi(mmp)3 indicates similar decomposition behaviour to the Sr-Ta and Sr-Nb alkoxides, demonstrating its suitability as a complementary source of Bi for SBT, SBN and SBTN.

Bismuth titanate thin films have been grown by liquid injection MOCVD using Bi(mmp)3 in combination with the new Ti precursors Ti(OPri)2(mmp)2 and Ti(mmp)4 (mmp = OCMe2CH2OMe). Films were grown on Si(100) substrates over the temperature range 300 – 600°C, and were shown to consist predominantly of the Bi4Ti3O12 phase at substrate temperatures > 500°C.
