Summary
Drug development is a long and arduous process that requires many researchers at different types of institutions. These include researchers in university settings, researchers in government settings, researchers in non-profit organizations and researchers in the pharmaceutical industry. The pharmaceutical industry itself is heterogeneous, ranging from tiny biotech companies to large multi-national organizations. This chapte emphasizes drug development efforts by the pharmaceutical industry but will also make note of the many collaborations between pharma and researchers at other types of institutions.

Mesotrione Activity on Crabgrass (Digitaria spp.) as Influenced by Nitrogen Fertilization Rate, Source, and Timing

Mesotrione, a 4-hydroxyphenylpyruvate dioxygenase-inhibiting herbicide, is labeled for PRE and POST crabgrass control. It has enhanced efficacy on smooth and large crabgrass when applied in conjunction with soil-applied nitrogen (N). The objectives of this study, using crabgrass as the weed species, were to (1) determine the influence of N rate and tissue N concentration on mesotrione activity, (2) determine the influence of N source on mesotrione activity, and (3) determine the influence of N application timing on mesotrione activity. Large crabgrass plants that received 12 kg N ha−1 or more before mesotrione application had more bleached and necrotic leaves compared with plants that received 0 kg N ha−1 7 d after treatment (DAT) in the greenhouse. Although N application rates as high as 98 kg N ha−1 were tested, 90% leaf bleaching and necrosis were observed with rates of 8.9 or 10.1 kg N ha−1 in Tennessee and Indiana, respectively. Nitrogen concentration in large crabgrass leaf and stem tissue on the day of the mesotrione application was closely related to the bleaching and necrosis symptoms observed 7 DAT. Although N rate influenced mesotrione activity, N source did not. Nitrogen application timing was also important, with N applications 3, 1, and 0 d before a mesotrione application having the highest percentage of bleached and necrotic leaves in greenhouse experiments. Both greenhouse and field trials support the finding that N applications in proximity to the mesotrione application enhance herbicide activity. Thus, practitioners can pair N and POST mesotrione applications together or in proximity to enhance crabgrass control.

Time-dependent adsorption of imazethapyr was studied in the laboratory on a sandy loam soil at 16% moisture for 30 d. Soil pH was adjusted to 4.5 to 6.8. Concentration of imazethapyr in soil water declined rapidly within the first day of incubation for all soil pH levels, indicating rapid initial adsorption. However, the concentration of imazethapyr in the soil water continued to decline slowly over time. Even at pH 6.8, where batch equilibrium studies at high solution to soil ratios indicated low adsorption, only 22% of the applied imazethapyr was in solution 10 d after treatment (DAT), and 10% 30 DAT. Soil column mobility studies also showed a rapid decline in mobility as time after application increased. The amount of imazethapyr remaining in soil columns rapidly increased within 4 to 10 DAT, even at pH 7.8, where low adsorption is predicted. Imazethapyr was stable during the time of these experiments, so degradation to less soluble or more tightly bound products is not the reason for the increased adsorption. These results indicate that batch equilibrium studies that use slurries with high solution to soil ratios (often 2:1 or higher) and short equilibration times may not accurately predict mobility of acidic herbicides in the field, where moisture content is lower.

The European pine shoot moth (Rhyacionia buoliana (Schiff.)) attacks pine species grown in Michigan for forestry, Christmas tree, and ornamental purposes. Much research has been carried out on life history and direct insecticide control, (Butcher and Haynes 1960; Donley 1960) in efforts to minimize injury. As a result of these studies, practical control methods have been developed for intensively managed stands such as Christmas trees and nursery stock. While the need for a better understanding of shoot moth biology and ecology is generally accepted, the inability of foresters and entomologists to obtain satisfactory shoot moth control with low gallonage insecticide applications under forestry conditions has given impetus to studies on host preference and the influence of the host on shoot moth success and development. Such information, when available, might conceivably suggest planting and management steps that foresters could take in order to minimize attack and injury by this pest in the future. In Michigan, the European pine shoot moth overwinters as a partially grown larva and feeding is resumed about April 20. By late May or early June, pupation takes place in the bud area and adults emerge in about 2 weeks. Eggs are laid on the bark and needles, and hatching occurs within 10 to 15 days. Newly hatched larvae feed at the base of the current year's needles, later moving to the terminal and lateral buds where they remain until the following spring. Pitch blisters or “tents” formed as a result of feeding. in the bud area are useful indicators of shoot moth presence and activity.

New poly(fluorene-thiophene) alternating copolymers are described in which either the dioctylfluorene or bithiophene units in poly(9,9-dioctylfluorene-alt-bithiophene) (F8T2) are replaced by other fluorene or thiophene-based groups, respectively. Improvements in solubility are realized when the bithiophene unit of F8T2 is replaced by dihexylterthiophene or dihexylpentathiophene units. Melting temperatures are also lowered by 50 – 100°C in these polymers when compared to F8T2. Replacement of the bithiophene unit of F8T2 with a dihexylpentathiophene unit also results in a significant improvement in hysteresis (< 2 V vs. 3.5 – 5 V for F8T2). Initial results are also reported on the thermal cleavage of the C8 side groups of F8T2, which yields an insoluble polymeric semiconductor film that continues to exhibit transistor switching characteristics as part of a bottom gate device.
