Background
Response to lithium in patients with bipolar disorder is associated with clinical and transdiagnostic genetic factors. The predictive combination of these variables might help clinicians better predict which patients will respond to lithium treatment.
AimsTo use a combination of transdiagnostic genetic and clinical factors to predict lithium response in patients with bipolar disorder.
MethodThis study utilised genetic and clinical data (n = 1034) collected as part of the International Consortium on Lithium Genetics (ConLi+Gen) project. Polygenic risk scores (PRS) were computed for schizophrenia and major depressive disorder, and then combined with clinical variables using a cross-validated machine-learning regression approach. Unimodal, multimodal and genetically stratified models were trained and validated using ridge, elastic net and random forest regression on 692 patients with bipolar disorder from ten study sites using leave-site-out cross-validation. All models were then tested on an independent test set of 342 patients. The best performing models were then tested in a classification framework.
ResultsThe best performing linear model explained 5.1% (P = 0.0001) of variance in lithium response and was composed of clinical variables, PRS variables and interaction terms between them. The best performing non-linear model used only clinical variables and explained 8.1% (P = 0.0001) of variance in lithium response. A priori genomic stratification improved non-linear model performance to 13.7% (P = 0.0001) and improved the binary classification of lithium response. This model stratified patients based on their meta-polygenic loadings for major depressive disorder and schizophrenia and was then trained using clinical data.
ConclusionsUsing PRS to first stratify patients genetically and then train machine-learning models with clinical predictors led to large improvements in lithium response prediction. When used with other PRS and biological markers in the future this approach may help inform which patients are most likely to respond to lithium treatment.

Background
Studying phenotypic and genetic characteristics of age at onset (AAO) and polarity at onset (PAO) in bipolar disorder can provide new insights into disease pathology and facilitate the development of screening tools.
AimsTo examine the genetic architecture of AAO and PAO and their association with bipolar disorder disease characteristics.
MethodGenome-wide association studies (GWASs) and polygenic score (PGS) analyses of AAO (n = 12 977) and PAO (n = 6773) were conducted in patients with bipolar disorder from 34 cohorts and a replication sample (n = 2237). The association of onset with disease characteristics was investigated in two of these cohorts.
ResultsEarlier AAO was associated with a higher probability of psychotic symptoms, suicidality, lower educational attainment, not living together and fewer episodes. Depressive onset correlated with suicidality and manic onset correlated with delusions and manic episodes. Systematic differences in AAO between cohorts and continents of origin were observed. This was also reflected in single-nucleotide variant-based heritability estimates, with higher heritabilities for stricter onset definitions. Increased PGS for autism spectrum disorder (β = −0.34 years, s.e. = 0.08), major depression (β = −0.34 years, s.e. = 0.08), schizophrenia (β = −0.39 years, s.e. = 0.08), and educational attainment (β = −0.31 years, s.e. = 0.08) were associated with an earlier AAO. The AAO GWAS identified one significant locus, but this finding did not replicate. Neither GWAS nor PGS analyses yielded significant associations with PAO.
ConclusionsAAO and PAO are associated with indicators of bipolar disorder severity. Individuals with an earlier onset show an increased polygenic liability for a broad spectrum of psychiatric traits. Systematic differences in AAO across cohorts, continents and phenotype definitions introduce significant heterogeneity, affecting analyses.

The Yellow Sea region is of high global importance for waterbird populations, but recent systematic bird count data enabling identification of the most important sites are relatively sparse for some areas. Surveys of waterbirds at three sites on the coast of southern Jiangsu Province, China, in 2014 and 2015 produced peak counts of international importance for 24 species, including seven globally threatened and six Near Threatened species. The area is of particular global importance for the ‘Critically Endangered’ Spoon-billed Sandpiper Calidris pygmaea (peak count across all three study sites: 62 in spring [2015] and 225 in autumn [2014] and ‘Endangered’ Spotted Greenshank Tringa guttifer (peak count across all three study sites: 210 in spring [2014] and 1,110 in autumn [2015]). The southern Jiangsu coast is therefore currently the most important migratory stopover area in the world, in both spring and autumn, for both species. Several serious and acute threats to waterbirds were recorded at these study sites. Paramount is the threat of large-scale land claim which would completely destroy intertidal mudflats of critical importance to waterbirds. Degradation of intertidal mudflat habitats through the spread of invasive Spartina, and mortality of waterbirds by entrapment in nets or deliberate poisoning are also real and present serious threats here. Collisions with, and displacement by, wind turbines and other structures, and industrial chemical pollution may represent additional potential threats. We recommend the rapid establishment of effective protected areas for waterbirds in the study area, maintaining large areas of open intertidal mudflat, and the urgent removal of all serious threats currently faced by waterbirds here.
Antarctic Ice-Sheet Volume at 18000 Years B.P. and Holocene Sea-Level Changes at the West Antarctic Margin

The Antarctic ice sheet has been reconstructed at 18000 years b.p. by Hughes and others (in press) using an ice-flow model. The volume of the portion of this reconstruction which contributed to a rise of post-glacial eustatic sea-level has been calculated and found to be (9.8±1.5) × 106 km3. This volume is equivalent to 25±4 m of eustatic sea-level rise, defined as the volume of water added to the ocean divided by ocean area. The total volume of the reconstructed Antarctic ice sheet was found to be (37±6) × 106 km3. If the results of Hughes and others are correct, Antarctica was the second largest contributor to post-glacial eustatic sea-level rise after the Laurentide ice sheet. The Farrell and Clark (1976) model for computation of the relative sea-level changes caused by changes in ice and water loading on a visco-elastic Earth has been applied to the ice-sheet reconstruction, and the results have been combined with the changes in relative sea-level caused by Northern Hemisphere deglaciation as previously calculated by Clark and others (1978). Three families of curves have been compiled, showing calculated relative sea-level change at different times near the margin of the possibly unstable West Antarctic ice sheet in the Ross Sea, Pine Island Bay, and the Weddell Sea. The curves suggest that the West Antarctic ice sheet remained grounded to the edge of the continental shelf until c. 13000 years b.p., when the rate of sea-level rise due to northern ice disintegration became sufficient to dominate emergence near the margin predicted otherwise to have been caused by shrinkage of the Antarctic ice mass. In addition, the curves suggest that falling relative sea-levels played a significant role in slowing and, perhaps, reversing retreat when grounding lines approached their present positions in the Ross and Weddell Seas. A predicted fall of relative sea-level beneath the central Ross Ice Shelf of as much as 23 m during the past 2000 years is found to be compatible with recent field evidence that the ice shelf is thickening in the south-east quadrant.

Predictions of global changes in relative sea level caused by retreat of the Antarctic Ice Sheet from its 18,000 yr B.P. maximum to its present size are calculated numerically. When combined with the global predictions of relative sea-level change resulting from retreat of the Northern Hemisphere ice sheets, the results may be compared directly to observations of sea-level change on the Antarctic continent as well as at distant localities. The comparison of predictions to the few observations of sea-level change on Antarctica supports the view that the Antarctic Ice Sheet was larger 18,000 years ago than at present. The contribution of the Antarctic Ice Sheet to the total eustatic sea-level rise is assumed to be 25 m (25% of the assumed total eustatic rise). If as little as 0.7 m of this 25-m rise occurred between 5000 yr B.P. and the present, few mid-oceanic islands would emerge. If the Antarctic Ice Sheet attained its present dimensions by 6000 yr B.P., however, and if the volume of the ocean has remained constant for the past 5000 years, numerous islands throughout the Southern Hemisphere would emerge. It is suggested that a thorough study of Pacific islands, believed by some to have slightly emerged shorelines of Holocene age, would yield useful information about ocean volume changes during the past 5000 years, and hence on the glacial history of the Antarctic Ice Sheet.

Results of stratigraphic charcoal analysis from thin sections of varved lake sediments have been compared with fire scars on red pine trees in northwestern Minnesota to determine if charcoal data accurately reflect fire regimes. Pollen and opaque-spherule analyses were completed from a short core to confirm that laminations were annual over the last 350 yr. A good correspondence was found between fossil-charcoal and fire-scar data. Individual fires could be identified as specific peaks in the charcoal curves, and times of reduced fire frequency were reflected in the charcoal data. Charcoal was absent during the fire-suppression era from 1920 A.D. to the present. Distinct charcoal maxima from 1864 to 1920 occurred at times of fire within the lake catchment. Fire was less frequent during the 19th century, and charcoal was substantially less abundant. Fire was frequent from 1760 to 1815, and charcoal was abundant continuously. Fire scars and fossil charcoal indicate that fires did not occur during 1730–1750 and 1670–1700. Several fires occurred from 1640 to 1670 and 1700 to 1730. Charcoal counted from pollen preparations in the area generally do not show this changing fire regime. Simulated “sampling” of the thin-section data in a fashion comparable to pollen-slide methods suggests that sampling alone is not sufficient to account for differences between the two methods. Integrating annual charcoal values in this fashion still produced much higher resolution than the pollen-slide method, and the postfire suppression decline of charcoal characteristic of my method (but not of pollen slides) is still evident. Consideration of the differences in size of fragments counted by the two methods is necessary to explain charcoal representation in lake sediments.

Proglacial lakes, formed during retreat of the Laurentide ice sheet, evolved quickly as outlets became ice-free and the earth deformed through glacial isostatic adjustment. With high-resolution digital elevation models (DEMs) and GIS methods, it is possible to reconstruct the evolution of surface hydrology. When a DEM deforms through time as predicted by our model of viscoelastic earth relaxation, the entire surface hydrologic system with its lakes, outlets, shorelines and rivers also evolves without requiring assumptions of outlet position. The method is applied to proglacial Lake Oshkosh in Wisconsin (13,600 to 12,900 cal yr BP). Comparison of predicted to observed shoreline tilt indicates the ice sheet was about 400 m thick over the Great Lakes region. During ice sheet recession, each of the five outlets are predicted to uplift more than 100 m and then subside approximately 30 m. At its maximum extent, Lake Oshkosh covered 6600 km2 with a volume of 111 km3. Using the Hydrologic Engineering Center-River Analysis System model, flow velocities during glacial outburst floods up to 9 m/s and peak discharge of 140,000 m3/s are predicted, which could drain 33.5 km3 of lake water in 10 days and transport boulders up to 3 m in diameter.

A numerical model of ice-stream retreat during the Holocene period of rising sea-level has been constructed, and applied to one of the ice streams draining the Ross Sea sector of the West Antarctic ice sheet. The purpose of the experiment is to assess the relative significance of eustatic sea-level rise, the response of the solid Earth to the changing ice load, and the formation of the Ross Ice Shelf on retreat of the ice-stream grounding line to its present position in the Ross embayment. An underlying hypothesis is that marine ice sheets (such as that in West Antarctica), the ocean, and the solid Earth form a coupled dynamic system in which changes, once initiated by an external event such as rising sea-level, can proceed independently of climatic change. Ice stream E (see maps by Rose 1979) was used as a test case during development of the model.
Particle-Size Evidence for Source Areas of Charcoal Accumulation in Late Holocene Sediments of Eastern North American Lakes

Two methods of analyzing charcoal in sediment reveal changes in charcoal accumulation across temperate eastern North America during the last several hundred years. In one method the analyst counts mostly small particles that reflect regional emissions; in the other, the analyst counts only larger particles derived mostly from such local sources as catchment fires. We used these methods to compare charcoal accumulation at 14 lakes from the prairie/forest border in Minnesota to eastern Maine. The two methods gave concordant accumulation rates for sediments of pre-1850 age at each of 4 lakes analyzed by both methods. This concordance is consistent with the interpretation that pre-1850 emissions were controlled by broad-scale factors, such as climatically controlled regional differences in fuels and moisture. Since 1900 large particles decreased greatly, and small particles decreased slightly, in Minnesota and Wisconsin. By contrast in the Northeast the large particle accumulation has remained at the low values measured in pre-1900 sediments at most sites, while small particles increased everywhere east of central New York and Pennsylvania. The observed patterns suggest that (1) large particles primarily reflect local fires that were common in the Midwest before fire suppression became effective, (2) large particles were rare in the Northeast, especially before extensive land clearance, (3) small particles reflect regional combustion that increased in the Northeast after extensive use of fire for land clearance and wood burning for industrial purposes of the 19th century, and (4) small particles remain abundant in the Midwest long after effective fire suppression, probably because these well-dispersed small particles have a large source area that extends beyond the local wildfires that account for large particles before European settlement.

Principles from particle-motion physics were applied to recurring problems of the interpretation of stratigraphic charcoal data: (1) fires within catchments of lakes often produce no record in fossil-charcoal curves and (2) periods characterized by no local fire (e.g., 20th-century fire suppression) often display as much charcoal as times when local fire was frequent. Quantitative theory on source area, transport, deposition, and sampling of charcoal shows the relationship between particle sizes counted by alternative methods of charcoal analysis (pollen slides for particles 5–80 μm in diameter, petrographic thin sections for particles 50–10,000 μm diameter) and charcoal diagrams. The relationship between diameter and critical and deposition velocities results in fundamental aerodynamic differences between the sizes of particles quantified by the two methods. Charcoal recorded on pollen slides is of a size that is difficult to lift, but once entrained it remains in suspension. Thin-section charcoal is lifted at relatively low wind velocities, but it is not suspended at normal surface wind speeds. Thermal buoyancy during fire lofts charcoal above the forest canopy, depending on particle size and wind speed. Pollen-slide charcoal sizes are underrepresented near the fire, because they remain in suspension and are preferentially exported from the burn area. Thin-section charcoal is convected to lower heights on average and is deposited nearby. Following fire, thin-section charcoal is redistributed locally by wind and thus may enter lakes. Because of cohesive forces and aerodynamics, more pollen-slide charcoal remains on the ground, and less enters lakes. Source areas for pollen-slide charcoal are subcontinental to global, and diagrams of pollen-slide charcoal are biased toward nonlocal charcoal. They can be used to interpret importance of fire for broad spatial and temporal scales. Thin-section charcoal represents, the catchment fire regime. Simulation models that generate charcoal during fire, mix sediments, and then sample at specified intervals indicate that (1) in the absence of sediment mixing the pollen-slide method should consistently resolve individual fires that occur with an expectation of >30–50 yr, (2) unless samples are continuous, neither method will produce useful estimates of fire frequency, and (3) even a modest amount of sediment mixing will obscure the signal.

Reducing the delay of backend interconnects is critical in delivering improved performance in next generation computer chips. One option is to implement interlayer dielectric (ILD) materials with increasingly lower dielectric constant (k) values. Despite industry need, there has been a recent decrease in study and production of these materials in academia and business communities. We have generated a backbone and porogen system that allows us to control porosity from 0 to 60% volume, achieve k-values from 3.4 to 1.6, maintain high chemical stability to various wet cleans, and deliver uniquely high mechanical strength at a given porosity. Finite element modeling and experimental results demonstrate that further improvements can be achieved through control of the pore volume into an ordered network. With hopes to spur more materials development, this paper discusses some molecular design and nanoscale hierarchical principles relevant to making next generation low-k ILD materials.

Introduction
Red Bull Stratos was a commercial program that brought a test parachutist, protected by a full-pressure suit, in a stratospheric balloon with pressurized capsule to over 127,582 ft (38,969 m), from which he free fell and subsequently parachuted to the ground. Given that the major risks to the parachutist included ebullism, negative Gz (toe-to-head) acceleration exposure from an uncontrolled flat spin, and trauma, a comprehensive plan was developed to recover the parachutist under nominal conditions and to respond to any medical contingencies that might have arisen. In this report, the project medical team describes the experience of providing emergency medical support and crew recovery for the manned balloon flights of the program.
MethodsThe phases of flight, associated risks, and available resources were systematically evaluated.
ResultsSix distinct phases of flight from an Emergency Medical Services (EMS) standpoint were identified. A Medical Support Plan was developed to address the risks associated with each phase, encompassing personnel, equipment, procedures, and communications.
DiscussionDespite geographical, communications, and resource limitations, the medical team was able to implement the Medical Support Plan, enabling multiple successful manned balloon flights to 71,615 ft (21,828 m), 97,221 ft (29,610 m), and 127,582 ft (38,969 m). The experience allowed refinement of the EMS and crew recovery procedures for each successive flight and could be applied to other high altitude or commercial space ventures.
