Effect of hydroxylamine on the subcellular distribution of arrestin (S-antigen) in rod photoreceptors

The immunocytochemical labeling of arrestin (S-antigen) in photoreceptors of the ovine retina was examined following incubation of the retina with hydroxylamine (NH2OH), an agent known to inhibit the phosphorylation of photoactivated rhodopsin. Intact, isolated retinas bathed in medium containing 20 mM NH2OH, or in control medium lacking NH2OH, were maintained in darkness or exposed to bright light for 3 min (dark-adapted and light-adapted conditions, respectively); further incubated in darkness for 10 min; and then fixed and prepared for cryosectioning. Cryosections were incubated with anti-S-antigen monoclonal antibody MAb A2G5; with secondary antibodies that were conjugated with horseradish peroxidase; and with either 3–amino-9–ethyl carbazole or diaminobenzidine as chromogen. Anti-arrestin labeling in cryosections was then analyzed densitometrically using a light-microscopic image processing system. In dark-adapted control retinas, labeling density of the photoreceptor outer segment (OS) layer (0.061 ± 0.004; average ± S.e.m.) was less than that of the inner segment (IS) layer (0.138 ± 0.011). In light-adapted control retinas, OS labeling density (0.139 ± 0.007) exceeded IS labeling density (0.095 ± 0.005). Incubation with NH2OH eliminated this light-dependent increase in labeling of the OS relative to that of the IS, i.e. eliminated the increase in relative OS/IS labeling. Densities of labeling were 0.110 ± 0.006 (OS) and 0.183 ± 0.006 (IS) in NH2OH-treated dark-adapted retinas vs. 0.078 ± 0.004 (OS) and 0.182 ± 0.008 (IS) in NH2OH-treated light-adapted retinas. Anti-arrestin labeling was also examined in retinas that were exposed to 3 min or 13 min of bright light and then immediately fixed. Among retinas incubated in the absence of NH2OH, an increase in OS/IS labeling density was evident after 3 min of illumination, and retinas illuminated for 13 min exhibited an even larger increase in OS/IS labeling. An increase in OS/IS labeling was also exhibited by NH2OH-treated retinas that had been illuminated for 3 min; by comparison with dark-adapted NH2OH-treated controls (average value of OS/IS labeling: 0.60), OS/IS labeling in these illuminated retinas was 0.97. However, OS/IS labeling in NH2OH-treated retinas that had been illuminated for 13 min (average value: 0.35) was lower than that of the dark-adapted controls. The results indicate that, within intact rods, NH2OH inhibits the light-dependent increase in OS/IS anti-arrestin labeling that is ordinarily expressed at long times (~10 min) after major bleaching of the visual pigment. Among the possible bases for the effect of NH2OH are a reduction in the driving force for the movement of arrestin from the inner to the outer segment and/or a facilitation of the degradation of arrestin in the outer segment.

This report studies the electromigration performance of W-plug via structures under the reservoir effect. The lifetime improvement factor M was observed to be a weak function of the stressing current and approximately equal to 2. A Simple model is included in the report to explain this observation. The model also predicts the most effective reservoir length for electromigration lifetime improvement.

Electromigration lifetime dependence on crystallographic texture for AlCu interconnects is determined. It is found that enhancement of AlCu texture at <111> orientation improves EM endurance. But this beneficial effect is limited after a certain level of texture enhancement is reached. The effect of lifetime improvement is proved to result from a decrease in atomic diffusivity. Saturation of the lifetime improvement effect for highly textured AlCu indicates a change in the main diffusion mechanism for electromigration, possibly from the regular grain boundary diffusion to diffusion through edge dislocations.
Effect of Via Etch Profile and Barrier Metal on Electromigration Performance of W-Filled Via Structure in TiN/AlCu/TiN Metallization

In this study, we have evaluated electromigration performance of W-plug vias, fabricated with process variations in via hole etching as well as in via barrier deposition, on a TiN(barrier)/AlCu/TiN metallization system. We found that via etch profile scheme as well as deposition conformity for via barrier can significantly affect W-plug via lifetimes. The results indicate that the quality of contact with AlCu layer of bottom level metal is a critical factor in determining via lifetime. Precise control of via etching or by use of conformal via barrier to ensure a good barrier between W-plug and the AlCu of bottom level metal is essential for achieving reliable via structures.

As device geometries and operating voltage continue to scale while functional density increases, it is imperative to reduce the RC time delay. The replacement of Si0 2 as an intermetal dielectric with an insulator of lower dielectric constant is a particularly attractive solution since it provides immediate performance improvement through reduction in capacitance. An embedded polymer integration scheme improves the interconnect performance through line-to-line capacitance reduction by using polymer only between tightly spaced lines. The gapfill polymeric materials do not degrade the electromigration performance of standard multilayered TiN/Al/TiN interconnects. Embedded polymers alleviate many of the integration and reliability problems associated with polymer integration, and can be easily adopted into a standard production process.

The optimization of electromigration (EM) and stress-induced voiding (SV) properties of advanced interconnects impacts many critical system design parameters. In particular, the choice of materials and manufacturing processes must be carefully planned during the early phases of product development. In layered metallizations, both the barrier and capping layers design can affect electromigration resistance and stress-relaxation behavior, while electrical performance often constitutes a trade-off. This is shown specifically in a study of titanium diffusion into Al-Cu from TiN barrier layers, and in initial stress-relaxation tests characterizing the effect of Ti addition in the capping layer. The development of advanced characterization techniques supports the trial-and-error experimental optimization process, but models predicting EM and SV reliability are needed and should include complex sets of microstructural and design parameters.

The study presented in this paper establishes correlations between key passivation morphological characteristics and metal electromigration performance. This was done by using test structures including extrusion monitors which provided narrow gaps along both sides of the test stripes. The parameters studied were: (1) the nature of the gap filling (oxide process and step coverage), (2) local oxide planarization, and (3) oxide thickness above the leads tested. Failure analysis was conducted by SEM cross-sections of cracked passivation areas. We observed that in the case of aluminum metallizations capped by a thin anti-reflective coating (ARC) layer, failure occurred in all cases by aluminum extrusion from the top corners of the leads, and at 45 degree angles into the passivation. Ultimately, the lifetime was dependent on oxide thickness above the test leads rather than gap-filling characteristics.

The International Centre for Diffraction Data (1CDD), formally the Joint Committee for Powder Diffraction Standards (JCPDS), recently published a compilation of metal and alloy indexes of powder diffraction and structural data. This compilation is designed to make the identification and characterization of metallic and related materials easier and far more accurate. Systematic searches for chemical analogs and greater use of partial chemical knowledge are now possible.
