Exploring the demography and conservation needs of hawksbill sea turtles Eretmochelys imbricata in north-west Mexico

The hawksbill sea turtle Eretmochelys imbricata is categorized as Critically Endangered on the IUCN Red List and its population has declined by over 80% in the last century. The Eastern Pacific population is one of the most threatened hawksbill populations globally. Western Mexico is the northern distribution limit for hawksbill sea turtles in the Eastern Pacific and recent research indicates that the Mexican Pacific portion of the population is a separate management unit because of the restricted movements of these turtles. Here we use the most complete database of sighting records in the north-west Pacific of Mexico to identify sites where hawksbill turtles are present. We also develop a conservation index to determine the conservation status of hawksbill turtle sites. Our results demonstrate the importance of this region for juveniles and the relevance of rocky reefs and mangrove estuaries as habitats for hawksbill turtles. We identified 52 sites with records of hawksbill turtles. Most of these sites (71%) are not protected; however, sites with high conservation value included islands and coastal sites along the Baja California peninsula that are established as marine protected areas. Reefs and mangrove estuaries relevant for hawksbill turtles are probably also significant fish nursery areas that are important for local fishing communities, creating opportunities for conservation strategies that combine science, local engagement and policy to benefit both local fishing communities and hawksbill sea turtle conservation.
Prevalence of adiposity-based chronic disease and its association with anthropometric and clinical indices: a cross-sectional study

The present study aimed to determine the prevalence of adiposity-based chronic disease (ABCD) and its association with anthropometric indices in the Mexican population. A cross-sectional study was conducted in 514 adults seen at a clinical research unit. The American Association of Clinical Endocrinology/AACE/ACE criteria were used to diagnose ABCD by first identifying subjects with BMI ≥ 25 kg/m2 and those with BMI of 23–24·9 kg/m2 and waist circumference ≥ 80 cm in women or ≥ 90 cm in men. The presence of metabolic and clinical complications associated with adiposity, such as factors related to metabolic syndrome, prediabetes, type 2 diabetes, dyslipidaemia and arterial hypertension, were subsequently evaluated. Anthropometric indices related to cardiometabolic risk factors were then determined. The results showed the prevalence of ABCD was 87·4 % in total, 91·5 % in men and 86 % in women. The prevalence of ABCD stage 0 was 2·4 %, stage 1 was 33·7 % and stage 2 was 51·3 %. The prevalence of obesity according to BMI was 57·6 %. The waist/hip circumference index (prevalence ratio (PR) = 7·57; 95 % CI 1·52, 37·5) and the conicity index (PR = 3·46; 95 % CI 1·34, 8·93) were better predictors of ABCD, while appendicular skeletal mass % and skeletal muscle mass % decreased the risk of developing ABCD (PR = 0·93; 95 % CI 0·90, 0·96; and PR = 0·95; 95 % CI 0·93, 0·98). In conclusion, the prevalence of ABCD in our study was 87·4 %. This prevalence increased with age. It is important to emphasise that one out of two subjects had severe obesity-related complications (ABCD stage 2).
Prevalence and molecular characterization of Sarcoptes scabiei from vicuñas (Vicugna vicugna) from Southern Peruvian Andes

Sarcoptic mange is a disease caused by an infectious parasite in the vicuñas (Vicugna vicugna) from South America. Although molecular studies have provided much information about the epidemiology of this disease, this information is still unknown in vicuñas. This study determined the prevalence and molecular characterization of Sarcoptes scabiei from vicuñas from Southern Peruvian Andes. During the 2018 shearing season, 181 vicuñas were clinically evaluated for lesions compatible with mange. Sarcoptes scabiei was detected in 35 (19.3%) vicuñas, and 50 mites from 25 vicuñas were selected for molecular analyses of the mitochondrial (cox1) and nuclear (ITS2) genetic markers. Molecular analyses of the cox1 and ITS2 sequences showed an identity of 94–99% and 99.8–100% with previous S. scabiei sequences registered in the GenBank, respectively. Sequence polymorphisms were more evident in the ITS2 than in the cox1, but only the cox1 had an association with the host. Phylogenetic analysis of S. scabiei cox1 sequences from vicuñas showed a cluster with S. scabiei cox1 sequences from canids, suggesting that the origin of S. scabiei from vicuña is associated with canid mites. This research is the first molecular analysis of S. scabiei from vicuñas. Future molecular studies will be necessary to determine the species variety, geographic segregation and host–parasite adaptation for this vicuña's mite.
Effect of the BCAT2 polymorphism (rs11548193) on plasma branched-chain amino acid concentrations after dietary intervention in subjects with obesity and insulin resistance

Branched-chain amino acids (BCAA) are considered markers of insulin resistance (IR) in subjects with obesity. In this study, we evaluated whether the presence of the SNP of the branched-chain aminotransferase 2 (BCAT2) gene can modify the effect of a dietary intervention (DI) on the plasma concentration of BCAA in subjects with obesity and IR. A prospective cohort study of adult subjects with obesity, BMI ≥ 30 kg/m2, homeostatic model assessment-insulin resistance (HOMA-IR ≥ 2·5) no diagnosed chronic disease, underwent a DI with an energy restriction of 3140 kJ/d and nutritional education for 1 month. Anthropometric measurements, body composition, blood pressure, resting energy expenditure, oral glucose tolerance test results, serum biochemical parameters and the plasma amino acid profile were evaluated before and after the DI. SNP were assessed by the TaqMan SNP genotyping assay. A total of eighty-two subjects were included, and fifteen subjects with a BCAT2 SNP had a greater reduction in leucine, isoleucine, valine and the sum of BCAA. Those subjects also had a greater reduction in skeletal muscle mass, fat-free mass, total body water, blood pressure, muscle strength and biochemical parameters after 1 month of the DI and adjusting for age and sex. This study demonstrated that the presence of the BCAT2 SNP promotes a greater reduction in plasma BCAA concentration after adjusting for age and sex, in subjects with obesity and IR after a 1-month energy-restricted DI.

Ladle refining plays a key role in achieving the quality of the steel since in this reactor temperature and chemical composition is adjusted, elimination of non-metallic inclusions is performed, and also deoxidation and desulphurization are operations taking place in the refining process. Specifically, the metal-slag mass exchanges have not received much attention through scientific studies. In this work, a rigorous study on the mass exchange between metal and slag is presented through a scaled water physical model. In the model, thymol (playing the role of a solute such as sulfur) is added to the water (playing the role of steel) and silicon oil (playing the role of slag) picks up the thymol, while the ladle is agitated with the central injection of gas. The evolution of thymol concentration in time was measured. Also, a mathematical model was developed and cast into the commercial CFD code Fluent Ansys to represent the fluid flow phenomena and the mass transfer through the solution of the continuity equation, the turbulent momentum conservation equations and the species mass conservation equation. There is a good agreement between the measured and the computed results regarding the thymol concentration evolution in water and consequently the mathematical model was validated regarding the mass species metal-slag exchanges and it may be used to study metal-slag exchanges in the steel ladle such as deoxidation or desulphurization.

The prohibition of commercial mineral resource extraction through the Antarctic Treaty System has removed one significant source of potential damage to Antarctica's geological and geomorphological values. However, given the on-going increase in Antarctic tourism and scientific footprint, some high-quality geological features may be vulnerable to human impact, such as damage due to the construction of logistical facilities, unregulated collection of geological specimens or oversampling for scientific purposes. The Protocol on Environmental Protection to the Antarctic Treaty puts in place a framework for the protection of Antarctica's environmental, scientific, historic, wilderness and aesthetic values. However, the Antarctic Protected Area system is still immature and further implementation of existing management tools may be required to protect the diverse range of vulnerabilities, qualities and spatial scales represented in the geology and geomorphology of the continent. At sites where high-quality mineralogical or palaeontological specimens exist in limited quantities, considerations of how best to prevent oversampling and manage access to remaining material may be supported by assessment of cumulative impacts. Examination of the level of Antarctic specimen loans from a selection of national geological collections suggested that existing publically accessible geological collections could be better utilized, which could reduce environmental impact and oversampling at vulnerable Antarctic sites.

The glycolytic enzyme triosephosphate isomerase catalyses the isomerization between glyceraldehyde 3-phosphate and dihydroxyacetone phosphate. Here we report that Trichomonas vaginalis contains 2 fully functional tpi genes. Both genes are located in separated chromosomal context with different promoter regulatory elements and encode ORFs of 254 amino acids; the only differences between them are the character of 4 amino acids located in α-helices 1, 2 and 8. Semi-quantitative RT-PCR assays showed that tpi2 transcript is approximately 3·3-fold more abundant than tpi1. Using an anti-TvTIM2 polyclonal antibody it was demonstrated that TIM proteins have a cytoplasmic localization and both enzymes are able to complement an Escherichia coli strain carrying a deletion of its endogenous tpi gene. Both TIM proteins assemble as dimers and their secondary structure assessment is essentially identical to TIM from Saccharomyces cerevisiae. The kinetic catalytic constants of the recombinant enzymes using glyceraldehyde-3-phosphate as substrate are similar to the catalytic constants of TIMs from other organisms including parasitic protozoa. As T. vaginalis depends on glycolysis for ATP production, we speculate 2 possible reasons to maintain a duplicated tpi copy on its genome: an increase in gene dosage or an early event of neofunctionalization of TIM as a moonlighting protein.

We determined the rate of nosocomial viral respiratory infection in infants and the effect of an infection control program during 4 winter seasons. The rate of nosocomial viral respiratory infection decreased from 6.09 episodes per 100 patients admitted during the first study year to 1.46 episodes per 100 patients admitted during the last study year.
Biocompatibility, Bioactivity and Mechanical Properties of Portland Cement and Portland Cement-Metakaolin Blends for Bone Tissue Engineering Applications

We studied the potential applications of Portland cement and Portland cement-Metakaolin blends as scaffolding materials for load bearing bone tissue engineering. Cementitious pastes were prepared by mixing Portland cement and Metakaolin at different ratios (100:0, 85:15), and hydrated under a concentrated CO2 atmosphere (carbonated pastes). Pastes fabricated similarly, but hydrated under normal atmospheric conditions were used for comparison (non-carbonated pastes). Compressive tests were run to evaluate the mechanical properties of the pastes. The bioactivity of the samples was tested in a simulated body fluid (SBF) solution for 1 and 4 days. Sample morphology and chemistry were characterized via scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS), respectively. The cytocompatibility was studied using human osteosarcoma (HOS) cell cultures and the direct contact assay. Mechanical characterization did not show significant differences in the compressive strength of the blends compared to pure cement controls. The bioactivity test revealed that the pastes induced surface precipitation of calcium phosphate (CaP) when exposed to the SBF solution (as confirmed by SEM and EDS). Non-carbonated pastes induced early CaP precipitation. Cytocompatibility experiments showed that the carbonated blends allowed adequate cell growth. Non-carbonated blends presented a highly cytotoxic behavior. The introduction of Metakaolin did not affect the cytocompatibility of the samples. These results show that Portland cement and Portland cement-Metakaolin blends could present suitable characteristics for applications as scaffolding materials in load bearing bone tissue engineering.
Pre-penetration events in fungal parasitism of nematode eggs

The present investigation deals with the main factors involved in early infection of nematode eggs by fungal parasites. We studied the effect of hydrophobicity on appressorium formation by germlings of Pochonia rubescens (syn. Verticillium suchlasporium), P. chlamydosporia (syn. V. chlamydosporium) and Lecanicillium lecanii (syn. V. lecanii). Appressoria were frequently formed on hydrophobic surfaces such as polyvinyl chloride or polystyrene and were infrequently formed on hydrophilic materials such as glass or aluminium. Infected eggs probed with the FITC-labelled lectin Concanavalin-A showed intense labelling corresponding to appressoria formed by fungal parasites on the eggshell surface. Proteolytic activity was found in extracts from conidia and germlings of fungal parasites (especially P. chlamydosporia) in the absence of nematode eggs. Addition of the serine proteinase inhibitors phenylmetylsulphonyl fluoride (PMSF) or diisopropyl fluorophosphonate (DFP) to the extracts reduced their proteolytic activity. PMSF was the most effective inhibitor. Zymography also revealed proteolytic activity in extracts from the three fungi tested. This activity mostly corresponded to bands of Rf's of substrate degradation similar to that of purified main protease (P32) from P. rubescens. Other bands with molecular weight higher than P32 (low Rf) were found especially for P. chlamydosporia extracts. For L. lecanii only bands of low Rf were found. Serum anti-P32 partially inhibited proteolytic activity of extracts from conidia and germlings. Application of PMSF and DFP to the inoculum, reduced egg penetration for the three species studied. PMSF caused the highest reduction in eggs infected by L. lecanii, while DFP significantly reduced egg infection by both P. chlamydosporia and L. lecanii. Our results therefore show hydrophobicity, appresorium formation and protease production as factors involved in early parasitism of nematode eggs.
