The impact of COVID-19 on disease epidemiology, family dynamics, and social justice in Minnesota: All that you cannot see

Objective:
The COVID-19 pandemic presented a challenge to established seed grant funding mechanisms aimed at fostering collaboration in child health research between investigators at the University of Minnesota (UMN) and Children’s Hospitals and Clinics of Minnesota (Children’s MN). We created a “rapid response,” small grant program to catalyze collaborations in child health COVID-19 research. In this paper, we describe the projects funded by this mechanism and metrics of their success.
Methods:Using seed funds from the UMN Clinical and Translational Science Institute, the UMN Medical School Department of Pediatrics, and the Children’s Minnesota Research Institute, a rapid response request for applications (RFAs) was issued based on the stipulations that the proposal had to: 1) consist of a clear, synergistic partnership between co-PIs from the academic and community settings; and 2) that the proposal addressed an area of knowledge deficit relevant to child health engendered by the COVID-19 pandemic.
Results:Grant applications submitted in response to this RFA segregated into three categories: family fragility and disruption exacerbated by COVID-19; knowledge gaps about COVID-19 disease in children; and optimizing pediatric care in the setting of COVID-19 pandemic restrictions. A series of virtual workshops presented research results to the pediatric community. Several manuscripts and extramural funding awards underscored the success of the program.
Conclusions:A “rapid response” seed funding mechanism enabled nascent academic-community research partnerships during the COVID-19 pandemic. In the context of the rapidly evolving landscape of COVID-19, flexible seed grant programs can be useful in addressing unmet needs in pediatric health.

Objectives: Glioblastoma is a lethal disease in the elderly population. We aimed to evaluate disease and treatment outcomes in the oldest-old patients. Methods: Patients >80 years old with histologically confirmed glioblastoma treated between 2004 and 2009 were identified. We included patients managed with best supportive care (BSC), temozolomide (TMZ) alone, radiotherapy (RT) alone, or concomitantly with TMZ (CRT). Survival outcomes were analyzed using the Kaplan–Meier method. Results: Ultimately, 48 patients were analyzed. Median age and Eastern Cooperative Oncology Group (ECOG) Performance Status were 82 years and 2, respectively. The median Age-Adjusted Charlson Index (AAC) was 6. Gross total and subtotal resections were performed in 16.7% and 18.8% of patients, respectively. Biopsy followed by RT alone was the treatment modality for 23/48 (47.9%), while 17/48 (35.4%) received surgery followed by RT alone or CRT. A total of 8 (16.7%) were managed with BSC after biopsy. Median overall survival (OS) and progression-free survival (PFS) were 4.1 (95% confidence interval [95% CI] 3.3-4.9) and 2.7 (95% CI 1.5-3.9) months, respectively. Improved median OS was observed in those treated with surgical resection followed by RT alone or CRT (7.1 months), compared to biopsy followed by RT alone (4.2 months) or BSC (2.0 months; p=0.002). Surgical resection, age≤85, and AAC<6 were associated with better OS (p=0.032, p=0.031, and p=0.02, respectively). Cause of death was neurological progression in 56% of cases. RT was well-tolerated. Conclusions: PFS and OS outcomes remain poor in the oldest-old patients (>80 years old). Younger age, lower AAC, surgical resection, and adjuvant treatment were associated with improved OS.

First Nearctic record of Diolcogaster claritibia (Hymenoptera: Braconidae: Microgastrinae), with notes on taxonomic status and natural history

The Microgastrinae wasp Diolcogaster claritibia (Papp, 1959) (Hymenoptera: Braconidae), a parasitoid of the diamondback moth, Plutella xylostella (Linnaeus, 1758) (Lepidoptera: Plutellidae), is recorded from the Nearctic region (Alberta, Manitoba, and Ontario, Canada) for the first time. In spite of some minor morphological and molecular (DNA barcoding) differences, the available data indicate that the European and New World specimens should be considered one species, but more specimens and loci need to be sampled to conclusively determine the phylogeographic history of the species. Diolcogaster claritibia seems to be widely distributed within the Holarctic, and the molecular data suggest that it was recently introduced to North America from Europe.
Improving Signal to Noise in Labeled Biological Specimens Using Energy-Filtered TEM of Sections with a Drift Correction Strategy and a Direct Detection Device

Energy filtered transmission electron microscopy techniques are regularly used to build elemental maps of spatially distributed nanoparticles in materials and biological specimens. When working with thick biological sections, electron energy loss spectroscopy techniques involving core-loss electrons often require exposures exceeding several minutes to provide sufficient signal to noise. Image quality with these long exposures is often compromised by specimen drift, which results in blurring and reduced resolution. To mitigate drift artifacts, a series of short exposure images can be acquired, aligned, and merged to form a single image. For samples where the target elements have extremely low signal yields, the use of charge coupled device (CCD)-based detectors for this purpose can be problematic. At short acquisition times, the images produced by CCDs can be noisy and may contain fixed pattern artifacts that impact subsequent correlative alignment. Here we report on the use of direct electron detection devices (DDD’s) to increase the signal to noise as compared with CCD’s. A 3× improvement in signal is reported with a DDD versus a comparably formatted CCD, with equivalent dose on each detector. With the fast rolling-readout design of the DDD, the duty cycle provides a major benefit, as there is no dead time between successive frames.

Hypovitaminosis D may be associated with diabetes, hypertension and CHD. However, because studies examining the associations of all three chronic conditions with circulating 25-hydroxyvitamin D (25(OH)D) and 1,25-dihydroxyvitamin D (1,25(OH)2D) are limited, we examined these associations in the US Prostate, Lung, Colorectal and Ovarian (PLCO) Cancer Screening Trial (n 2465). Caucasian PLCO participants selected as controls in previous nested case–control studies of 25(OH)D and 1,25(OH)2D were included in this analysis. Diabetes, CHD and hypertension prevalence, risk factors for these conditions and intake of vitamin D and Ca were collected from a baseline questionnaire. Results indicated that serum levels of 25(OH)D were low ( < 50 nmol/l) in 29 % and very low ( < 37 nmol/l) in 11 % of subjects. The prevalence of diabetes, hypertension and CHD was 7, 30 and 10 %, respectively. After adjustment for confounding by sex, geographical location, educational level, smoking history, BMI, physical activity, total dietary energy and vitamin D and Ca intake, only diabetes was significantly associated with lower 25(OH)D and 1,25(OH)2D levels. Caucasians who had 25(OH)D ≥ 80 nmol/l were half as likely to have diabetes (OR 0·5 (95 % CI 0·3, 0·9)) compared with those who had 25(OH)D < 37 nmol/l. Those in the highest quartile of 1,25(OH)2D ( ≥ 103 pmol/l) were less than half as likely to have diabetes (OR 0·3 (95 % CI 0·1, 0·7)) than those in the lowest quartile ( < 72 pmol/l). In conclusion, the independent associations of 25(OH)D and 1,25(OH)2D with diabetes prevalence in a large population are new findings, and thus warrant confirmation in larger, prospective studies.

Transparent conducting oxides (TCOs) have been known and employed technologically for more than 50 years, primarily in the form of doped single-cation oxides such as In2O3 and SnO2. Beginning in the 1990s, however, multi-cation oxide TCOs began to be developed in Japan (see the article by Minami in this issue and the references therein) and at the former Bell Laboratories. Since then, new TCO phases are being reported with increasing frequency as technological interest in this area heightens. At the same time, our fundamental understanding of the chemical and structural origins of transparent conductivity continues to expand and promises a pathway to dramatically improved materials for a host of applications. This article describes a collaborative, multi-investigator bulk an d thin-film research effort at Northwestern University aimed at the synthesis, characterization, and enhanced understanding of multi-cation (compound and solidsolution) TCOs, and provides a brief account of what we are discovering about this important class of materials.

Layered copper-oxide superconductors exhibit the highest critical transition temperatures of any materials. Yet all of the known double perovskites A′A″B′B″O6 containing copper have a random or rock salt distribution of the B cations with the exception of the unique layered arrangement found in La2CuSnO6. Only the layered arrangement contains the CuO22- planes which are necessary for high-temperature superconductivity. The occurrence of layered or two dimensional structures increases markedly when vacancies are introduced on the oxygen sublattice, as evidenced in Ln2AEmCu2TimO5+3m (Ln = lanthanide, Y: AE = Ba, Ca: 2 ≤ m ≤ 4). Similarities among oxygen-deficient structures, especially those with two-dimensional solid-state features, are discussed. Combined conductivity and thermopower analysis are presented to elucidate their unique internal chemistry, defect structure, and conduction parameters. In particular, data for La2-xSrxCuSnO6 are presented and related to the crystal chemistry of the copper-oxygen layer. These data are compared with La2Ba2Cu2Sn2O11 and La2Ba2Cu2Ti2O11 to illustrate the significance of oxygen vacancies on the properties of the copper oxygen planes. New layered cuprates are discussed including the mixed A-site stoichiometries Ln′Ln″AEmCu2TimO5+3m (Ln = lanthanide, Y: AE = Ba, Ca: 2 ≤ m ≤ 4) which contain the smaller lanthanide (Ln″) ordered between the closely spaced, facing sheets of Cu-O square pyramids.

Reaction of boehmite, [AI(O)(OH)]n, with an excess of carboxylic acid (HO2CR) results in the formation of the carboxy substituted alumoxanes, [AI(O)x(OH)y(O2CR)z)]n where 2x + y + z = 3 and R = alkyl substituents. The alumoxanes have been fully characterized by SEM, elemental analysis, IR and multinuclear NMR spectroscopy. The physical properties of the alumoxanes are highly dependent on the identity of R, and range from insoluble crystalline powders, e.g. R = CH3, to powders which readily form solutions or gels in hydrocarbon solvents, e.g. R = C5H11. All of the alumoxanes decompose under mild thermolysis to yield γ-alumina.
