Vitamin D receptor genotype influences risk of upper respiratory infection

SNP in the vitamin D receptor (VDR) gene is associated with risk of lower respiratory infections. The influence of genetic variation in the vitamin D pathway resulting in susceptibility to upper respiratory infections (URI) has not been investigated. We evaluated the influence of thirty-three SNP in eleven vitamin D pathway genes (DBP, DHCR7, RXRA, CYP2R1, CYP27B1, CYP24A1, CYP3A4, CYP27A1, LRP2, CUBN and VDR) resulting in URI risk in 725 adults in London, UK, using an additive model with adjustment for potential confounders and correction for multiple comparisons. Significant associations in this cohort were investigated in a validation cohort of 737 children in Manchester, UK. In all, three SNP in VDR (rs4334089, rs11568820 and rs7970314) and one SNP in CYP3A4 (rs2740574) were associated with risk of URI in the discovery cohort after adjusting for potential confounders and correcting for multiple comparisons (adjusted incidence rate ratio per additional minor allele ≥1·15, Pfor trend ≤0·030). This association was replicated for rs4334089 in the validation cohort (Pfor trend=0·048) but not for rs11568820, rs7970314 or rs2740574. Carriage of the minor allele of the rs4334089 SNP in VDR was associated with increased susceptibility to URI in children and adult cohorts in the United Kingdom.

Long half-life biologically active fission products, such as technetium-99, present particular problems for the disposal of spent nuclear fuel (SNF). Technetium is present in relatively high concentrations in fuel (approx. 1kg tonne-1 SNF) and has very high mobility in oxidizing environments. Technetium is therefore generally removed from SNF either by solvent extraction and reduction, during the PUREX process, or by sorption via ion exchange processes. Historically technetium has been disposed of via dilution and dispersion in the sea but stringent regulations now mean that the preferred long term option is immobilization in a highly stable and durable matrix. In this contribution we have looked at the synthesis of fluorite derivative crystalline host phases based on the zirconolite structure. Samples have been characterised by X-ray diffraction (XRD), scanning electron microscopy (SEM), energy dispersive spectroscopy (EDX), thermo-gravimetric analysis (TG), and mass spectroscopy (MS). We have used Mo as an inactive surrogate for Tc.

Effect of polyphenol-rich grape seed extract on ambulatory blood pressure in subjects with pre- and stage I hypertension

Dietary polyphenols, such as those from grape products, may exert beneficial effects on cardiovascular health, including anti-hypertensive effects. We investigated the effect of a specific grape seed extract (GSE) rich in low-molecular-weight polyphenolic compounds on ambulatory blood pressure (ABP) in untreated subjects with pre- and stage I hypertension. In addition, potential mechanisms that could underlie the hypothesised effect of GSE on blood pressure (BP), and platelet aggregation, were explored. The study was designed as a double-blind, placebo-controlled, randomised, parallel-group intervention study including seventy healthy subjects with systolic BP between 120 and 159 mmHg. A 1-week run-in period was followed by an 8-week intervention period, during which subjects consumed capsules containing either 300 mg/d of GSE or a placebo (microcrystalline cellulose). Before and after the intervention, daytime ABP readings, 24 h urine samples and fasting and non-fasting blood samples were taken. The mean baseline systolic BP was 135·8 (se 1·3) mmHg and diastolic BP was 81·5 (se 0·9) mmHg. BP values were modestly, but not significantly, affected by the polyphenol-rich GSE treatment v. placebo with an effect of − 3·0 mmHg for systolic BP (95 % CI − 6·5, 0·5) and − 1·4 mmHg for diastolic BP (95 % CI − 3·5, 0·6). Vasoactive markers including endothelin-1, NO metabolites and asymmetric dimethylarginine, plasma renin activity and platelet aggregation were not affected by the GSE intervention. Our findings show that consumption of polyphenol-rich GSE does not significantly lower ABP in untreated subjects with pre- and stage I hypertension.

We investigated postprandial changes in transcript abundance following a single satiating meal in juvenile Atlantic salmon (Salmo salar L.) (about 70 g body mass) following fasting for 1 week at 12°C. The expression of twenty-three growth-related genes was determined in fast myotomal muscle using quantitative real-time PCR at the following postprandial time points: − 12, 0, 1, 3, 6, 12, 24, 48 and 96 h. The gut was fullest 1–6 h after feeding and emptied within 48–96 h. IGF-I, MyoD1c, MRF4 and myf5 transcripts were sharply up-regulated within 1 h of refeeding and are promising candidate genes involved in a fast-response signalling system that regulates fish myotomal muscle growth. These genes clustered together with MyoD1b and suggest a coordinated regulation to favour resumption of myogenesis as an early response to feeding. Insulin-like growth factor (IGF)-II and the ubiquitin ligase MAFbx/atrogin-1 were initially down-regulated but restored to initial values after 12 h. It is also suggested that local production of IGF-I within the muscle might suppress catabolic pathways depressing MAFbx/atrogin-1.

The effects of incremental exercise on appetite, energy intake (EI), expenditure (EE) and balance (EB) in lean men and women were examined. Six men (age 29·7 (sd 5·9) years, weight 75·2 (sd 15·3) kg, height 1·75 (sd 0·11) m) and six women (age 24·7 (sd 5·9) years, weight 66·7 (sd 9·10) kg, height 1·70 (sd 0·09) m) were each studied three times during a 16 d protocol, corresponding to no additional exercise (Nex), moderate-intensity exercise (Mex; 1·5–2·0 MJ/d) and high-intensity exercise (Hex; 3·0–4·0 MJ/d) regimens. Subjects were fed to EB during days 1–2, and during days 3–16 they fed ad libitum from a medium-fat diet of constant composition. Daily EE, assessed using the doubly labelled water method, was 9·2, 11·6 and 13·7 MJ/d (P < 0·001; sed 0·45) for the women and 12·2, 14·0 and 16·7 MJ/d (P = 0·007; sed 1·11) for the men on the Nex, Mex and Hex treatments, respectively. EI was 8·3, 8·6 and 9·9 MJ/d (P = 0·118; sed 0·72) for the women and 10·6, 11·6 and 12·0 MJ/d (P = 0·031; sed 0·47) for the men, respectively. On average, subjects compensated for about 30 % of the exercise-induced energy deficit. However, the degree of compensation varied considerably among individuals. The present study captured the initial compensation in EI for exercise-induced energy deficits. Total compensation would take a matter of weeks.
Topology: a novel method to describe branching patterns in Peronospora viciae colonies

Topology provides a novel means to describe branching patterns and has not been applied to fungal colonies previously. For any branched structure, various topologies are possible, and these lie between two extremes, a herringbone pattern (main axis with primary laterals) and a dichotomous pattern (highly branched system). We applied topological methods to colonies of Peronospora viciae 48 h after inoculation of Pisum sativum leaves. The methods are based on two simulations, one developed for channel networks such as found in river systems and another for biological systems. Although not a true herringbone form, the Peronospora viciae colonies have a strong herringbone element within their growth pattern. All 25 colonies analysed fell into the random distribution according to the confidence limits calculated from simulations for biological systems. These confidence limits, however, represent the percentile distribution of all simulated networks, and only those structures with a perfect herringbone or dichotomous topology fall outside the range. The tendency of P. viciae colonies towards herringbone growth is reflected by the topological indices for altitude and external pathlength (a(obs)/E(a) and pe(obs)/E(pe), where a=altitude, pe=external pathlength, obs=observed for the P. viciae colonies and E=expected values for random growth), and the slope of the regression analysis for a(obs) and pe(obs). We consider this trend as significant because it was consistent for all but one of the colonies, and implies that growth can be envisaged as an intermediate between random and herringbone topology. It is proposed that initial herringbone growth may reflect a strategy that is aimed at overcoming host resistance, achieving rapid colonisation of infected tissue and maximising the potential for nutrient acquisition. This topology would also increase the likelihood of finding a compatible mating type for reproduction between heterothallic isolates.

Excavations at two sites in Gwynedd have produced new evidence for highland zone rural settlement during the Romano-British period. At Bryn Eryr this activity was the culmination of a long sequence of development during the Iron Age. At Bush Farm, similarly, a Romano-British phase succeeded earlier, probably late prehistoric occupation of the site. At both locations significant structural changes took place during the Romano-British period as small stone-walled houses replaced larger, clay-walled buildings. Changes in the local farming economy have been identified and the proximity of the vicus at Segontium provided access to Roman pottery vessels. The quality and range of pottery from Bryn Eryr and Bush Farm are considerable in comparison with other ‘Highland Zone’ settlements and the material is discussed in detail.

With the financial services industries in the member countries of the European Union coming under increasing attention resulting in relaxation of cross border controls, this paper addresses some pensions matters in this European context and how they may relate to the U.K. The main theme of the paper is the book reserve approach to pensions provision. Details of the German book reserve method are provided before developing possible ways in which this philosophy could be introduced into the U.K. These possibilities are assessed and consideration given to the effect they may have on the economy, commerce, the pension fund industry and, not least, the actuarial profession in the U.K.
