Improving the quality and conduct of multi-center clinical trials is essential to the generation of generalizable knowledge about the safety and efficacy of healthcare treatments. Despite significant effort and expense, many clinical trials are unsuccessful. The National Center for Advancing Translational Science launched the Trial Innovation Network to address critical roadblocks in multi-center trials by leveraging existing infrastructure and developing operational innovations. We provide an overview of the roadblocks that led to opportunities for operational innovation, our work to develop, define, and map innovations across the network, and how we implemented and disseminated mature innovations.

The search for life in the Universe is a fundamental problem of astrobiology and modern science. The current progress in the detection of terrestrial-type exoplanets has opened a new avenue in the characterization of exoplanetary atmospheres and in the search for biosignatures of life with the upcoming ground-based and space missions. To specify the conditions favourable for the origin, development and sustainment of life as we know it in other worlds, we need to understand the nature of global (astrospheric), and local (atmospheric and surface) environments of exoplanets in the habitable zones (HZs) around G-K-M dwarf stars including our young Sun. Global environment is formed by propagated disturbances from the planet-hosting stars in the form of stellar flares, coronal mass ejections, energetic particles and winds collectively known as astrospheric space weather. Its characterization will help in understanding how an exoplanetary ecosystem interacts with its host star, as well as in the specification of the physical, chemical and biochemical conditions that can create favourable and/or detrimental conditions for planetary climate and habitability along with evolution of planetary internal dynamics over geological timescales. A key linkage of (astro)physical, chemical and geological processes can only be understood in the framework of interdisciplinary studies with the incorporation of progress in heliophysics, astrophysics, planetary and Earth sciences. The assessment of the impacts of host stars on the climate and habitability of terrestrial (exo)planets will significantly expand the current definition of the HZ to the biogenic zone and provide new observational strategies for searching for signatures of life. The major goal of this paper is to describe and discuss the current status and recent progress in this interdisciplinary field in light of presentations and discussions during the NASA Nexus for Exoplanetary System Science funded workshop ‘Exoplanetary Space Weather, Climate and Habitability’ and to provide a new roadmap for the future development of the emerging field of exoplanetary science and astrobiology.

The Carnmenellis granite and its aureole contain the only recorded thermal groundwaters (up to 52 °C) in British granites. They occur as springs in tin mines at depths between 200 and 700 m and most are saline (maximum mineralization 19 310 mg 1−1). Mining activity has disturbed the groundwater circulation pattern developed over a geological time-scale and levels of bomb-produced tritium (> 4 TU) indicate that a significant component (up to 65 %) of the most saline waters are of recent origin. All components of all the mine waters are of meteoric origin. Radiogenic 4He contents, 40Ar/36Ar ratios, and uranium series geochemistry suggest that the thermal component has a likely residence time of at least 5 × 104 years and probably of order 106 years.
The thermal waters have molar Na+/Cl− ratios considerably less than 1 but they are enriched relative to sea water in all major cations except Mg. The groundwater is also particularly enriched in Li with contents ranging up to 125 mg 1−1. The groundwater salinity, which may reach a maximum of 30 000 mg 1−1, is shown to result from weathering reactions of biotite (probably through a chloritization step) and plagioclase feldspar, to kaolinite. On volumetric considerations, fluid inclusions cannot contribute significantly to the groundwater salinity, and stable isotope ratios rule out any contribution from sea water.
Groundwater silica contents and molar Na+/K+ ratios suggest that the likely equilibration temperature is 54°C, which would imply a depth of circulation of about 1.2 km.

An efficient and robust method to measure vitamin D (25-hydroxy vitamin D3 (25(OH)D3) and 25-hydroxy vitamin D2 in dried blood spots (DBS) has been developed and applied in the pan-European multi-centre, internet-based, personalised nutrition intervention study Food4Me. The method includes calibration with blood containing endogenous 25(OH)D3, spotted as DBS and corrected for haematocrit content. The methodology was validated following international standards. The performance characteristics did not reach those of the current gold standard liquid chromatography-MS/MS in plasma for all parameters, but were found to be very suitable for status-level determination under field conditions. DBS sample quality was very high, and 3778 measurements of 25(OH)D3 were obtained from 1465 participants. The study centre and the season within the study centre were very good predictors of 25(OH)D3 levels (P<0·001 for each case). Seasonal effects were modelled by fitting a sine function with a minimum 25(OH)D3 level on 20 January and a maximum on 21 July. The seasonal amplitude varied from centre to centre. The largest difference between winter and summer levels was found in Germany and the smallest in Poland. The model was cross-validated to determine the consistency of the predictions and the performance of the DBS method. The Pearson’s correlation between the measured values and the predicted values was r 0·65, and the sd of their differences was 21·2 nmol/l. This includes the analytical variation and the biological variation within subjects. Overall, DBS obtained by unsupervised sampling of the participants at home was a viable methodology for obtaining vitamin D status information in a large nutritional study.

Global climate change is expected to affect the frequency, intensity and duration of extreme water-related weather events such as excessive precipitation, floods, and drought. We conducted a systematic review to examine waterborne outbreaks following such events and explored their distribution between the different types of extreme water-related weather events. Four medical and meteorological databases (Medline, Embase, GeoRef, PubMed) and a global electronic reporting system (ProMED) were searched, from 1910 to 2010. Eighty-seven waterborne outbreaks involving extreme water-related weather events were identified and included, alongside 235 ProMED reports. Heavy rainfall and flooding were the most common events preceding outbreaks associated with extreme weather and were reported in 55·2% and 52·9% of accounts, respectively. The most common pathogens reported in these outbreaks were Vibrio spp. (21·6%) and Leptospira spp. (12·7%). Outbreaks following extreme water-related weather events were often the result of contamination of the drinking-water supply (53·7%). Differences in reporting of outbreaks were seen between the scientific literature and ProMED. Extreme water-related weather events represent a risk to public health in both developed and developing countries, but impact will be disproportionate and likely to compound existing health disparities.

Oxides, nitrides, carbides and silicides contain more than one type of atoms that gives one extra degree of freedom in the grain boundary and interfacial structure. This degree of freedom is the choice of the boundary plane containing different type of atoms which results of several structural multiplicities in the boundary. Coexistence of the structural multiplicities (or boundary domains) in the interface and grain boundary were reported [1, 2]. The structural information in high frequency domain is usually blurred and lost in the lens system of a high resolution transmission electron microscopes (HRTEM) such that the HRTEM image may not truly represent the real structure. Conventionally, the structures of grain boundary and interface were solved using forward method. The forward method involves matching the simulated images from guessed possible models with the through focal series experimental images. Recently, rereconstruction methods such as focal variation holography [3, 4], tiltbeam holography [5] and Gerchberg-Saxton algorithm [6] provide a backward (direct) way to recover the structural information lost in a modern microscope.

EFTEM has become a very useful tool in characterizing material properties, because it allows studying local chemical and electronic properties of a specimen with nanometer level spatial resolution. There are two spectroscopic imaging methods have been established to obtain both the spatial and the spectra information, I (x, y, ΔE), which can be quantitatively evaluated. One technique, as known as spectrum-image method, is based on scanning the electron beam across the sample to acquire the EELS spectrum with two-dimensional information. Conversely, the other method retrieves the spectra in a specific position from a series of energy loss images, as known as imaging-spectrum method. The spectrum-image method can be achieved with better energy resolution, however, a long acquisition time. Compared to spectrum-image method, which acquires a series of spectrum with scanning probe, the advantage of imaging-spectrum method is the capability of simultaneously recording abundant information in a series of maps.
