An isometric transducer system has been used to record spontaneous motor activity in Diclidophora merlangi in vitro. Motility took the form of either continuous irregular contractions or bursts of activity with intermittent quiescent periods. Maximal activity was recorded from specimens at 5–8 °C in artificial sea water (ASW). Decerebration induced a period of enhanced motility which subsided within 1 h. Water turbulence elicited large, rapid contractions of the longitudinal body musculature which did not habituate or fatigue. Induced water turbulence at frequencies higher than 3/min inhibited spontaneous movements, resulting in a progressive reduction in contraction amplitude and, eventually (30/min), the abolition of any response to water movement. The neurotoxin tetrodotoxin failed to modify worm activity. Excitatory responses from both intact worms and strip preparations were obtained with 5-hydroxytryptamine, dopamine and noradrenaline, but aminergic antagonists failed to block their effects. Atropine stimulated contractility, whereas the effects of acetylcholine or carbachol were inconsistent. Nicotine increased muscle tone but the effect was unaltered by atropine, mecamylamine or d–tubocurarine. Muscarine, hemicholinium, neostigmine and eserine were without apparent effect.

An in vitro method for studying radiorespiration has been adapted to single macrofilariae. Using this method viable (but not heat-killed) Dipetalonema viteae and Onchocera gibsoni macrofilariae evolved measurable amounts of 14CO2 from L-[U-14C]glutamine. Nonlinear and less uniform rates of 14CO2 evolution were demonstrated with D-[U-14C]glucose, [1-14C]acetate and [1-14C]octanoate. These findings led us to develop an in vitro screen in which inhibition of 14CO2 evolution from L-[U-14C]glutamine was used as a parameter for gauging macrofilaricidal activity. Using this screen we have examined the activity of 17 ‘so-called’ antifilarial standards and found a greater degree of sensitivity than shown by other biochemical criteria. Other data obtained suggest a role for radiorespirometry in determining the viability of fragments of Onchocerca tissue.

125I-Schistosoma mansoni schistosomulum surface antigens were immunoprecipitated with human antibodies from individual Egyptian patients diagnosed as being either acutely or chronically infected with S. mansoni. Both sets of patients were found to have IgG antibodies in their sera capable of immunoprecipitating the major Mr > 200, 38 and 32K antigens. However, the immunoprecipitation of the Mr 200K antigen was found to constitute a significantly greater proportion of the total precipitate achieved with acute sera than with chronic sera. The Mr 38 and 32K antigens were more variably precipitated by the acute sera than the chronic sera but the proportion of the total precipitation that these two antigens constituted was not found to be significantly different between the two sets of sera. Immunoprecipitation with pooled antibodies absorbed with egg and adult worm homogenates which had been treated to remove either carbohydrate or polypeptide epitopes demonstrated that the Mr > 200K antigen was the principal target of egg-cross-reactive anti-carbohydrate antibody amongst the antigens detected. The Mr 38 and 32K antigens were found to be precipitated by antibodies to protease-sensitive and periodate-insensitive polypeptide epitopes. These results are consistent with egg-cross-reactive anti-carbohydrate IgG antibody making a greater contribution to schistosomulum surface recognition in acute infection than in chronic infection. Indeed the presence of a higher level of egg-cross-reactive and anti-carbohydrate antibody directed against schistosomulum surface epitopes in an acute serum pool than in a chronic serum pool was confirmed by measurement of antibody binding to whole schistosomula.

This study provides the first quantitative assessment of the cutaneous reaction elicited in naive and sensitized rats by invading larvae of Strongyloides ratti. Parasites were identified in skin samples harvested from both naive and sensitized rats between 0·5 and 8 h following percutaneous infection, but were no longer visible at the site of invasion at 24 h. Mast cells, neutrophils, eosinophils and mononuclear cells recruited into the dermis in response to invading larvae were recorded and quantified. Cellular infiltrates were observed as early as 1 h post-invasion in naive/challenged skin, where total cell numbers were up to 3 times greater than in naive control tissue. Peak numbers of all cell types were recorded from 3 to 8 h in both naive/challenged and sensitized/challenged hosts. Interestingly, the reaction in sensitized/challenged rats was neither enhanced nor accelerated, a feature perhaps attributable to lack of recognition of parasite antigens by the host. This possibility was investigated by immunofluorescent labelling, which clearly demonstrated changes in the surface antigen profile of the parasite following penetration of the host skin both in vitro and in vivo. It is proposed that these changes in surface antigenicity constitute an evasive stratagem used by the parasite to deter the host from mounting a potentially lethal inflammatory response.

Population outbreaks of house mice (Mus domesticus) occur periodically in the wheatlands of southeastern Australia. This paper uses mathematical models to assist in the evaluation of the potential of a nematode, Capillaria hepatica, as a biological control agent to reduce the severity of these ‘plagues’. C. hepatica is unique amongst helminths of mammals in that its eggs are released only upon the death of an infected host. The major goal of the modelling in this paper is to determine the impact of this feature on the population dynamics of the host-parasite interaction. Simple differential equation models are used to examine the general properties of the system and determine which population parameters are most crucial to the outcome of the interaction. These models are supplemented by age-structured models which investigate the initial behaviour of the system after introduction of the parasite. The necessity of host death for transmission is a strongly destabilizing factor, suggesting that C. hepatica cannot regulate most populations stably in the absence of strong resource limitation, although it has the potential to depress mouse populations below infection-free levels. Although C. hepatica influences mouse fecundity at lower burdens than it affects mortality, the age-structured models show that parasite-induced host death cannot be neglected. Because transmission requires host death, the parasite life-cycle operates on a time-scale similar to that of the hosts, and introduction of the parasite as early as possible in the development period of an outbreak will therefore be necessary to achieve substantial reductions in plague intensity.

All polyopisthocotylean monogeneans previously studied, including representatives of the Polystomatidae infecting anuran amphibians, feed on host blood. However, the present analysis of species of Polystomoides, Polystomoidella and Neopolystoma, polystomatids which infect chelonian reptiles, has shown that this group has diverged nutritionally from related parasites. Histochemical tests failed to demonstrate haemoglobin in the gut caeca, and X-ray microanalysis confirmed the absence of haematin (or high concentrations of bound iron) in the gastrodermis. The chelonian poly-stomatids (and also the single monogenean which infects a mammal, Oculotrema hippopotami) feed on epithelial cells and mucus, the diet typical of monopisthocotyleans. Transmission electron microscopy revealed the same gastrodermal architecture in representatives of Polystomoides from Africa, N. America and S.E. Asia. The organization of the caecal epithelium conforms with that of blood-feeding polyopisthocotyleans, with two components: lamellated cells responsible for intracellular digestion interspersed with elements of a non-lamellated connecting syncytium. In other polyopistho-cotyleans, the syncytium probably has a skeletal, supportive role, related to the problems of intracellular accumulation of haematin, but in polystomatids infecting chelonians the syncytium is extremely reduced and its presence probably reflects an ancestry amongst blood-feeding relatives. The utilization of the presumably more primitive monogenean diet of epithelial cells and mucus by chelonian polystomatids may be related to the scarcity of superficial blood vessels in their oral and urinary bladder habitats.

Oogenesis in Brugia pahangi has been studied by means of the aceto-orcein chromosomal squash technique and light-microscope autoradiography. The use of colchicine has demonstrated a 2–3 mm terminal germinative zone within the ovary, in which continuous and rapid mitotic division of germ cells occurs. In 80% of the gonads, oocytes within a 1–2 mm length of the ovary proximal to the germinative zone were at the prophase of meiosis I. Primary oocytes with markedly less condensed chromatin, apparently interphase cells, were observed in the corresponding region of the ovary in the remaining 20% of material examined. A cyclical or phased development of primary oocytes is suggested. Autoradiographic studies, concerned with the incorporation of [5-3H]uridine into germ cells of B. pahangi in vitro, further suggest that the onset of meiotic prophase is associated with the initiation of high RNA synthetic activity. Following meiotic prophase, oocytes complete meiosis I before entering a period of growth during which the chromatin material is decondensed. Recondensation of chromosomes prior to meiosis II is only observed after fertilization within the seminal receptacle. On completion of meiosis II, with the extrusion of a polar body, the haploid chromosome complement of the female unites with that of the male, re-establishing the diploid number of the zygote (2n = 10).

Predisposition to trichuriasis in mice is reflected in the inability of certain strains, or certain individuals within strains, to express protective immunity. Poor responders fail to expel worms and harbour chronic patent infections. The mechanisms underlying this phenomenon were studied in poor responder mice challenged after abbreviated or prolonged primary infections. Mice exposed to a complete primary infection were fully susceptible when challenged after the removal of the primary infection by anthelmintic. Failure to expel either infection suggests (a) that non-responsiveness to a primary infection does not reflect an inability to expel worms of a certain size, i.e. is not a consequence of the speed of the immune response in relation to parasite growth and (b) that non-responsiveness is long-lasting. Challenge after abbreviation of primary infections at different stages of worm development showed that persistence of larvae beyond day 21 was critical in determining poor response to reinfection. By inference the same conclusion can be drawn about the inability of such mice to expel primary infections. Serological analysis suggested a relationship between low antibody titres, restricted antigen recognition profiles and resistance to infection. It is suggested that the later stages of parasite development are immunosuppressive; the implications for human trichuriasis are discussed.

The parameters of species richness, abundance and diversity of the intestinal helminth infracommunities of a sample of 269 Common gulls, Larus canus, were examined over one season at Agdenes, Norway. The communities developed rapidly in unfledged gulls and the diversity parameters were of similar magnitude in immature and adult birds. All parameters were low in the early part of the season, reached maxima in June or July and fluctuated erratically or declined thereafter. Comparison of summary parameters with those from other species of gulls indicated that helminth community diversity in Common gulls is fairly typical of gulls in general. In gulls as a group, helminth communities show common features of high species richness, low abundance, few or no core species and a number of species that occur rarely and in low numbers. Expectations of particularly high parasite community diversity were not fully realised because species lists and diversity of the component community are not a very good basis for predicting diversity at the infracommunity level.

The survival of Heligmosomoides polygyrus was monitored during primary infections in female C57Bl10, NIH and BALB/c mice at low and high intensities of infection. Survivorship curves were fitted for each data set and analysed. C57Bl10 mice, given either low or high intensities of infection, harboured parasites for 28–37 weeks, heavier infections surviving marginally but significantly longer. Essentially the survivorship curves of H. polygyrus in C57Bl10 mice could be accounted for by senility, the increased probability of worms with a longer life-span occurring at high infection intensities and, possibly, by a contribution from host-protective immune mechanisms in the terminal stages of infection. The pattern of survivorship was different in NIH and BALB/c mice. NIH mice showed weak but significant density-dependent suppression of parasite loss and infections in this strain did not exceed 27·5 weeks in duration. Primary infections in BALB/c mice were briefer still and showed marked dependence on parasite density. Thus low-level infections lasted 10–15 weeks whereas heavier infections survived for 21–34 weeks. The data suggested that both strains developed host-protective responses to adult H. polygyrus and that parasite survival was curtailed earlier than would be expected if senility alone was involved. The hybrid strains (C57Bl10 × NIH)F1 and (B10G × NIH)F1 both expelled H. polygyrus in a dose-dependent manner, worm loss commencing within 10 weeks of infection. In some experiments worm loss was clearly evident by weeks 4 and 6. These hybrid strains showed gene complementation in that adult worms were cleared considerably earlier than in parental strains.

Distribution of Leptorhynchoides thecatus (Acanthocephala: Rhadinorhynchidae) among the pyloric caeca and the relationship between site and rate of maturation were studied in laboratory infectons of 10, 25 and 40 cystacanths fed to green sunfish, Lepomis cyanellus. After 1 week fish fed at each intensity had significantly different numbers of worms. By the 3rd week post-infection, parasites disappeared from the anterior portion of the intestine. At this time the mean numbers of worms recovered from 25 and 40-cystacanth infections were not significantly different. At the end of the 1st week, the area where caeca join the alimentary tract (between caecal area) and caeca numbered 6 and 7 contained significantly more worms than the other sites. By the 3rd week post-infection only caecum 7 contained significantly more worms, and at 5 weeks there was no significant difference between the number of worms present in any caecum or the between caecal area. Initially worms in the more intense infections matured more slowly, but by the 3rd week post-infection there was no significant difference in the states of maturity. The rate of maturation was not related to the site occupied.

The distribution of Ascaris lumbricoides within a community was examined at an initial mass anthelmintic treatment programme (January 1984) and following an 11-month period of reinfection (November 1984). Similar patterns of the negative binomial parameter k (an inverse measure of parasite aggregation) and the proportion of parasites within the most heavily infected quartile of the community were recorded at the two dates. The pattern of parasite aggregation within individuals, measured by parameter k, appears to be a stable characteristic of this host-parasite relationship. Significant variation in the intensity of infection was observed between households in the community. The number of family members living in the house strongly influenced the mean Ascaris burden and proportion of relatively heavy infections within adults and children. This finding suggests that the density of people in a house positively influences the frequency of exposure to infective stages of Ascaris, which in turn plays a major role in determining which individuals will harbour heavy infections. Positive correlations were recorded between the initial and reinfection burdens of individuals, relative to others in the community. The correlations were strongest in the youngest and oldest age groups and were more frequently significant among age-stratified groups of females, compared to males. A comparative examination of hypothetical treatment strategies suggests that for Ascaris infections in this community, targetting age groups with anthelmintic treatment would probably be more cost-effective in the long term in reducing the abundance of this parasite than selective treatment of individually identified heavy infections.

The course of primary infection with a hamster-adapted strain of Ancylostoma ceylanicum was studied in inbred DSN and randomly bred WO/GD and WO/CR hamsters. Infective larvae were administered orally and began to develop in the small intestine without embarking on a tissue migration. Only the occasional larva was detected in other organ sites. It was concluded that the developing larvae moulted on days 3–4 and again to the pre-adult stage about 9–11 days post-infection. Worm burdens in infected hamsters were stable for at least 11 weeks after infection. There was no sudden expulsive phase and some adult worms survived for over 200 days. Overall the sex ratio of worms in groups of hamsters killed concurrently was about 50% although occasionally the ratio was biased in favour of one sex in individual animals. The blood packed cell volume (PCV) was significantly depressed 2 weeks following infection and continued to decline until a point of stability was achieved 4–5 weeks post-infection. The PCV subsequently remained depressed throughout the period of observation. Infected hamsters lost weight if kept in groups, but not when housed in separate cages. Groups of animals which lost weight did not recover to normal values within 11 weeks of infection. It is suggested that this model of hookworm infection has scope for exploring aspects of the host-parasite relationship which the canine models cannot fulfill adequately.

The in vivo activity of two cyclosporins, cyclosporin A (CsA) and a non-immunosuppressive derivative of dihydrocylosporin A (DHCsA-d) against three tapeworms, Hymenolepis microstoma, H. diminuta and Mesocestoides corti, has been assessed. CsA reversibly reduced the dry weight of H. microstoma in the mouse, briefly delayed oviposition and had a statistically significant effect on worm numbers recovered. Oral and subcutaneous treatments of both CsA and DHCsA-d were effective in reducing worm weight; juvenile worms were most susceptible but worms of all ages responded to drug by a dramatic reduction in weight from which they recovered. Multiple courses of CsA were no more active than single courses of treatment but dose response suggested that a threshold level of drug was necessary to evoke activity. By contrast, H. diminuta in the rat was completely unaffected by CsA but no explanation for the differences in drug response by these two closely related helminths is forthcoming. Mesocestoides corti responded reversibly to CsA in the mouse by a reduction in asexual proliferation of both liver and peritoneal cavity tetrathyridia. The data presented argue in favour of a range of anti-parasitic activities by cyclosporins but the details of the various putative modes of action remain to be defined.

The results are presented of an experimental study of the population biology of chronic Trichuris muris (Nematoda) infection in cortisone-treated CBA/Ca mice. Attention is focused upon both the validity of the common use of faecal egg counts to demonstrate density dependence in helminth fecundity, and the identification of other possible density-dependent mechanisms that may regulate worm numbers in chronic trichuriasis. The results show that faecal egg counts, although demonstrating high daily variation, are not an artefact of host faecal output but a significant density-dependent function of worm burden. This finding contrasts with the observations on Heligmosomoides polygyrus infection in outbred MF1 mice, but accords with similar studies in a wide variety of host - helminth systems. Worm establishment in the murine host is found to be a density related function of infection dose. This is attributed to the probable existence of a physical gut-carrying capacity in the murine host for T. muris. Worm distribution in the gut is also shown to be density dependent, with worms being displaced from the caecum to the colon at increasing intensities of infection. The sex ratio of the adult parasites, however, is found to be both unitary and independent of worm burden. Evidence for a significant density-dependent decline in female T. muris growth or size is presented. The results also show a significant positive association between female T. muris weight and per capita fecundity. These findings indicate that the stunted growth of individual worms at high parasite densities may be a potential mechanism underlying density dependence in helminth fecundity.

A total of 28 components were detected in the free amino acid (FAA) pool of hydatid fluid from primary and secondary equine cysts, secondary ovine cysts and host plasma. Examination of data from equine cysts revealed that the majority of FAAs were present in significantly greater concentrations in secondary cysts, glycine being over 30 times more concentrated. Values for total carbohydrates and glucose did not, however, differ significantly and total protein content was greater in primary cysts. Comparison of the (FAA) pool of secondary equine and ovine cysts revealed strain variation. It was demonstrated that most FAAs were more concentrated in hydatid fluid than in the corresponding host plasma, many concentration ratios exceeding 10. The possible contribution that mediated amino acid transport across the cyst wall and parasite amino acid metabolism makes to the composition of the FAA pool was discussed. No significant plasma aminoacidaemia was associated with infection.

The activity of the gluconeogenic enzyme, alanine-amino-transferase (ALT), in a preparation from the liver of rats was studied by means of an in vitro assay throughout the course of a primary infection of Nippostrongylus brasiliensis, established by a subcutaneous injection of approximately 4000 3rd-stage larvae. The activity was measured on days 1–14 p.i. in both uninfected and infected rats and a marked pattern in the enzyme's activity was observed. In infected rats, the activity increased from 1·46±0·19 U/g liver on day 1 p.i. to a peak on day 4 p.i. of 10·75±1·62 U/g liver, then decreased to a trough of 0·44±0·18 U/g liver on day 10 p.i. before returning to original levels by day 14 p.i., by which time the infection had been largely eliminated. In uninfected rats the activity of the liver enzyme remained constant throughout this period with a value of 2·54±0·12 U/g liver. The activity of the enzyme in vitro was found to be related to the size of the inoculum on days 4 and 10 p.i. It was proposed that these observations could be due to either (1) a direct effect of the parasite, or (2) a consequence of the host immune response to the infection. In order to investigate the second proposition more fully, liver ALT activity was investigated by in vitro assay on selected days p.i. in rats experiencing a secondary N. brasiliensis infection. In these rats the liver ALT activity was observed to reach a peak on day 2 p.i., with an activity of 3·87 ± 0-28 U/g liver, and a trough on day 4 p.i. with an activity of 0·11 ±0·03 U/g liver, returning to similar levels to those measured in uninfected rats by day 7 p.i. When serum prepared from rats having secondary N. brasiliensis infections collected on day 4 p.i. was added to the assay, a reduction in the activity of liver ALT activity from both the infected and uninfected rats was measured by in vitro assay. The results are discussed in relation to protein metabolism and gluconeogenesis in rats infected with N. brasiliensis, and also in relation to the host’s immune response to the parasitic infection.

Mean species richness and diversity of the helminth infracommunity of Apodemus sylvaticus in woodland areas of Co. Down, Northern Ireland, varied in time and space. Variation in infracommunity structure among individual hosts, however, always accounted for more than 60% of the variation in the data from different places or different times. Helminth species richness increased with increasing population density, the percentage of the host population 16 weeks old or older, and the proportion of the host population with animal material in their stomachs, at two sites monitored over 33 months. The basis for spatial variation in infracommunity structure is less certain but host dynamics and differences in diet are likely to play some role. It is concluded that analysis at the infracommunity level focuses closely on the potential for species interactions and overlap in resource utilization. Infracommunity structure, at least in the case of A. sylvaticus, varies markedly in time and space and between individual hosts. Such variation should not be ignored in comparative studies.

The time-course of low and high intensity primary infections with Heligmosomoides polygyrus was monitored in SJL and SWR mice, both of which usually expel worms within 7 weeks of larval administration. Worm expulsion in these strains was not dependent on the intensity of infection, with low and high intensity worm burdens being lost within the same period of time. The ability to expel worms rapidly was inherited in a dominant manner in F1 offspring of SJL or SWR mice mated with C57Bl10 mice; the latter being a strain in which no loss of worms was evident within 10 weeks of infection. However, neither (SJL × C57Bl10)F1 nor (SWR × C57Bl10)F1 mice expelled worms as rapidly as the parental SJL and SWR strains. (SWR × B10G)F1 [H-2q] mice eliminated worms faster than (SWR × C57Bl10)F1 [H-2bq], suggesting that the b haplotype had a moderating influence on the expulsion process. In fact (SWR × B10G)F1 mice showed a significant reduction in worm burdens by week 4 but by weeks 6–8 the rate of worm loss had slowed considerably. In contrast, SJL and SWR mice, whilst initiating rejection slightly later, (after week 4) expelled all worms within the following 2 weeks. Thus two distinct patterns of response were observed among the fast responder strains as exemplified by SWR and SJL mice on the one hand and (SWR × B10G)F1 on the other. Our results support the hypothesis that the course of a primary infection with H. polygyrus is influenced by multiple host gene loci, some of which are encoded within the MHC. SJL and SWR mice probably have similar if not identical gene combinations at loci which determine a fast responder phenotype, distinguishing them from the other mouse strains which have been studied.

The DNA from a number of free-living and parasitic nematode species was examined to determine the genomic number and distribution of DNA sequences encoding two evolutionary conserved proteins; the major sperm protein (MSP) and nematode actin. Ascaris and Caenorhabditis MSP cDNA sequences and Ascaris genomic actin sequences were used to probe Southern blots of Eco RI and Hin d III digested nematode DNA. The number of MSP genes varied widely between the 1 MSP gene in Ascaris and the 60 MSP genes in Caenorhabditis. Filarial nematodes appeared to have 1—4 MSP genes while the plant and insect parasitic species showed from 5–12 MSP-hybridizing restriction fragments. Mammalian intestinal parasites showed between 1 and 13 bands hybridizing with the MSP probes. Blots probed to estimate the number of actin genes showed that, with the exception of Ascaris which contains more than 20 germ line sequences that encode actin, all of the nematodes tested had between 3 and 9 bands that hybridized to the Ascaris genomic actin probe. The possible use of highly conserved sequences such as MSP and actin to differentiate between nematode species in diagnostic and taxonomic studies is discussed.
