Mental health before and during the COVID-19 pandemic in two longitudinal UK population cohorts

Background
The COVID-19 pandemic and mitigation measures are likely to have a marked effect on mental health. It is important to use longitudinal data to improve inferences.
AimsTo quantify the prevalence of depression, anxiety and mental well-being before and during the COVID-19 pandemic. Also, to identify groups at risk of depression and/or anxiety during the pandemic.
MethodData were from the Avon Longitudinal Study of Parents and Children (ALSPAC) index generation (n = 2850, mean age 28 years) and parent generation (n = 3720, mean age 59 years), and Generation Scotland (n = 4233, mean age 59 years). Depression was measured with the Short Mood and Feelings Questionnaire in ALSPAC and the Patient Health Questionnaire-9 in Generation Scotland. Anxiety and mental well-being were measured with the Generalised Anxiety Disorder Assessment-7 and the Short Warwick Edinburgh Mental Wellbeing Scale.
ResultsDepression during the pandemic was similar to pre-pandemic levels in the ALSPAC index generation, but those experiencing anxiety had almost doubled, at 24% (95% CI 23–26%) compared with a pre-pandemic level of 13% (95% CI 12–14%). In both studies, anxiety and depression during the pandemic was greater in younger members, women, those with pre-existing mental/physical health conditions and individuals in socioeconomic adversity, even when controlling for pre-pandemic anxiety and depression.
ConclusionsThese results provide evidence for increased anxiety in young people that is coincident with the pandemic. Specific groups are at elevated risk of depression and anxiety during the COVID-19 pandemic. This is important for planning current mental health provisions and for long-term impact beyond this pandemic.

Background
It is not clear to what extent associations between schizophrenia, cannabis use and cigarette use are due to a shared genetic etiology. We, therefore, examined whether schizophrenia genetic risk associates with longitudinal patterns of cigarette and cannabis use in adolescence and mediating pathways for any association to inform potential reduction strategies.
MethodsAssociations between schizophrenia polygenic scores and longitudinal latent classes of cigarette and cannabis use from ages 14 to 19 years were investigated in up to 3925 individuals in the Avon Longitudinal Study of Parents and Children. Mediation models were estimated to assess the potential mediating effects of a range of cognitive, emotional, and behavioral phenotypes.
ResultsThe schizophrenia polygenic score, based on single nucleotide polymorphisms meeting a training-set p threshold of 0.05, was associated with late-onset cannabis use (OR = 1.23; 95% CI = 1.08,1.41), but not with cigarette or early-onset cannabis use classes. This association was not mediated through lower IQ, victimization, emotional difficulties, antisocial behavior, impulsivity, or poorer social relationships during childhood. Sensitivity analyses adjusting for genetic liability to cannabis or cigarette use, using polygenic scores excluding the CHRNA5-A3-B4 gene cluster, or basing scores on a 0.5 training-set p threshold, provided results consistent with our main analyses.
ConclusionsOur study provides evidence that genetic risk for schizophrenia is associated with patterns of cannabis use during adolescence. Investigation of pathways other than the cognitive, emotional, and behavioral phenotypes examined here is required to identify modifiable targets to reduce the public health burden of cannabis use in the population.

Active galactic nuclei (AGN) are believed to regulate star formation inside their host galaxies through “AGN feedback”. We summarise our on-going study of luminous AGN (z ∼ 0.2−3; LAGN,bol 1043 erg s−1), which is designed to search for observational signatures of feedback by combining observed star-formation rate (SFR) measurements from statistical samples with cosmological model predictions. Using the EAGLE hydrodynamical cosmological simulations, in combination with our Herschel + ALMA surveys, we show that – even in the presence of AGN feedback – we do not necessarily expect to see any relationships between average galaxy-wide SFRs and instantaneous AGN luminosities. We caution that the correlation with stellar mass for both SFR and AGN luminosity can contribute to apparent observed positive trends between these two quantities. On the other hand, the EAGLE simulations, which reproduce our observations, predict that a signature of AGN feedback can be seen in the wide specific SFR distributions of all massive galaxies (not just AGN hosts). Overall, whilst we can not rule out that AGN have an immediate small-scale impact on in-situ star-formation, all of our results are consistent with a feedback model where galaxy-wide in-situ star formation is not rapidly suppressed by AGN, but where the feedback likely acts over a longer timescale than a single AGN episode.

Seven half-day regional listening sessions were held between December 2016 and April 2017 with groups of diverse stakeholders on the issues and potential solutions for herbicide-resistance management. The objective of the listening sessions was to connect with stakeholders and hear their challenges and recommendations for addressing herbicide resistance. The coordinating team hired Strategic Conservation Solutions, LLC, to facilitate all the sessions. They and the coordinating team used in-person meetings, teleconferences, and email to communicate and coordinate the activities leading up to each regional listening session. The agenda was the same across all sessions and included small-group discussions followed by reporting to the full group for discussion. The planning process was the same across all the sessions, although the selection of venue, time of day, and stakeholder participants differed to accommodate the differences among regions. The listening-session format required a great deal of work and flexibility on the part of the coordinating team and regional coordinators. Overall, the participant evaluations from the sessions were positive, with participants expressing appreciation that they were asked for their thoughts on the subject of herbicide resistance. This paper details the methods and processes used to conduct these regional listening sessions and provides an assessment of the strengths and limitations of those processes.

Herbicide resistance is ‘wicked’ in nature; therefore, results of the many educational efforts to encourage diversification of weed control practices in the United States have been mixed. It is clear that we do not sufficiently understand the totality of the grassroots obstacles, concerns, challenges, and specific solutions needed for varied crop production systems. Weed management issues and solutions vary with such variables as management styles, regions, cropping systems, and available or affordable technologies. Therefore, to help the weed science community better understand the needs and ideas of those directly dealing with herbicide resistance, seven half-day regional listening sessions were held across the United States between December 2016 and April 2017 with groups of diverse stakeholders on the issues and potential solutions for herbicide resistance management. The major goals of the sessions were to gain an understanding of stakeholders and their goals and concerns related to herbicide resistance management, to become familiar with regional differences, and to identify decision maker needs to address herbicide resistance. The messages shared by listening-session participants could be summarized by six themes: we need new herbicides; there is no need for more regulation; there is a need for more education, especially for others who were not present; diversity is hard; the agricultural economy makes it difficult to make changes; and we are aware of herbicide resistance but are managing it. The authors concluded that more work is needed to bring a community-wide, interdisciplinary approach to understanding the complexity of managing weeds within the context of the whole farm operation and for communicating the need to address herbicide resistance.

The surfaces of fresh and tarnished (through exposure to the atmosphere) bornite have been studied by measurement of reflectance and quantitative colour parameters in the visible region, X-ray photoelectron and Auger electron spectroscopy, and conversion electron Mössbauer spectroscopy. The results of these studies, in combination with those of previous workers, indicate that a surface coating of an iron hydroxy-oxide forms on bornite, leaving beneath it a layer with a copper sulphide composition. A model to explain this alteration process is proposed based on a suggested surface reconstruction in bornite, leading to metals in essentially trigonal planar coordination. Resulting destabilization of iron relative to copper in this crystal chemical environment is suggested to account for its preferential oxidation.

The Driggith vein is representative of the lead-zinc mineralization in the Caldbeck Fells area, with a primary mineral assemblage consisting of quartz, baryte, calcite, chalcedony, pyrite, arsenopyrite, galena, sphalerire, bournonite, argentian tetrahedrite, and native antimony. Broadly similar assemblages are found in Pb-Zn mineralization of Upper Devonian to Permian age else-where in the Lake District. The paragenetic sequence appears to have been early pyrite and minor arsenopyrite associated with quartz; with later baryte, calcite, sphalerite, and chalcopyrite enclosed by galena. Within the galena occur inclusions of tetrahedrite, bournonite, and native antimony. A later stage of baryte mineralization was followed by the alteration of primary sulphides to secondary minerals. Elsewhere in the Lake District, galena occurring in this association also contains inclusions of native antimony, bournonite, and other sulphosalts, and in this respect differs from galena associated with earlier veins composed dominantly of chalcopyrite, pyrite, and arsenopyrite. The inclusions make a major contribution to the high antimony values reported in trace element analyses of galena from the Lake District. The occurrence of these inclusions, which in some cases display preferred orientation within the host galena, is discussed in terms of phase relations involving PbS and AgSbS2.
2460: Change in duration of postoperative antibiotic prophylaxis in esophagectomy patients: Outcomes in a single academic institution

OBJECTIVES/SPECIFIC AIMS: Ivor-Lewis esophagectomy (ILE) is an invasive surgical procedure with a high incidence of postoperative pneumonia. Antibiotic prophylaxis could reduce respiratory infections but increase Clostridium difficile and antibiotic resistance. Our institution reduced the duration of piperacillin-tazobactam prophylaxis following ILE from 4 to 1 day or less in January 2015. We evaluated short-term outcomes in ILE patients before and after this institutional change. METHODS/STUDY POPULATION: Retrospective cohort study of all ILE patients from 2012 to 2016. We confirmed antibiotic duration directly from nursing medication administration records. The primary outcomes of this study were rates of C. difficile and postoperative pneumonia. Secondary outcomes include other infection, length of hospital stay, and readmission within 30 days. We used logistic regression to analyze impact of days of antibiotics and χ2 or Fischer exact tests for categorical variables. RESULTS/ANTICIPATED RESULTS: Of 104 ILE patients, 40.4% (n=42) were after January 2015, 11.5% developed pneumonia and 5.8% developed C. difficile colitis. ILE patients received more days of antibiotics before the institutional change compared with after (6.1 vs. 2.9 d, p<0.01). For a 1-day increase in antibiotic duration, the odds of acquiring C. difficile increased significantly by 1.2 (p=0.03). Before compared with after the institutional change, rates of C. difficile were 8.1% Versus 2.4% (p>0.2), rates of pneumonia were 11.3% Versus 11.9% (p>0.2), and length of stay was 10.9 Versus 10.5 days (p>0.2), respectively. DISCUSSION/SIGNIFICANCE OF IMPACT: Institutional policy can have an impact on patient outcomes. Antibiotic stewardship is associated with reduced rates of inpatient C. difficile. Our study suggests reduced antibiotics are not associated with pneumonia, although larger studies are necessary to confirm this finding. Surgeons should consider the benefit of decreased rates of C. difficile before administering prolonged antibiotic prophylaxis following esophagectomies.
Palmer Amaranth (Amaranthus palmeri) Management in Dicamba-Resistant Cotton

Cotton growers rely heavily upon glufosinate and various residual herbicides applied preplant, PRE, and POST to control Palmer amaranth resistant to glyphosate and acetolactate synthase-inhibiting herbicides. Recently deregulated in the United States, cotton resistant to dicamba, glufosinate, and glyphosate (B2XF cotton) offers a new platform for controlling herbicide-resistant Palmer amaranth. A field experiment was conducted in North Carolina and Georgia to determine B2XF cotton tolerance to dicamba, glufosinate, and glyphosate and to compare Palmer amaranth control by dicamba to a currently used, nondicamba program in both glufosinate- and glyphosate-based systems. Treatments consisted of glyphosate or glufosinate applied early POST (EPOST) and mid-POST (MPOST) in a factorial arrangement of treatments with seven dicamba options (no dicamba, PRE, EPOST, MPOST, PRE followed by [fb] EPOST, PRE fb MPOST, and EPOST fb MPOST) and a nondicamba standard. The nondicamba standard consisted of fomesafen PRE, pyrithiobac EPOST, and acetochlor MPOST. Dicamba caused no injury when applied PRE and only minor, transient injury when applied POST. At time of EPOST application, Palmer amaranth control by dicamba or fomesafen applied PRE, in combination with acetochlor, was similar and 13 to 17% greater than acetochlor alone. Dicamba was generally more effective on Palmer amaranth applied POST rather than PRE, and two applications were usually more effective than one. In glyphosate-based systems, greater Palmer amaranth control and cotton yield were obtained with dicamba applied EPOST, MPOST, or EPOST fb MPOST compared with the standard herbicides in North Carolina. In contrast, dicamba was no more effective than the standard herbicides in the glufosinate-based systems. In Georgia, dicamba was as effective as the standard herbicides in a glyphosate-based system only when dicamba was applied EPOST fb MPOST. In glufosinate-based systems in Georgia, dicamba was as effective as standard herbicides only when dicamba was applied twice.
Quaternary depositional patterns and sea-level fluctuations, Northeastern North Carolina

A detailed record of late Quaternary sea-level oscillations is preserved within the upper 45 m of deposits along an eight km transect across Croatan Sound, a drowned tributary of the Roanoke/Albemarle drainage system, northeastern North Carolina. Drill-hole and seismic data reveal nine relatively complete sequences filling an antecedent valley comprised of discontinuous middle and early Pleistocene deposits. On interfluves, lithologically similar marine deposits of different sequences occur stacked in vertical succession and separated by ravinement surfaces. Within the paleo-drainage, marine deposits are separated by fluvial and/or estuarine sediments deposited during periods of lowered sea level. Foraminiferal and molluscan fossil assemblages indicate that marine facies were deposited in a shallow-marine embayment with open connection to shelf waters. Each sequence modifies or truncates portions of the preceding sequence or sequences. Sequence boundaries are the product of a combination of fluvial, estuarine, and marine erosional processes. Stratigraphic and age analyses constrain the ages of sequences to late Marine Isotope Stage (MIS) 6 and younger (∼ 140 ka to present), indicating multiple sea-level oscillations during this interval. Elevations of highstand deposits associated with late MIS 5 and MIS 3 imply that sea level was either similar to present during those times, or that the region may have been influenced by glacio-isostatic uplift and subsidence.
Rapid Holocene coastal change revealed by high-resolution micropaleontological analysis, Pamlico Sound, North Carolina, USA

Foraminiferal analyses of 404 contiguous samples, supported by diatom, lithologic, geochronologic and seismic data, reveal both rapid and gradual Holocene paleoenvironmental changes in an 8.21-m vibracore taken from southern Pamlico Sound, North Carolina. Data record initial flooding of a latest Pleistocene river drainage and the formation of an estuary 9000 yr ago. Estuarine conditions were punctuated by two intervals of marine influence from approximately 4100 to 3700 and 1150 to 500 cal yr BP. Foraminiferal assemblages in the muddy sand facies that accumulated during these intervals contain many well-preserved benthic foraminiferal species, which occur today in open marine settings as deep as the mid shelf, and significant numbers of well-preserved planktonic foraminifera, some typical of Gulf Stream waters. We postulate that these marine-influenced units resulted from temporary destruction of the southern Outer Banks barrier islands by hurricanes. The second increase in marine influence is coeval with increased rate of sea-level rise and a peak in Atlantic tropical cyclone activity during the Medieval Climate Anomaly. This high-resolution analysis demonstrates the range of environmental variability and the rapidity of coastal change that can result from the interplay of changing climate, sea level and geomorphology in an estuarine setting.
Annual Grass Control in Peanut (Arachis hypogaea) with Clethodim and Imazapic

Field experiments were conducted to evaluate possible interactions of clethodim with imazapic applied as mixtures or sequentially for control of broadleaf signalgrass, fall panicum, goosegrass, and large crabgrass. Imazapic at 70 g ai/ha alone controlled grass weeds inconsistently, whereas clethodim at 140 g ai/ha alone controlled the same weeds at least 99%. Imazapic did not affect broadleaf signalgrass control by clethodim. Reduced control of fall panicum, goosegrass, and large crabgrass was observed when clethodim and imazapic were applied in mixture. Antagonism of clethodim occurred when clethodim was applied 1 d before or up to 3 d after application of imazapic (fall panicum and large crabgrass). Antagonism of goosegrass control was noted when imazapic was applied 3 d before or up to 7 d after application of clethodim. In other experiments, large crabgrass and Texas panicum control by clethodim (70 and 140 g/ha) applied alone or with imazapic (70 g/ ha) or bentazon (1.1 kg ai/ha) plus 2,4-DB (0.28 kg ai/ha) either with or without ammonium sulfate (2.8 kg/ha) was evaluated. Texas panicum control by clethodim was reduced by imazapic regardless of the ammonium sulfate rate. However, large crabgrass control by imazapic was not affected in these experiments. Control of both grasses by clethodim was reduced substantially by bentazon plus 2,4-DB, although in some instances ammonium sulfate improved control when in mixture. Ammonium sulfate improved control by clethodim in some instances irrespective of the broadleaf–sedge herbicide treatments.

Exploring cryptic diversity in publicly available strains of the model diatom Thalassiosira pseudonana (Bacillariophyceae)

The model diatom Thalassiosira pseudonana is believed to be a single species with a global distribution, but it has not been confirmed previously whether isolates from different environmental and geographic origins are genotypically and phenotypically identical. In the present study, a polyphasic approach was employed to characterize nine clonal isolates, plus an additional replicate of one of the isolates, of the diatom T. pseudonana from culture collections to investigate whether there was any cryptic speciation in the publicly available strains of this species. Morphological analysis using scanning electron microscopy concluded that the strains were indistinguishable. Furthermore, conventional DNA barcoding genes (SSU rDNA, ITS1 and ITS2 rDNA and rbcL), revealed no nucleotide variation among the strains tested. On employing a whole genome fingerprinting technique, Amplified Fragment Length Polymorphism (AFLP), three clusters were revealed, although the level of variation between the clusters was surprisingly low. These findings indicate a low level of diversity among these cultured T. pseudonana strains, despite their wide spatial and temporal distribution and the salinity range of their original habitats. Based on the limited number of available strains, this suggests that T. pseudonana is a highly conserved diatom that nevertheless has an ability to tolerate wide ranges of salinity and populate varied geographic locations.
