Parsing components of auditory predictive coding in schizophrenia using a roving standard mismatch negativity paradigm

Background
Mismatch negativity (MMN) is an event-related potential (ERP) component reflecting auditory predictive coding. Repeated standard tones evoke increasing positivity (‘repetition positivity’; RP), reflecting strengthening of the standard's memory trace and the prediction it will recur. Likewise, deviant tones preceded by more standard repetitions evoke greater negativity (‘deviant negativity’; DN), reflecting stronger prediction error signaling. These memory trace effects are also evident in MMN difference wave. Here, we assess group differences and test-retest reliability of these indices in schizophrenia patients (SZ) and healthy controls (HC).
MethodsElectroencephalography was recorded twice, 2 weeks apart, from 43 SZ and 30 HC, during a roving standard paradigm. We examined ERPs to the third, eighth, and 33rd standards (RP), immediately subsequent deviants (DN), and the corresponding MMN. Memory trace effects were assessed by comparing amplitudes associated with the three standard repetition trains.
ResultsCompared with controls, SZ showed reduced MMNs and DNs, but normal RPs. Both groups showed memory trace effects for RP, MMN, and DN, with a trend for attenuated DNs in SZ. Intraclass correlations obtained via this paradigm indicated good-to-moderate reliabilities for overall MMN, DN and RP, but moderate to poor reliabilities for components associated with short, intermediate, and long standard trains, and poor reliability of their memory trace effects.
ConclusionMMN deficits in SZ reflected attenuated prediction error signaling (DN), with relatively intact predictive code formation (RP) and memory trace effects. This roving standard MMN paradigm requires additional development/validation to obtain suitable levels of reliability for use in clinical trials.

Background: Despite its inhibitory control requirements, antisaccade deficits have been consistently associated with working memory impairments in schizophrenia. We investigated whether variance in antisaccade performance could be better accounted for in terms of a specific inhibitory function. Method: We assessed 48 clinically stable out-patients with schizophrenia on an antisaccade task, as well as on measures of spatial and verbal working memory, sustained selective attention, and a simple motoric go/no-go measure of response inhibition. Results: In a stepwise multiple regression analysis, go/no-go task performance accounted for a considerably greater percentage of variance in antisaccade performance (25.3%) than either working memory (8.4%) or sustained selective attention task (9.1%). Discussion: We conclude that antisaccade deficits in schizophrenia appear to be better understood in terms of a specific deficit of inhibitory control than in terms of more general difficulties with context maintenance or goal neglect. Respite for Dementia Caregivers: The Effects of Adult Day Service Use on Caregiving Hours and Care Demands

The objective of this study was to determine whether adult day service use was related to decreases in primary caregiving hours (i.e., the time caregivers spent on activities of daily living/instrumental activities of daily living and behavior problems for care recipients) and care recipient function for these domains. Three-month longitudinal data from the Adult Day Care Collaborative Study (N = 400) were used. Adult day service users reported greater decreases in hours spent on behavior problems when compared to nonusers, even after controlling for baseline differences between the two groups. In addition, adult day service users reported decreased frequency of behavior problems in their relatives who attended adult day programs. The findings suggest that adult day services, if used over time, are effective in restructuring caregiving time and may offer potential benefits not only to family caregivers but to community-residing older adults who have dementia as well.

The first Welshman: excavations at Pontnewydd

Pontnewydd Cave lies in the Elwy Valley, near the western edge of the Vale of Clywd (FIG. 1), six km north-west of the modern town of Denbigh. The site first became well-known through the successive investigations of the nineteenth-century geologists Professors William Boyd Dawkins and T. H. McKenny Hughes, the latter working with the Reverend D. R. Thomas, then Vicar of Cefn. Boyd Dawkins' s excavations at the site (1874, 286–7; 1880, 192) yielded a fauna which included hippopotamus, but no artifacts were observed. The work of Hughes & Thomas (1874; Hughes, 1887) seems to have been confined principally to scouring Boyd Dawkins' s dumps, and yielded not only fauna but also evidence of the presence of man in the form both of artifacts and of a human molar tooth, since lost. This molar has long excited interest on account of its description by Busk (in Hughes & Thomas, 1874, 390) as being ‘of very large size’ and looking ‘quite as ancient as the rest [of the fauna]’. One of the many research aims of the current investigation was, therefore, to examine the possibility that the cave was a site where human remains might be found in association with an archaeological industry.
