Background:
Obesity is highly prevalent and disabling, especially in individuals with severe mental illness including bipolar disorders (BD). The brain is a target organ for both obesity and BD. Yet, we do not understand how cortical brain alterations in BD and obesity interact.
Methods:We obtained body mass index (BMI) and MRI-derived regional cortical thickness, surface area from 1231 BD and 1601 control individuals from 13 countries within the ENIGMA-BD Working Group. We jointly modeled the statistical effects of BD and BMI on brain structure using mixed effects and tested for interaction and mediation. We also investigated the impact of medications on the BMI-related associations.
Results:BMI and BD additively impacted the structure of many of the same brain regions. Both BMI and BD were negatively associated with cortical thickness, but not surface area. In most regions the number of jointly used psychiatric medication classes remained associated with lower cortical thickness when controlling for BMI. In a single region, fusiform gyrus, about a third of the negative association between number of jointly used psychiatric medications and cortical thickness was mediated by association between the number of medications and higher BMI.
Conclusions:We confirmed consistent associations between higher BMI and lower cortical thickness, but not surface area, across the cerebral mantle, in regions which were also associated with BD. Higher BMI in people with BD indicated more pronounced brain alterations. BMI is important for understanding the neuroanatomical changes in BD and the effects of psychiatric medications on the brain.

Background
Studying phenotypic and genetic characteristics of age at onset (AAO) and polarity at onset (PAO) in bipolar disorder can provide new insights into disease pathology and facilitate the development of screening tools.
AimsTo examine the genetic architecture of AAO and PAO and their association with bipolar disorder disease characteristics.
MethodGenome-wide association studies (GWASs) and polygenic score (PGS) analyses of AAO (n = 12 977) and PAO (n = 6773) were conducted in patients with bipolar disorder from 34 cohorts and a replication sample (n = 2237). The association of onset with disease characteristics was investigated in two of these cohorts.
ResultsEarlier AAO was associated with a higher probability of psychotic symptoms, suicidality, lower educational attainment, not living together and fewer episodes. Depressive onset correlated with suicidality and manic onset correlated with delusions and manic episodes. Systematic differences in AAO between cohorts and continents of origin were observed. This was also reflected in single-nucleotide variant-based heritability estimates, with higher heritabilities for stricter onset definitions. Increased PGS for autism spectrum disorder (β = −0.34 years, s.e. = 0.08), major depression (β = −0.34 years, s.e. = 0.08), schizophrenia (β = −0.39 years, s.e. = 0.08), and educational attainment (β = −0.31 years, s.e. = 0.08) were associated with an earlier AAO. The AAO GWAS identified one significant locus, but this finding did not replicate. Neither GWAS nor PGS analyses yielded significant associations with PAO.
ConclusionsAAO and PAO are associated with indicators of bipolar disorder severity. Individuals with an earlier onset show an increased polygenic liability for a broad spectrum of psychiatric traits. Systematic differences in AAO across cohorts, continents and phenotype definitions introduce significant heterogeneity, affecting analyses.
Sea-ice thickness in the coastal northeastern Chukchi Sea from moored ice-profiling sonar

Time series ice-draft data were obtained from moored ice-profiling sonar (IPS), in the coastal northeastern Chukchi Sea during 2009/10. Time series data show seasonal growth of sea-ice draft, occasionally interrupted by coastal polynya. The sea-ice draft distribution indicates a slightly lower abundance of thick, deformed ice compared with the eastern Beaufort Sea. In January, a rapid increase in the abundance of thick ice coincided with a period of minimal drift indicating compaction again the coast and dynamical thickening. The overall mean draft and corresponding derived thickness are 1.27 and 1.38 m, respectively. The evolution of modal ice thickness observed can be explained mostly by thermodynamic growth. The derived ice thicknesses are used to estimate heat losses based on ERA-interim data. Heat losses from the raw, 1 s IPS data are ~50 and 100% greater than those calculated using IPS data averaged over spatial scales of ~20 and 100 km, respectively. This finding demonstrates the importance of subgrid-scale ice-thickness distribution for heat-loss calculation. The heat-loss estimate based on thin ice data derived from AMSR-E data corresponds well with that from the 1 s observed ice-thickness data, validating heat-loss estimates from the AMSR-E thin ice-thickness algorithm.

Data from the Seasonal Ice Zone Observing Network (SIZONet) acquired near Barrow, Alaska, during the 2009/10 ice season allow novel comparisons between measurements of ice thickness and velocity. An airborne electromagnetic survey that passed over a moored Ice Profiling Sonar (IPS) provided coincident independent measurements of total ice and snow thickness and ice draft at a scale of 10 km. Once differences in sampling footprint size are accounted for, we reconcile the respective probability distributions and estimate the thickness of level sea ice at 1.48 ± 0.1 m, with a snow depth of 0.12 ± 0.07 m. We also complete what we believe is the first independent validation of radar-derived ice velocities by comparing measurements from a coastal radar with those from an under-ice acoustic Doppler current profiler (ADCP). After applying a median filter to reduce high-frequency scatter in the radar-derived data, we find good agreement with the ADCP bottom-tracked ice velocities. With increasing regulatory and operational needs for sea-ice data, including the number and thickness of pressure ridges, coordinated observing networks such as SIZONet can provide the means of reducing uncertainties inherent in individual datasets.
Observations of supercooled water and frazil ice formation in an Arctic coastal polynya from moorings and satellite imagery

Formation of supercooled water and frazil ice was studied in the Chukchi Sea coastal polynya off Barrow, Alaska, USA, in winter 2009/10, using moored salinity/temperature sensors and Ice Profiling Sonar (IPS) data along with satellite data. Oceanographic data from two moorings revealed episodic events of potential supercooling at 30–40m depth, including the possibility of in situ supercooling, while the polynya was open. We identified frazil ice-like signals in the IPS data down to 5–15 m depth, associated with large heat loss and windy, turbulent conditions in an active polynya. This likely represents the first IPS observation of frazil ice in the marine environment. On the day of the maximum signal of frazil ice, spaceborne synthetic aperture radar shows streaks of high backscatter within the polynya, indicating active frazil ice formation just downwind of the mooring sites. In addition, the longer-term potential supercooling that persisted for 1–3 weeks occurred twice despite the absence of polynya activity at the mooring sites. These events occurred during periods dominated by the northeastward current. A series of coastal polynyas had formed southwest of the mooring sites prior to these events. Thus, the water masses with potential supercooling were likely advected from these polynyas.

It has been postulated that aging is the consequence of an accelerated accumulation of somatic DNA mutations and that subsequent errors in the primary structure of proteins ultimately reach levels sufficient to affect organismal functions. The technical limitations of detecting somatic changes and the lack of insight about the minimum level of erroneous proteins to cause an error catastrophe hampered any firm conclusions on these theories. In this study, we sequenced the whole genome of DNA in whole blood of two pairs of monozygotic (MZ) twins, 40 and 100 years old, by two independent next-generation sequencing (NGS) platforms (Illumina and Complete Genomics). Potentially discordant single-base substitutions supported by both platforms were validated extensively by Sanger, Roche 454, and Ion Torrent sequencing. We demonstrate that the genomes of the two twin pairs are germ-line identical between co-twins, and that the genomes of the 100-year-old MZ twins are discerned by eight confirmed somatic single-base substitutions, five of which are within introns. Putative somatic variation between the 40-year-old twins was not confirmed in the validation phase. We conclude from this systematic effort that by using two independent NGS platforms, somatic single nucleotide substitutions can be detected, and that a century of life did not result in a large number of detectable somatic mutations in blood. The low number of somatic variants observed by using two NGS platforms might provide a framework for detecting disease-related somatic variants in phenotypically discordant MZ twins.

The scores of 269 depressed patients on the Depressive Category-Type Scale were analysed. The distributions of component and total scores were found to be bimodal, indicating that most depressed patients can be divided into an endogenous group and another characterized by the presence of neurotic features and the absence of endogenous ones. These findings were discussed in relation to those of other investigators. The relationship between the differentiation of syndromes and the classification of patients into corresponding diagnostic groups was considered. Finally, it was suggested that it should be interesting to try to estimate, from published data, the correlation between endogenous and neurotic depression in the general population.

An abattoir survey was undertaken to determine the prevalence of foodborne zoonotic organisms colonizing cattle, sheep and pigs at slaughter in Great Britain. The study ran for 12 months from January 2003, involved 93 abattoirs and collected 7703 intestinal samples. The design was similar to two previous abattoir surveys undertaken in 1999–2000 allowing comparisons. Samples were examined for VTEC O157, Salmonella, thermophilic Campylobacter and Yersinia enterocolitica. The prevalence of VTEC O157 faecal carriage was 4·7% in cattle, 0·7% in sheep and 0·3% in pigs. A significant decrease in sheep was detected from the previous survey (1·7%). Salmonella carriage was 1·4% in cattle, a significant increase from the previous survey of 0·2%. In sheep, faecal carriage was 1·1% a significant increase from the previous survey (0·1%). In pigs, carriage was 23·4%, consistent with the previous study. Thermophilic Campylobacter spp. were isolated from 54·6% of cattle, 43·8% of sheep and 69·3% of pigs. Y. enterocolitica was isolated from 4·5% of cattle, 8·0% of sheep and 10·2% of pigs.

The performance of Hereford × Friesian calves sired by Hereford bulls selected for either lean growth rate (LGR) or lean food conversion ratio (LFCR), or by unselected Hereford bulls, was evaluated on 327 calves on three farms over 2 years. Animals started test at 130 days of age on average and remained on test for approximately 300 days, whereupon all animals were slaughtered and carcass dissections were undertaken. Individual food intake was measured on tioo of the farms (189 animals), but only intake per pen of animals was measured on the third farm and individual food intake had to be estimated. LGR, LFCR and other traits describing performance were calculated from the growth, food intake and carcass composition data.
If no breed or environment interactions exist it is expected that proportionately 0-5 of the genetic differences between selected and control line bulls would be transmitted to their offspring. For the LGR and LFCR line bulls these values were 0·38 and 0·44, respectively, however the LGR value had a much smaller confidence interval. Genetic correlations derived from regressing breeding values predicted from offspring performance on breeding values predicted from the bulk's own performance in the selection experiment were 0·62 (s.e. 0·28) and 0·96 (s.e. 0·28) for LGR and LFCR, respectively. Heritabilities were: live-weight gain on test, 0·27; daily gain, 0·48; food intake, 0·06; food conversion ratio, 0·46; predicted carcass lean content, 0·10; killing-out proportion, 0·10; LGR, 0·36 and LFCR, 0·48.

The development of resistance to insecticides in insect pests is an important and increasing problem. As insecticide use increases in tropical areas, the numbers of resistant insect strains will increase.
The adverse effects of insecticide resistance include crop losses, increased production costs, increased environmental hazards, and socio-economic problems. The increasing difficulties and costs of discovering and developing new insecticides together with the widespread occurrence of resistant strains of insect pests means that insecticides are a limited resource that must be conserved. Methods of conserving insecticides include sensible management of their use and the implementation of IPM programmes.
The problems of insecticide resistance and strategies for minimizing and managing resistance in tropical areas are discussed using examples of insect pests of stored products, cotton, rice and bananas.
The effect of altering the hormone status of ewes on the outflow rate of protein supplements from the rumen and so on protein degradability

Four experiments were carried out on individually penned Finn Dorset and Suffolk × Finn Dorset ewes receiving complete diets of milled hay and concentrates and rationed at various levels below voluntary intake. In the first experiment, 20 non-pregnant non-lactating ewes were each given daily 1·6 kg food and injected intramuscularly with either isotonic saline (control), progesterone plus oestrogen (20 mg + 80 ng per 24 h) to mimic hormone status in pregnancy, bromocriptine (1 mg per 12 h) to suppress plasma prolactin, or thyrotrophin releasing factor (TRF) to elevate plasma concentrations of prolactin and the iodothyronines. Fractional outflow rates of chromium-mordanted fish meal from the rumen were estimated from the rate of excretion of chromium in faeces and were proportionately 0·25 higher for ewes given TRF than for controls (P < 0·025). None of the other treatments had a significant effect on outflow rate.
In experiment 2, fractional outflow rates from the rumen were determined during July (long daylength) and again during December (short daylength) in 12 ewes that had their seasonal pattern of plasma prolactin modified by removal of their anterior cervical sympathetic ganglia. Another 12 sham-operated ewes were used as controls. Neither the seasonal changes in prolactin nor their modification by cervical ganglionectomy significantly affected outflow rate.
Twelve non-pregnant non-lactating ewes were used in the third experiment to test the effects of mid-winter shearing (mean ambient temperature −4°C) on fractional outflow rates from the rumen and on the plasma concentrations of prolactin, thyroxine (T4) and cortisol. Shearing caused a drop in plasma prolactin, gave proportional increases (P < 0·05) of 0·47 and 0·16 in T4 and cortisol respectively and gave a 0·12 increase (P < 0·01) in the outflow rate of chromium-mordanted fish meal from the rumen. The increases in T4, and outflow rate were sustained for at least 2 weeks.
For experiment 4, 22 ewes each suckling two lambs were used in three trials to see if variation between ewes in milk production and in lamb growth rate was correlated with variation in outflow rate from the rumen. The only significant correlation (r = 0·68, P < 0·01) was between outflow rate and lamb growth rate for the eight ewes in trial 1. The results of all four experiments are discussed in relation to the mechanisms by which non-dietary factors, via their effect on the rate of outflow of digesta from the rumen, may alter the quantity of protein reaching the abomasum and the likely significance of this alteration on animal performance.
School Phobia: A Therapeutic Trial with Clomipramine and Short-Term Outcome

A double-blind trial failed to demonstrate any significant short-term effects of clomipramine in doses recommended for use in general practice (in addition to the usual range of psychotherapeutic help) in the treatment of children with school refusal and neurotic disorder. Patterns of improvement were also studied for the sample as a whole irrespective of treatment. Neither age nor sex were significantly related to improvement, except on one behavioural measure where girls initially did better than boys. In addition, it was found that there was a rapid relief of depression but neurotic symptomatology tended to persist.
The performance of beef cattle given diets of barley supplemented with either vegetable protein or non-protein nitrogen

1. Four growth trials were carried out with 297 group-fed steers given diets based on barley, minerals and vitamins, unsupplemented or supplemented with either soya bean meal or various sources of non-protein nitrogen.
2. Steers given unsupplemented barley (11% crude protein in dry matter) had inferior growth rates up to 300 kg live weight but thereafter were equal to those given barley supplemented with urea in the drinking water or soya bean meal.
3. There were no significant differences in the growth rates of steers given barley supplemented with soya bean meal and those given barley supplemented with ammonium and mineral salts of formic acid or a mixture of acids (predominantly acetic).
4. In two out of three trials, tenth-rib joints from steers given barley supplemented with ammonium salts of mixed acids contained less fat and more protein and water than those from steers given barley supplemented with soya bean meal or proprietary protein supplements.
5. A number of possible explanations for the apparent carcass differences is discussed.
