Introduction
COMP360 is a synthetic, proprietary, purified form of psilocybin in development for treatment-resistant depression (TRD) with FDA Breakthrough Therapy designation. In a recent phase IIb study, COMP360 psilocybin 25mg was superior to 1mg on change from baseline (CFB) to Week 3 on the Montgomery-Åsberg Depression Rating Scale (MADRS) total score (primary efficacy endpoint), when administered alongside psychological support. Quick Inventory of Depressive Symptomatology-Self Rated (QIDS-SR16) total score (exploratory efficacy endpoint) showed similar results.
ObjectivesTo analyse changes in specific depression symptoms after psilocybin treatment in the aforementioned study, as measured by individual item scores on the MADRS and QIDS-SR16 (range 0-6 and 0-3).
MethodsParticipants with TRD were randomised to single doses of psilocybin 25mg (n=79), 10mg (n=75), or 1mg (n=79). A remote, blinded rater assessed the MADRS at Baseline, Day 2 (the day post-psilocybin), and Weeks 1, 3, 6, 9, and 12. The QIDS-SR16 was self-rated at Baseline, Day 1, Day 2, and Weeks 1, 2, 3, 6, 9, and 12. At each time point, descriptive statistics were calculated for each MADRS and QIDS-SR16 individual item score.
ResultsAt Week 3, MADRS items with the largest differences in mean CFB in the 25mg arm were Inability to Feel, Apparent Sadness, Lassitude, and Reported Sadness. Greater improvement in the 25mg arm was apparent from Day 2 and remained to Week 12 (Lassitude remained to Week 6 only). On the QIDS-SR16, the item with the largest difference in mean CFB at Week 3 in the 25mg arm was in Feeling Sad and remained evident to Week 12 (Table 1).

As anhedonia is predictive of poorer treatment response, and improvements in anhedonia correlate with improvements in functioning, it is important to understand the impact of treatments on this symptom.

Objective:
The coronavirus disease 2019 (COVID-19) pandemic has resulted in shortages of personal protective equipment (PPE), underscoring the urgent need for simple, efficient, and inexpensive methods to decontaminate masks and respirators exposed to severe acute respiratory coronavirus virus 2 (SARS-CoV-2). We hypothesized that methylene blue (MB) photochemical treatment, which has various clinical applications, could decontaminate PPE contaminated with coronavirus.
Design:The 2 arms of the study included (1) PPE inoculation with coronaviruses followed by MB with light (MBL) decontamination treatment and (2) PPE treatment with MBL for 5 cycles of decontamination to determine maintenance of PPE performance.
Methods:MBL treatment was used to inactivate coronaviruses on 3 N95 filtering facepiece respirator (FFR) and 2 medical mask models. We inoculated FFR and medical mask materials with 3 coronaviruses, including SARS-CoV-2, and we treated them with 10 µM MB and exposed them to 50,000 lux of white light or 12,500 lux of red light for 30 minutes. In parallel, integrity was assessed after 5 cycles of decontamination using multiple US and international test methods, and the process was compared with the FDA-authorized vaporized hydrogen peroxide plus ozone (VHP+O3) decontamination method.
Results:Overall, MBL robustly and consistently inactivated all 3 coronaviruses with 99.8% to >99.9% virus inactivation across all FFRs and medical masks tested. FFR and medical mask integrity was maintained after 5 cycles of MBL treatment, whereas 1 FFR model failed after 5 cycles of VHP+O3.
Conclusions:MBL treatment decontaminated respirators and masks by inactivating 3 tested coronaviruses without compromising integrity through 5 cycles of decontamination. MBL decontamination is effective, is low cost, and does not require specialized equipment, making it applicable in low- to high-resource settings.

Summary: In this paper we build on work investigating the feasibility of human immunodeficiency virus (HIV) testing in emergency departments (EDs), estimating the prevalence of hepatitis B, C and HIV infections among persons attending two inner-London EDs, identifying factors associated with testing positive in an ED. We also undertook molecular characterisation to look at the diversity of the viruses circulating in these individuals, and the presence of clinically significant mutations which impact on treatment and control.
Blood-borne virus (BBV) testing in non-traditional settings is feasible, with emergency departments (ED) potentially effective at reaching vulnerable and underserved populations. We investigated the feasibility of BBV testing within two inner-London EDs. Residual samples from biochemistry for adults (⩾18 years) attending The Royal Free London Hospital (RFLH) or the University College London Hospital (UCLH) ED between January and June 2015 were tested for human immunodeficiency virus (HIV)Ag/Ab, anti-hepatitis C (HCV) and HBsAg. PCR and sequence analysis were conducted on reactive samples. Sero-prevalence among persons attending RFH and UCLH with residual samples (1287 and 1546), respectively, were 1.1% and 1.0% for HBsAg, 1.6% and 2.3% for anti-HCV, 0.9% and 1.6% for HCV RNA, and 1.3% and 2.2% for HIV. For RFH, HBsAg positivity was more likely among persons of black vs. white ethnicity (odds ratio 9.08; 95% confidence interval 2.72–30), with anti-HCV positivity less likely among females (0.15, 95% CI 0.04–0.50). For UCLH, HBsAg positivity was more likely among non-white ethnicity (13.34, 95% CI 2.20–80.86 (Asian); 8.03, 95% CI 1.12–57.61 (black); and 8.11, 95% CI 1.13–58.18 (other/mixed)). Anti-HCV positivity was more likely among 36–55 year olds vs. ⩾56 years (7.69, 95% CI 2.24–26.41), and less likely among females (0.24, 95% CI 0.09–0.65). Persons positive for HIV-markers were more likely to be of black vs. white ethnicity (4.51, 95% CI 1.63–12.45), and less likely to have one ED attendance (0.39, 95% CI 0.17–0.88), or female (0.12, 95% CI 0.04–0.42). These results indicate that BBV-testing in EDs is feasible, providing a basis for further studies to explore provider and patient acceptability, referral into care and cost-effectiveness.

Slender liquid jets that have a curved trajectory have been examined in a range of papers using a method introduced in Wallwork et al. (Proc. IUTAM Symp. on Free-Surface Flows, 2000, Kluwer; J. Fluid Mech., vol. 459, 2002, pp. 43–65) and Decent et al. (J. Engng Maths, vol. 42, 2002, pp. 265–282), for jets that emerge from an orifice on the surface of a rotating cylindrical container, successfully comparing computational results to measurements arising from laboratory experiments. Wallwork et al. (2000, 2002) and Decent et al. (2002) based their analyses on the slenderness of the jet, and neglected the torsion of the centreline of the jet, which is valid since in most situations examined the torsion is zero or small. Shikhmurzaev & Sisoev (J. Fluid Mech., vol. 819, 2017, pp. 352–400) used differential geometry and incorporated the torsion. This paper shows that these two methods produce identical results at leading order when the torsion is zero or when the torsion is
$O(1)$, in an asymptotic framework based upon the slenderness of the jet, and shows that the method of Wallwork et al. (2000, 2002) and Decent et al. (2002) is accurate for parameters corresponding to scenarios previously examined and also when the torsion is
$O(1)$. It is shown that the method of Shikhmurzaev & Sisoev (2017) should be used when the torsion is asymptotically large or when the jet is not slender.

Field experiments were conducted in 1993, 1994, and 1995 to determine the effects of glufosinate rate and application timing on giant foxtail, common lambsquarters, common cocklebur, and Pennsylvania smartweed control in absence of a crop. Glufosinate at 140 g ai/ha controlled less than 80% of the weed species evaluated. When glufosinate rate was increased to 420 g/ha and applied to 10-cm giant foxtail, control was greater than 80% all 3 yr of the study. Applications made to 10-cm plants resulted in 80% or greater control for common cocklebur all 3 yr and Pennsylvania smartweed 2 of the 3 yr with 420 and 560 g/ha, respectively. Common lambsquarters was the most tolerant species evaluated and was not consistently controlled acceptably (> 80%), even with glufosinate at rates of 560 g/ha. Control with glufosinate at 420 or 560 g/ha was most effective when applied at the 10-cm weed height compared either to the 5- or 15-cm weed height.
Early life stress and the anxious brain: evidence for a neural mechanism linking childhood emotional maltreatment to anxiety in adulthood

Background
Childhood emotional maltreatment (CEM) increases the likelihood of developing an anxiety disorder in adulthood, but the neural processes underlying conferment of this risk have not been established. Here, we test the potential for neuroimaging the adult brain to inform understanding of the mechanism linking CEM to adult anxiety symptoms.
MethodOne hundred eighty-two adults (148 females, 34 males) with a normal-to-clinical range of anxiety symptoms underwent structural and functional magnetic resonance imaging while completing an emotion-processing paradigm with facial expressions of fear, anger, and happiness. Participants completed self-report measures of CEM and current anxiety symptoms. Voxelwise mediation analyses on gray-matter volumes and activation to each emotion condition were used to identify candidate brain mechanisms relating CEM to anxiety in adulthood.
ResultsDuring processing of fear and anger faces, greater amygdala and less right dorsolateral prefrontal (dlPFC) activation partially mediated the positive relationship between CEM and anxiety symptoms. Greater right posterior insula activation to fear also partially mediated this relationship, as did greater ventral anterior cingulate (ACC) and less dorsal ACC activation to anger. Responses to happy faces in these regions did not mediate the CEM-anxiety relationship. Smaller right dlPFC gray-matter volumes also partially mediated the CEM-anxiety relationship.
ConclusionsActivation patterns of the adult brain demonstrate the potential to inform mechanistic accounts of the CEM conferment of anxiety symptoms. Results support the hypothesis that exaggerated limbic activation to negative valence facial emotions links CEM to anxiety symptoms, which may be consequent to a breakdown of cortical regulatory processes.
The roles of actuaries in UK life offices: changes and challenges

We examine the roles of actuaries in UK life offices, along with trends, challenges to and opportunities for actuaries. We carry out an analysis of senior roles in life offices, a questionnaire survey and interviews with relevant senior personnel. We find that actuaries occupy many important roles in life offices and are regarded as having good industry knowledge and technical skills, especially in financial modelling. There are fewer executive directors and more non-executive directors of life offices who are actuaries compared with the position in 1990. A higher proportion of reserved roles is outsourced to consultants than was the case in 1990. Only a small number of Actuarial Function Holders are directors. Actuaries are more siloed than was the case in the past, although actuaries are well represented in the finance and risk functions of many offices. Although actuarial work in connection with the preparation for Solvency II will decline, there will be important ongoing requirements for actuaries following Solvency II implementation. We also see opportunities for actuaries in four areas: in risk management, in financial analysis and management based on Solvency II and international financial reporting standards, in connection with “big data”, and in product development and the customer proposition. There are implications for the examination syllabus, continuing professional development and research.

Background
Brain-derived neurotrophic factor (BDNF) Val66Met polymorphism contributes to the development of depression (major depressive disorder, MDD), but it is unclear whether neural effects observed in healthy individuals are sustained in MDD.
AimsTo investigate BDNF Val66Met effects on key regions in MDD neurocircuitry: amygdala, anterior cingulate, middle frontal and orbitofrontal regions.
MethodMagnetic resonance imaging scans were acquired in 79 persons with MDD (mean age 49 years) and 74 healthy volunteers (mean age 50 years). Effects on surface area and cortical thickness were examined with multiple comparison correction.
ResultsPeople who were Met allele carriers showed reduced caudal middle frontal thickness in both study groups. Significant interaction effects were found in the anterior cingulate and rostral middle frontal regions, in which participants in the MDD group who were Met carriers showed the greatest reduction in surface area.
ConclusionsModulatory effects of the BDNF Val66Met polymorphism on distinct subregions in the prefrontal cortex in MDD support the neurotrophin model of depression.

Background
White matter abnormalities have been implicated in the aetiology of major depressive disorder; however, the relationship between the severity of symptoms and white matter integrity is currently unclear.
AimsTo investigate white matter integrity in people with major depression and healthy controls, and to assess its relationship with depressive symptom severity.
MethodDiffusion tensor imaging data were acquired from 66 patients with recurrent major depression and a control group of 66 healthy individuals matched for age, gender and IQ score, and analysed with tract-based spatial statistics. The relationship between white matter integrity and severity of depression as measured by the Beck Depression Inventory was examined.
ResultsDepressive illness was associated with widespread regions of decreased white matter integrity, including regions in the corpus callosum, superior longitudinal fasciculus and anterior corona radiata, compared with the control group. Increasing symptom severity was negatively correlated with white matter integrity, predominantly in the corpus callosum.
ConclusionsWidespread alterations in white matter integrity are evident in major depressive disorder. These abnormalities are heightened with increasing severity of depressive symptoms.

Pupae of the onion maggot, Hylemya antiqua (Meigen), did not complete their development when irradiated with 3.0 kiloroentgens within five days of pupal formation. Older pupae were not adversely affected. Females from treated pupae were sterile and laid no eggs when caged with normal males. The males from treated pupae were sterilized to a high degree, yet normal females mated with them and laid normal complements of eggs. Less than 0.2% of 10,616 eggs laid by normal females caged with sterile males hatched. A 6:1 ratio of sterile to normal flies reduced egg hatch to 1.7%.

Two XRD specimen holders we designed for use with highly radioactive specimens are described. An injection mold was fabricated to allow inexpensive production of one of the holders. These holders are suitable for single-time use. The ease of use and disposable nature of this holder resulted in a dramatic reduction in personnel exposure and an uncontaminated diffraction unit. A second holder, based on the design of the first, is used to obtain XRD patterns from powders where preferred orientation is an issue and from clays. Both holders result in negligible background, since the specimen is essentially levitated in the X-ray beam. This is a benefit over other methods, such as collodion, that introduce significant background to the XRD pattern.

INTRODUCTION
Arid and semi-arid climates are mainly characterised as those areas where precipitation is less (and often considerably less) than potential evapotranspiration. These climate regions are ideal environments for salt to accumulate in natural soil and groundwater settings since evaporation and transpiration essentially remove freshwater from the system, leaving residual salts behind. Similarly, the characteristically low precipitation rates reduce the potential for salt to be diluted by rainfall. Thus arid and semi-arid regions make ideal ‘salt concentrator’ hydrologic environments. Indeed, salt flats, playas, sabkhas and saline lakes, for example, are ubiquitous features of arid and semi-arid regions throughout the world (Yechieli and Wood,2002). In such settings, variable density flow phenomena are expected to be important, especially where hypersaline brines overlie less dense groundwater at depth. In contrast, seawater intrusion in coastal aquifers is a global phenomenon that is not constrained to only arid and semi-arid regions of the globe and is inherently a variable density flow problem by its very nature. These two examples make it clear that variable density flow problems occur in, but importantly extend beyond, arid and semi-arid regions of the globe. The intention of this chapter is therefore not to limit ourselves to modelling arid zone hydrological systems, but rather to present a more general treatment of variable density groundwater flow and solute transport phenomena and modelling. The concepts presented in this chapter are therefore not climatologically constrained to arid or semi-arid zones of the world, although they do apply equally there.
