To investigate the manipulation of electromagnetic properties of two-dimensional materials, this effort characterizes charge transfer behavior of colloidal COF-5 (covalent organic framework) in the presence of various metal ions. A series of metal chloride compounds was introduced to COF-5 in solution and solid film phases and the interaction of the material with electromagnetic radiation was monitored across the visible region using electronic absorption spectroscopy. Notable changes were observed, quantified, and discussed for copper (II) chloride (CuCl2), chromium (III) chloride (CrCl3), and iron (III) chloride (FeCl3) with COF-5. Ligand-to-metal and metal-to-ligand charge transfer are explored as a possible mechanism for the observed electronic behaviors.

OBJECTIVES/SPECIFIC AIMS: The objective of this study was to determine if trimethylamine N-oxide (TMAO) alone could acutely alter cardiac contractile function on a beat-to-beat basis. METHODS/STUDY POPULATION: CD1 adult mouse hearts were extracted, attached to a force transducer, oxygenated, and paced within an organ bath. Changes in contractility were measured after pipetting or reverse perfusing TMAO through the aorta via a modified Langendorff apparatus to facilitate TMAO delivery into the myocardium. To determine if our findings translated to the human heart, we performed contractility experiments using human right atrial appendage biopsy tissue retrieved during cardiopulmonary bypass procedures. To investigate whether TMAO alters contractile rate, in a separate series of experiments, the atria and sinoatrial node of isolated hearts were kept intact to allow for spontaneous beating without artificial pacing and were treated with TMAO or vehicle. In addition, intracellular calcium measurements were performed on spontaneously beating embryonic rat cardiomyocytes after TMAO or vehicle treatment. RESULTS/ANTICIPATED RESULTS: We found acute exposure to TMAO, diluted into the organ bath, increased average contraction amplitude 20% and 41% at 300 µM and 3000 µM, respectively (p<0.05, n=6). Langendorff reverse perfusion of mouse hearts ex vivo with 300 µM TMAO generated an even greater response than nonperfusion peripheral exposure and increased isometric force 32% (p<0.05, n=3). Consistent with what we observed in mouse hearts, incubation of human atrial muscle tissue with TMAO at 3000 µM increased isometric tension 31% compared with vehicle (p<0.05, n=4). TMAO treatment (3000 µM) also increased average beating frequency of ex vivo mouse hearts by 27% compared with vehicle (p<0.05, n=3) and increased the spontaneous beating frequency of primary rat cardiomyocytes by 13% compared with vehicle treatment (p<0.05, n=4). DISCUSSION/SIGNIFICANCE OF IMPACT: TMAO, at pathological concentrations, directly increases the force and rate of cardiac contractility. Initially, these inotropic and chronotropic effects may be adaptive during CKD; however, chronic increases in isometric tension and beating frequency can promote cardiac remodeling and heart failure. Further translational studies are needed to understand the intricate relationship between the microbiome, kidneys, and heart and to examine if TMAO represents a therapeutic target for reducing cardiovascular mortality in CKD patients.

Space weathering affects reflectance spectra of the Moon and S-complex asteroids by spectral bluing (increasing reflectance with decreasing wavelength) of their surface materials at UV/blue (less than 400 nm) wavelengths. This spectral bluing is attributed to a degradation of the UV absorption feature seen in spectral reflectance of olivine as a result of the creation of nanophase (npFe0) iron. We have modeled the effect of the addition of small amounts of npFe0 intimately mixed with particles from a hypothetical material and a terrestrial basalt. The addition of 0.0001% npFe0 affects the reflectance at these UV/blue wavelengths, while the addition of 0.01% is required to see the visible/near infrared reddening and diminution of VNIR absorption features. Thus, the UV/blue spectral reflectance characteristics allow earlier detection of the onset of space weathering effects.

The Monte Carlo software CASINO has been expanded with new modules for the simulation of complex beam scanning patterns, for the simulation of cathodoluminescence (CL), and for the calculation of electron energy deposition in subregions of a three-dimensional (3D) volume. Two examples are presented of the application of these new capabilities of CASINO. First, the CL emission near threading dislocations in gallium nitride (GaN) was modeled. The CL emission simulation of threading dislocations in GaN demonstrated that a better signal-to-noise ratio was obtained with lower incident electron energy than with higher energy. Second, the capability to simulate the distribution of the deposited energy in 3D was used to determine exposure parameters for polymethylmethacrylate resist using electron-beam lithography (EBL). The energy deposition dose in the resist was compared for two different multibeam EBL schemes by changing the incident electron energy.

The reflectance of Saturn's moon Enceladus has been measured at far ultraviolet (FUV) wavelengths (115–190 nm) by Cassini's UltraViolet Imaging Spectrograph (UVIS). At visible and near infrared (VNIR) wavelengths Enceladus' reflectance spectrum is very bright, consistent with a surface composed primarily of H2O ice. At FUV wavelengths, however, Enceladus is surprisingly dark – darker than would be expected for pure water ice. We find that the low FUV reflectance of Enceladus can be explained by the presence of a small amount of NH3 and a small amount of a tholin in addition to H2O ice on the surface.

An evaluation programme is presented for determining activity of unknown compounds on Ascaris suum larvae migrating into pulmonary tissues of animals. Data on gross lung lesions due to migrating ascarid larvae were obtained in a series of 44 screening tests consisting of 223 group observations from 1115 mice orally infected with A. suum. Lesion scores were graded in ascending order from 0 to 5 depending upon number and extent of pulmonary haemorrhages. In general, there was no significant difference in lesion scores between untreated mice and mice treated with unknown compounds in their ration. The programme is based on an initial 95% rejection of compounds with a 5% ability to retest compounds in groups of mice.
The grand mean score for 44 tests was 2·88. Ninety per cent confidence limits for means among tests established scores at 2·21 and 3·55, and among groups of mice at 1·76 and 4·00, When the test mean was less than 2·2, the ascarid ova used for infection were considered non-virulent. Compounds selected as active were in groups of mice having a mean lesion score of 1·76 or less. Banminth, Styquin and Tetramisole showed larvicidal activity. A reasonable compromise exists in this system wherein false leads are tolerated to avoid screening out effective materials. The host-parasite relationship is proposed as a valuable tool for anthelmintic evaluation.

INTRODUCTION
It appears that one of the first things that occurred to Felix d'Herelle when he discovered bacteriophages in 1917 was that these mysterious objects might provide a means of killing bacteria that are pathogenic to humans (Summers, 1999). The still ongoing story of phage therapy, as this approach was called, has been told elsewhere and will not be retold here, but it serves to point out that scientists have been interested in the effects of phages on their hosts since their discovery. d'Herelle believed, and eventually established, that phages are viruses that infect bacteria. However, it was not until the experimental investigations of phages at the dawn of molecular biology in the 1940s and 1950s that it became clear that phages - and for that matter their bacterial hosts - are genetic organisms (Luria and Delbrück, 1943; Hershey and Rotman, 1949; Hershey and Chase, 1952; Stent, 1963), just like fruit flies, corn, and humans, and so could be expected to mutate and evolve.
Although some work was done on the evolution of phages in the 1960s, 1970s, and 1980s, a more detailed understanding of the genetic mechanisms of phage evolution had to wait until the advent of high-throughput DNA sequencing in the 1990s. This is because the genetic history of a phage, while it is to a significant extent encoded in the phage's genome sequence, is largely invisible to our analysis until we can compare that sequence to the genome sequences of other phages.

RNAs are modular biomolecules, composed largely of conserved structural subunits, or motifs. These structural motifs comprise the secondary structure of RNA and are knit together via tertiary interactions into a compact, functional, three-dimensional structure and are to be distinguished from motifs defined by sequence or function. A relatively small number of structural motifs are found repeatedly in RNA hairpin and internal loops, and are observed to be composed of a limited number of common ‘structural elements’. In addition to secondary and tertiary structure motifs, there are functional motifs specific for certain biological roles and binding motifs that serve to complex metals or other ligands. Research is continuing into the identification and classification of RNA structural motifs and is being initiated to predict motifs from sequence, to trace their phylogenetic relationships and to use them as building blocks in RNA engineering.
Coping With Dementia: Evaluation of Four Nonpharmacologic Interventions

To evaluate nonpharmacologic interventions, caregivers (65 women, 38 men) and their dementia-diagnosed spouses (patients) were randomized to one of four treatment programs (cognitive stimulation, dyadic counseling, dual supportive seminar, and early-stage day care) or to a wait-list control group. Assessments occurred initially and at postintervention (3 months). Patients were evaluated on memory, verbal fluency, and problem-solving ability, and caregivers were assessed on marital interaction, emotional status, and physical health, along with stress, coping, and social support. Caregivers also completed a program evaluation. Repeated measures procedures showed that patients in the cognitive stimulation group demonstrated more improvement over time in cognitive outcomes, and caregivers decreased in depressive symptoms. Early-stage day-care and dual supportive seminar group caregivers reported a decrease in hostility and a decrease in use of negative coping strategies, respectively. Although qualitatively derived benefits differed across groups, similarities in program content reduced the potential for quantitative differentiation among the groups.
