Target-site and non–target site mechanisms of pronamide resistance in annual bluegrass (Poa annua) populations from Mississippi golf courses

The mitotic-inhibiting herbicide pronamide controls susceptible annual bluegrass (Poa annua L.) pre- and postemergence, but in some resistant populations, postemergence activity is compromised, hypothetically due to a target-site mutation, lack of root uptake, or an unknown resistance mechanism. Three suspected pronamide-resistant (LH-R, SC-R, and SL-R) and two pronamide-susceptible (BS-S and HH-S) populations were collected from Mississippi golf courses. Dose–response experiments were conducted to confirm and quantify pronamide resistance, as well as resistance to flazasulfuron and simazine. Target sites known to confer resistance to mitotic-inhibiting herbicides were sequenced, as were target sites for herbicides inhibiting acetolactate synthase (ALS) and photosystem II (PSII). Pronamide absorption and translocation were investigated following foliar and soil applications. Dose–response experiments confirmed pronamide resistance of LH-R, SC-R, and SL-R populations, as well as instances of multiple resistance to ALS- and PSII-inhibiting herbicides. Sequencing of the α-tubulin gene confirmed the presence of a mutation that substituted isoleucine for threonine at position 239 (Thr-239-Ile) in LH-R, SC-R, SL-R, and BS-S populations. Foliar application experiments failed to identify differences in pronamide absorption and translocation between the five populations, regardless of harvest time. All populations had limited basipetal translocation—only 3% to 13% of the absorbed pronamide—across harvest times. Soil application experiments revealed that pronamide translocation was similar between SC-R, SL-R, and both susceptible populations across harvest times. The LH-R population translocated less soil-applied pronamide than susceptible populations at 24, 72, and 168 h after treatment, suggesting that reduced acropetal translocation may contribute to pronamide resistance. This study reports three new pronamide-resistant populations, two of which are resistant to two modes of action (MOAs), and one of which is resistant to three MOAs. Results suggest that both target site– and translocation-based mechanisms may be associated with pronamide resistance. Further research is needed to confirm the link between pronamide resistance and the Thr-239-Ile mutation of the α-tubulin gene.

We describe the incidence of suicidality (2007–2017) in people with depression treated by secondary mental healthcare services at South London and Maudsley NHS Trust (n = 26 412). We estimated yearly incidence of ‘suicidal ideation’ and ‘high risk of suicide’ from structured and free-text fields of the Clinical Record Interactive Search system. The incidence of suicidal ideation increased from 0.6 (2007) to 1 cases (2017) per 1000 population. The incidence of high risk of suicide, based on risk forms, varied between 0.06 and 0.50 cases per 1000 adult population (2008–2017). Electronic health records provide the opportunity to examine suicidality on a large scale, but the impact of service-related changes in the use of structured risk assessment should be considered.

Public involvement in service change has been identified as a key facilitator of health care transformation (Foley et al., 2017) but little is known about how health policy influences whether and how organisations involve the public in change processes. This qualitative study compares policy and practice for involving the public in major service changes across the UK's four health systems (England, Northern Ireland, Wales and Scotland). We analysed policy documents, and conducted interviews with officials, stakeholders, NHS staff and public campaigners (total number of interviewees = 47). Involving the public in major service change was acknowledged as a policy challenge in all four systems. Despite ostensible similarities, there were some clear differences between the four health systems' processes for involving patients and the public in major changes to health services. The extent of central Government oversight, the prescriptiveness of Government guidance, the role for intermediary bodies and arrangements for independent scrutiny of contentious decisions all vary. We analyse how health policy in the four systems has used ‘sticks’ and ‘sermons’ to promote particular approaches, and conclude that both policy and the wider system context within which health care organisations try to effect change are significant, and understudied aspect of contemporary practice.
45 Long-term Treatment with Deutetrabenazine Is Associated with Continued Improvement in Tardive Dyskinesia: Results from an Open-label Extension Study

Study Objective
To evaluate long-term efficacy of deutetrabenazine in patients with tardive dyskinesia (TD) by examining response rates from baseline in Abnormal Involuntary Movement Scale (AIMS) scores. Preliminary results of the responder analysis are reported in this analysis.
BackgroundIn the 12-week ARM-TD and AIM-TD studies, the odds of response to deutetrabenazine treatment were higher than the odds of response to placebo at all response levels, and there were low rates of overall adverse events and discontinuations associated with deutetrabenazine.
MethodPatients with TD who completed ARM-TD or AIM-TD were included in this open-label, single-arm extension study, in which all patients restarted/started deutetrabenazine 12mg/day, titrating up to a maximum total daily dose of 48mg/day based on dyskinesia control and tolerability. The study comprised a 6-week titration and a long-term maintenance phase. The cumulative proportion of AIMS responders from baseline was assessed. Response was defined as a percent improvement from baseline for each patient from 10% to 90% in 10% increments. AlMS score was assessed by local site ratings for this analysis.
Results343 patients enrolled in the extension study (111 patients received placebo in the parent study and 232 patients received deutetrabenazine). At Week 54 (n=145; total daily dose [mean±standard error]: 38.1±0.9mg), 63% of patients receiving deutetrabenazine achieved ≥30% response, 48% of patients achieved ≥50% response, and 26% achieved ≥70% response. At Week 80 (n=66; total daily dose: 38.6±1.1mg), 76% of patients achieved ≥30% response, 59% of patients achieved ≥50% response, and 36% achieved ≥70% response. Treatment was generally well tolerated.
ConclusionsPatients who received long-term treatment with deutetrabenazine achieved response rates higher than those observed in positive short-term studies, indicating clinically meaningful long-term treatment benefit.

Study Objective
To evaluate the long-term safety and tolerability of deutetrabenazine in patients with tardive dyskinesia (TD) at 2years.
BackgroundIn the 12-week ARM-TD and AIM-TD studies, deutetrabenazine showed clinically significant improvements in Abnormal Involuntary Movement Scale scores compared with placebo, and there were low rates of overall adverse events (AEs) and discontinuations associated with deutetrabenazine.
MethodPatients who completed ARM-TD or AIM-TD were included in this open-label, single-arm extension study, in which all patients restarted/started deutetrabenazine 12mg/day, titrating up to a maximum total daily dose of 48mg/day based on dyskinesia control and tolerability. The study comprised a 6-week titration period and a long-term maintenance phase. Safety measures included incidence of AEs, serious AEs (SAEs), and AEs leading to withdrawal, dose reduction, or dose suspension. Exposure-adjusted incidence rates (EAIRs; incidence/patient-years) were used to compare AE frequencies for long-term treatment with those for short-term treatment (ARM-TD and AIM-TD). This analysis reports results up to 2 years (Week106).
Results343 patients were enrolled (111 patients received placebo in the parent study and 232 received deutetrabenazine). There were 331.4 patient-years of exposure in this analysis. Through Week 106, EAIRs of AEs were comparable to or lower than those observed with short-term deutetrabenazine and placebo, including AEs of interest (akathisia/restlessness [long-term EAIR: 0.02; short-term EAIR range: 0–0.25], anxiety [0.09; 0.13–0.21], depression [0.09; 0.04–0.13], diarrhea [0.06; 0.06–0.34], parkinsonism [0.01; 0–0.08], somnolence/sedation [0.09; 0.06–0.81], and suicidality [0.02; 0–0.13]). The frequency of SAEs (EAIR 0.15) was similar to those observed with short-term placebo (0.33) and deutetrabenazine (range 0.06–0.33) treatment. AEs leading to withdrawal (0.08), dose reduction (0.17), and dose suspension (0.06) were uncommon.
ConclusionsThese results confirm the safety outcomes seen in the ARM-TD and AIM-TD parent studies, demonstrating that deutetrabenazine is well tolerated for long-term use in TD patients.

Taylor blast wave (TBW) theory and geometrical shock dynamics (GSD) theory describe a radially expanding shock wave front through an inert material, typically an ideal gas, in the strong blast wave limit and weak acoustic limit respectively. We simulate a radially expanding blast shock in air using a hydrodynamic simulation code and numerically describe the intermediate region between these two limits. We test our description of the intermediate shock phase through a two-dimensional simulation of the Bryson and Gross experiment. We then apply the principles of GSD to materials that follow the Mie–Gruneisen equation of state, such as plastics and metals, and derive an equation that accurately relates the acceleration, velocity and curvature of the shock through these materials. Along with detonation shock dynamics (DSD), which describes detonation shock propagation through high explosive fluids, we develop a hybrid DSD/GSD model for the simulation of heterogeneous explosives. This model enables computationally efficient simulation of the shock front in high explosive/inert mixtures consisting of simple or complex geometric configurations. We simulate an infinite two-dimensional slab consisting of one half explosive, PBXN-9, and one half aluminium and model the boundary angle conditions using shock polar analysis. We also simulate a series of high explosive unit cells embedded with aluminium spherical particles, and we compare the propagation of the detonation shock front with a direct numerical simulation performed with the ALE3D code.
Corn Response to POST-Applied HPPD-Inhibitor Based Premix Herbicides with In-Furrow and Foliar-Applied Insecticides

In recent years injury has been reported in numerous Tennessee corn fields treated with an organophosphate (OP) insecticide and either a mesotrione- or tembotrione-based herbicide premix. Research was conducted with the objective to determine if corn treated with an in-furrow application of chlorpyrifos or a foliar application of chlorpyrifos or bifenthrin, or a combination of in-furrow and foliar treatment, would be more predisposed to injury when either a premix of S-metolachlor, mesotrione, and glyphosate (meso premix) or a premix of tembotrione plus thiencarbazone (tembo premix) was applied. The main effects of insecticide or herbicide, the two-way interaction of insecticide by herbicide, and the three-way interaction of herbicide by insecticide by application type of insecticide were all significant for injury as well as yield. When chlorpyrifos was used both in-furrow and foliarly on corn treated with the tembo premix, injury was increased to 56% and yield was reduced 58% compared with corn that had not been treated with that insecticide. This use pattern of chlorpyrifos utilized in-furrow followed by chlorpyrifos applied foliarly with the tembo premix essentially doubled the injury (29 to 56%) and increased yield loss from 41% where chlorpyrifos was only utilized foliarly to 49% when chlorpyrifos was applied in-furrow and foliarly. Corn injury was negligible (< 6%) and yield was similar where the meso premix was examined in combination of both types of insecticide applications. This study clearly demonstrated the phytotoxic interaction between these two herbicide premixes and the OP insecticide chlorpyrifos. Therefore, growers need to be mindful of which herbicides are utilized when OP insecticides are used for insect management.

We present an asymptotic theory for the dynamics of detonation when the radius of curvature of the detonation shock is large compared to the one-dimensional, steady, Chapman-Jouguet (CJ) detonation reaction-zone thickness. The analysis considers additional time-dependence in the slowly varying reaction zone to that considered ill previous works. The detonation is assumed to have a sonic point in the reaction- zone structure behind the shock, and is referred to as an eigenvalue detonation. A new, iterative method is used to calculate the eigenvalue relation, which ultimately is expressed as an intrinsic, partial differential equation (PDE) for the motion of the shock surface. Two cases are considered for an ideal equation of state. The first corresponds to a model of a condensed-phase explosive, with modest reaction rate sensitivity, and the intrinsic shock surface PDE is a relation between the normal detonation shock velocity, Dn, the first normal time derivative of the normal shock velocity, Dn, and the shock curvature, K. The second case corresponds to a gaseous explosive mixture, with the large reaction rate sensitivity of Arrhenius kinetics, and the intrinsic shock surface PDE is a relation between the normal detonation shock velocity, Dn, its first and second normal time derivatives of the normal shock velocity, Dn, Dn, and the shock curvature, K, and its first normal time derivative of the curvature, k. For the second case, one obtains a one-dimensional theory of pulsations of plane CJ detonation and a theory that predicts the evolution of self-sustained cellular detonation. Versions of the theory include the limits of near-CJ detonation, and when the normal detonation velocity is significantly below its CJ value. The curvature of the detonation can also be of either sign, corresponding to both diverging and converging geometries.

A theory of time-dependent two-dimensional detonation is developed for an explosive with a finite-thickness reaction zone. A representative initial–boundary-value problem is treated that illustrates how the planar shock of an initially one-dimensional detonation becomes non-planar in response to the action of an edge rarefaction that is generated at the explosive's lateral surface. The solution of this time-dependent problem has a wave-hierarchy structure that at late times includes a weakly two-dimensional hyperbolic region and a fully two-dimensional parabolic region. The wave head of the rarefaction is carried by the hyperbolic region. We show that the shock locus is analytic at the wave head. The dynamics of the final approach to two-dimensional steady-state detonation is controlled by Burgers’ equation for the shock locus. We also present some results concerning the stability of the solutions to our problem.

Experiments in the HMX-based condensed explosive PBX-9501 were carried out to validate a reduced, asymptotically derived description of detonation shock dynamics (DSD) where it is assumed that the normal detonation shock speed is determined by the total shock curvature. The passover experiment has a lead disk embedded in a right circular cylindrical charge of PBX-9501 and is initiated from the bottom. The subsequent detonation shock experiences a range of dynamic states with both diverging (convex) and converging (concave) configurations as the detonation shock passes over the disk. The time of arrival of the detonation shock at the top surface of the charge is recorded and compared against DSD simulation and direct multi-material simulation. A new wide-ranging equation of state (EOS) and rate law that is constrained by basic explosive characterization experiments is introduced as a constitutive description of the explosive. This EOS and rate law is used to compute the theoretical normal shock velocity, curvature relation of the explosive for the reduced description, and is also used in the multi-material simulation. The time of arrival records are compared against the passover experiment and the dynamic motion of the shock front and states within the explosive are analysed. The experiment and simulation data are in excellent agreement. The level of agreement, both qualitative and quantitative, of theory and simulation with experiment is encouraging because it indicates that descriptions such as the wide-ranging EOS/rate law and the corresponding reduced DSD description can be used effectively to model real explosives and predict complex dynamic behaviors.

Based on a general theory of detonation waves with an embedded sonic locus that we have previously developed, we carry out asymptotic analysis of weakly curved slowly varying detonation waves and show that the theory predicts the phenomenon of detonation ignition and failure. The analysis is not restricted to near Chapman–Jouguet detonation speeds and is capable of predicting quasi-steady, normal detonation shock speed versus curvature ($D$–$\kappa$) curves with multiple turning points. An evolution equation that retains the shock acceleration, $\skew2\dot{D}$, namely a $\skew2\dot{D}$–$D$–$\kappa$ relation is rationally derived which describes the dynamics of pre-existing detonation waves. The solutions of the equation for spherical detonation are shown to reproduce the ignition/failure phenomenon observed in both numerical simulations of blast wave initiation and in experiments. A single-step chemical reaction described by one progress variable is employed, but the kinetics is sufficiently general and is not restricted to Arrhenius form, although most specific calculations are performed for Arrhenius kinetics. As an example, we calculate critical energies of direct initiation for hydrogen–oxygen mixtures and find close agreement with available experimental data.

We investigate hydrodynamic instability of a steady planar detonation wave propagating in a circular tube to three-dimensional linear perturbations, using the normal mode approach. Spinning instability is identified and its relevance to the well-known spin detonation is discussed. The neutral stability curves in the plane of heat release and activation energy exhibit bifurcations from low-frequency to high-frequency spinning modes as the heat release is increased at fixed activation energy. With a simple Arrhenius model for the heat release rate, a remarkable qualitative agreement with experiment is obtained with respect to the effects of dilution, initial pressure and tube diameter on the behaviour of spin detonation. The analysis contributes to the explanation of spin detonation which has essentially been absent since the discovery of the phenomenon over seventy years ago.

The stability of an overdriven planar detonation wave is examined for a one-step Arrhenius reaction model with an order-one post-shock temperature-scaled activation energy θ in the limit of a small post-shock temperature-scaled heat release β. The ratio of specific heats, γ is taken such that (γ−1)=O(1). Under these assumptions, which cover a wide range of realistic physical situations, the steady detonation structure can be evaluated explicitly, with the reactant mass fraction described by an exponentially decaying function. The analytical representation of the steady structure allows a normal-mode description of the stability behaviour to be obtained via a two-term asymptotic expansion in β. The resulting dispersion relation predicts that for a finite overdrive f, the detonation is always stable to two-dimensional disturbances. For large overdrives, the identification of regimes of stability or instability is found to depend on a choice of distinguished limit between the heat release β and the detonation propagation Mach number D*. Regimes of instability are found to be characterized by the presence of a single unstable oscillatory mode over a finite range of wavenumbers.

A detailed investigation of the hydrodynamic stability to transverse linear disturbances of a steady, one-dimensional detonation in an ideal gas undergoing an irreversible, unimolecular reaction with an Arrhenius rate constant is conducted via a normal-mode analysis. The method of solution is an iterative shooting technique which integrates between the detonation shock and the reaction equilibrium point. Variations in the disturbance growth rates and frequencies with transverse wavenumber, together with two-dimensional neutral stability curves and boundaries for all unstable low- and higher frequency modes, are obtained for varying detonation bifurcation parameters. These include the detonation overdrive, chemical heat release and reaction activation energy. Spatial perturbation eigenfunction behaviour and phase and group velocities are also obtained for selected sets of unstable modes. Results are presented for both Chapman–Jouguet and overdriven detonation velocities. Comparisons between the earlier pointwise determination of stability and interpolated neutral stability boundaries obtained by Erpenbeck are made. Possible physical mechanisms which govern the wavenumber selection underlying the initial onset of either regular or irregular cell patterns are also discussed.
