Effects of physical form of β-lactoglobulin and calcium ingestion on GLP-1 secretion, gastric emptying and energy intake in humans: a randomised crossover trial

The aim of this study was to assess whether adding Ca2+ to aggregate or native forms of β-lactoglobulin alters gut hormone secretion, gastric emptying rates and energy intake in healthy men and women. Fifteen healthy adults (mean ± sd: 9M/6F, age: 24 ± 5 years) completed four trials in a randomised, double-blind, crossover design. Participants consumed test drinks consisting of 30 g of β-lactoglobulin in a native form with (NATIVE + MINERALS) and without (NATIVE) a Ca2+-rich mineral supplement and in an aggregated form both with (AGGREG + MINERALS) and without the mineral supplement (AGGREG). Arterialised blood was sampled for 120 min postprandially to determine gut hormone concentrations. Gastric emptying was determined using 13C-acetate and 13C-octanoate, and energy intake was assessed with an ad libitum meal at 120 min. A protein × mineral interaction effect was observed for total glucagon-like peptide-1 (GLP-1TOTAL) incremental AUC (iAUC; P < 0·01), whereby MINERALS + AGGREG increased GLP-1TOTAL iAUC to a greater extent than AGGREG (1882 ± 603 v. 1550 ± 456 pmol·l−1·120 min, P < 0·01), but MINERALS + NATIVE did not meaningfully alter the GLP-1 iAUC compared with NATIVE (1669 ± 547 v. 1844 ± 550 pmol·l−1·120 min, P = 0·09). A protein × minerals interaction effect was also observed for gastric emptying half-life (P < 0·01) whereby MINERALS + NATIVE increased gastric emptying half-life compared with NATIVE (83 ± 14 v. 71 ± 8 min, P < 0·01), whereas no meaningful differences were observed between MINERALS + AGGREG v. AGGREG (P = 0·70). These did not result in any meaningful changes in energy intake (protein × minerals interaction, P = 0·06). These data suggest that the potential for Ca2+ to stimulate GLP-1 secretion at moderate protein doses may depend on protein form. This study was registered at clinicaltrials.gov (NCT04659902).
Clinical pathology and cardiovascular parameters are not influenced by housing rats under increased environmental complexity

Since the release of the revised Appendix A from the Council of Europe for housing of laboratory animals there have been claims that laboratory animals should be housed under more complex conditions; known popularly as enrichment. A number of studies have expressed concerns that this may increase uncontrollable variation in the animals, thereby creating the need for greater numbers of animals. Within neurobiology there would appear to be a scientific basis for such concern. However, even though this may be used as an argument for denying the animal environmental enrichment, it is unclear whether there is any basis for concern within other research areas. The aim of this study, therefore, was to explore whether clinical pathology and cardiovascular parameters were influenced by housing rats under environmentally enriched conditions. Male, Sprague-Dawley rats were housed under three different regimes: non-enriched, standard-enriched (according to the guidelines of the Council of Europe) and extra-enriched with a shelf and higher cages. All housing forms were based upon commercially available, standardised equipment. A total of 41 different parameters were monitored via clinical pathology, telemetry and coagulation tests and virtually no differences were observed in relation to the manner in which the rats were housed. The uncontrollable variation observed in our study was compared to within-strain variation data supplied from the breeder and was relatively low in all three types of housing. We conclude, based upon our studies in male, Sprague-Dawley rats, that so far there is no basis for concern that enriched housing will lead to increased group sizes when using animals for research within this field and, as such, there is no reason not to enrich the environment of such rats.
Plasma glucagon-like peptide-1 responses to ingestion of protein with increasing doses of milk minerals rich in calcium

A high dose of whey protein hydrolysate fed with milk minerals rich in calcium (Capolac®) results in enhanced glucagon-like peptide-1 (GLP-1) concentrations in lean individuals; however, the effect of different calcium doses ingested alongside protein is unknown. The present study assessed the dose response of calcium fed alongside 25 g whey protein hydrolysate on GLP-1 concentrations in individuals with overweight/obesity. Eighteen adults (mean ± sd: 8M/10F, 34 ± 18 years, 28·2 ± 2·9 kgm−2) completed four trials in a randomised, double-blind, crossover design. Participants consumed test solutions consisting of 25 g whey protein hydrolysate (CON), supplemented with 3179 mg (LOW), 6363 mg (MED) or 9547 mg (HIGH) Capolac® on different occasions, separated by at least 48 h. The calcium content of test solutions equated to 65, 892, 1719 and 2547 mg, respectively. Arterialised-venous blood was sampled over 180 min to determine plasma concentrations of GLP-1TOTAL, GLP-17–36amide, insulin, glucose, NEFA, and serum concentrations of calcium and albumin. Ad libitum energy intake was measured at 180 min. Time–averaged incremental AUC (iAUC) for GLP-1TOTAL (pmol·l−1·min−1) did not differ between CON (23 ± 4), LOW (25 ± 6), MED (24 ± 5) and HIGH (24 ± 6). Energy intake (kcal) did not differ between CON (940 ± 387), LOW (884 ± 345), MED (920 ± 334) and HIGH (973 ± 390). Co-ingestion of whey protein hydrolysate with Capolac® does not potentiate GLP-1 release in comparison with whey protein hydrolysate alone. The study was registered at clinical trials (NCT03819972).

In 2019, a 42-year-old African man who works as an Ebola virus disease (EVD) researcher traveled from the Democratic Republic of Congo (DRC), near an ongoing EVD epidemic, to Philadelphia and presented to the Hospital of the University of Pennsylvania Emergency Department with altered mental status, vomiting, diarrhea, and fever. He was classified as a “wet” person under investigation for EVD, and his arrival activated our hospital emergency management command center and bioresponse teams. He was found to be in septic shock with multisystem organ dysfunction, including circulatory dysfunction, encephalopathy, metabolic lactic acidosis, acute kidney injury, acute liver injury, and diffuse intravascular coagulation. Critical care was delivered within high-risk pathogen isolation in the ED and in our Special Treatment Unit until a diagnosis of severe cerebral malaria was confirmed and EVD was definitively excluded.
This report discusses our experience activating a longitudinal preparedness program designed for rare, resource-intensive events at hospitals physically remote from any active epidemic but serving a high-volume international air travel port-of-entry.

Toca 511 (vocimagene amiretrorepvec) is an investigational retroviral replicating vector that selectively infects dividing cancer cells, integrates into the genome and replicates due to immune defects in tumors. Toca 511 spreads through tumors and stably delivers the gene encoding an optimized yeast cytosine deaminase that converts the prodrug Toca FC (investigational, extended-release of 5-fluorocytosine) into 5-fluorouracil. In preclinical models, 5-fluorouracil kills infected dividing cancer cells, myeloid derived suppressor cells and tumor associated macrophages, enabling immune activation against the tumor. In this dose ascending Ph1 trial (NCT01470794), Toca 511 was injected into the resection cavity wall of patients with rHGG, followed by courses of oral Toca FC. Additional cohorts included combination with bevacizumab or lomustine. Across the Ph1 program, the safety profile remains favorable. Objective responses (ORs) were assessed by IRR using MRI scans prior to Toca FC treatment as baseline. ORs occurred 6-19 months after Toca 511 administration, suggesting an immunologic mechanism. The ORs were observed in 4 patients with IDH1-wildtype and 2 patients with IDH1-mutant tumors, including 5 complete responses (CRs) with the investigational therapy alone, and 1 CR in combination with bevacizumab. The median duration of response (mDoR) was 35.1+ months. As of AUG2017, all responders were CR and remain alive. In a 23-patient subgroup who received high doses of Toca 511 and met Ph3 trial criteria, mOS was 14.4 months, 3-year survival rate was 26.1%, and mDoR was 35.7+ months with a durable response rate of 21.7%. Data suggest a positive association of durable response with OS.

Objective
Optimal nutrition for hospital patients is crucial and routine monitoring of patients’ nutrient intake is imperative. However, personalised monitoring and customised intervention using traditional methods is challenging and labour-intensive, consequently it is often neglected in hospital settings. The present pilot study aimed to examine the reliability and validity of the Dietary Intake Monitoring System (DIMS) against the weighed food method (WFM).
DesignThe DIMS 2.0 is composed of an integrated digital camera, weighing scale, radio-frequency identification sensor and WIFI connection for real-time image and weight dietary data acquisition and analysis. The DIMS equipment was used to collect data for a paired set of meals both before and after meal consumption at lunchtime.
SettingOdense University Hospital, Denmark.
SubjectsPhotos and weights of seventeen patient meals were captured.
ResultsThe results showed a significant correlation between DIMS and WFM for energy (r=0·99, P<0·01) and protein intake (r=0·98, P<0·01). Intraclass correlation coefficients (ICC) revealed a high degree of agreement among the four non-trained assessors for estimates of portion size of each food item before (0·88, P<0·01) and after consumption (0·99, P<0·01). The ICC for energy and protein intake were 0·99 (P<0·01) and 0·99 (P<0·01), respectively. Bland–Altman plots revealed no systematic bias.
ConclusionsConsidering the huge benefits associated with routine monitoring, technological advances have made it possible to develop a novel, easy-to-use DIMS that, according to the findings, is a valid alternative for use in hospital settings.
Pre- and post-operative dizziness, tinnitus, and taste disturbances among cochlear implant recipients

Objective:
To determine the pre- and post-operative prevalence of dizziness, tinnitus and taste disturbances in adult cochlear implant recipients.
Methods:A questionnaire regarding pre- and post-operative dizziness, tinnitus and taste disturbances was sent to 170 cochlear implant recipients implanted between January 2003 and March 2009. Seventy-seven patients (41 per cent) responded.
Results:Pre-operatively, 20 per cent of the participants experienced dizziness, 52 per cent experienced tinnitus and 3 per cent experienced taste disturbances. Post-operative dizziness developed in 46 per cent of patients and resolved in the majority of these; however, 15 per cent reported dizziness more than six months after implantation. Tinnitus worsened in 25 per cent of patients, whereas 73 per cent reported attenuation or termination of tinnitus. Post-operatively, tinnitus developed in 12 per cent and taste disturbances developed in 17 per cent of the patients.
Conclusion:The high prevalence of dizziness, tinnitus and taste disturbances reported by cochlear implant recipients necessitates that assessment of symptoms related to inner ear and chorda tympani damage are included when evaluating operative results.

Recurrent glioblastoma (GBM) has an unmet need for effective therapies. Toca 511 (vocimagene amiretrorepvec), a retroviral replicating vector with the transgene cytosine deaminase, selectively infects, persists and spreads in tumor. Subsequent oral administration of 5-fluorocytosine (Toca FC) produces 5-fluorouracil (5-FU) within infected cells. 5-FU kills cancer cells and myeloid derived suppressor cells, inducing robust antitumor immune responses in animal models. In 2 Phase 1 studies, Toca 511 was administered into the cavity wall after surgical resection (NCT01470794) or intratumoral injection by biopsy needle (NCT01156584). To provide context to the results observed, subjects were compared to an external lomustine treated control (Courtesy Denovo Biopharma; Wick 2010). Treatment with Toca 511/Toca FC from 2 Phase I studies showed significant improvement in OS HR equals to 0.48, p less than 0.001, with similar effect in the surgical resection (OS HR 0.45, p equals to 0.003) and intratumoral injection (OS HR 0.56, p equals to 0.060). Fewer related greater or equal to Grade 3 adverse events (AEs) were reported for Toca 511/Toca FC (2.5 percent) vs. lomustine (36.9 percent). There was a virtual absence of hematologic toxicity for Toca 511/Toca FC vs. lomustine (Grade greater or equal to 3 thrombocytopenia 23.8 percent). Discontinuations for AEs occurred in 0percent for Toca 511/Toca FC vs. 4.8 percent for lomustine. Toca 511 is surgically delivered and treatment-emergent AEs regardless of attribution included incision site pain (20 percent), procedural pain (12.5 percent), and wound infection (5 percent) vs. 0percent, 1.2 percent, 1.2 percent respectively for lomustine. Toca 511/Toca FC significantly improved survival and safety relative to lomustine. A Phase 2/3 trial has launched (NCT02414165).

Locations of gamma dose rate sensors have often been chosen by administrative or geometrical criteria. Nowadays computational capacity allows for a more realistic basis. We use simulations of potential radioactive plumes based on weather data of one year to investigate the threats to regions without own nuclear power plants and to find good numbers and locations of sensors to detect such plumes. We optimise sensor locations by minimising a cost function that can take into account numbers of undetected plumes, their dose to the region in general, or on the population. Besides we assess the effect of administrative constraints, be it that sensors have to cover administrative units, or that optimisation is done for sub-regions separately. Finally we evaluate the robustness of the approach if less or other plumes are used. The main findings are that sensors at boundaries are often best, but also typical paths of plumes may be important, and that administrative constraints may necessitate much more sensors. The small numbers of sensors actually deployed in these regions seem sufficient. However, the latter may be an artefact of the low number of plumes we considered. Altogether, combined with other considerations, this approach can contribute to better decisions about gamma dose rate sensor locations.
On the requirements to establish a European radiological preparedness for malicious airborne dispersion scenarios

European computerised decision support systems are currently targeted for large accidental atmospheric contaminant releases from nuclear installations. To make these systems applicable also for malicious dispersion events, such as ‘dirty bomb’ blasts, a series of modifications and extensions are necessary. Also European decision support handbooks need supplementary sections to cover the features of these types of scenarios. An overview is given of the requirements.

Within the EURANOS project data assimilation (DA) approaches have been successfully applied in two areas to improve the predictive power of simulation models used in the RODOS and ARGOS decision support systems. For the areas of atmospheric dispersion modelling and of modelling the fate of radio-nuclides in urban areas the results of demonstration exercises are presented here. With the data assimilation module of the RIMPUFF dispersion code, predictions of the gamma dose rate are corrected with simulated readings of fixed detector stations. Using the DA capabilities of the IAMM package for mapping the radioactive contamination in inhabited areas, predictions of a large scale deposition model have been combined with hypothetical measurements on a local scale. In both examples the accuracy of the model predictions has been improved and the uncertainties have been reduced.
Effect of moderate intakes of different tea catechins and caffeine on acute measures of energy metabolism under sedentary conditions

Green tea may stimulate energy metabolism; however, it is unclear if acute effects are caused by specific catechins, caffeine or their combination. The objective of the present study was to examine the separate and combined effects of different catechins and caffeine on energy expenditure (EE) and fat oxidation over a single day. Fifteen healthy, normal-weight males received capsules containing placebo, caffeine alone (150 mg), or caffeine plus a catechin mixture (600 mg) enriched in either epigallocatechin-3-gallate (EGCG), epigallocatechin or a mix of catechins, in a randomised cross-over double-blinded design. On each test day EE, respiratory quotient (RQ) and substrate oxidation were measured under sedentary conditions in a respiratory chamber for 13·5 h. We found no significant treatment effect on EE (P = 0·20) or RQ (P = 0·68). EGCG with caffeine insignificantly raised EE and fat oxidation v. caffeine-only and placebo (EE 5·71 (se 0·12) v. 5·68 (se 0·14) v. 5·59 (se 0·13) MJ/12·5 h, respectively; fat oxidation 84·8 (se 5·2) v. 80·7 (se 4·7) v. 76·8 (se 4·0) g/12·5 h). Catechin/caffeine combinations at these dosages and mode of application had non-significant acute effects on EE and fat oxidation. The maximum observed effect on EE of about 2 % could still be meaningful for energy balance over much longer period of exposure. However, higher short-term effects reported in the literature may reflect variations in green tea extracts, added caffeine, or synergies with physical activity. The specific mechanisms and conditions that may underpin observed longer-term benefits of catechin-enriched green tea consumption on body composition remain to be confirmed.

Particle diffusion is investigated in a strictly two-dimensional collisionless guiding-centre model for a strongly magnetized plasma. An analytical expression is presented for the entire time variation of the mean square test-particle displacement in the limit of low-frequency, strongly turbulent, electric field fluctuations. The analysis relies on an explicit integral expression for the Lagrangian autocorrelation function in terms of the Eulerian wavenumber spectrum and a time-varying weight function. Bohm diffusion is discussed by means of a simple model spectrum. The analysis applies for turbulent transport associated with electrostatic convective cells, magnetostatic cells and drift wave turbulence with the assumption of local homogeneity and isotropy in two dimensions.

The European Model for Inhabited Areas (ERMIN) was developed to allow a user to explore different recovery options following the contamination of an urban environment with radioactive material and to refine an appropriate strategy for the whole region affected. The input data include a description of the environment, initial deposition of radionuclides on to a reference surface and a description of countermeasures. Output information includes the average doses to members of the public from external exposure to gamma and beta radiation from deposited radionuclides and inhalation of resuspended radioactivity, the contamination on urban surfaces, the activity concentration in air from resuspension, the doses to workers undertaking the recovery work, the quantity and activity of waste generated and the cost and work required to implement the countermeasure. ERMIN has been designed to be implemented as a tool that supports the approach of decision-makers and allows the area to be broken down into smaller regions where different conditions prevail and different countermeasure packages are enacted.

Interface motion in a vibrated granular layer
Granular materials are now recognized as a distinct state of matter, and studies of their behavior form a fascinating interdisciplinary branch of science. The intrinsic dissipative nature of the interactions between the constituent macroscopic particles gives rise to several basic properties specific to granular substances, setting granular matter apart from the conventional gaseous, liquid, or solid states.
Thin layers of granular materials subjected to vertical vibration exhibit a diversity of patterns. The particular pattern is determined by the interplay between driving frequency f and the acceleration amplitude Γ. Interfaces in vibrated granular layers, existing for large enough amplitude of vibration, separate large domains of flat layers oscillating with opposite phase. These two phases are related to the period-doubling character of the flat layer motion at large plate acceleration. Interfaces are either smooth or “decorated” by periodic undulations depending on parameters of vibration. An additional subharmonic driving results in a controlled displacement of the interface with respect to the center of the experimental cell. The speed and the direction of the interface motion are sensitive to the phase and amplitude of the subharmonic driving.
The image sequence above shows interface nucleation and propagation towards the center of the cell, with dimensionless time tf labeled in each image. The interface forms at the right side wall of the cell due to small-amplitude phase-shifted subharmonic driving. After the additional driving stops, the interface moves towards the center, creating small-scale localized structures in the process.
Effect of fermented liquid food and zinc bacitracin on microbial metabolism in the gut and sensoric profile of m. longissimus dorsi from entire male and female pigs

The aim of this work in pigs was to evaluate the effect of fermented liquid food (FLF) and the combination of FLF plus short-term addition of an antibiotic food additive zinc bacitracin (FLF + ZB) compared with fed non-pelleted dry food (NPDF), on microbial metabolism in the gut and the effects on flavour and odour attributes and profiles of pig meat as well as boar odour from entire male and female pigs.
At an average start weight of 60 kg, 108 pigs (54 males and 54 females) were equally allocated to three treatments according to pen-replicate, litter and sex. Microbial metabolism in the gut was studied in 24 pigs, eight from each treatment. For sensory profile evaluation (flavour, odour and tenderness) of pork loins from m. longissimus dorsi, 48 pigs (24 entire males and 24 females) were selected. The sensory evaluation of different qualities of flavour and odour as well as tenderness of cooked pork loins (LD) was done by a boar taint (skatole and androstenone) trained taste panel. To evaluate the odour and the flavour, a sensory profile analysis was performed which involved the following attributes: total off-odour/off-flavour, pig, urine, manure, naphthalene, rancid, sweet and sweat in the whole sample including both meat and fat.
Giving FLF to pigs significantly changed the microbiota in the gastro-intestinal tract compared with NPDF. In particular, the density of coliform bacteria was reduced in the gastro-intestinal tract of the pigs on FLF. Giving FLF demonstrated no effects on skatole concentration in caecum, colon, blood and backfat and boar odour attributes, whereas administration of FLF + ZB decreased the skatole concentrations and the typical boar odours, pig and manure odour, compared especially with the pigs on NPDF. However, meat from pigs given FLF either with or without zinc bacitracin had smaller but significantly worsened scores for three flavour attributes — pig flavour, rancid flavour and total off-flavour — compared with meat from NPDF pigs and the three flavour attributes were equally affected in both sexes. This seems to point to a significant worsening of meat flavour in pigs given FLF that is independent of sex and boar odour problems (skatole and androstenone).
Reactive Ion Etching Damage to Shallow Junctions

The damage of reactive ion etching to shallow junctions is an important consideration in advanced technology.In this paper, the damage incurred during contact etch is studied, with emphasis on those defects responsible for junction leakage of shallow junctions.Shallow p+/n and n+/p junctions have been prepared with depths of 160 nm.Junction leakage measurements have been made for various amounts of silicon loss up to within 20 nm of the junctions by using a CHF3 + CO2 plasma.The degree of chemical and structural damage has been characterized by using photoluminescence, SIMS, and spreading carrier profiling.
The leakage current density was found to depend strongly on contact area and increase rapidly with junction etch depth after the etched surface has extended to within 80 nm of the junction boundary.The concentration and depth of damage increases with increasing plasma exposure until saturation.Etching induced defects are observed in photoluminescence, and one such defect is identified as a carbon interstitialcy.Enhanced diffusion effects were observed for both chemical contamination from the etch gas and the junction dopants.The spatial distribution of the chemical and structural damage has been found to correlate with the junction leakages.The annealing behavior of damage has also been investigated.
