Congenital transmission of Chagas disease plays an important role in endemic countries because it is not a diagnosis that is encountered frequently in prenatal care. Due to limited information regarding congenital transmission of Trypanosoma cruzi in Mexico, the present study aimed to investigate protozoan infectivity and modulation of immune responses in human placental explants infected with T. cruzi Ia Mexican strains. The Inc-5 strain showed increased infectivity and modulated IL-1β, IL-10 and TLR-4, decreasing their expression after 24 h of infection. Both strains (Inc-5 and Ninoa) stimulated the production of TNF-α and decreased IL-6 levels 96 h after infection. An important detachment of the syncytiotrophoblast caused by infection with T. cruzi was observed after 24 h of infection. In this study, ex vivo infection of human placental villi was performed to better understand interactions involving parasitic T. cruzi and human placental tissue. It was concluded that the strains of TcIa present parasitism in placental tissue, modulation of the innate immune system of the placenta, and cause intense detachment of the syncytiotrophoblast, a fact that may be more associated with abortion and premature birth events than the congenital transmission itself, justifying the low rate of this transmission mechanism by this genotype.

Visceral leishmaniasis is an endemic protozoonosis observed in over 60 countries, with over 500 000 new cases recorded annually. Although the diagnostic procedure of its symptomatic forms is well established, for asymptomatic patients, who represent about 85% of those infected, there is no consensus on the best method for its identification. Recent studies have presented molecular techniques as viable identification methods, with good sensitivity and specificity indices in asymptomatic individuals. Therefore, we aimed to use molecular methods to assess their effectiveness in identifying the presence of asymptomatic infection by Leishmania infantum (L. infantum) individuals from endemic regions of Brazil. Screening was performed by real-time polymerase chain reaction (qPCR) and confirmed by sequencing the cytochrome B gene. Of the 127 samples [from 608 blood donors who had participated in a previous study, of which 34 were positive by the enzyme-linked immunosorbent assay (ELISA) rK39] tested by qPCR, 31 (24.4%) were positive. In the sequencing of 10 qPCR-positive samples, five were identified as L. infantum. Complimentary samples of the ELISA rK39 and conventional PCR showed only reasonable and low agreement with qPCR, respectively. The qPCR confirmed the presence of infection in five of the 10 sequenced samples, ELISA confirmed three, and the conventional PCR confirmed none.
Analysis of regulatory T cells and CTLA-4 expression in pregnant women according to seropositivity to Toxoplasma gondii

Pregnancy is considered a period in which immunomodulation occurs, although it is important for the maintenance of the foetus, could contribute to infections as Toxoplasma gondii. Immune response cells such as regulatory T cells participate in this immunomodulation, and surface molecules such as CTLA-4 develop an immunosuppressive role, could contribute to the establishment of the parasite. This study aimed to evaluate the presence of regulatory T cells and the expression of CTLA-4 in parturient and non-pregnant seropositive and seronegative for anti-T. gondii antibodies. Sixty-two participants were evaluated, 14 parturient with negative serology, 23 parturient with positive serology, 16 non-pregnant women seronegative and 9 non-pregnant women seropositive. Immunophenotyping was performed for characterize TCD4+Foxp3+ cells, T CD4+CD25-Foxp3+, TCD4+CD25highFoxp3+, TCD4+CTLA-4+, TCD4+CD25-CTLA-4+ and TCD4+CD25highCTLA-4+. We observed a lower level of CD4+CD25highFoxp3+ cells from seropositive parturient compared with seropositive non-pregnant cells. Significative levels of CD4+CD25-Foxp3+ cells from seronegative pregnant were observed compared with seropositive pregnant cells. Furthermore, the higher level of CD4+CD25-CTLA-4+ cells populations was detected in seropositive pregnant cells compared with seropositive non-pregnant. Although a significant increase in CTLA-4 cells was observed in pregnant women positive for anti-T. gondii antibodies, this increase did not cause a risk of reactivation of the infection.

Chagas Disease is a zoonosis caused by the parasite Trypanosoma cruzi. Several high-resolution markers have subdivided T. cruzi taxon into at least seven lineages or Discrete Typing Units (DTUs) (TcI-TcVI and TcBat). Trypanosoma cruzi I is the most diverse and geographically widespread DTU. Recently a TcI genotype related to domestic cycles was proposed and named as TcIDOM. Herein, we combined traditional markers and housekeeping genes and applied a Multispecies Coalescent method to explore intra-TcI relationships, lineage boundaries and genetic diversity in a random set of isolates and DNA sequences retrieved from Genbank from different countries in the Americas. We found further evidence supporting TcIDOM as an independent and emerging genotype of TcI at least in Colombia and Venezuela. We also found evidence of high phylogenetic incongruence between parasite's gene trees (including introgression) and embedded species trees, and a lack of genetic structure among geography and hosts, illustrating the complex dynamics and epidemiology of TcI across the Americas. These findings provide novel insights into T. cruzi systematics and epidemiology and support the need to assess parasite diversity and lineage boundaries through hypothesis testing using different approaches to those traditionally employed, including the Bayesian Multispecies coalescent method.
Glacier recession and water resources in Peru’s Cordillera Blanca

The tropical glaciers of the Cordillera Blanca, Peru, are rapidly retreating, resulting in complex impacts on the hydrology of the upper Río Santa watershed. The effect of this retreat on water resources is evaluated by analyzing historical and recent time series of daily discharge at nine measurement points. Using the Mann-Kendall nonparametric statistical test, the significance of trends in three hydrograph parameters was studied. Results are interpreted using synthetic time series generated from a hydrologic model that calculates hydrographs based on glacier retreat sequences. The results suggest that seven of the nine study watersheds have probably crossed a critical transition point, and now exhibit decreasing dry-season discharge. Our results suggest also that once the glaciers completely melt, annual discharge will be lower than present by 2-30% depending on the watershed. The retreat influence on discharge will be more pronounced during the dry season than at other periods of the year. At La Balsa, which measures discharge from the upper Río Santa, the glacier retreat could lead to a decrease in dry-season average discharge of 30%.
Leishmania infection in bats from a non-endemic region of Leishmaniasis in Brazil

Leishmaniasis is a complex of zoonotic diseases caused by parasites of the genus Leishmania, which can develop in domestic as well as wild animals and humans throughout the world. Currently, this disease is spreading in rural and urban areas of non-endemic regions in Brazil. Recently, bats have gained epidemiological significance in leishmaniasis due to its close relationship with human settlements. In this study, we investigated the presence of Leishmania spp. DNA in blood samples from 448 bats belonging to four families representing 20 species that were captured in the Triangulo Mineiro and Alto Paranaiba areas of Minas Gerais State (non-endemic areas for leishmaniasis), Brazil. Leishmania spp. DNA was detected in 8·0% of the blood samples, 41·6% of which were Leishmania infantum, 38·9% Leishmania amazonensis and 19·4% Leishmania braziliensis. No positive correlation was found between Leishmania spp. and bat food source. The species with more infection rates were the insectivorous bats Eumops perotis; 22·2% (4/18) of which tested positive for Leishmania DNA. The presence of Leishmania in the bat blood samples, as observed in this study, represents epidemiological importance due to the absence of Leishmaniasis cases in the region.
Psychometric Properties of the Spanish Adaptation of the Health Care Communication Questionnaire (HCCQ)

This study’s aim is to adapt the Health Care Communication Questionnaire in a Spanish sample, and then test the psychometric properties of the adapted instrument. To do so, the questionnaire was adapted for the Spanish context and then applied in a pilot study as well as a final study. The final sample consisted of 200 patients at Morales Meseguer Hospital in Murcia, Spain. The results show that this adaptation’s psychometric properties were similar to those of the original questionnaire. As for item analysis, all items obtained discriminant indices > .30. Confirmatory factor analysis revealed the same structure as that of the original questionnaire (χ2/df = 1.345; CFI = .983; IFI = .983; TLI = .977; RMSEA = .042), with indices reflecting adequate goodness of fit. Also, results from the analysis of each dimension’s internal consistency had coefficients between .71 and .86. We conclude that the Spanish version of the HCCQ has adequate psychometric properties, is useful, and will serve its purpose in the context in which it will be used.
Morphometric trait differentiation between a wild and a mass-reared population of Anastrepha fraterculus (Diptera: Tephritidae)

The South American fruit fly Anastrepha fraterculus (Wiedemann) is an important pest in many countries. The sterile insect technique is an effective method of controlling Ceratitis capitata (Wiedemann) in Argentina and has been proposed for use against A. fraterculus. Because this technique relies on sterile mass-reared males mating with wild females, it is essential to verify that artificial rearing does not reduce male mating competitiveness. Several morphometric characters were evaluated to detect differences between a wild population and a laboratory strain that was derived from it and reared artificially since 1997. Eight morphometric traits were analysed as indicators of body size, head shape and potential mobility: Thorax Length, Head Width, Face Width, Eye Length, Wing Length, Wing Width, Third Tibia Length and Femur Length. The results were analysed using multivariate analysis of variance, linear multiple regression and logistic multiple regression. In general, laboratory flies were larger than wild ones (possibly because the larval diet was supplied ad libitum). Laboratory males had significantly larger Head Width and Eye Length and a smaller Wing Width than wild males. Laboratory females differed from wild ones only by having narrower wings. These results could be due to environmental and genetic factors, or as a consequence of genetic drift (for the latter) during colony establishment plus gradual adaptation to laboratory conditions, where flight ability is most likely less important (resources are found easily at close distances). Also, short-distance interactions among individuals are more frequent in a colony, possibly favouring increased facial trait sizes by sexual selection. Because long-term morphological changes could represent the beginning of intraspecific differentiation, they should probably be worthy of some consideration if a large mass-rearing colony is established.

The catalytic activity of disordered binary alloy metal surfaces is investigated for the oxygen reduction reaction (ORR) by generating free energy diagrams and performing calculations on d-band centers of alloys. The disorder was simulated using virtual crystal approximation; then, based on periodic, self-consistent density functional theory (DFT) methods, we calculated adsorption energies of reaction intermediates. Alternative pathway for ORR mechanism, involving proton/electron transfer to adsorbed oxygen and hydroxyl, is considered. The methodology was applied to (111) surface of PdxCu1-x disordered binary alloys, with different values of x concentration. This study found that at the ORR equilibrium potential of 1.23 V, the reactivity of all surfaces is shown to be limited by the rate of OH removal from the surface. Among the surfaces studied, the surface of Pd0.80Cu0.20 shows the highest reactivity and is more active than other non-Pt alloys. These results are in excellent agreement with earlier experimental and theoretical work.

New findings of the scleractinian coral Dendrophyllia laboreli in various localities off the European coast in the Gulf of Cadiz and the Strait of Gibraltar, as well as the Atlantic coast of Morocco, are reported. These new findings redefine the current northernmost known limit of distribution of this species, previously known in African localities from Ghana to the Canary Islands. Some ecological and biogeographical remarks are also included.

The Vulnerable Antillean manatee Trichechus manatus manatus (Mammalia, Sirenia) is threatened by hunting, entanglement in fishing nets and habitat degradation but most knowledge of the species comes from coastal and estuarine populations. We therefore studied an Antillean manatee population located >1,100 km from the Caribbean coast, in the Orinoco River of Colombia, during 2001–2005. To verify the occurrence of the manatee and to investigate its habitat use throughout the annual hydrological cycle, we carried out surveys for manatees and manatee feeding signs over c. 40 km of the Orinoco River, between El Burro and Bachaco. We made 870 sightings in 1,003 hours of observation, and found 69 feeding areas in 776 hours of surveys. From 82 interviews in a total of 28 locations in Venezuela and Colombia we compiled 90 reports of manatees injured or killed in the Orinoco during 1980–2004. Both habitat use by the manatee and anthropogenic impacts on the species vary with the annual hydrological cycle. Reproduction appears to occur during the low water period in restricted areas that retain deep water. Improved knowledge of the dynamics of manatee migration and regulation of human activities, especially those related to use of nets and boat traffic, are required for conservation of the Antillean manatee in this freshwater habitat.

Most of the tests, questionnaires, and neuropsychological batteries for the assessment of dementia have been translated and adapted for use in the Hispanic population without having normative data, which results in a high number of false positives when age and educational level are not considered. The Short Cognitive Performance Test (SKT) is a psychometric instrument evaluating memory and attention deficits that has been developed and standardized in Germany (Erzigkeit, 1989a, 1989b). The objective of this study was to adapt the SKT, to establish normative criteria that take into consideration age and educational level, and to establish its concurrent validity in comparison to other neuropsychological tests: Brief Neuropsychological Evaluation for Spanish-Speaking Subjects (Ostrosky-Solís et al., 1994), the Mini-Mental State Examination (Folstein et al., 1975), and the Blessed Functional Scale (Blessed et al., 1968) in 238 neurologically intact subjects and 97 subjects with mild to moderate dementia. The SKT showed adequate sensitivity (80.5%) and specificity (80.3%) in subjects with medium and high educational level; however, the sensitivity and specificity diminished (75% and 56.7%) in subjects with no education or low educational level. The adapted and validated version of the SKT in the Mexican population has been shown to be a psychometric instrument that in subjects with medium educational level can detect cognitive alterations and is able to determine the severity of deterioration; however, in subjects with low educational level and severe dementia, the SKT cannot be usefully administered.
