SCN2A encodes a voltage-gated sodium channel (designated NaV1.2) vital for generating neuronal action potentials. Pathogenic SCN2A variants are associated with a diverse array of neurodevelopmental disorders featuring neonatal or infantile onset epilepsy, developmental delay, autism, intellectual disability and movement disorders. SCN2A is a high confidence risk gene for autism spectrum disorder and a commonly discovered cause of neonatal onset epilepsy. This remarkable clinical heterogeneity is mirrored by extensive allelic heterogeneity and complex genotype-phenotype relationships partially explained by divergent functional consequences of pathogenic variants. Emerging therapeutic strategies targeted to specific patterns of NaV1.2 dysfunction offer hope to improving the lives of individuals affected by SCN2A-related disorders. This Element provides a review of the clinical features, genetic basis, pathophysiology, pharmacology and treatment of these genetic conditions authored by leading experts in the field and accompanied by perspectives shared by affected families. Background:
The current research sought to characterize current mood state profiles in healthy young versus older adults using 100-point visual analogue mood scales (VAMS), provide within-sample and new sample replication of age-group differences, assess sex differences, and compare with commonly used standardized symptom measures.
Methods:In two studies, six word-only VAMS (happy, sad, calm, tense, energetic, and sleepy) were administered in a laboratory setting. In Study 1, 22 young and 29 older males completed the VAMS six times (twice per day at weekly intervals). In Study 2, 60 young (30 males) and 60 older (30 males) adults completed on one occasion the VAMS, Beck Depression Inventory-II, State-Trait Anxiety Inventory, and Pittsburgh Sleep Quality Index.
Results:VAMS scores showed that older adults had a tendency to indicate feeling happier, less sad, calmer, less tense, more energetic, and less sleepy than young adults. This pattern occurred across assessment points and irrespective of sex, except for the tense VAMS, which showed higher scores in females than males in young but not older adults. The standardized measures showed significant age-group differences for Trait Anxiety only (lower in older than young adults).
Conclusions:These findings establish current mood state differences in young versus older adults. The absence of age-group differences in past studies may relate to the limited precision of the scales (only 7 points, in contrast to the 100-point scales used here).
Crop Tolerance and Weed Management Systems in Imidazolinone-Tolerant Corn (Zea mays L.)

Field studies were conducted in 1997 and 1998 to evaluate the efficacy of imidazolinone weed management systems and crop tolerance of imidazolinone-tolerant (IT) corn to imazapic. Imazapic (36 and 72 g/ha) was evaluated when applied PRE; early postemergence (EPOST), when corn was at the two- to three-leaf stage; and late postemergence (LPOST), when corn was at the six- to eight-leaf stage. Imazapyr + imazethapyr EPOST and metolachlor + atrazine followed by (fb) primisulfuron LPOST were evaluated as commercial standards. Imazapic at 36 g/ha EPOST controlled johnsongrass, Texas panicum, smellmelon, and ivyleaf and entireleaf morningglory at least 93% when adequate rainfall occurred. Devil's claw was controlled at least 85% with imazapic EPOST or LPOST at either rate. Imazapic at 36 g/ha EPOST and LPOST controlled eclipta 88 and 91%, respectively. Yellow nutsedge was controlled >91% with imazapic LPOST, which was superior to imazapic applied EPOST. Broadleaf signalgrass was controlled 94% with imazapic at 72 g/ha LPOST, which was significantly higher than other herbicide treatments. Imazapic at 36 and 72 g/ha applied PRE under moisture stress resulted in reduced weed control; but when adequate rainfall occurred, weed control was generally similar to that of EPOST and LPOST applications. Imazapic crop response at 72, 105, 140, and 211 g/ha applied at EPOST and LPOST was evaluated in two IT corn hybrids. Crop response varied with soil and environmental conditions and application timings. Imazapic at 72, 105, and 140 g/ha EPOST resulted in crop injury 33 to 55% at 6 wk after planting (WAP) in a coarse soil; however, crop injury decreased to <20% at 12 WAP. Low crop injury (<9%) was observed at 12 WAP in a clay soil. Imazapic applied LPOST resulted in lower crop injury than EPOST application. Corn plant height and yield were not affected by any imazapic treatment.

Maternal nutrition during pregnancy has a significant influence in establishing patterns of metabolism and postnatal behaviours in offspring, and therefore shapes their risk of developing disorders in later life. Although it is well established that a mismatch between food consumption and energy expenditure leads to obesity and metabolic dysregulation, little research has investigated the biological origin of such behaviour. We conducted the present experiments to investigate effects of early-life nutrition on preference between wheel running and lever pressing for food during adult life. To address this issue we employed a well-established experimental approach in the rat which has shown that offspring of mothers undernourished during pregnancy develop obesity and metabolic disorders when kept under standard laboratory conditions. Using this experimental approach, two studies were conducted where offspring of ad libitum-fed dams and dams undernourished throughout pregnancy were given the choice between wheel running and pressing a response lever for food. Across subsequent conditions, the rate at which the response lever provided food was varied from 0·22 to 6·0 (study 1) and 0·19 to 3·0 (study 2) pellets per min. Compared with the control group, offspring from dams undernourished during pregnancy showed a consistently greater preference for running over lever pressing for food throughout both experiments of the study. The results of the present study provide experimental evidence that a mother's nutrition during pregnancy can result in a long-term shift in her offspring's lifestyle choices that are relevant to obesity prevention. Such a shift, if endorsed, will have substantial and wide-ranging health consequences throughout the lifespan.

There is great potential for the use of nanostructures in numerous applications. Investigation of nanoparticle films and structures is an important area of research for the production of nanoengineered devices. However, for these devices to become a reality, a production method that can yield high-rate synthesis of nanostructured powders is necessary. The hypersonic plasma particle deposition (HPPD) process has been shown to be capable of such high-rate production of nanoparticle films and structures. Versatile in its ability to manufacture nanoparticles of different chemistries HPPD also has the capability of in situ particle consolidation and assembly. In this study, chemically diverse films and structures have been produced by HPPD on a variety of substrates. Using novel specimen preparation techniques, these nanoparticles have been characterized by TEM. Fundamental issues of importance have been investigated for both the nanoparticle structure and the constituent nanoparticles. These issues include nanoparticle crystallinity and defect structure. The chemical homogeneity and structural characteristics of the deposition are also investigated. This application of microscopy to aid process development has resulted in insights into the nanoparticle formation process and the dynamics of the HPPD process.
Synthesis of surface-metallized polymeric films by in situ reduction of (4,4,4-trifluoro-1-(2-thienyl)-1,3-butanedionato) silver(I) in a polyimide matrix

Thermal curing of the (4,4,4-trifluoro-1-(2-thienyl)-1,3-butanedionato)silver(I)-containing poly(amic acid) formed from 3,3′,4,4′-benzophenone tetracarboxylic acid dianhydride (BTDA) and 4,4′-oxydianiline (4,4′-ODA) in dimethylacetamide gives both polyimide films via cyclodehydration and reduction of silver(I) to the native metal. Silver(0) migrates to the surface resulting in surface metallized composite films, which can have excellent reflectivity, but do not exhibit surface electrical conductivity. The films retain mechanical and thermal properties similar to those of the parent polyimide. X-ray diffraction shows crystalline face-centered-cubic silver in the films after thermal curing. Microscopy data show that the surface particle sizes are in the range of approximately 50–100 nm. Significant silver remains in the bulk of the polyimide film with varying particles sizes generally less than approximately 15 nm. The interior of the metallized films is not electrically conducting. Films were characterized by x-ray diffraction, differential scanning calorimetry, thermal gravimetric analysis, x-ray photoelectron spectroscopy, transmission electron microscopy, scanning electron microscopy, and mechanical measurements.
