The emerging role of NOTCH3 receptor signalling in human lung diseases

The mammalian respiratory system or lung is a tree-like branching structure, and the main site of gas exchange with the external environment. Structurally, the lung is broadly classified into the proximal (or conducting) airways and the distal alveolar region, where the gas exchange occurs. In parallel with the respiratory tree, the pulmonary vasculature starts with large pulmonary arteries that subdivide rapidly ending in capillaries adjacent to alveolar structures to enable gas exchange. The NOTCH signalling pathway plays an important role in lung development, differentiation and regeneration post-injury. Signalling via the NOTCH pathway is mediated through activation of four NOTCH receptors (NOTCH1-4), with each receptor capable of regulating unique biological processes. Dysregulation of the NOTCH pathway has been associated with development and pathophysiology of multiple adult acute and chronic lung diseases. This includes accumulating evidence that alteration of NOTCH3 signalling plays an important role in the development and pathogenesis of chronic obstructive pulmonary disease, lung cancer, asthma, idiopathic pulmonary fibrosis and pulmonary arterial hypertension. Herein, we provide a comprehensive summary of the role of NOTCH3 signalling in regulating repair/regeneration of the adult lung, its association with development of lung disease and potential therapeutic strategies to target its signalling activity.
Defining Regions of Pre-Colonial Africa: A Controlled Vocabulary for Linking Open-Source Data in Digital History Projects

Regionalizing pre-colonial Africa aids in the collection and interpretation of primary sources as data for further analysis. This article includes a map with six broad regions and 34 sub-regions, which form a controlled vocabulary within which researchers may geographically organize and classify disparate pieces of information related to Africa’s past. In computational terms, the proposed African regions serve as data containers in order to consolidate, link, and disseminate research among a growing trend in digital humanities projects related to the history of the African diasporas before c. 1900. Our naming of regions aims to avoid terminologies derived from European slave traders, colonialism, and modern-day countries.
What do community paramedics assess? An environmental scan and content analysis of patient assessment in community paramedicine

Objectives
Patient assessment is a fundamental feature of community paramedicine, but the absence of a recognized standard for assessment practices contributes to uncertainty about what drives care planning and treatment decisions. Our objective was to summarize the content of assessment instruments and describe the state of current practice in community paramedicine home visit programs.
MethodsWe performed an environmental scan of all community paramedicine programs in Ontario, Canada, and used content analysis to describe current assessment practices in home visit programs. The International Classification on Functioning, Disability, and Health (ICF) was used to categorize and compare assessments. Each item within each assessment form was classified according to the ICF taxonomy.
ResultsA total of 43 of 52 paramedic services in Ontario, Canada, participated in the environmental scan with 24 being eligible for further investigation through content analysis of intake assessment forms. Among the 24 services, 16 met inclusion criteria for content analysis. Assessment forms contained between 13 and 252 assessment items (median 116.5, IQR 134.5). Most assessments included some content from each of the domains outlined in the ICF. At the subdomain level, only assessment of impairments of the functions of the cardiovascular, hematological, immunological, and respiratory systems appeared in all assessments.
ConclusionAlthough community paramedicine home visit programs may differ in design and aim, all complete multi-domain assessments as part of patient intake. If community paramedicine home visit programs share similar characteristics but assess patients differently, it is difficult to expect that the resulting referrals, care planning, treatments, or interventions will be similar.

In recent years, an increasing number of online archival databases of primary sources related to the history of the African diaspora and slavery have become freely and readily accessible for scholarly and public consumption. This proliferation of digital projects and databases presents a number of challenges related to aggregating data geographically according to the movement of people in and out of Africa across time and space. As a requirement to linking data of open-source digital projects, it has become necessary to delimit the entire continent of precolonial Africa during the era of the slave trade into broad regions and sub-regions that can allow the grouping of data effectively and meaningfully.
Evaluating risks of plant growth regulator–resistant soybean technologies to horseradish production

Off-target movement of dicamba and 2,4-D may injure and reduce the yield of many fruit and vegetable crops, impacting specialty crop producers and herbicide applicators alike. Two field experiments were established, using plant growth regulator–resistant soybean herbicide technologies, to evaluate drift and carryover risks to horseradish production. The drift experiment was conducted in 2015 and 2016 to evaluate impact of dicamba and 2,4-D simulated drift on horseradish production with a mid-POST application in soybean. Simulated drift rates were 1/10,000X, 1/1,000X, and 1/100X, with 1/2X, 1X, and 2X of standard application rates. Injury and yield loss was greater following application of 2,4-D than with dicamba. Yield reductions were observed beginning at the 1/1,000X rate of 2,4-D, with complete crop loss occurring when rates exceed 1/2X. In comparison, dicamba only reduced yields when applied at the 1X and 2X rates. Only horseradish roots from plants treated with dicamba at the 2X rate had greater dicamba residue than the nontreated control, and the amount detected, 0.32 parts per billion (ppb), was lower than the EPA tolerance of 100 ppb in root crops. There was little to no harvestable tissue for 2,4-D residue analysis for plants treated with 2,4-D at rates above 1/2X. The carryover experiment was a 2-yr rotational evaluation conducted in 2014, 2015, and 2016 to assess dicamba carryover to horseradish following application to dicamba-resistant soybean the previous season. Observations taken at 4, 6, and 8 wk after planting indicated no significant horseradish injury, nor was height, stand, or root weight reduced. These results suggest that horseradish growers should have few concerns about injury from dicamba drift or carryover. While 2,4-D applicators may need to be cautious when making applications near horseradish fields, 2,4-D may be an effective tool for controlling volunteer horseradish in 2,4-D–resistant soybean.
Unwell in hospital but not incapable: cross-sectional study on the dissociation of decision-making capacity for treatment and research in in-patients with schizophrenia and related psychoses

Background
Consent to research with decision-making capacity for research (DMC-R) is normally a requirement for study participation. Although the symptoms of schizophrenia and related psychoses are known to affect decision-making capacity for treatment (DMC-T), we know little about their effect on DMC-R.
AimsWe aimed to determine if DMC-R differs from DMC-T in proportion and associated symptoms in an in-patient sample of people with schizophrenia and related psychoses.
MethodCross-sectional study of psychiatric in-patients admitted for assessment and/or treatment of schizophrenia and related psychoses. We measured DMC-R and DMC-T using ‘expert judgement’ clinical assessment guided by the MacArthur Competence Assessment Tool for Clinical Research, the MacArthur Competence Assessment Tool for Treatment and the legal framework of the Mental Capacity Act (2005), in addition to symptoms of psychosis.
ResultsThere were 84 participants in the study. Half the participants had DMC-R (51%, 95% CI 40–62%) and a third had DMC-T (31%, 95% CI 21–43%) and this difference was statistically significant (P < 0.01). Thought disorder was most associated with lacking DMC-R (odds ratio 5.72, 95% CI 2.01–16.31, P = 0.001), whereas lack of insight was most associated with lacking DMC-T (odds ratio 26.34, 95% CI 3.60–192.66, P = 0.001). With the exception of improved education status and better DMC-R, there was no effect of sociodemographic variables on either DMC-R or DMC-T.
ConclusionsWe have shown that even when severely unwell, people with schizophrenia and related psychoses in in-patient settings commonly retain DMC-R despite lacking DMC-T. Furthermore, different symptoms have different effects on decision-making abilities for different decisions. We should not view in-patient psychiatric settings as a research ‘no-go area’ and, where appropriate, should recruit in these settings.

Control of Volunteer Horseradish and Palmer Amaranth (Amaranthus palmeri) with Dicamba and Glyphosate

Management of volunteer horseradish is a challenge when it is grown in rotation with other crops, such as corn and soybean. Although volunteer horseradish may not cause yield loss, these plants serve as hosts for various soilborne pathogens that damage subsequent horseradish crops. In addition to volunteer horseradish, glyphosate-resistant Palmer amaranth is becoming difficult to control in southwestern Illinois, as a consequence of the plant’s ability to withstand glyphosate and drought, produce many seeds, and grow rapidly. Field studies were conducted to evaluate the effect of glyphosate and dicamba on volunteer horseradish and Palmer amaranth control in 2014 and 2015. Glyphosate alone (1,265 and 1,893 g ae ha−1) and glyphosate plus dicamba at the high rate (1,680 g ae ha−1) provided the greatest volunteer horseradish control, ranging from 81% to 89% and 90% to 93%, respectively. Measures of root biomass reduction followed similar trends. Glyphosate alone provided the greatest reduction in volunteer horseradish root viability (79% to 100%) but was similar in efficacy to applications of glyphosate plus dicamba in most comparisons. Efficacy of PRE-only applications on Palmer amaranth control ranged from 92% to 99% control in 2014 and 68% to 99% in 2015. However, PRE-only applications were often similar in efficacy to PRE followed by (fb) glyphosate plus dicamba applied POST. Treatments containing flumioxazin did not control Palmer amaranth as well as other treatments. POST applications alone were not effective in managing Palmer amaranth. Many of the PRE fb POST treatment options tested will improve resistance management over PRE-only treatments, provide control of Palmer amaranth, and allow horseradish to be planted the following spring.

With advances in therapeutics for rare, genetic and syndromic diseases, there is an increasing need for objective assessments of phenotypic endpoints. These assessments will preferentially be high precision, non-invasive, non-irradiating, and relatively inexpensive and portable. We report a case of a child with an extensive lymphatic vascular malformation of the head and neck, treated with an mammalian target of Rapamycin (mTOR) inhibitor that was assessed using 3D facial analysis. This case illustrates that this technology is prospectively a cost-effective modality for treatment monitoring, and it supports that it may also be used for novel explorations of disease biology for conditions associated with disturbances in the mTOR, and interrelated, pathways.

Phonon density of states of model ferroelectrics

First principles density functional calculations and inelastic neutron scattering measurements have been used to study the variations of the phonon density of states of PbTiO3 and SrTiO3 as a function of temperature. The phonon spectra of the quantum paraelectric SrTiO3 is found to be fundamentally distinct from those of ferroelectric PbTiO3 and BaTiO3. SrTiO3 has a large 70-90 meV phonon band-gap in both the low temperature antiferrodistortive tetragonal phase and in the high temperature cubic phase.
Key bonding changes in these perovskites lead to spectacular differences in their observed phonon density of states.
