Background
Studying phenotypic and genetic characteristics of age at onset (AAO) and polarity at onset (PAO) in bipolar disorder can provide new insights into disease pathology and facilitate the development of screening tools.
AimsTo examine the genetic architecture of AAO and PAO and their association with bipolar disorder disease characteristics.
MethodGenome-wide association studies (GWASs) and polygenic score (PGS) analyses of AAO (n = 12 977) and PAO (n = 6773) were conducted in patients with bipolar disorder from 34 cohorts and a replication sample (n = 2237). The association of onset with disease characteristics was investigated in two of these cohorts.
ResultsEarlier AAO was associated with a higher probability of psychotic symptoms, suicidality, lower educational attainment, not living together and fewer episodes. Depressive onset correlated with suicidality and manic onset correlated with delusions and manic episodes. Systematic differences in AAO between cohorts and continents of origin were observed. This was also reflected in single-nucleotide variant-based heritability estimates, with higher heritabilities for stricter onset definitions. Increased PGS for autism spectrum disorder (β = −0.34 years, s.e. = 0.08), major depression (β = −0.34 years, s.e. = 0.08), schizophrenia (β = −0.39 years, s.e. = 0.08), and educational attainment (β = −0.31 years, s.e. = 0.08) were associated with an earlier AAO. The AAO GWAS identified one significant locus, but this finding did not replicate. Neither GWAS nor PGS analyses yielded significant associations with PAO.
ConclusionsAAO and PAO are associated with indicators of bipolar disorder severity. Individuals with an earlier onset show an increased polygenic liability for a broad spectrum of psychiatric traits. Systematic differences in AAO across cohorts, continents and phenotype definitions introduce significant heterogeneity, affecting analyses.

Cultural transmission biases such as prestige are thought to have been a primary driver in shaping the dynamics of human cultural evolution. However, few empirical studies have measured the importance of prestige relative to other effects, such as content biases present within the information being transmitted. Here, we report the findings of an experimental transmission study designed to compare the simultaneous effects of a model using a high- or low-prestige regional accent with the presence of narrative content containing social, survival, emotional, moral, rational, or counterintuitive information in the form of a creation story. Results from multimodel inference reveal that prestige is a significant factor in determining the salience and recall of information, but that several content biases, specifically social, survival, negative emotional, and biological counterintuitive information, are significantly more influential. Further, we find evidence that reliance on prestige cues may serve as a conditional learning strategy when no content cues are available. Our results demonstrate that content biases serve a vital and underappreciated role in cultural transmission and cultural evolution.

Background
Recently, artificial intelligence-powered devices have been put forward as potentially powerful tools for the improvement of mental healthcare. An important question is how these devices impact the physician-patient interaction.
AimsAifred is an artificial intelligence-powered clinical decision support system (CDSS) for the treatment of major depression. Here, we explore the use of a simulation centre environment in evaluating the usability of Aifred, particularly its impact on the physician–patient interaction.
MethodTwenty psychiatry and family medicine attending staff and residents were recruited to complete a 2.5-h study at a clinical interaction simulation centre with standardised patients. Each physician had the option of using the CDSS to inform their treatment choice in three 10-min clinical scenarios with standardised patients portraying mild, moderate and severe episodes of major depression. Feasibility and acceptability data were collected through self-report questionnaires, scenario observations, interviews and standardised patient feedback.
ResultsAll 20 participants completed the study. Initial results indicate that the tool was acceptable to clinicians and feasible for use during clinical encounters. Clinicians indicated a willingness to use the tool in real clinical practice, a significant degree of trust in the system's predictions to assist with treatment selection, and reported that the tool helped increase patient understanding of and trust in treatment. The simulation environment allowed for the evaluation of the tool's impact on the physician–patient interaction.
ConclusionsThe simulation centre allowed for direct observations of clinician use and impact of the tool on the clinician–patient interaction before clinical studies. It may therefore offer a useful and important environment in the early testing of new technological tools. The present results will inform further tool development and clinician training materials.

OBJECTIVES/SPECIFIC AIMS: To evaluate the NIH-sponsored Best Practices for Social and Behavioral Research e-learning course. METHODS/STUDY POPULATION: Four universities partnered in a pilot study to evaluate this new course. Outcomes from 294 participants completing the course included efficient progress through the training, perceived relevance of the course to current work, level of engagement with the course material, intent to work differently as a result of the course, and downloading digital resources. RESULTS/ANTICIPATED RESULTS: Participants rated the course as relevant and engaging (6.4 and 5.8 on a 7-point Likert scale) and 96% of respondents said they would recommend the course to colleagues. Qualitative analysis of participant testimonials suggested that most respondents had a readiness to change in the way they worked as a result of the course. Overall, results suggest participants completed the course efficiently, perceived outcomes positively and worked differently after the training. DISCUSSION/SIGNIFICANCE OF IMPACT: These results will inform new guidelines for future participants (e.g., average time to complete, expectations for knowledge checks in the training). Future studies should include larger samples and closer coordination and communication between study sites.

Introduction
The Best Practices in Social and Behavioral Research Course was developed to provide instruction on good clinical practice for social and behavioral trials. This study evaluated the new course.
MethodsParticipants across 4 universities took the course (n=294) and were sent surveys following course completion and 2 months later. Outcomes included relevance, how engaging the course was, and working differently because of the course. Open-ended questions were posed to understand how work was impacted.
ResultsParticipants rated the course as relevant and engaging (6.4 and 5.8/7 points) and reported working differently (4.7/7 points). Participants with less experience in social and behavioral trials were most likely to report working differently 2 months later.
DiscussionThe course was perceived as relevant and engaging. Participants described actions taken to improve rigor in implementing trials. Future studies with a larger sample and additional participating sites are recommended.
Lower Respiratory Symptoms Associated With Environmental and Reconstruction Exposures After Hurricane Sandy

Objective
In a population with prior exposure to the World Trade Center (WTC) disaster, this study sought to determine the relationship between Hurricane Sandy-related inhalation exposures and post-Sandy lower respiratory symptoms (LRS).
MethodsParticipants included 3835 WTC Health Registry enrollees who completed Wave 3 (2011-2012) and Hurricane Sandy (2013) surveys. The Sandy-related inhalational exposures examined were: (1) reconstruction exposure; (2) mold or damp environment exposure; and (3) other respiratory irritants exposure. LRS were defined as wheezing, persistent cough, or shortness of breath reported on ≥1 of the 30 days preceding survey completion. Associations between LRS and Sandy exposures, controlling for socio-demographic factors, post-traumatic stress disorder, and previously reported LRS and asthma were examined using multiple logistic regression.
ResultsOver one-third of participants (34.4%) reported post-Sandy LRS. Each of the individual exposures was also independently associated with post-Sandy LRS, each having approximately twice the odds of having post-Sandy LRS. We found a dose-response relationship between the number of types of Sandy-related exposures reported and post-Sandy LRS.
ConclusionsThis study provides evidence that post-hurricane clean-up and reconstruction exposures can increase the risk for LRS. Public health interventions should emphasize the importance of safe remediation practices and recommend use of personal protective equipment. Experiments were conducted in 1991 and 1992 to evaluate annual grass and broadleaf weed control with bromoxynil applied alone and with graminicides. Bromoxynil controlled velvetleaf better than entireleaf morningglory and controlled both species better than sicklepod. Mixing bromoxynil with fluazifop-P, quizalofop-P, and sethoxydim reduced control of barnyardgrass, green foxtail, large crabgrass, and shattercane. Control of broadleaf signalgrass with quizalofop-P was reduced by bromoxynil. Annual grass control with clethodim was not affected by bromoxynil.
Moisture Stress Effects on the Absorption, Translocation, and Metabolism of Haloxyfop in Johnsongrass (Sorghum halepense) and Large Crabgrass (Digitaria sanguinalis)

Glasshouse studies were undertaken to determine the effect of imposed moisture stress on the phytotoxicity of haloxyfop; the absorption, translocation, and metabolism of 14C-haloxyfop; and 14C-photoassimilate partitioning in johnsongrass and large crabgrass. Following foliar applications of haloxyfop at 30 and 25 g ai ha–1 to large crabgrass and johnsongrass, respectively, control 15 days after treatment was 92% for nonstressed plants and less than 12% for water-stressed plants. Foliar absorption of 14C-haloxyfop was reduced by moisture stress 1, 3, 5, and 24 h after treatment (HAT) in large crabgrass and 1, 3, 5, 48, and 72 HAT in johnsongrass. Regardless of stress treatment, absorption in both species reached a maximum by 24 HAT. Translocation of the radiolabel from the treated leaf to plant parts above and below the node of the treated leaf was inhibited by moisture stress in large crabgrass and johnsongrass at all harvest intervals beginning 5 and 24 HAT, respectively. Metabolism of 14C-haloxyfop was not altered by moisture stress. Fixation of 14CO2 and subsequent distribution of the 14C-photoassimilates were reduced by moisture stress. Decreases in photoassimilate translocation were similar to reductions in 14C-haloxyfop translocation. Moisture stress reduced the phytotoxicity of haloxyfop in the two grasses, and the reduced activity of haloxyfop appeared to be partially related to changes in herbicide absorption and translocation.

In 2010, a grower survey was administered to 1,299 growers in 22 states to determine changes in weed management in the United States from 2006 to 2009. The majority of growers had not changed weed management practices in the previous 3 yr; however, 75% reported using weed management practices targeted at glyphosate-resistant (GR) weeds. Growers were asked to rate their efforts at controlling GR weeds and rate the effectiveness of various practices for controlling/preventing GR weeds regardless of whether they were personally using them. Using the herbicide labeled rate, scouting fields, and rotating crops were among the practices considered by growers as most effective in managing GR weeds. Sixty-seven percent of growers reported effective management of GR weeds. Between the 2005 and 2010 Benchmark surveys, the frequency of growers using specific actions to manage GR weeds increased markedly. Although the relative effectiveness of practices, as perceived by growers, remained the same, the effectiveness rating of tillage and the use of residual and POST herbicides increased.
Seedbank and Field Emergence of Weeds in Glyphosate-Resistant Cropping Systems in the United States

A segment of the debate surrounding the commercialization and use of glyphosate-resistant (GR) crops focuses on the theory that the implementation of these traits is an extension of the intensification of agriculture that will further erode the biodiversity of agricultural landscapes. A large field-scale study was initiated in 2006 in the United States on 156 different field sites with a minimum 3-yr history of GR-corn, -cotton or -soybean in the cropping system. The impact of cropping system, crop rotation, frequency of using the GR crop trait, and several categorical variables on seedbank weed population density and diversity was analyzed. The parameters of total weed population density of all species in the seedbank, species richness, Shannon's H′ and evenness were not affected by any management treatment. The similarity between the seedbank and aboveground weed community was more strongly related to location than management; previous year's crops and cropping systems were also important while GR trait rotation was not. The composition of the weed flora was more strongly related to location (geography) than any other parameter. The diversity of weed flora in agricultural sites with a history of GR crop production can be influenced by several factors relating to the specific method in which the GR trait is integrated (cropping system, crop rotation, GR trait rotation), the specific weed species, and the geographical location. Continuous GR crop, compared to fields with other cropping systems, only had greater species diversity (species richness) of some life forms, i.e., biennials, winter annuals, and prostrate weeds. Overall diversity was related to geography and not cropping system. These results justify further research to clarify the complexities of crops grown with herbicide-resistance traits to provide a more complete characterization of their culture and local adaptation to the weed seedbank.

Almost 1,650 corn, cotton, and soybean growers in 22 states participated in a 2010 telephone survey to determine their attitudes with regard to which weed species were most problematic in glyphosate-resistant (GR) crop production systems for corn, cotton, and soybean. The survey is a follow-up to a previous 2005 to 2006 survey that utilized a smaller set of growers from fewer states. In general, growers continued to estimate weed populations as low and few challenges have been created following adoption of GR cropping systems. Pigweed and foxtail species were dominant overall, whereas other species were more commodity and state specific. Corn, cotton, and soybean growers cited velvetleaf, annual morningglory, and waterhemp, respectively, as predominant weeds. Growers in the South region were more likely to report pigweed and waterhemp (Amaranthus spp.), whereas growers in the East and West reported horseweed. When growers were asked with which GR weeds they had experienced personally, horseweed was reported in all regions, but growers in the South more frequently reported pigweed, whereas growers in the East and West regions more frequently reported waterhemp. Comparisons with the previous 2005 survey indicated that more growers believed they were experiencing GR weeds and were more aware of specific examples in their state. In particular, the Amaranthus complex was of greatest concern in continuously cropped soybean and cotton.

A 2010 survey of 1,299 corn, cotton, and soybean growers was conducted to determine their attitudes and awareness regarding glyphosate-resistant (GR) weeds and resultant implications on weed management practices. An additional 350 growers included in the current study participated in a 2005 survey, and these answers were compared across time so that cross-sectional and longitudinal comparisons of responses could be made. Most growers surveyed in 2010 were aware of the potential for weeds to evolve resistance to glyphosate; however, many growers were not aware of glyphosate resistance in specific weeds in their county or state. Growers in the South were different from growers in other geographic regions and were significantly more aware of local cases of GR weeds. Awareness of GR weeds did not increase appreciably from 2005 to 2010, but the percentage who reported GR weeds as problematic was significantly higher. Grower reports of GR weeds on-farm in 2010 were up considerably from 2005, with growers in the South reporting significantly more instances than growers in other regions. Growers in the South were also more likely to consider glyphosate resistance a serious problem. Overall, 30% of growers did not consider GR weeds to be a problem. It appears that most growers received information about glyphosate resistance from farm publications, although in the South this percentage was less than for other geographic regions. Growers in the South received more information from universities and extension sources.

Approximately 1,300 growers from 22 states were surveyed during 2010 to determine herbicide use. Cropping systems included continuous glyphosate-resistant corn, cotton, and soybean, and various combinations of these crops and rotations with non–glyphosate-resistant crops. The most commonly used herbicide for both fall and spring applications was glyphosate followed by synthetic auxin herbicides. Herbicide application in spring was favored over application in the fall. The percentage of growers in a glyphosate-only system was as high as 69% for some cropping systems. Excluding glyphosate, the most frequently used herbicides included photosystem II, mitotic, and protoporphyrinogen oxidase inhibitors. A higher percentage of growers integrated herbicides other than glyphosate during 2010 compared with 2005. Extensive educational efforts have promoted resistance management by increasing the diversity of herbicides in glyphosate-resistant cropping systems. However, a considerable percentage of growers continued use of only glyphosate from the period of 2005 to 2010, and this practice most likely will continue to exert a high level of selection for evolved glyphosate-resistant weed species.

Amorphization of Crystalline Phase Change Material by Ion Implantation

Germanium ion implantation at an energy of 30 keV was used as a different method to re-amorphize thin films of crystalline phase change material Ge2Sb2Te5 (GST). It was found that rather low doses of 5×1013 cm-2 were sufficient to re-amorphize GST. Amorphization was determined by X-ray diffraction (XRD) as well as reflectivity measurements. Re-crystallization characteristics of ion-implantation-amorphized samples was studied using time-resolved XRD. It showed that samples re-crystallize at an increased crystallization temperature with increasing dose compared to as-deposited material. A static laser tester was applied to measure the crystallization times of material that was (1) as–deposited amorphous; (2) crystallized by annealing and re-amorphized by melt-quenching using a laser pulse; and (3) crystallized by annealing and re-amorphized by ion implantation. It was found that as-deposited amorphous and high-dose ion implanted samples had longer crystallization times while melt-quenched amorphous and low-dose ion implanted samples had shorter crystallization times.
