Selection of Rattus norvegicus cumulus–oocyte complex for vitrification by brilliant cresyl blue

The influence of the method of evaluating developmentally competent oocytes on their viability after cryopreservation still needs to be better understood. The objective of this study was to determine the cleavage and embryo developmental rates after parthenogenetic activation of cumulus–oocyte complexes (COCs) selected by different concentrations of brilliant cresyl blue (BCB) and cryopreservation. In the first experiment, COCs were separated into groups and incubated for 1 h in medium containing BCB (13 μM, 16 μM, or 20 μM). The control group was not exposed to BCB staining. In the second experiment, COCs were divided into four groups: 13 μM BCB(+), 13 μM BCB(−), fresh control (selected by morphologic observation and immediately in vitro matured) and vitrified control (selected by morphologic evaluation, vitrified, and in vitro matured). In the first experiment, the 13 μM BCB group displayed greater development rates at the morula stage (65.45%, 36/55) when compared with the other groups. In the second experiment, cleavage (47.05%, 72/153) and morula development (33.55%, 51/153) of the control group of fresh COCs were increased compared with the other groups. However, when comparing morula rates between vitrified COC control and BCB(+) groups, the BCB(+) group had better results (19.23%, 5/26 and 64.7%, 11/17, respectively). Our best result in rat COC selection by BCB staining was obtained using a concentration of 13 μM. This selection could be a valuable tool to improve vitrification outcomes, as observed by the BCB(+) group that demonstrated better results compared with the vitrified COC control.
From social networks to bird enthusiasts: reporting interactions between plastic waste and birds in Peru

Peru has the second-highest diversity of birds in the world, but little is known about the interactions between birds and plastic waste. To fill this knowledge gap, we searched the scientific literature, collected information from social networks such as Facebook and databases such as Macaulay Library and iNaturalist and solicited records through messaging with researchers and bird enthusiasts. We found 119 bird interactions with plastic debris involving 39 species from 20 families, with the red-legged cormorant Phalacrocorax gaimardi and the neotropical cormorant Phalacrocorax brasilianus being the most affected species. By type of interaction category, plastic waste in nests was the most abundant, followed by entanglement, capture and handling and ingestion. Ropes, nets and soft plastics such as bags were the most frequently reported types of waste. As our methodology has limitations, it is probable that other species that also interact with plastic waste have not been reported, so we recommend further study.
Distribution of daily protein intake across meals and all-cause mortality in community-dwelling older adults

Recent findings suggest that the distribution of protein intake throughout the day has an impact on various health outcomes in older adults, independently of the amount consumed. We evaluated the association between the distribution of dietary protein intake across meals and all-cause mortality in community-dwelling older adults. Data from 3225 older adults aged ≥ 60 years from the Seniors-ENRICA-1 cohort were examined. Habitual dietary protein consumption was collected in 2008–2010 and in 2012 through a validated diet history. Protein distribution across meals was calculated for each participant as the coefficient of variation (CV) of protein intake per meal, in sex-specific tertiles. Vital status was obtained from the National Death Index up to 30 January 2020. Cox proportional hazards regression was performed to determine the hazard ratios (HR) and their 95 % CI for the association between the distribution of daily protein intake across meals and all-cause mortality. Over a median follow-up of 10·6 years, 591 deaths occurred. After adjustment for potential confounders, the CV of total protein intake was not associated with all-cause mortality (HR and 95 % CI in the second and third tertile v. the lowest tertile: 0·94 (0·77, 1·15) and 0·88 (0·72, 1·08); Ptrend = 0·22). Similarly, the HR of all-cause mortality when comparing extreme tertiles of CV for types of protein were 0·89 (0·73, 1·10) for animal-protein intake and 1·02 (0·82, 1·25) for plant-protein intake. Dietary protein distribution across meals was not associated with all-cause mortality, regardless of protein source and amount, among older adults. Further studies should investigate whether this picture holds for specific causes of death.
Prolonged nightly fasting and lower-extremity functioning in community-dwelling older adults

It is unknown if time-restricted feeding confers a protective effect on the physical function of older adults. The aim of this study was to assess prolonged nightly fasting in association with performance-based lower-extremity function (LEF) in a large population of community-dwelling older adults. A cross-sectional study was carried out among 1226 individuals ≥64 years from the Seniors-ENRICA-II (Study on Nutrition and Cardiovascular Risk in Spain) cohort. Habitual diet was assessed through a validated diet history. Fasting time was classified into the following categories: ≤9, 10–11 and ≥12 h/d (prolonged nightly fasting). Performance-based LEF was assessed with the Short Physical Performance Battery (SPPB). After adjusting for potential confounders, a longer fasting period was associated with a higher likelihood of impaired LEF (OR for the second and third categories v. ≤ 9 h/d fasting: 2·27 (95 % CI 1·56, 3·33) and 2·70 (95 % CI 1·80, 4·04), respectively; P trend < 0·001). Fasting time showed a significant association with the SPPB subtests balance impairment (OR for highest v. shortest fasting time: 2.48; 95 % CI 1·51, 4·08; P trend = 0·001) and difficulty to rise from a chair (OR 1·47; 95 % CI 1·05, 2·06; P trend = 0·01). The risk associated with ≥12 h fasting among those with the lowest levels of physical activity was three times higher than among those with ≤9 h fasting with the same low level of physical activity. Prolonged nightly fasting was associated with a higher likelihood of impaired LEF, balance impairment, and difficulty to rise from a chair in older adults, especially among those with low levels of physical activity.

The objective of this study was to identify attitudes towards the scientific search for extraterrestrial life among students from public and private universities in Peru. This research was inspired by similar studies, realized in Sweden, which used the same instrument adapting it to our reality. The process consisted of a survey of the Peruvian student population by targeting it in different regions of Peru. The sample consisted of 1237 students from different academic areas. The findings show that 92% of the students believe in the existence of life outside our planet, with differences between the subgroups surveyed.
Are legume-based recipes an appropriate source of nutrients for healthy ageing? A prospective cohort study

Although legumes are rich in protein and fibre, and low in saturated fat and Na, traditional legume-based recipes include substantial amounts of processed meat, salt and potatoes, which could counteract the potential benefits of legumes. This prospective study aimed to assess the longitudinal association of consumption of different types of legumes, and traditional legume-based recipes, with unhealthy ageing in older adults. Data were taken from 2505 individuals aged ≥60 years from the Seniors-ENRICA cohort. Habitual legume consumption was assessed in 2008–2010 with a validated diet history. Unhealthy ageing was measured in the 2013, 2015 and 2017 follow-up waves, with a fifty-two-item multidimensional health deficit accumulation index (DAI) which ranges from 0 (best) to 100 (worst health). The mean age was 68·7 years, with 53·1 % of women. Among study participants, 78·4 % reported consumption of legumes, with a mean intake of 57·9 g/d. Multivariable-adjusted linear regression models did not show an association between total legume consumption and the DAI over a 7-year follow-up (non-standardised coefficient for the second and highest v. the lowest tertile of consumption: 0·94 (95 % CI −0·30, 2·17) and 0·18 (95 % CI −1·07, 1·43), respectively; Ptrend = 0·35). Similar results were observed for the 3-year and 5-year follow-ups and, separately, for lentils, beans, chickpeas and traditional legume-based recipes. According to the results obtained, consumption of legumes and traditional legume-based recipes is not associated with unhealthy ageing and can be part of a healthy diet in old age.
Prevalence of scars of anthropogenic origin in coastal bottlenose dolphin in Ecuador

The prevalence of scars and wounds of anthropogenic origin was assessed in seven different coastal bottlenose dolphin communities (Tursiops truncatus) on the south-western coast of Ecuador. Between 2011 and 2017 a total of 117 trips were conducted representing a total sampled distance of 6281 km. Twenty-five of the 189 (13.2%) free-ranging photo-identified dolphins were recorded with dorsal fin damage, V-shaped wounds, sawed edges and deformities in the caudal region. The scarring prevalence ranged from 0 to 44.4% and was associated with either fishing interactions or vessel strikes. Dolphin scarring increased five times in the last 25 years from 2.2 to 11.1% and was correlated with a decrease in population in the inner estuary of the Gulf of Guayaquil. Damaged dorsal fins are associated mainly with fishing gear. V-shaped wounds were recorded in three different communities, Posorja, Estero Salado and Salinas, and given their severity associated with either fishing gear or vessel strikes. During the study period three dolphins were found entangled in fishing gears, two in gillnets and one in a long-line, emphasizing the threats posed by current fishing practice to the species. More effective management measures are urgently needed to reverse the observed population decline. This may include reduction of fishing effort, implementation of area-based approaches to coastal planning (including Marine Protected Area designations) and support for further research to understand the problem. Given the difficulty in taking direct observations, scarring prevalence is proposed as a proxy for estimating boat traffic and fishing gear impacts upon cetaceans.

The objective of this work was to evaluate the rate of meiosis resumption and nuclear maturation of rat (Rattus norvegicus) oocytes selected for in vitro maturation (IVM) after staining of cumulus–oocyte complexes (COCs) with blue cresyl brilliant (BCB) using different protocols: exposure for 30, 60 or 90 min at 26 μM BCB (Experiment 1), and exposure for 60 min at 13, 20 or 26 μM BCB (Experiment 2). In Experiment 1, the selection of oocytes exposed to BCB for 60 min was found to be the most suitable, as meiosis resumption rates in the BCB+ group (n = 35/61; 57.37%) were the closest to the observed in the control (not exposed) group (n = 70/90; 77.77%) and statistically higher than the values observed for the BCB− group (n = 3/41; 7.32%). Additionally, the more effective evaluation of diagnostic tests (sensitivity and negative predictive value 100%) was observed in COCs exposed for 60 min. In Experiment 2, the 13 μM BCB+ group presented rates of meiosis resumption (n = 57/72; 72.22%) similar to the control group (n = 87/105; 82.86%) and higher than other concentration groups. However, this results of the analysis between BCB− oocytes was also higher in the 13 μM BCB group (n = 28/91; 30.78%) when compared with BCB− COCs exposed to 20 μM (n = 3/62; 4.84%) or 26 μM (n = 3/61; 4.92%) BCB. The nuclear maturation rate in the 13 μM BCB group was similar between BCB+ or BCB− oocytes. The 20 μM BCB group had a lower rate of nuclear maturation of BCB− oocytes than other groups. Thus, our best results in the selection of Rattus norvegicus oocytes by staining with BCB were obtained using the concentration of 13 μM and 20 μM, and an incubation period of 60 min.

The theory of algebraic module specifications and modular systems was developed initially mainly on the basis of equational algebraic specifications. We show that it is in fact almost independent of what kind of underlying specification framework is chosen. More specifically, we present a formulation where this framework appears as an indexed category or, equivalently, specification frame. The ensuing theory is called the theory of abstract module specifications. We are able to prove main results concerning the correctness and compositionality of abstract module specifications in a purely categorical way, assuming the existence of pushouts of morphisms between abstract specifications that allow model amalgamation, functor extension and/or suitable free constructions. Then, by instantiating the theory of abstract module specifications to the behaviour specification frame in the sense of Nivela and Orejas, we obtain a theory of behaviour module specifications.
Detection of prothymosin alpha in oocytes and embryos of Bufo arenarum

Prothymosin alpha (PTA) was detected by immunocytological and biochemical methods in oocytes at different stages of oogenesis, and in early embryos of the amphibian Bufo arenarum. In all cases PTA was detected in the nucleus and was absent from the cytoplasm. This indicates that this protein could act at the level of regulating transcription. Western blots were carried out using polyclonal antibodies with extracts of embryos at different stages of development from early fertilisation up to neural tube. With this method PTA was detected in all the samples under study.

The aim of this paper is to present an enumerative survey of abuse of heroin and methadone by persons of 30 years of age and under, known to have resided at some time within the Oxford City boundaries during the year 1969.
