As the main global terrestrial ecosystem component, grasslands are extremely sensitive to global climate change. With increasing human activities over the last century, grassland ecosystems have been degraded to different degrees. However, the evolution of lake-grassland ecosystems in recent centuries remains unclear due to the dearth of high-resolution records. Here, we present high-resolution lacustrine sediment grain size, pollen (Artemisia, Myriophyllum), Pediastrum, and n-alkane records from Ganggeng Nur Lake to investigate vegetation, lake evolution, and human effects in semiarid northern China. Four stages were identified from the last ca. 150 years: (1) the natural evolution stage (AD 1870–1945), in which there was a wet climate around Ganggeng Nur and the lake level rose from increased runoff; (2) the human disturbance stage (AD 1945–1967), in which the regional climate got drier and human activities began having a detectable effect on the grassland ecosystem; (3) the human transformation stage (AD 1967–2005), in which a completely arid climate coupled with the implementation of a series of land reclamation policies resulted in a large reduction in grassland areas, extensive soil erosion, exacerbated climate change, and shrinking lake areas; and (4) the posttreatment stage (AD 2005–2018), in which soil erosion was alleviated by policy implementation and a favorable humid climate.

The association between time to positivity (TTP) of blood culture and the clinical prognosis of patients with Klebsiella pneumoniae bloodstream infection (BSI) remains unclear. A retrospective study of 148 inpatients with BSI caused by K. pneumoniae was performed at Shanghai Tongji Hospital, China, from October 2016–2020. The total in-hospital fatality rate was 32%. The median TTP was 11.0 (7.7–16.1) h and the optimal cutoff for prediction of in-hospital mortality was 9.4 h according to the ROC curve. Early TTP (<9.4 h) was a risk factor for in-hospital mortality by univariate analysis (OR = 2.5, 95% CI 1.2–5.0, P = 0.01), but not by multivariate analysis (OR = 2.7, 95% CI 1.0–7.4, P = 0.06). Old age, serum creatinine, white blood cells, and C-reactive protein values were risk factors for in-hospital mortality by multivariate analysis. Early TTP was not a risk factor for septic shock (OR = 1.8, 95% CI 0.6–5.1, P = 0.27) or ICU admission (OR = 1.0, 95% CI 1.0–1.0, P = 0.32). In conclusion, the in-hospital fatality rate of patients with K. pneumoniae BSI was relatively high and associated with an early TTP of blood cultures. However, no increased risk of mortality, septic shock or ICU admission was evident in early TTP patients.

The poultry red mite (PRM), Dermanyssus gallinae, is one of the most detrimental ectoparasite on poultry farms worldwide. The blood fed on birds provides the mites with nutrition and energy for their activities, development and reproduction. In the evaluation of the efficacy of novel drugs or vaccines against PRMs, their effects on blood digestion are generally used as a key parameter. The blood digestion of haematophagous arthropods (including D. gallinae) is usually assessed by weighing; however, this method shows some limitations. The main objective of the present study was to develop a scoring method that can quickly and visually evaluate the blood digestion status of PRMs. A 0–4 point scoring criterion was established to describe the blood digestion status of D. gallinae based on the changes in appearance in the intestinal tract of PRMs during the blood digestion process. There was a good consistency between the results obtained by the blood digestion scoring and the weighing, indicating the reliability of this new method. The results obtained from volunteers were consistent with the results from researchers with low coefficient of variation, indicating that the scoring method has good practicability. The applicability of the scoring method was confirmed in an efficacy study, where it was found that doramectin could significantly inhibit the blood digestion of PRMs, lowering the blood digestion score.

The dynamic mechanical force transmitted through microenvironments during tissue formation and regeneration continuously impacts the mechanics of cells and thereby regulates gene and protein expression. The mechanical properties are altered during the process of stem cells differentiating into different lineages. At different stages of differentiation, stem cells display different mechanical properties in response to surrounding microenvironments, which depend on the subcellular structures, especially the cytoskeleton and nucleus. The mechanical properties of the cell nucleus affect protein folding and transport as well as the condensation of chromatin, through which the cell fate is regulated. These findings raise the question as to how cell mechanics change during differentiation. In this study, the mechanical properties of human bone marrow mesenchymal stem cells (hBMSCs) were determined during adipogenic and osteogenic differentiation by atomic force microscopy (AFM). The cytoskeletal structure and the modification of histone were investigated using laser confocal microscope and flow cytometry. The mechanical properties of cell nuclei at different stages of cell differentiation were compared. The stiffness of nuclei increased with time as osteogenesis was induced in hBMSCs. The H3K27me3 level increased during osteogenesis and adipogenesis according to flow cytometry analysis. Our results show conclusively that AFM is a facile and effective method to monitor stem cell differentiation. The measurement of cell mechanical properties by AFM improves our understanding on the connection between mechanics and stem cell fate.

Wildfires are sensitive to climate change, but their response to changes in temperature and precipitation on long timescales is still disputed. In this study, we present a ~9.4 ka black carbon mass sedimentation rate (BCMSR) record from Lake Ximenglongtan (XMLT), southwestern China, to elucidate the Holocene fire regime and its linkages to climatic conditions. The results indicate that the regional fire activity was low during the early Holocene (before 7.6 cal ka BP), increased notably at 7.6 cal ka BP, and continued to increase gradually during the mid- to late Holocene until 2.2 ka. The episodes of higher fire occurrence reflected by higher BCMSR over the last 2.2 ka might be more likely related to the intensified human activities. The cool and humid climate during the early Holocene limited the spread of fire, while warming and drying at ~7.6 cal ka BP triggered higher fire occurrence. Instead of temperature, changes in precipitation dominated fire regime variation during the mid- to late Holocene. On millennial timescales, we suggest that Holocene fire variability has been predominantly controlled by the combined effects of Northern Hemisphere (NH) summer and winter insolation that influenced monsoonal precipitation and fire season temperature, respectively. Indian Ocean Dipole (IOD) events may also have affected fire incidence through influencing monsoon intensity.

A 328.58 m drill core (XK12) was recovered from lacustrine–alluvial sediments in the Xingkai Basin, northeast China, with the aim of obtaining a high-resolution pollen record of East Asian winter monsoon (EAWM) evolution since 3.6 Ma. An index based on the pollen record of thermophilous trees and terrestrial herbs is used as an indicator of winter temperature conditions controlled by the EAWM, at the glacial–interglacial scale. Primary age control was established based on lithostratigraphy and magnetostratigraphy, and then the pollen index was correlated to the LR04 global benthic δ18O record and finally tuned to Earth orbital obliquity to produce a high-resolution astronomical time scale. The pollen record indicates that the EAWM underwent two stepwise enhancements at 2.8 and 1.6 Ma. These events are consistent with paleoclimatic records of mean quartz grain size from the Chinese Loess Plateau, and they are also in accord with the initiation and intensification of Northern Hemisphere glaciation. Our findings suggest that the variability of the EAWM since 3.6 Ma was primarily controlled by changes in global ice volume and climatic cooling.

Altered gut microbial ecology contributes to the development of metabolic diseases including obesity. However, studies based on different populations have generated conflicting results due to diet, environment, methodologies, etc. The aim of our study was to explore the association between gut microbiota and BMI in Chinese college students. The 16S next-generation sequencing (NGS) was used to test the gut microbiota of nine lean, nine overweight/obesity and ten normal-weight male college students. The differences in gut microbiota distribution among three groups were compared, and the relationship between the richness, diversity, composition of gut microbiota and BMI were analysed. The predominant phyla Bacteroidetes and Firmicutes were further confirmed by real-time PCR. Metagenomic biomarker discovery was conducted by linear discriminant analysis (LDA) effect size (LEfSe). NGS revealed that gut microbiota composition was different among three groups, but there was no difference in the abundance ratio of Firmicutes:Bacteroidetes. Several bacterial taxa were in linear relationship with BMI (positive relationship: uncultured bacterium (Bacteroides genus); negative relationship: Porphyromonadaceae, Acidaminococcaceae, Rikenellaceae, Desulfovibrionaceae, Blautia, Anaerotruncus, Parabacteroides, Alistipes). Moreover, gut microbiota diversity decreased with the increase in BMI. And LEfSe analysis indicated that Blautia, Anaerotruncus and its uncultured species were significantly enriched in the lean group (LDA score ≥ 3), Parasuterella and its uncultured species were significantly enriched in the overweight/obese groups (LDA score ≥ 3). In general, gut microbiota composition and microbial diversity were associated with BMI in Chinese male college students. Our results might enrich the understanding between gut microbiota and obesity.

A 336-cm-long sediment core spanning the last 130 ka was recovered from Lake Xingkai on the northeastern margin of the East Asian summer monsoon domain to reveal the linkage between lacustrine depositional processes and environmental changes. Bayesian end member modeling analysis was conducted to partition and interpret the grain-size distributions of Lake Xingkai sediments. Our results suggest that the sedimentary system is characterized by three end members (EMs). EM1 and EM2, with a modal value of 13 and 10 μm, respectively, indicate the variation of local hydraulic conditions. EM3, with a modal value of 5 μm, reflects the background atmospheric dust loading. High atmospheric dust concentration generally occurred during Marine Isotope Stage (MIS) 5d, MIS 4, and early MIS 3, when the climate in the Asian dust source region was cold and dry. In contrast, low dust concentration prevailed during MIS 2, likely due to the southward shift of the westerlies driven by maximum ice volume in the high latitudes.

Stable carbon isotope (δ13C) values of organic matter in lacustrine sediments are commonly used to trace past changes in terrestrial and aquatic carbon cycles. Here we use a high-resolution, well-dated δ13C record from Lake Tengchongqinghai (TCQH) in southwestern China, together with other proxy indices, to reconstruct the paleolimnological history over the past 18.5 ka. Organic matter in the sediments of Lake TCQH is derived predominately from aquatic macrophytes. The lacustrine primary productivity is closely linked with lake-level changes affected by variations in the strength of the Asian summer monsoon and modified by evapotranspiration. Similar to lake sediments world-wide, a ca. − 3‰ shift occurred in the δ13C values of Lake TCQH in response to the significant increase in atmospheric CO2 concentration during the last deglaciation. In the Holocene, the availability of dissolved CO2 in the lake water of Lake TCQH was determined by variations in hydraulic energy: low water turbulence creates a thick, stagnant boundary layer around aquatic plants, which will restrict the rate of CO2 diffusion and result in more positive δ13C values of aquatic plants. In contrast, significant water turbulence dramatically reduces the boundary layer thickness leading to more negative δ13C values of aquatic plants.

Acute phase response (APR) in developing embryos/larvae remains poorly studied in fish, and information regarding the expression and role of acute phase proteins (APPs) in developing embryos/larvae is rather limited in teleosts. In this study we investigated the APR in zebrafish (Danio rerio) embryos/larvae challenged with LPS by examining the expression of APP genes encoding CRP, SAA, LECT2, HAMP and HP and APP inducer genes encoding IL-1β and TNF-α. Expression of all the seven genes was up-regulated in embryos/larvae after challenge with LPS as revealed by q-RT-PCR. Whole-mount in situ hybridization (WISH) showed that all the genes displayed strong maternal expression in the cleavage and blastula stages, and ubiquitous expression in the gastrula and segmentation stage embryos, and then they were expressed differently in specific tissues in later developmental stages. Interestingly, challenge with LPS resulted in de novo expression of the SAA gene in the yolk sac and intestine, the LECT2 gene in the yolk sac, and the IL-1β gene in the yolk sac and the dispersed neutrophils of caudal vein, and visible enhanced expression of the LECT2 gene in the yolk sac. These are the first such data reported in teleosts, showing that LPS challenge is able to modify the expression patterns of APP and APP inducer genes. Altogether, these data suggest that zebrafish embryos/larvae are able to respond to acute infection though their immune system remains in a developing and immature state.

The paper is concerned with the unconditional stability and error estimates of fully discrete Galerkin-Galerkin FEMs for the equations of incompressible miscible flows in porous media. We prove that the optimal L2 error estimates hold without any time-step (convergence) conditions, while all previous works require certain time-step restrictions. Theoretical analysis is based on a splitting of the error into two parts: the error from the time discretization of the PDEs and the error from the finite element discretization of the corresponding time-discrete PDEs, which was proposed in our previous work [26, 27]. Numerical results for both two and three-dimensional flow models are presented to confirm our theoretical analysis.

The paper is concerned with heat and sweat transport in porous textile media with a non-local thermal radiation and phase change. The model, based on a combination of these classical heat transfer mechanisms (convection, conduction and radiation), is governed by a nonlinear, degenerate and strongly coupled parabolic system. The thermal radiative flow is described by a radiation transport equation and characterized by the thermal absorptivity and emissivity of fibre. A conservative boundary condition is introduced to describe the radiative heat flux interacting with environment. With the conservative boundary condition, we prove the global existence of positive/non-negative weak solutions of a nonlinear parabolic system. A typical clothing assembly with a polyester batting material sandwiched in two laminated covers is investigated numerically. Numerical results show that the contribution of radiative heat transfer is comparable with that of conduction/convection in the sweating system.

The combination of density functional theory in local density approximation and dynamical mean field theory (LDA+DMFT) was employed in a preliminary study of the strong electron correlation effects in a promising nuclear fuel—uranium mononitride (UN). For the ferromagnetic phase, the effective impurity problem arising in the LDA+DMFT [1-3] cycle is solved with the spin-polarized T-matrix fluctuation exchange (SPTF) solver, which includes spin–orbit interactions. Concerning the paramagnetic phase, the disordered local moment (DLM) approach was used, based on both standard local density approximation (LDA) and LDA+U. Basic spectral properties and material properties, such as the spin, orbital and total magnetic moments on U atom were calculated for various values of the Hubbard parameter U with a fixed exchange parameter J. Our main focus was to compare the calculated spectral functions (density of states) for different magnetic phases and different methods to the experimental XPS data [4]. On top of that, the total moments of the paramagnetic and ferromagnetic phases are compared with the measured values by neutron spectroscopy [4, 5].

We present a new composite quadrature rule which is exact for polynomials of degree 2N + K – 1 with N abscissas at each subinterval and K boundary conditions. The corresponding orthogonal polynomials are introduced and the analytic formulae for abscissas and weight functions are presented. Numerical results show that the new quadrature rule is more efficient, compared with classical ones.

Allelic expression of the rice yield-related gene, leucine-rich receptor-like kinase 6 (LRK6), in the hybrid of 93-11 (Oryza sativa L. subsp. Indica var. 93-11) and Nipponbare (O. sativa L. subsp. Japonica var. Nipponbare) is determined by allelic promoter cis-elements. Using deletion analysis of the LRK6 promoter, we identified two distinct regions that might contribute to LRK6 expression. Sequence alignment revealed differences in these LRK6 promoter regions in 93-11 and Nipponbare. One of the segments, named differential sequence of LRK6 promoter 2 (DSLP2), contains potential transcription factor binding sites. Using a yeast one-hybrid assay, we isolated an ethylene-responsive factor (ERF) protein that binds to DSLP2. Sequence analysis and a GCC-box assay showed that the ERF gene, O. sativa ERF 3 (OsERF3), which belongs to ERF subfamily class II, has a conserved ERF domain and an ERF-associated amphiphilic repression repressor motif. We used an in vivo mutation assay to identify a new motif (5′-TAA(A)GT-3′) located in DSLP2, which interacts with OsERF3. These results suggest that OsERF3, an AP2 (APETALA 2 Gene)/ERF transcription factor, binds the LRK6 promoter at this new motif, which might cause differential expression of LRK6 in the 93-11/Nipponbare hybrid.
