Biological activity of the non-microbial fraction of kefir: antagonism against intestinal pathogens

Kefir is a fermented milk obtained by the activity of kefir grains which are composed of lactic and acetic acid bacteria, and yeasts. Many beneficial health effects have been associated with kefir consumption such as stimulation of the immune system and inhibition of pathogenic microorganisms. The biological activity of kefir may be attributed to the presence of a complex microbiota as well as the microbial metabolites that are released during fermentation. The aim of this work was to characterise the non-microbial fraction of kefir and to study its antagonism against Escherichia coli, Salmonella spp. and Bacillus cereus. During milk fermentation there was a production of organic acids, mainly lactic and acetic acid, with a consequent decrease in pH and lactose content. The non-microbial fraction of kefir added to nutrient broth at concentrations above 75% v/v induced a complete inhibition of pathogenic growth that could be ascribed to the presence of un-dissociated lactic acid. In vitro assays using an intestinal epithelial cell model indicated that pre-incubation of cells with the non-microbial fraction of kefir did not modify the association/invasion of Salmonella whereas pre-incubation of Salmonella with this fraction under conditions that did not affect their viability significantly decreased the pathogen's ability to invade epithelial cells. Lactate exerted a protective effect against Salmonella in a mouse model, demonstrating the relevance of metabolites present in the non-microbial fraction of kefir produced during milk fermentation.
Persistence of Dinitroaniline Herbicides in Soil

An oat (Avena sativa L. ‘Dal’) primary root bioassay procedure was used to study the persistence of eight dinitroaniline herbicides in soil. Sensitivity of the bioassay to the herbicides depended on the length of time roots were exposed to the herbicide treatments. Root inhibition increased with time of exposure. Herbicide persistence was studied under growth chamber, greenhouse, and field conditions. The herbicides remained biologically active the longest under dry and cool soil conditions. At 5 C, little change in herbicide activity was observed over an 8-month period. Under greenhouse conditions, the herbicides in wet soil dissipated rapidly, as compared with those in dry soil. Soil samples taken from depths of 0 to 7.5 cm and from 7.5 to 15 cm in the field showed that the herbicides were retained primarily in the surface 7.5 cm. Small amounts were found from 7.5 to 15 cm only shortly after the herbicides were applied. Dinitramine (N4,N4-diethyl-α,α,α-trifluoro-3,5-dinitrotoluene-2,4-diamine) was the only herbicide tested of which soil residues from spring field applications were not detected the following fall.

Effects of soil water and herbicide vapors on the phytotoxicity of eight dinitroaniline herbicides to oats (Avena sativa L. ‘Dal’) were studied. Oat primary root length was inhibited more by the herbicides than was shoot length or shoot fresh weight. All of the herbicides, except oryzalin (3,5-dinitro-N4,N4-dipropylsulfanilamide), inhibited primary root length through vapor activity. Vapors of dinitramine (N4,N4-diethyl-α,α,α-trifluoro-3,5-dinitrotoluene-2,4-diamine) were most inhibitory. Herbicide vapor inhibition increased with temperature. Soil water affected oryzalin activity more than it did that of the other herbicides. Oryzalin phytotoxicity to the oat primary root was reduced more at the low soil water than was that of the other herbicides. Its low vapor activity apparently reduced its effectiveness in dry soil. Difference in magnitude of herbicide phytotoxicity between two soil moisture levels generally was more pronounced at lower herbicide concentration.

Adsorption of benefin (N-butyl-N-ethyl-α,α,α-trifluoro-2,6-dinitro-p-toluidine), dinitramine (N4,N4-diethyl-α,α,α-trifluoro-3,5-dinitrotoluene-2,4-diamine), fluchloralin [N-(2-chloroethyl)-2,6-dinitro-N-propyl-4-(trifluoromethyl)aniline], oryzalin (3,5-dinitro-N4,N4-dipropylsulfanilamide), profluralin [N-(cyclopropylmethyl)-α,α,α-tri-fluoro-2,6-dinitro-N-propyl-p-toluidine], and trifluralin (α,α,α-trifluoro-2,6-dinitro-N,N-dipropyl-p-toluidine) was studied in 10 Wisconsin soils. Ratios of the quantity of each herbicide adsorbed and quantities remaining in the soil solution at equilibrium (Kd value) on a Piano silt loam (Typic Argiudoll fine-silty, mixed, mesic) remained relatively constant over a range of concentrations. Herbicide adsorption by the soils was related more closely to soil organic matter than to the other soil chemical and physical properties. Diffusion of the herbicides in Piano silt loam was affected by soil water. Diffusion of trifluralin, profluralin and benefin decreased as soil water increased. Diffusion of dinitramine and fluchloralin did not change significantly with change in water content. Diffusion of oryzalin increased at the highest soil water content. None of the herbicides moved more than 10 mm in the soil during a 17-day period. In unsaturated Piano silt loam, relative mobility of the herbicides was trifluralin ≥benefin>profluralin>fluchloralin>dinitramine≥oryzalin. Oryzalin reached highest mobility in water-saturated soil.

Roots or shoots of oats (Avena sativa L. ‘Dal’) and peas (Pisum sativum L. ‘Early Perfection’) were exposed to vapors of 14C-labeled benefin (N-butyl-N-ethyl-α,α,α-trifluoro-2,6-dinitro-p-toluidine), dinitramine (N4,N4-diethyl-α,α,α-trifluoro-3,5-dinitrotoluene-2,4-diamine), fluchloralin [N-(2-chloroethyl)-2,6 dinitro-N-propyl-4-(trifluoromethyl)aniline], oryzalin (3,5-dinitro-N4,N4-dipropylsulfanilamide), profluralin [N-(cyclopropylmethyl)-α,α,α-trifluoro-2,6-dinitro-N-propyl-p-toluidine], and trifluralin (α,α,α-trifluoro-2,6-dinitro-N,N-dipropyl-p-toluidine). All of the dinitroaniline herbicides except oryzalin were absorbed by roots and shoots of germinating oats and peas. Some root-shoot translocation of the herbicides was observed in peas, but no shoot-root transport could be detected in either peas or oats. In peas, 14C from root-absorbed benefin, fluchloralin, profluralin and trifluralin was detected in shoots, and 14C from benefin and trifluralin was detected in cotyledons. In general, vapor absorption was correlated with rates of herbicide volatilization.

Glutamine may be a precursor for NO synthesis, which may play a crucial role in bacterial translocation (BT). The goal of the present study was to investigate the potential effects of glutamine on BT and the immunological response in an experimental model of NO synthase inhibition by NG-nitro-L-arginine methyl ester (l-NAME). Mice were randomly assigned to four groups: sham; intestinal obstruction (IO); IO+500 mg/kg per d glutamine (GLN); IO+GLN plus 10 mg/kg per d l-NAME (GLN/LN). The groups were pretreated for 7 d. BT was induced by ileal ligation and was assessed 18 h later by measuring the radioactivity of 99mTc-Escherichiacoli in the blood and organs. Mucosal damage was determined using a histological analysis. Intestinal permeability (IP) was assessed by measuring the levels of 99mTc-diethylenetriaminepentaacetic acid in the blood at 4, 8 and 18 h after surgery. IgA and cytokine concentrations were determined by ELISA in the intestinal fluid and plasma, respectively. BT was increased in the GLN/LN and IO groups than in the GLN and sham groups. IP and intestinal mucosa structure of the sham, GLN and GLN/LN groups were similar. The GLN group had the highest levels of interferon-γ, while IL-10 and secretory IgA levels were higher than those of the IO group but similar to those of the GLN/LN group. The present results suggest that effects of the glutamine pathway on BT were mediated by NO. The latter also interferes with the pro-inflammatory systemic immunological response. On the other hand, IP integrity preserved by the use of glutamine is independent of NO.

Caves and rockshelters are a key component of the archaeological record but are often regarded as natural places conveniently exploited by human communities. Archaeomorphological study shows however that they are not inert spaces but have frequently been modified by human action, sometimes in ways that imply a strong symbolic significance. In this paper the concept of ‘aménagement’, the re-shaping of a material space or of elements within it, is applied to Chauvet Cave in France and Nawarla Gabarnmang rockshelter in Australia. Deep within Chauvet Cave, fallen blocks were moved into position to augment the natural structure known as The Cactus, while at Nawarla Gabarnmang, blocks were removed from the ceiling and supporting pillars removed and discarded down the talus slope. These are hence not ‘natural’ places, but modified and socially constructed.
Fall determinants in older long-term care residents with dementia: a systematic review

Background: Persons with dementia are two to three times more likely to fall compared to persons without dementia. In long-term care settings, the dementia prevalence is highest. Therefore, older long-term care residents with dementia can be considered a high-risk group for falls. Nevertheless, no systematic evaluation of fall determinants in this population was found. The purpose of this study was to identify fall determinants among older long-term care residents with dementia or cognitively impaired persons in long-term care, by conducting a systematic literature review.
Methods: We searched English, French, Dutch, and German articles listed in: CINAHL, Cochrane Library, EMBASE, MEDLINE, PsycINFO, PubMed, and Web of Science. Additionally, references of included articles were screened. Studies were included if determinants or circumstances of falls in older persons with dementia living in long-term care were assessed.
Results: Eight studies met the inclusion criteria. Three studies were excluded from detailed analysis because of insufficient quality. Use of psychotropic drugs, a “fair or poor” general health, gait impairments, and age were associated with an increased fall risk. Also trunk restraints were associated with an increased number of falls while full bedrails and wandering behavior were protective against falls.
Conclusions: Fall risk factors known from other populations, e.g. use of psychotropic drugs, physical restraints, and health conditions, are found in long-term care residents with dementia as well. Due to the limited evidence available, future studies with adequate sample sizes and prospective designs are required to determine specific fall risk factors and verify existing results in this population.

Since publication of the Society for Healthcare Epidemiology of America position paper on Clostridium difficile infection in 1995, significant changes have occurred in the epidemiology and treatment of this infection. C. difficile remains the most important cause of healthcare-associated diarrhea and is increasingly important as a community pathogen. A more virulent strain of C. difficile has been identified and has been responsible for more-severe cases of disease worldwide. Data reporting the decreased effectiveness of metronidazole in the treatment of severe disease have been published. Despite the increasing quantity of data available, areas of controversy still exist. This guideline updates recommendations regarding epidemiology, diagnosis, treatment, and infection control and environmental management.

This study has determined the response of a muscle strip preparation from Fasciola hepatica to 5-HT and a range of agonists that distinguish between the vertebrate receptor 5-HT subtypes. 5-HT increased the resting tone and the rhythmic activity of the muscle strip. Of the 19 compounds tested, only 10 had an effect similar to 5-HT and all but 2 of these were tryptamine compounds. 5-HT was more potent than tryptamine whilst 4-OH-tryptamine had no effect, suggesting that the response is mediated by a 5-HT rather than a tryptamine receptor. 5-Fluorotryptamine and 5-carboxyamidotryptamine were the most potent agonists. 8-OH-DPAT also mimicked the effect of 5-HT, though less potently. Assuming that these agents elicit their response through a common receptor, this suggests the presence of a 5-HT receptor with similar properties in terms of agonist recognition as the vertebrate 5-HT1 class of receptor involved in controlling Fasciola muscle motility.

This paper determines the asymptotic solution of certain initial-boundary value problems for singularly-perturbed reaction-diffusion equations, including the Allen–Cahn and Cahn–Hilliard equations, on bounded one-dimensional spatial domains for r[ges ]0. Attention is focused on the metastable evolution of a transition layer over an asymptotically exponentially-long time interval.
