Ageing modifies acute resting blood pressure responses to incremental consumption of dietary nitrate: a randomised, cross-over clinical trial

Beetroot (BR) is a rich source of nitrate (NO3-) that has been shown to reduce blood pressure (BP). Yet, no studies have examined the vascular benefits of BR in whole-food form and whether the effects are modified by age. This study was a four-arm, randomised, open-label, cross-over design in twenty-four healthy adults (young n 12, age 27 ± 4 years, old n 12, age 64 ± 5 years). Participants consumed whole-cooked BR at portions of (NO3- content in brackets) 100 g (272 mg), 200 g (544 mg) and 300 g (816 mg) and a 200-ml solution containing 1000 mg of potassium nitrate (KNO3) on four separate occasions over a 4-week period (≥7-d washout period). BP, plasma NO3- and nitrite (NO2-) concentrations, and post-occlusion reactive hyperaemia via laser Doppler, were measured pre- and up to 5-h post-intervention. Data were analysed by repeated-measures ANOVA. Plasma NO2- concentrations were higher in the young v. old at baseline and post-intervention (P < 0·05). All NO3- interventions decreased systolic and diastolic BP in young participants (P < 0·05), whereas only KNO3 (at 240–300 min post-intake) significantly decreased systolic (–4·8 mmHg, −3·5 %, P = 0·024) and diastolic (–5·4 mmHg, −6·5 %, P = 0·007) BP in older participants. In conclusion, incremental doses of dietary NO3- reduced systolic and diastolic BP in healthy young adults whereas in the older group a significant decrease was only observed with the highest dose. The lower plasma NO2- concentrations in older participants suggest that there may be mechanistic differences in the production of NO from dietary NO3- in young and older populations.
HPPD-resistant cotton response to isoxaflutole applied preemergence and postemergence

Studies were conducted in 2019 and 2020 in Lewiston, NC, to determine the crop response of 4-hydroxyphenylpyrivate dioxygenase (HPPD)-resistant cotton to isoxaflutole (IFT) and other cotton herbicides as part of a cotton weed management program that included herbicides applied preemergence, early postemergence (EPOST), and mid-postemergence (MPOST). IFT was applied PRE at 105 g ha−1 alone and in various combinations with acetochlor, diuron, fluometuron, fluridone, fomesafen, pendimethalin, and pyrithiobac. EPOST treatments included IFT at 53 or 105 g ha−1 alone or in combination with glyphosate or glufosinate, or dimethenamid-P + glufosinate. Glyphosate + glufosinate was applied MPOST to all treatments except the nontreated control. Cotton injury from IFT applied PRE was minimal (0% to 3%). Injury following EPOST application of dimethenamid-P + glufosinate ranged from 3% to 5% and 6% to 9% in 2019 and 2020, respectively. In both years, injury from IFT applied PRE followed by IFT applied EPOST never exceeded injury from IFT applied PRE followed by dimethenamid-P + glufosinate. Isoxaflutole applied PRE followed by IFT applied EPOST at 105 g ha−1 resulted in 0% to 2% cotton injury, indicating that IFT can be applied either PRE or EPOST with minimal risk to cotton. Late-season cotton height and cotton lint yield were not affected by any herbicide treatment. The experimental HPPD-resistant cotton cultivar was minimally injured by IFT applied PRE and EPOST, it tolerated standard cotton herbicides, and yield loss was not observed. Given these results, HPPD-resistant cotton and IFT may be integrated into cotton weed management systems with minimal risk for cotton injury and provide an additional effective mechanism of action for managing troublesome weeds in cotton.

Aims
To conduct a meta-analysis of the effect of ketamine on psychopathology in healthy volunteers and patients with schizophrenia, and the experimental factors affecting this.
BackgroundKetamine is increasingly used to treat depression and other psychiatric disorders but can induce schizophrenia-like symptoms. Despite this, the consistency and magnitude of symptoms induced by ketamine, or what factors influence the effects of ketamine on these remain unknown.
MethodMEDLINE, EMBASE and PsychINFO databases were searched for within-subject placebo controlled studies reporting symptoms using the Brief Psychiatric Rating Scale (BPRS) or Positive and Negative Syndrome Scale (PANSS) in response to an acute ketamine challenge in healthy participants or people with schizophrenia. Two independent investigators extracted study-level data for a random-effects meta-analysis. Total, positive and negative BPRS and PANSS scores were extracted. Sub-group analyses were conducted examining the effect of: blinding status, ketamine preparation, infusion method and time between ketamine and placebo condition. Standardized mean change scores were used as effect sizes for individual studies. Standardized mean changes between ketamine and placebo for total, positive and negative BPRS and PANSS were calculated.
ResultOf 7819 citations retrieved, 36 studies involving healthy participants were included. The overall sample included 725 healthy volunteers exposed to both the ketamine and placebo condition. Ketamine induced a significant increase in transient psychopathology in healthy participants, for total (Standardized mean change (SMC) = 1.50 (95% CI = 1.23 to 1.77), p < 0.0001), positive (SMC = 1.55 (95% CI = 1.29 to 1.81), p < 0.0001) and negative (SMC = 1.16, (95% CI = 0.96 to 1.35), p < 0.0001) symptom ratings, relative to the placebo condition. This effect was significantly greater for positive symptoms than negative symptoms (p = 0.004). Bolus followed by constant infusion increased ketamine's effect on positive symptoms relative to infusion alone (p = 0.006). Single-day study design increased ketamine's effect on total symptoms (p = 0.007), but age and gender did not moderate effects. There were insufficient studies for meta-analysis of studies in schizophrenia. Of these studies, two found a significant increase in symptoms with ketamine administration in total and positive symptoms. Only one study found an increase in negative symptom severity with ketamine.
ConclusionThese findings show that acute ketamine administration induces schizophrenia-like symptomatology with large effect sizes but there is a greater increase in positive than negative symptoms, and when a bolus is used. These findings suggest bolus doses should be avoided in its therapeutic use to minimize the risk of inducing transient positive psychotic symptoms.

In their 1960 book on finite Markov chains, Kemeny and Snell established that a certain sum is invariant. The value of this sum has become known as Kemeny’s constant. Various proofs have been given over time, some more technical than others. We give here a very simple physical justification, which extends without a hitch to continuous-time Markov chains on a finite state space. For Markov chains with denumerably infinite state space, the constant may be infinite and even if it is finite, there is no guarantee that the physical argument will hold. We show that the physical interpretation does go through for the special case of a birth-and-death process with a finite value of Kemeny’s constant.

The polyplacophoran Matthevia Walcott, 1885 has been reported previously only from Cambrian rocks. Recovery of specimens of two different species of Matthevia from the top of the cherty lower Gasconade Dolomite in Missouri extends the known range of the genus into the Ordovician. The presence of the Lower Ordovician trilobite Praepatokephalus Lochman, 1964 in one collection provides the biostratigraphic evidence that the new specimens of Matthevia are Ordovician in age. Matthevia erecta n. sp. is described. An emended diagnosis for the trilobite Praepatokephalus is provided; two species of the genus are briefly described but are left in open nomenclature as P. sp. A and P. sp. B.
Disability but not social support predicts cognitive deterioration in late-life depression

Background:
Depression in late life is a risk factor for cognitive decline. Depression is also associated with increased disability and social support deficits; these may precede conversion to dementia and inform risk. In this study, we examined if baseline or one-year change in disability and social support predicted later cognitive deterioration.
Methods:299 cognitively intact depressed older adults were followed for an average of approximately seven years. Participants received antidepressant treatment according to a standardized algorithm. Neuropsychological testing and assessment of disability and social support were assessed annually. Cognitive diagnosis was reviewed annually at a consensus conference to determine if participants remained cognitively normal, or if they progressed to either dementia or cognitively impaired, no dementia (CIND).
Results:During study participation, 167 individuals remained cognitively normal (56%), 83 progressed to CIND (28%), and 49 progressed to dementia (16%). Greater baseline instrumental activities of daily living (IADL) deficits predicted subsequent conversion to a cognitive diagnosis (CIND or dementia). However, neither baseline measures nor one-year change in basic ADLs (BADLs) and social support predicted cognitive conversion. In post hoc analyses, two IADL measures (managing finances, preparing meals) significantly increased the odds of cognitive conversion.
Conclusions:Greater IADL deficits predicted increased risk of cognitive conversion. Assessment of IADL deficits may provide clues about risk of later cognitive decline.

A Qualitative Perspective on Multiple Health Behaviour Change: Views of Smoking Cessation Advisors Who Promote Physical Activity

There are mixed views on whether smoking cessation advisors should focus only on quitting smoking or also promote simultaneous health behaviour changes (e.g., diet, physical activity), but no studies have qualitatively examined the views and vicarious experiences of such health professionals. Semi-structured interviews were conducted with 11 trained smoking cessation advisors who promote physical activity to their clients. The data were categorised into themes using thematic analysis supported by qualitative data analysis software. We report themes that were related to why advisors promote multiple health behaviour change and issues in timing. Physical activity could be promoted as a cessation aid and also as part of a holistic lifestyle change consistent with a nonsmoker identity, thereby increasing feelings of control and addressing fear of weight gain. Multiple changes were promoted pre-quit, simultaneously and post-quit, and advisors asserted that it is important to focus on the needs and capabilities of individual clients when deciding how to time multiple changes. Also, suggesting that PA was a useful and easily performed cessation aid rather than a new behaviour (i.e., structured exercise that may seem irrelevant) may help some clients to avoid a sense of overload.

GI/M/1-type Markov chains make up a class of two-dimensional Markov chains. One dimension is usually called the level, and the other is often called the phase. Transitions from states in level k are restricted to states in levels less than or equal to k+1. For given transition probabilities in the interior of the state space, we show that it is always possible to define the boundary transition probabilities in such a way that the level and phase are independent under the stationary distribution. We motivate our analysis by first considering the quasi-birth-and-death process special case in which transitions from any state are restricted to states in the same, or adjacent, levels.
A High-Resolution Radiocarbon Calibration Between 11,700 and 12,400 Calendar Years Bp Derived from 230Th Ages of Corals from Espiritu Santo Island, Vanuatu

This paper presents radiocarbon results from a single Diploastrea heliopora coral from Vanuatu that lived during the Younger Dryas climatic episode, between ca. 11,700 and 12,400 calendar yr bp. The specimen has been independently dated with multiple 230Th measurements to permit calibration of the 14C time scale. Growth bands in the coral were used to identify individual years of growth. 14C measurements were made on each year. These values were averaged to achieve decadal resolution for the 14C calibration. The relative uncertainty of the decadal 14C data was below 1% (2σ). The data are in good agreement with the existing dendrochronology and allow for high-resolution calibration for most years. Variations in the fine structure of the 14C time series preserved in this specimen demonstrate sporadic rapid increases in the Δ14C content of the surface ocean and atmosphere. Certain sharp rises in Δ14C are coincident with gaps in coral growth evidenced by several hiatuses. These may be related to rapid climatic changes that occurred during the Younger Dryas. This is the first coral calibration with decadal resolution and the only such data set to extend beyond the dendrochronology-based 14C calibration.
