The isotope values of fossil snail shells can be important archives of climate. Here, we present the first carbon (δ13C) and oxygen (δ18O) isotope values of snail shells in interior Alaska to explore changes in vegetation and humidity through the late-glacial period. Snail shell δ13C values were relatively consistent through the late glacial. However, late-glacial shell δ13C values are 2.8‰ higher than those of modern shells. This offset is best explained by the Suess effect and changes in the δ13C values of snail diet. Snail shell δ18O values varied through the late glacial, which can be partially explained by changes in relative humidity (RH). RH during the snail growing period was modeled based on a published flux balance model. Results suggest a dry period toward the beginning of the Bølling–Allerød (~14 ka) followed by two distinct stages of the Younger Dryas, a wetter stage in the early Younger Dryas from 12.9 to 12.3 ka, and subsequent drier stage in the late Younger Dryas between 12.3 and 11.7 ka. The results show that land snail isotopes in high-latitude regions may be used as a supplementary paleoclimate proxy to help clarify complex climate histories, such as those of interior Alaska during the Younger Dryas.

OBJECTIVES/GOALS: The innate immune responses to Multisystem Inflammatory Syndrome in Children (MIS-C) are not fully known. Using samples from MIS-C, we will assess the cellular responses and develop a novel Tri-Specific Killer Engager (TRiKE) that engages innate immune cells to improve those responses. METHODS/STUDY POPULATION: We collected blood samples from 60 pediatric patients from which we isolated plasma and peripheral blood mononuclear cells. We received blood samples from 13 MIS-C, 32 severe acute COVID, 5 COVID-19 asymptomatic, and 15 COVID-19 negative patients. Using plasma, we then performed ELISAs to determine IgG antibody levels against SARS-CoV-2 and plaque reduction neutralization tests to determine neutralizing antibody functions. We isolated DNA to look at Fc receptor genetics. We also utilized utilize flow cytometry assays determine the phagocytosis and killing abilities of the innate cells from these patients. This data will be correlated with clinical outcomes. Additionally, we have developed a novel SARS-CoV-2 TRiKE which directs natural killer (NK) cell killing specifically to of COVID-19 infected cells. RESULTS/ANTICIPATED RESULTS: MIS-C patients had higher IgG antibody titers against SARS-CoV-2 compared to children with symptomatic or asymptomatic COVID. MIS-C patients also neutralized SARS-CoV-2 more effectively than children with acute symptomatic or asymptomatic COVID-19. We found natural killer cells and monocytes are dysfunctional in MIS-C patients and do not kill SARS-CoV-2 infected cells as well. Specifically, NK cells do not kill COVID-19 infected cells as well. To combat this, we have successfully generated and are now testing a Tri-Specific Killer engager (TRiKE) which binds one ends to NK cells, one end to the Spike protein on COVID-19 infected cells and contains IL-15 to improve NK cell function. We anticipate that we can improve NK cell killing of COVID-19 infected cells with this TRiKE. DISCUSSION/SIGNIFICANCE: We found that MIS-C patients have antibodies that can neutralize SARS-CoV-2 but that that innate immune cells that engage antibodies are dysfunctional. We are have successfully developed and are targeting this response with a TRiKE to improve innate immune cell functional; this may serve as an adjunctive therapeutic if proven successful.

Background and Purpose:
The impact of dementia-related stressors and strains have been examined for their potential to threaten the well-being of either the person with dementia or the family care partner, but rarely have studies considered the dyadic nature of well-being in dementia. The purpose of this study was to examine the dyadic effects of multiple dimensions of strain on the well-being of dementia care dyads.
Methods:Using multilevel modeling to account for the inter-relatedness of individual well-being within dementia care dyads, we examined cross-sectional responses collected from 42 dyads comprised of a hospitalized patient diagnosed with a primary progressive dementia (PWD) and their family care partner (CP). Both PWDs and CPs self-reported on their own well-being using measures of quality of life (QOL-Alzheimer’s Disease scale) and depressive symptoms (Center for Epidemiological Studies Depression Scale).
Results:In adjusted models, the PWD’s well-being (higher QOL and lower depressive symptoms) was associated with significantly less strain in the dyad’s relationship. The CP’s well-being was associated with significantly less care-related strain and (for QOL scale) less relationship strain.
Conclusions:Understanding the impact of dementia on the well-being of PWDs or CPs may require an assessment of both members of the dementia care dyad in order to gain a complete picture of how dementia-related stressors and strains impact individual well-being. These results underscore the need to assess and manage dementia-related strain as a multi-dimensional construct that may include strain related to the progression of the disease, strain from providing care, and strain on the dyad’s relationship quality.

The geologic setting of the Ziegler Reservoir fossil site is somewhat unusual — the sediments containing the Pleistocene fossils were deposited in a lake on top of a ridge. The lake basin was formed near Snowmass Village, Colorado (USA) when a glacier flowing down Snowmass Creek Valley became thick enough to overtop a low point in the eastern valley wall and entered the head of Brush Creek Valley. When the glacier retreated at about 155–130 ka, near the end of Marine Oxygen Isotope Stage 6, the Brush Creek Valley lobe left behind a moraine that impounded a small alpine lake. The lake was initially ~ 10 m deep and appears to have been highly productive during most of its existence, based on the abundant and exquisitely preserved organic material present in the sediments. Over time, the basin slowly filled with (mostly) eolian sediment such that by ~ 87 ka it contained a marsh or wetland rather than a true lake. Open-water conditions returned briefly between ~ 77 and 55 ka before the impoundment was finally breached to the east, establishing ties with the Brush Creek drainage system and creating an alpine meadow that persisted until historic times.

Studies of terrestrial biotic and environmental dynamics of Marine Oxygen Isotope Stage (MIS) 5, also called the Last Interglacial Period, provide insight into the effects of long-term climate change on Pleistocene ecosystems. In North America, however, there are relatively few fossil sites that definitively date to MIS 5. Even fewer contain multiple ecosystem components (vertebrates, invertebrates, plants) that have been studied in detail, and none are located at high elevation. Thus, our view of North American ecosystems during MIS 5 is, at best, an incomplete composite view, and alpine ecosystems are entirely undocumented.

In North America, terrestrial records of biodiversity and climate change that span Marine Oxygen Isotope Stage (MIS) 5 are rare. Where found, they provide insight into how the coupling of the ocean–atmosphere system is manifested in biotic and environmental records and how the biosphere responds to climate change. In 2010–2011, construction at Ziegler Reservoir near Snowmass Village, Colorado (USA) revealed a nearly continuous, lacustrine/wetland sedimentary sequence that preserved evidence of past plant communities between ~140 and 55 ka, including all of MIS 5. At an elevation of 2705 m, the Ziegler Reservoir fossil site also contained thousands of well-preserved bones of late Pleistocene megafauna, including mastodons, mammoths, ground sloths, horses, camels, deer, bison, black bear, coyotes, and bighorn sheep. In addition, the site contained more than 26,000 bones from at least 30 species of small animals including salamanders, otters, muskrats, minks, rabbits, beavers, frogs, lizards, snakes, fish, and birds. The combination of macro- and micro-vertebrates, invertebrates, terrestrial and aquatic plant macrofossils, a detailed pollen record, and a robust, directly dated stratigraphic framework shows that high-elevation ecosystems in the Rocky Mountains of Colorado are climatically sensitive and varied dramatically throughout MIS 5.

OBJECTIVE
To determine the clinical diagnoses associated with the National Healthcare Safety Network (NHSN) pneumonia (PNEU) or lower respiratory infection (LRI) surveillance events
DESIGNRetrospective chart review
SETTINGA convenience sample of 8 acute-care hospitals in Pennsylvania
PATIENTSAll patients hospitalized during 2011–2012
METHODSMedical records were reviewed from a random sample of patients reported to the NHSN to have PNEU or LRI, excluding adults with ventilator-associated PNEU. Documented clinical diagnoses corresponding temporally to the PNEU and LRI events were recorded.
RESULTSWe reviewed 250 (30%) of 838 eligible PNEU and LRI events reported to the NHSN; 29 reported events (12%) fulfilled neither PNEU nor LRI case criteria. Differences interpreting radiology reports accounted for most misclassifications. Of 81 PNEU events in adults not on mechanical ventilation, 84% had clinician-diagnosed pneumonia; of these, 25% were attributed to aspiration. Of 43 adult LRI, 88% were in mechanically ventilated patients and 35% had no corresponding clinical diagnosis (infectious or noninfectious) documented at the time of LRI. Of 36 pediatric PNEU events, 72% were ventilator associated, and 70% corresponded to a clinical pneumonia diagnosis. Of 61 pediatric LRI patients, 84% were mechanically ventilated and 21% had no corresponding clinical diagnosis documented.
CONCLUSIONSIn adults not on mechanical ventilation and in children, most NHSN-defined PNEU events corresponded with compatible clinical conditions documented in the medical record. In contrast, NHSN LRI events often did not. As a result, substantial modifications to the LRI definitions were implemented in 2015.

Summary
Introduction
Marine protected areas (MPAs) are increasingly used as tools to conserve biodiversity, manage fishing effort, and facilitate recovery of degraded ecosystems (Roberts et al., 2001; Sale, 2002; Hastings and Botsford, 2003; Cinner et al., 2006). Marine protected areas are expected to produce long-lasting local increases in the density, size, diversity, and/or productivity of marine organisms within MPA boundaries (Russ and Alcala 1996; Claudet et al., 2006; Chapter 2), as well as regional increases outside of the MPA via spillover from the MPA to sites that continue to be fished (Chapter 3). Assessment of the actual effects of MPAs relative to these goals is essential for adaptive management and decision-making. Despite a large number of assessments, however, there remains considerable uncertainty about the actual effects of MPAs (e.g., Osenberg et al., 2006).
The central question (and challenge) that underlies the assessment of any MPA is easy to express but difficult to measure: “How does the state of the system (e.g., density or size of a target organism) within the MPA compare to the state that would have existed had the MPA never been established?” The former can be directly observed; however, the latter cannot and must therefore be estimated through indirect means (Stewart-Oaten et al., 1986; Osenberg and Schmitt, 1996; Stewart-Oaten, 1996a, 1996b; Stewart-Oaten and Bence, 2001; Osenberg et al., 2006). Several different approaches have been proposed to estimate this baseline (Box 6.1; see also Osenberg et al., 2006), yet, considerable controversy still exists (Box 6.2).

Studies of press coverage afforded women running for public office indicate that historically, women tend to garner less coverage overall and that the coverage they do receive tends to focus disproportionately on their appearance, personality, and family status at the expense of their qualifications and issue positions. This study examines newspaper coverage of U.S. Senator Hillary Rodham Clinton's campaign for the 2008 Democratic presidential nomination. Notably, Clinton did not allege that she was receiving too little coverage or coverage that focused disproportionately on her clothing or appearance. Rather, she charged that she was being treated negatively relative to her chief rival, U.S. Senator Barack Obama. More than 6,000 articles from 25 leading newspapers from across the country were content-coded from Labor Day through Super Tuesday in order to assess Clinton's coverage on two dimensions: traditional and tonal. On a range of traditional indicators of bias, such as coverage amount and mentions of candidate appearance, Clinton's coverage clearly broke established patterns typically afforded women presidential candidates. However, the tone of Clinton's coverage was decidedly negative relative to her male competitors. Normative implications of this mixed bag of fairness and bias are discussed.
