Effect of double-premilking teat disinfection protocols on bacterial counts on teat skin of cows and milker gloves in a free-stall-housed dairy herd

This research communication addresses the hypothesis that double premilking teat disinfection (DD) is more effective in reducing soiling and bacterial count on the cows' teat skin and milkers' gloves than conventional disinfection (CONV). The design was a 3 × 3 Latin square (three groups of cows and three treatments) with conventional teat disinfection (CONV, lactic acid application after forestripping), double teat disinfection using the same disinfectant (DD1D, lactic acid application before and after forestripping), and double teat disinfection using two different disinfectants (DD2D, application of lactic acid before and chlorine-based disinfectant after forestripping). All groups were assigned for six days for each treatment, and the evaluations and samples were collected on the last day, before and after treatment. We evaluated the teat cleanliness score (TCS), count of Gram-negative bacteria (coliforms and noncoliforms), Staphylococcus spp., Streptococcus spp., and total bacterial count (TBC) on the cows' teats and TBC on the milkers' gloves. TCS after premilking was lower in DD than CONV treatment. The reduction of Staphylococcus spp. count was greater in the DD treatment and tended to be higher in the DD2D. The TBC reduction on the cows' teats was greater in the DD treatments. The TBC on the milkers' gloves was lower for DD before and after premilking. In conclusion, DD can be an alternative for reducing some bacterial populations on cow teats and preventing the transmission of microorganisms between cows via the milkers' hands.

Background:
The early clinical predictors of respiratory failure in Latin Americans with Guillain–Barré syndrome (GBS) have scarcely been studied. This is of particular importance since Latin America has a high frequency of axonal GBS variants that may imply a worse prognosis.
Methods:We studied 86 Mexican patients with GBS admitted to the Instituto Nacional de Ciencias Médicas y Nutrición Salvador Zubirán, a referral center of Mexico City, to describe predictors of invasive mechanical ventilation (IMV).
Results:The median age was 40 years (interquartile range: 26–53.5), with 60.5% men (male-to-female ratio: 1.53). Most patients (65%) had an infectious antecedent (40.6% gastrointestinal). At admission, 38% of patients had a Medical Research Council (MRC) sum score <30. Axonal subtypes predominated (60.5%), with acute motor axonal neuropathy being the most prevalent (34.9%), followed by acute inflammatory demyelinating polyneuropathy (32.6%), acute motor sensory axonal neuropathy (AMSAN) (25.6%), and Fisher syndrome (7%). Notably, 15.1% had onset in upper limbs, 75.6% dysautonomia, and 73.3% pain. In all, 86% received either IVIg (9.3%) or plasma exchange (74.4%). IMV was required in 39.5% patients (72.7% in AMSAN). A multivariate model without including published prognostic scores yielded the time since onset to admission <15 days, axonal variants, MRC sum score <30, and bulbar weakness as independent predictors of IMV. The model including grading scales yielded lower limbs onset, Erasmus GBS respiratory insufficiency score (EGRIS) >4, and dysautonomia as predictors.
Conclusion:These results suggest that EGRIS is a good prognosticator of IMV in GBS patients with a predominance of axonal electrophysiological subtypes, but other early clinical data should also be considered.
Diagnostic application of sensitive and specific phage-exposed epitopes for visceral leishmaniasis and human immunodeficiency virus coinfection

The diagnosis of visceral leishmaniasis (VL) has improved with the search of novel antigens; however, their performance is limited when samples from VL/human immunodeficiency virus (HIV)-coinfected patients are tested. In this context, studies conducted to identify more suitable antigens to detect both VL and VL/HIC coinfection cases should be performed. In the current study, phage display was performed using serum samples from healthy subjects and VL, HIV-infected and VL/HIV-coinfected patients; aiming to identify novel phage-exposed epitopes to be evaluated with this diagnostic purpose. Nine non-repetitive and valid sequences were identified, synthetized and tested as peptides in enzyme-linked immunosorbent assay experiments. Results showed that three (Pep2, Pep3 and Pep4) peptides showed excellent performance to diagnose VL and VL/HIV coinfection, with 100% sensitivity and specificity values. The other peptides showed sensitivity varying from 50.9 to 80.0%, as well as specificity ranging from 60.0 to 95.6%. Pep2, Pep3 and Pep4 also showed a potential prognostic effect, since specific serological reactivity was significantly decreased after patient treatment. Bioinformatics assays indicated that Leishmania trypanothione reductase protein was predicted to contain these three conformational epitopes. In conclusion, data suggest that Pep2, Pep3 and Pep4 could be tested for the diagnosis of VL and VL/HIV coinfection.

To evaluate the impact of adding food education sessions to an exercise programme on cardiovascular risk factors in middle-aged and older patients with type 2 diabetes (T2D), a randomised parallel-group study was performed. Glycated haemoglobin, body mass index (BMI), waist circumference, fat mass (FM) and blood pressure were assessed at baseline and after 9 months. The recruitment was made in three primary healthcare centres from Vila Real, Portugal. Thirty-three patients (65⋅4 ± 5⋅9 years old) were engaged in a 9-month community-based lifestyle intervention programme: a supervised exercise programme (EX; n = 15; combined aerobic, resistance, agility/balance and flexibility exercise; three sessions per week; 75 min per session); or the same exercise programme plus concomitant food education sessions (EXFE; n = 18; 15-min lectures and dual-task strategies during exercise (answer nutrition questions while walking); 16 weeks). Significant differences between groups were identified in the evolution of BMI (P < 0.001, $\eta _{\rm p}^2 = 0{\cdot} 150$) and FM (P < 0.001, $\eta _{\rm p}^2 = 0{\cdot}130$), with best improvements observed in the EXFE group. The addition of a simple food education dietary intervention to an exercise programme improved body weight and composition, but not glycaemic control and blood pressure in middle-aged and older patients with T2D.
Mid-season sweet oranges for fresh and processing markets in Brazil

Pera sweet orange (Citrus sinensis L. Osbeck) is the most important mid-season sweet orange cultivar in Brazil, not only because of its desirable fruit quality for both juice processing and the fresh fruit market, but also because there is a lack of adequate mid-season alternative sweet orange cultivars. This study aimed to evaluate the horticultural performance of other sweet oranges in an attempt to identify similar or superior selections to the standard cultivar, Pera IAC sweet orange. Seventeen sweet oranges selections, including the standard Pera IAC, were budded on Sunki mandarin [C. sunki (Hayata) hort. ex Tanaka]. The experiment was planted in 2007, using 12-month-old nursery trees, with 6.5 × 2.5 m spacing in the Southern region of São Paulo state, Brazil. The experimental orchard was maintained with standard commercial management practices, and without supplementary irrigation. Data collection started three years after planting and was carried out until seven years after the experiment had been installed. Vegetative growth, plant yield, fruit drop, fruit quality, and fruit maturation regressions were assessed. Averages of the alternative selections were compared with those of the standard cultivar by Dunnett test. Performance indexes were calculated in order to identify potential superior selections for the fresh fruit market and for juice processing. Results indicated significant differences among the selections evaluated when compared with the standard cultivar, suggesting better alternatives to be exploited. Five selections (Biondo, Finike, Pera Alexandre Maróstica, Seleta Rio, and Vaccaro Blood) offered superior performance in terms of attributes required for the fresh fruit market, while one selection (Pera Alexandre Maróstica) was superior for juice processing traits. These alternatives to Pera IAC represent an opportunity for more efficient and productive scions with higher fruit quality, and with a slight shift within the mid-season harvest time of sweet oranges in Brazil.
An immunoproteomics approach to identify Leishmania infantum proteins to be applied for the diagnosis of visceral leishmaniasis and human immunodeficiency virus co-infection

The co-infection between visceral leishmaniasis (VL) and human immunodeficiency virus (HIV) has increased in several countries in the world. The current serological tests are not suitable since they present low sensitivity to detect the most of VL/HIV cases, and a more precise diagnosis should be performed. In this context, in the present study, an immunoproteomics approach was performed using Leishmania infantum antigenic extracts and VL, HIV and VL/HIV patients sera, besides healthy subjects samples; aiming to identify antigenic markers for these clinical conditions. Results showed that 43 spots were recognized by antibodies in VL and VL/HIV sera, and 26 proteins were identified by mass spectrometry. Between them, β-tubulin was expressed, purified and tested in ELISA experiments as a proof of concept for validation of our immunoproteomics findings and results showed high sensitivity and specificity values to detect VL and VL/HIV patients. In conclusion, the identified proteins in the present work could be considered as candidates for future studies aiming to improvement of the diagnosis of VL and VL/HIV co-infection.

There is no suitable vaccine against human visceral leishmaniasis (VL) and available drugs are toxic and/or present high cost. In this context, diagnostic tools should be improved for clinical management and epidemiological evaluation of disease. However, the variable sensitivity and/or specificity of the used antigens are limitations, showing the necessity to identify new molecules to be tested in a more sensitive and specific serology. In the present study, an immunoproteomics approach was performed in Leishmania infantum promastigotes and amastigotes employing sera samples from VL patients. Aiming to avoid undesired cross-reactivity in the serological assays, sera from Chagas disease patients and healthy subjects living in the endemic region of disease were also used in immunoblottings. The most reactive spots for VL samples were selected, and 29 and 21 proteins were identified in the promastigote and amastigote extracts, respectively. Two of them, endonuclease III and GTP-binding protein, were cloned, expressed, purified and tested in ELISA experiments against a large serological panel, and results showed high sensitivity and specificity values for the diagnosis of disease. In conclusion, the identified proteins could be considered in future studies as candidate antigens for the serodiagnosis of human VL.

Ternary Al–15 wt% Cu–7 wt% Si and Al–22 wt% Cu–7 wt% Si alloy specimens were generated by transient directional solidification (DS) and rapid solidification (RS) techniques. The microstructures are constituted by an α-Al dendritic matrix surrounded by two eutectic, that is, a binary eutectic (Si + α-Al) and a bimodal eutectic, consisting of cellular-type binary eutectic colonies (α-Al + Al2Cu) in a ternary eutectic matrix consisting of α-Al + Al2Cu + Si. The bimodal eutectic exists at cooling rates from 0.5 to 250 K/s. The secondary dendritic spacing, λ2, of the DS samples varied from 5 to 20 μm and from 10 to 18 μm for both examined alloys. The λ2 from 2.7 to 4.0 μm characterized the RS samples. Mechanical properties have been determined for various samples related to different dendritic spacing values. Based on the evaluation of the rapidly solidified microstructures, it was possible to assess the cooling rates.
High-through identification of T cell-specific phage-exposed mimotopes using PBMCs from tegumentary leishmaniasis patients and their use as vaccine candidates against Leishmania amazonensis infection

In the current study, phage-exposed mimotopes as targets against tegumentary leishmaniasis (TL) were selected by means of bio-panning cycles employing sera of TL patients and healthy subjects, besides the immune stimulation of peripheral blood mononuclear cells (PBMCs) collected from untreated and treated TL patients and healthy subjects. The clones were evaluated regarding their specific interferon-γ (IFN-γ) and interleukin-4 (IL-4) production in the in vitro cultures, and selectivity and specificity values were calculated, and those presenting the best results were selected for the in vivo experiments. Two clones, namely A4 and A8, were identified and used in immunization protocols from BALB/c mice to protect against Leishmania amazonensis infection. Results showed a polarized Th1 response generated after vaccination, being based on significantly higher levels of IFN-γ, IL-2, IL-12, tumour necrosis factor-α (TNF-α) and granulocyte-macrophage colony-stimulating factor (GM-CSF); which were associated with lower production of specific IL-4, IL-10 and immunoglobulin G1 (IgG1) antibodies. Vaccinated mice presented significant reductions in the parasite load in the infected tissue and distinct organs, when compared with controls. In conclusion, we presented a strategy to identify new mimotopes able to induce Th1 response in PBMCs from TL patients and healthy subjects, and that were successfully used to protect against L. amazonensis infection.
Antigenicity, immunogenicity and protective efficacy of a conserved Leishmania hypothetical protein against visceral leishmaniasis

In this study, a Leishmania hypothetical protein, LiHyS, was evaluated regarding its antigenicity, immunogenicity and protective efficacy against visceral leishmaniasis (VL). Regarding antigenicity, immunoblottings and an enzyme-linked immunosorbent assay using human and canine sera showed high sensitivity and specificity values for the recombinant protein (rLiHyS) in the diagnosis of VL. When evaluating the immunogenicity of LiHyS, which is possibly located in the parasite's flagellar pocket, proliferative assays using peripheral blood mononuclear cells from healthy subjects or VL patients showed a high proliferative index in both individuals, when compared to the results obtained using rA2 or unstimulated cultures. Later, rLiHyS/saponin was inoculated in BALB/c mice, which were then challenged with Leishmania infantum promastigotes. The vaccine induced an interferon-γ, interleukin (IL)-12 and granulocyte-macrophage colony-stimulating factor production, which was maintained after infection and which was associated with high nitrite and IgG2a antibody levels, as well as low IL-4 and IL-10 production. Significant reductions in the parasite load in liver, spleen, bone marrow and draining lymph nodes were found in these animals. In this context, the present study shows that the rLiHyS has the capacity to be evaluated as a diagnostic marker or vaccine candidate against VL.

An experimental and theoretical study of laminar vortical flows driven by oscillating electromagnetic forces that act in orthogonal directions in a shallow electrolytic fluid layer is presented. Forces are generated by the interaction of the field of a dipolar permanent magnet and two imposed alternating electric currents perpendicular to each other with independent frequencies varying in the range of 10–30 mHz. Velocity fields of the time-dependent flow are obtained using particle image velocimetry, while particle tracking allows exploration of the Lagrangian trajectories and time maps. An approximate two-dimensional analytical solution is obtained for the laminar creeping regime so that Lagrangian trajectories are integrated explicitly. These trajectories resemble Lissajous figures with the usual property that, when the ratio of the frequencies of the imposed currents is rational, closed paths are found, while non-closed paths occur when this ratio is irrational. Deviations of this regime that account for slight increase of inertial effects are explored through a quasi-two-dimensional numerical simulation. In this case, non-closed paths are found even for rational frequency ratios. This case was observed in the experiment. Lagrangian trajectories calculated numerically show a qualitative agreement with experimental particle tracking. Furthermore, numerical time maps obtained for increasing inertial effects and rational frequency ratios reveal a chaotic behaviour. Some features of the Lagrangian trajectories are validated experimentally. In particular, topological properties of the calculated and observed time maps are in qualitative agreement. In a characteristic case, a partial time map calculated numerically is compared with the section acquired from the experimental tracking of one particle.

OBJECTIVE
To identify risk factors related to Mycobacterium abscessus subsp. bolletii infection during an outbreak, associated with laparoscopic surgery and to propose recommendations for preventing new cases.
DESIGNA retrospective cohort study.
SETTINGA private hospital in Manaus, Brazil.
PATIENTSA cohort of 222 patients who underwent laparoscopic surgery between July 2009 and August 2010 by a single surgical team.
METHODSWe collected information about the patients and the surgical procedure using a standard form. We included sex, age, and variables with P≤0.2 in the bivariate analysis in a logistic regression model. Additionally, we reviewed the procedures for reprocessing the laparoscopic surgery equipment, and the strains obtained with culture were identified by molecular methods.
RESULTSWe recorded 60 (27%) cases of infection. After multivariate analysis, the duration of surgery beyond 1 hour (odds ratio [OR] 2.4; 95% confidence interval [CI] 1.2–4.5), not to have been the first operated patient on a given day (OR, 2.7; 95% CI, 1.4–5.2), and the use of permanent trocar (OR, 2.2; 95% CI, 1.1–4.2) were associated with infection. We observed that the surgical team attempted to sterilize the equipment in glutaraldehyde solution when sanitary authorities had already prohibited it. Eleven strains presented 100% DNA identity with a single strain, known as BRA100 clone.
CONCLUSIONSBecause contaminated material can act as vehicle for infection, ensuring adequate sterilization processing of video-assisted surgery equipment was crucial to stopping this single clonal outbreak of nonturbeculous mycobacteria in Brazil.

The solar absorptance αs of nanostructured selective surface (NSS) for solar thermal energy is improved. The NSS are prepared by AC electrochemical impregnation of metal inclusions (MI) into porous anodized aluminum oxide (AAO). The dependence of the NSS performance with composition depth profile and MI is studied by numeric simulations based in a gradient index model and effective medium theory. The results are compared with experimental NSS prepared varying three control parameters and MI (Ni, Cu, Ag). The αs is improved to > 85% (keeping thermal emittance εT relatively low) for Ni MI, mainly by increasing MI content. Increasing AAO thickness or MI molecular weight (for a given experimental composition profile) also improves the performance. For Ag the αs was further improved to 90%.

Summary
There is considerable interest in determining the exact route(s) taken by populations that entered the Americas, which would be expected to be genetically linked to many Asian lineages. However, some of the lineages of these American ancestors that were likely lost by drift through time may be unearthed by mitochondrial DNA (mtDNA) sequencing of the ancient populations that are revealed in a growing body of fossils in America, including Mexico. The most recent common ancestors in America are identifiable because their lineages survived from one generation to the next and have started their differentiation by means of accumulated mutations, defining the founder mtDNA. Consequently, the genetic studies of human Mexican populations is a research area will inform us about genetic markers specific to each population, their movement, and their relationship through time (Davis et al., 2011; Ebenesersdóttir et al., 2011; Martins et al., 2011).
DNA is passed from one generation to the next; most of it is admixture, making each person unique from his or her parents. However, mtDNA passes from the mother to the child with no changes (with few exceptions). Mitochondrial DNA allows men and women to trace their maternal lineages; the non-recombinant region of the Y chromosome that passed intact only from father to son without any modification allows men to trace their paternal lineage. Both Y-chromosome DNA and mtDNA are subject to occasional mutations that become inheritable as genetic markers and then, after several generations, a particular genetic marker is carried by almost all male and female inhabitants of the region where it arose (Terreros et al., 2011; Wilder et al., 2004). When people leave this region, they carry the marker with them, allowing the tracing of migration patterns of a given person’s ancient ancestors. In addition, anthropological studies will complement genetic studies in answering the question of why people migrate. Because of the complexity of choosing every population in our study, we focus on the Mexican population, including the pre-Hispanic populations and the contemporary Mestizo populations. Accordingly, studying mitochondrial DNA will allow us to reveal the pattern of pre-Hispanic population migration. This is of particular importance since the American continent has been populated as a consequence of different migrations through time (Tarazona-Santos and Santos, 2002).

Four distinct Franciscana Management Areas (FMAs) have been proposed based on several lines of evidence including genotype, phenotype, population response and distribution. To determine if differences in external morphology fit this division, a canonical variate analysis was carried out for males and/or females from FMAs I to IV using up to 14 characters. A total of 78 adult specimens were analysed. More than 90% of the differences between groups were summarized by three canonical variates. Females were larger than males in all areas. Females from FMA IV were of intermediate length between those from FMA I and FMA III and individuals from FMA II were smaller than those from all other areas. Position of dorsal fin and morphology of the anterior body region, differentiate individuals from FMA I and FMA III. Morphological differences found in this study give additional support for the proposed FMAs. Since habitat characteristics and franciscana feeding ecology vary regionally, it is possible that observed morphological differences are due to ecological divergence for niche occupation. The indication of a discontinuous distribution, consistency between genetic and morphological evidence, and a short time genetic divergence, might indicate that franciscanas inhabiting FMA I represent a distinct subspecies.

Factors Associated with Time to Acquisition of Bloodstream Infection in a Pediatric Intensive Care Unit

Objective.
To evaluate the risk factors that influence time to acquisition of a laboratory-confirmed bloodstream infection (LCBI).
Design.Prospective cohort study with an 18-month follow-up.
Setting.A 16-bed medical and/or surgical pediatric intensive care unit that cares for patients of the Brazilian Public Health System exclusively.
Patients.We included children from 0 to 18 years old who were represented by 875 consecutive admissions to the pediatric intensive care unit from January 1, 2005, through June 30, 2006. The children from all but 5 (0.6%) of the admissions were followed up until discharge or death. The majority (506 [58.2%]) were hospitalized for surgical pathology, and 254 (29.2%) underwent heart surgery.
Methods.We used a standardized questionnaire and data collection from daily charts. Information on risk factors was collected before the onset of first LCBI. Survival analysis was performed using the Kaplan-Meier method. The effect of the variables on the risk of LCBI each day was estimated through a Cox model fitting.
Results.Fifty-seven children (6.6%) developed an LCBI, 54 (94.7%) of whom made use of a central venous catheter. LCBI incidence was 11.27 episodes/1,000 patient-days and 17.92 episodes/1,000 patient-days when associated with a central venous catheter. Factors associated with time to the first LCBI in the Cox model were age less than 2 years (hazard ratio [HR], 1.99; 95% confidence interval [CI], 1.02–3.89), malnutrition (HR, 1.74; 95% CI, 1.01–3.00), use of a central venous catheter (HR, 4.36; 95% CI, 1.30–14.64), use of antibiotics before admission (HR, 0.58; 95% CI, 0.33–0.98), and use of transfused blood products (HR, 0.40; 95% CI, 0.22–0.74).
Conclusion.Factors associated with time to acquisition of LCBI were age less than 2 years, weight-for-age z score less than −2, and the use of a central venous catheter. Therefore, intensification of LCBI prevention efforts in patients with these characteristics is fundamental.
Electrically driven vortices in a weak dipolar magnetic field in a shallow electrolytic layer

Steady dipolar vortices continuously driven by electromagnetic forcing in a shallow layer of an electrolytic fluid are studied experimentally and theoretically. The driving Lorentz force is generated by the interaction of a dc uniform electric current injected in the thin layer and the non-uniform magnetic field produced by a small dipolar permanent magnet (0.33 T). Laminar velocity profiles in the neighbourhood of the zone affected by the magnetic field were obtained with particle image velocimetry in planes parallel and normal to the bottom wall. Flow planes at different depths of the layer were explored for injected currents ranging from 10 to 100 mA. Measurements of the boundary layer attached to the bottom wall reveal that owing to the variation of the field in the normal direction, a slightly flattened developing profile with no shear stresses at the free surface is formed. A quasi-two-dimensional magnetohydrodynamic numerical model that introduces the non-uniformity of the magnetic field, particularly its decay in the normal direction, was developed. Vertical diffusion produced by the bottom friction was modelled through a linear friction term. The model reproduces the main characteristic behaviour of the electromagnetically forced flow.
