Nitrogen and fatty acid rumen metabolism in cattle offered high or low polyphenol oxidase red clover silage

Polyphenol oxidase (PPO) in red clover (RC) has been shown to reduce both lipolysis and proteolysis in silo and implicated (in vitro) in the rumen. However, all in vivo comparisons have compared RC with other forages, typically with lower levels of PPO, which brings in other confounding factors as to the cause for the greater protection of dietary nitrogen (N) and C18 polyunsaturated fatty acids (PUFA) on RC silage. This study compared two RC silages which when ensiled had contrasting PPO activities (RC+ and RC−) against a control of perennial ryegrass silage (PRG) to ascertain the effect of PPO activity on dietary N digestibility and PUFA biohydrogenation. Two studies were performed the first to investigate rumen and duodenal flow with six Hereford×Friesian steers, prepared with rumen and duodenal cannulae, and the second investigating whole tract N balance using six Holstein-Friesian non-lactating dairy cows. All diets were offered at a restricted level based on animal live weight with each experiment consisting of two 3×3 Latin squares using big bale silages ensiled in 2010 and 2011, respectively. For the first experiment digesta flow at the duodenum was estimated using a dual-phase marker system with ytterbium acetate and chromium ethylenediaminetetraacetic acid as particulate and liquid phase markers, respectively. Total N intake was higher on the RC silages in both experiments and higher on RC− than RC+. Rumen ammonia-N reflected intake with ammonia-N per unit of N intake lower on RC+ than RC−. Microbial N duodenal flow was comparable across all silage diets with non-microbial N higher on RC than the PRG with no difference between RC+ and RC−, even when reported on a N intake basis. C18 PUFA biohydrogenation was lower on RC silage diets than PRG but with no difference between RC+ and RC−. The N balance trial showed a greater retention of N on RC+ over RC−; however, this response is likely related to the difference in N intake over any PPO driven protection. The lack of difference between RC silages, despite contrasting levels of PPO, may reflect a similar level of protein-bound-phenol complexing determined in each RC silage. Previously this complexing has been associated with PPOs protection mechanism; however, this study has shown that protection is not related to total PPO activity.
The impact of floor type on lameness and hoof health of dairy origin bulls

This study aimed to evaluate the effect of using different floor types to accommodate growing and finishing beef cattle on lameness. In all, 80 dairy origin bulls were blocked according to live weight and breed into 20 groups, and randomly allocated within groups to one of four treatments. The floor types studied were fully slatted flooring throughout the entire experimental period (CS); fully slatted flooring covered with rubber strips throughout the entire experimental period (RS); fully slatted flooring during the growing period and then moved to a solid floor covered with straw bedding during the finishing period (CS-S) and fully slatted flooring during the growing period and then moved to fully slatted flooring covered with rubber strips during the finishing period (CS-RS). The total duration of the study was 204 days. The first 101 days was defined as the growing period, with the remainder of the study defined as the finishing period. During the growing period, there was a tendency for bulls accommodated on CS to have a higher locomotion score compared with those accommodated on RS (P=0.059). However, floor type had no significant effect on locomotion score during the finishing period. There was also no significant effect of floor type on digital dermatitis during both the growing or finishing period. Floor type had no significant effect on swelling at the leg joints at the end of the finishing period. Bulls accommodated on RS had the least probability of bruised soles during both the growing and finishing period (P<0.01). Growing bulls accommodated on CS had significantly greater front heel height net growth compared with those accommodated on RS (P<0.05). However, bulls accommodated on RS had a tendency to have greater front toe net growth compared with those accommodated on CS (P=0.087). Finishing bulls accommodated on CS-RS had the greatest front toe net growth (P<0.001). Heel height net growth was greatest in bulls accommodated on CS-S (P<0.001). Floor type had no significant effect on mean maximum hoof temperature during the growing period. Finishing bulls accommodated on CS-S had a significantly lower mean maximum hoof temperature compared with those accommodated on any other floor type (P<0.001). The study concluded that rubber flooring is a suitable alternative to fully slatted flooring, reducing the prevalence of bruised soles. Despite greater toe net growth in bulls accommodated on rubber flooring, there was no effect of floor type on locomotion score, suggesting that increased toe net growth does not adversely affect walking ability. In addition, although mean maximum hoof temperature was lowest in bulls accommodated on straw bedding, there was no evidence to suggest this is indicative of improved hoof health.

The aim of this study was to evaluate the effect of using different floor types to accommodate growing and finishing beef cattle on their performance, cleanliness, carcass characteristics and meat quality. In total, 80 dairy origin young bulls (mean initial live weight 224 kg (SD=28.4 kg)) were divided into 20 blocks with four animals each according to live weight. The total duration of the experimental period was 204 days. The first 101 days was defined as the growing period, with the remainder of the study defined as the finishing period. Cattle were randomly assigned within blocks to one of four floor type treatments, which included fully slatted flooring throughout the entire experimental period (CS); fully slatted flooring covered with rubber strips throughout the entire experimental period (RS); fully slatted flooring during the growing period and moved to a solid floor covered with straw bedding during the finishing period (CS-S) and fully slatted flooring during the growing period and moved to fully slatted flooring covered with rubber strips during the finishing period (CS-RS). Bulls were offered ad libitum grass silage supplemented with concentrates during the growing period. During the finishing period, bulls were offered concentrates supplemented with chopped barley straw. There was no significant effect of floor type on total dry matter intake (DMI), feed conversion ratio, daily live weight gain or back fat depth during the growing and finishing periods. Compared with bulls accommodated on CS, RS and CS-RS, bulls accommodated on CS-S had a significantly lower straw DMI (P<0.01). Although bulls accommodated on CS and CS-S were significantly dirtier compared with those accommodated on RS and CS-RS on days 50 (P<0.05) and 151 (P<0.01), there was no effect of floor type on the cleanliness of bulls at the end of the growing and finishing periods. There was also no significant effect of floor type on carcass characteristics or meat quality. However, bulls accommodated on CS-S had a tendency for less channel, cod and kidney fat (P=0.084) compared with those accommodated on CS, RS and CS-RS. Overall, floor type had no effect on the performance, cleanliness, carcass characteristics or meat quality of growing or finishing beef cattle.
High-resolution ice cores from US ITASE (West Antarctica): development and validation of chronologies and determination of precision and accuracy

Shallow ice cores were obtained from widely distributed sites across the West Antarctic ice sheet, as part of the United States portion of the International Trans-Antarctic Scientific Expedition (US ITASE) program. The US ITASE cores have been dated by annual-layer counting, primarily through the identification of summer peaks in non-sea-salt sulfate (nssSO42–) concentration. Absolute dating accuracy of better than 2 years and relative dating accuracy better than 1 year is demonstrated by the identification of multiple volcanic marker horizons in each of the cores, Tambora, Indonesia (1815), being the most prominent. Independent validation is provided by the tracing of isochronal layers from site to site using high-frequency ice-penetrating radar observations, and by the timing of mid-winter warming events in stable-isotope ratios, which demonstrate significantly better than 1 year accuracy in the last 20 years. Dating precision to ±1 month is demonstrated by the occurrence of summer nitrate peaks and stable-isotope ratios in phase with nssSO42–, and winter-time sea-salt peaks out of phase, with phase variation of <1 month. Dating precision and accuracy are uniform with depth, for at least the last 100 years.

Local rates of change in ice-sheet thickness were calculated at 15 sites in West Antarctica using the submergence velocity technique. This method entails a comparison of the vertical velocity of the ice sheet, measured using repeat global positioning system surveys of markers, and local long-term rates of snow accumulation obtained using firn-core stratigraphy. Any significant difference between these two quantities represents a thickness change with time. Measurements were conducted at sites located ~100–200km apart along US ITASE traverse routes, and at several isolated locations. All but one of the sites are distributed in the Siple Coast and the Amundsen Sea basin along contours of constant elevation, along flowlines, across ice divides and close to regions of enhanced flow. Calculated rates of thickness change are different from site to site. Most of the large rates of change in ice thickness (~10cm a–1 or larger) are observed in or close to regions of rapid flow, and are probably related to ice-dynamics effects. Near-steady-state conditions are calculated mostly at sites in the slow-moving ice-sheet interior and near the mainWest Antarctic ice divide. These results are consistent with regional estimates of ice-sheet change derived from remote-sensing measurements at similar locations in West Antarctica

We present a new surface-balance and ice-motion dataset derived from high-precision GPS measurements from a network of steel poles within three icefields of the Allan Hills blue-ice area, Antarctica. The surveys were conducted over a 14 year time period. Ice-flow velocities and mass- balance estimates for the main icefield (MIF) are consistent with those from pre-GPS era measurements but have much smaller uncertainties. The current study also extends these measurements through the near-western icefield (NWIF) to the eastern edge of the mid-western icefield (MWIF). The new dataset includes, for the first time, well-constrained evidence of upward motion within the Allan Hills MIF, indicating that old ice should be present at the surface. These data and terrestrial meteorite ages suggest that paleoclimate reconstructions using the surface record within the Allan Hills MIF could potentially extend the ice-core-based record beyond the 800 000 years currently available in the EPICA Dome C core.

The effect of silver nanoparticles showing localised plasmonic resonances on the efficiency of thin film silicon solar cells is studied. Silver (Ag) nanodiscs were deposited on the surface of silicon cells grown on highly doped silicon substrates, through hole-mask colloidal lithography, which is a low-cost and bottom-up technique. The cells have no back reflector in order to exclusively study the effect of the front surface on their properties. Cells with nanoparticles were compared with both bare silicon cells and cells with an antireflection coating. We optically observe a resonance showing an absorption increase controllable by the disc radius. We also see an increase in efficiency with respect to bare cells, but we see a decrease in efficiency with respect to cells with an antireflection coating due to losses at wavelengths below the plasmon resonance. As the material properties are not notably affected by the particles deposition, the loss mechanism is an important absorption in the nanoparticles. We confirm this by numerical simulations.

Background
Psychiatric phenotypes are currently defined according to sets of descriptive criteria. Although many of these phenotypes are heritable, it would be useful to know whether any of the various diagnostic categories in current use identify cases that are particularly helpful for biological–genetic research.
AimsTo use genome-wide genetic association data to explore the relative genetic utility of seven different descriptive operational diagnostic categories relevant to bipolar illness within a large UK case–control bipolar disorder sample.
MethodWe analysed our previously published Wellcome Trust Case Control Consortium (WTCCC) bipolar disorder genome-wide association data-set, comprising 1868 individuals with bipolar disorder and 2938 controls genotyped for 276 122 single nucleotide polymorphisms (SNPs) that met stringent criteria for genotype quality. For each SNP we performed a test of association (bipolar disorder group v. control group) and used the number of associated independent SNPs statistically significant at P<0.00001 as a metric for the overall genetic signal in the sample. We next compared this metric with that obtained using each of seven diagnostic subsets of the group with bipolar disorder: Research Diagnostic Criteria (RDC): bipolar I disorder; manic disorder; bipolar II disorder; schizoaffective disorder, bipolar type; DSM–IV: bipolar I disorder; bipolar II disorder; schizoaffective disorder, bipolar type.
ResultsThe RDC schizoaffective disorder, bipolar type (v. controls) stood out from the other diagnostic subsets as having a significant excess of independent association signals (P<0.003) compared with that expected in samples of the same size selected randomly from the total bipolar disorder group data-set. The strongest association in this subset of participants with bipolar disorder was at rs4818065 (P = 2.42 × 10–7). Biological systems implicated included gamma amniobutyric acid (GABA)A receptors. Genes having at least one associated polymorphism at P<10–4 included B3GALTS, A2BP1, GABRB1, AUTS2, BSN, PTPRG, GIRK2 and CDH12.
ConclusionsOur findings show that individuals with broadly defined bipolar schizoaffective features have either a particularly strong genetic contribution or that, as a group, are genetically more homogeneous than the other phenotypes tested. The results point to the importance of using diagnostic approaches that recognise this group of individuals. Our approach can be applied to similar data-sets for other psychiatric and non-psychiatric phenotypes.
Schistosoma mansoni: analysis of the humoral and cellular basis of resistance in guinea-pigs vaccinated with radiation-attenuated cercariae

This study addresses the humoral and cellular basis of specific acquired immunity in the guinea-pig irradiated vaccine model of schistosomiasis mansoni. Rodents vaccinated with 500 gamma-irradiated cercariae and then splenectomized 4·5 weeks later showed a 33% reduction in resistance to challenge as compared to vaccinated animals or vaccinated/sham splenectomized controls. Serum harvested from once vaccinated individuals conferred modest levels of resistance upon naive recipients in some experiments, but transfer was not achieved consistently. Serum from vaccinated and thrice boosted rodents (Vbbb) routinely transferred around 45% immunity, however, provided it was given in 4 ml aliquots on day 9 post-challenge; Vbbb serum thus transferred 50% of donor immunity. Interestingly, multiple doses of this protective serum given on and either side of day 9 did not enhance the protection achieved with a single 4 ml aliquot. Neither peripheral lymph node cells nor splenocytes from the polyvaccinated serum donors were able to transfer resistance to recipient guinea-pigs and they failed to augment the protection achieved with Vbbb serum. Food-pad testing revealed no correlation between delayed hypersensitivity responses and immunity to challenge in vaccinated guinea-pigs. Although polyvaccine guinea-pig serum successfully protected homologous recipients, it failed to protect mice when administered either at the time of challenge (the optimal schedule for transfer of polyvaccine mouse serum), or around day 9 (the optimal schedule for guinea-pigs). Similarly, guinea-pigs could not be protected with polyvaccine rat serum that conferred 75% resistance upon naive recipient rats.

Early results from the SAGE-SMC (Surveying the Agents of Galaxy Evolution in the tidally-disrupted, low-metallicity Small Magellanic Cloud) Spitzer legacy program are presented. These early results concentrate on the SAGE-SMC MIPS observations of the SMC Tail region. This region is the high H i column density portion of the Magellanic Bridge adjacent to the SMC Wing. We detect infrared dust emission and measure the gas-to-dust ratio in the SMC Tail and find it similar to that of the SMC Body. In addition, we find two embedded cluster regions that are resolved into multiple sources at all MIPS wavelengths.
Investigation of growth rate variation between commercial pig herds

The study was designed to provide quantifiable information on both within- and between-herd variation in pig growth rate from birth to slaughter and to examine how this was influenced by moving pigs at a common age to a common environment. Five litters were selected from each of eight pig herds in Northern Ireland with varying growth performance. All eight herds were offered the same nutritional regime. Five pigs (three boars and two gilts) were selected from each litter. In each herd, 22 pigs (12 boars and 10 gilts) were weighed individually, every 4 weeks, from 4 to 20 weeks of age. At 4 weeks of age (weaning) three non-sibling boars were taken from each herd and brought to a common environment where they received medication, were housed individually from 6 weeks of age and offered the same dietary regime. They were weighed and feed intakes were recorded twice weekly. A growth rate difference of 61 g/day (P < 0.001), 112 g/day (P < 0.01) and 170 g/day (P < 0.001) was observed on farm, between the top and bottom quartile of herds during 4 to 8, 8 to 12 and 12 to 20 weeks of age, respectively. This difference in growth rate equated to an average difference in cost of production of ¢13/kg carcass on a birth to bacon unit. When pigs from the different herds were housed in the common environment, large variation in growth performance (143 g/day (P < 0.01) and 243 g/day (P < 0.001) for 8 to 12 and 12 to 20 weeks, respectively) was also observed between the top and bottom quartile of herds. Although feed efficiency was similar, a significant feed intake difference of 329 g/day (P < 0.01) and 655 g/day (P < 0.001) between 8 to 12 and 12 to 20 weeks of age was observed. The variation in growth rate between pigs whether managed on farm or in the common environment was similar (variation in days to 100 kg on farm and in the common environment was 18 and 19 days, respectively). When housed in the common environment, although the top and bottom quartile of pigs converted feed equally efficiently, pigs in the top quartile had significantly higher feed intakes suggesting greater appetites. It is difficult to assess the extent to which these differences can be attributed to genetic effects or pre-weaning environment, and how much the effects of management, disease or genetics contributed to the variation between and within herds.
The Shear Modulus of Vit 1 in the Supercooled Liquid and Glassy State

Using a non-contact electromagnetic-acoustic transformation (EMAT) technique, we have measured the temperature dependence of the infinite frequency shear modulus of Vit 1 at constant heating rate in the glassy and supercooled liquid states. Values of the shear softening fragility parameter -dln(G/Gg/d(T/Tg) are compared with those obtainable from specific heat and viscosity measurements, using the interstitialcy theory of condensed matter states. There is overall agreement found between these independently measured values.
