Opioid antagonism mitigates antipsychotic-associated weight gain: focus on olanzapine

Background
The endogenous opioid system affects metabolism, including weight regulation. Evidence from preclinical and clinical studies provides a rationale for targeting this system to mitigate weight-related side effects of antipsychotics. This review describes the role of the opioid system in regulating weight and metabolism, examines the effects of opioid receptor antagonism on those functions, and explores the use of opioid antagonists to mitigate antipsychotic-associated weight gain and/or metabolic effects.
MethodsA PubMed literature search was conducted to identify representative opioid antagonists and associated preclinical and clinical studies examining their potential for the regulation of weight and metabolism.
ResultsThe mu opioid receptor (MOR), delta opioid receptor (DOR), and kappa opioid receptor (KOR) types have overlapping but distinct patterns of central and peripheral expression, and each contributes to the regulation of body weight and metabolism. Three representative opioid antagonists (eg, naltrexone, samidorphan, and LY255582) were identified for illustration. These opioid antagonists differed in their receptor binding and pharmacokinetic profiles, including oral bioavailability, systemic clearance, and half-life, and were associated with varying effects on food intake, energy utilization, and metabolic dysregulation.
ConclusionsPreclinical and clinical data suggest that antagonism of the endogenous opioid system is a mechanism to address antipsychotic-associated weight gain and metabolic dysregulation. However, evidence suggests that the differing roles of MOR, DOR, and KOR in metabolism, together with the differences in receptor binding, pharmacokinetic, and functional activity profiles of the opioid receptor antagonists discussed in this review, likely contribute to their differential pharmacodynamic effects and clinical outcomes observed regarding antipsychotic-associated weight gain.

Since its inception in 2010, the Network for Public Health Law (Network) has aligned with federal, state, tribal, and local public health practitioners to assess how law can promote and protect the public’s health. In 2013, Network authors illustrated major trends in public health laws and policies emanating from an internal assessment of thousands of requests for technical assistance nationally. More recently, the Robert Wood Johnson Foundation (RWJF) has invited the Network and other partners to consider new ideas and strategies toward building a “culture of health.” Per Figure 1, RWJF’s conception of a culture of health emphasizes key action areas essential to the promotion of health across all sectors and diverse populations.

In letters to Mrs. Ernest Benzon and Mrs. Thomas FitzGerald, Browning claims affinity with the great philosopher of the Will, Schopenhauer, and asserts that elements of vitalism are the “substratum” of his life and work. These letters confirm the poet's place in the line of vitalist thought shaped by Schopenhauer, the English Romantics, and Carlyle and further developed by Nietzsche, George Bernard Shaw, Henri Bergson, and D. H. Lawrence. Vitalism resists precise definition; each theorist advances a singular terminology and application. Schopenhauer's vitalism may be understood from his concept of cosmic Will; Carlyle's from the essential presence of energy, movement, and change in the world. Bergson used the term élan vital and Lawrence such characteristically vague phrases as “sense of truth” and “supreme impulse” to express faith in forces operating beneath or hovering above the surface of life. Broadly put, when a rational orientation to the world ceased to be adequate, when rationalism devolved into a falsification of reality's authentic energy, major vitalists came into existence and posited as the true reality a primitive, universal force of which everything in that reality is an objectification. Unlike other vitalists in the English tradition, such as Blake and Lawrence, Browning was not comfortable with cosmic images. His vitalism breaks from the main line to focus on the individual human will, which he saw as an intuitive impulse and as a means to realize the self and locate its place in the world. For Browning, the comprehension of life's vital movement lay in the dynamic energy of willed action.
Lowering mechanical degradation of drag reducers in turbulent flow

Drag reduction (DR) agents are used in several ppm concentrations to accelerate significantly the flow through conduits in oil pipelines, oil well operations, flood water disposal, fire fighting, field irrigation, transport of suspensions and slurries, sewage systems, water heating and cooling systems, airplane tank filling, marine systems, and also in biomedical systems including blood flow. The drag reduction agents are typically high molecular mass polymers; in industrial applications they undergo mechanical degradation in turbulent flow. We provide an equation that describes quantitatively the degradation, thus predicting drag reduction as a function of time and of the concentration of the drag reduction agent. We report how grafting a polymer on the backbone of a different polymer affects the drag reduction efficacy. Our grafted polymer undergoes degradation by flow turbulence more slowly and also provides high levels of drag reduction efficacy at much lower concentrations than homopolymers do.

Two patients (G01, J02) with chronic nonfluent aphasia and sentence production deficits received syntactic mapping treatment to improve sentence production. The patients had dramatically different outcomes in that improved syntax production generalized to nontreatment tasks for G01, but not for JO2. To learn how treatment influenced the neural substrates for syntax production, both patients underwent pre- and posttreatment functional magnetic resonance imaging (fMRI) of sentence generation. G01 showed more robust activity posttreatment than pretreatment in Broca's area; ventral temporal activity decreased slightly from pre- to posttreatment. Comparison of J02's pretreatment and posttreatment images revealed little change, although activity was more diffuse pre- than posttreatment. Findings suggest that for G01, rehabilitation led to engagement of an area (Broca's area) used minimally during the pretreatment scan, whereas for J02, rehabilitation may have led to more efficient use of areas already involved in sentence generation during the pretreatment scan. fMRI findings are discussed in the context of sentence-production outcome and generalization. How are cultural similarities in non-contiguous regions to be explained? This has been an important question in ethnology for many years and still remains so to this day. E. B. Tylor termed it the ‘ great problem, the solution of which will alone bring the study of civilization into its full development as a science’. It has been important for two reasons : first because it involves a theory of, or attitude towards, culture in general; culture is thought of as having one kind of nature if these similarities are explained in one way, but having a different nature if explained in another. Secondly, the question is important because we wish to know in the case of particular instances whether similarities are due to independent or to a common origin; quite apart from general theory we wish to know, and feel that it is important to know, whether, for example, the practice of mummification in aboriginal Peru originated indigenously or whether it had its origin, as some have believed, in ancient Egypt.

Lewis Henry Morgan (1818–1881), the “Father of American Anthropology,” was one of the most distinguished scientists of the nineteenth century. After graduating from Union College in 1840 he studied law, practising this profession in Rochester until 1862 when he gave it up to devote himself to scientific labors. His first work in anthropology was with the Iroquois. His The League of the Iroquois,” the first scientific account of an Indian tribe ever given to the world,” appeared in 1851. This was followed by years of exhaustive research in the field of kinship and social organization, culminating in his monumental Systems of Consanguinity and Affinity of the Human Family (1871). His greatest work, Ancient Society, appeared in 1877. Translated into virtually all the languages of Europe and into Chinese and Japanese, this work has exerted a profound and enduring influence upon social science throughout the world. Morgan's last work, Houses and House Life of the American Aborigines, was published in 1881, shortly before his death.

In July, 1939, a celebration was held at Leland Stanford University to commemorate the hundredth anniversary of the discovery that the cell is the basic unit of all living tissue. Today we are beginning to realize and to appreciate the fact that the symbol is the basic unit of all human behavior and civilization.

Science is not merely a collection of facts and formulas. It is preëminently a way of dealing with experience. The word may be appropriately used as a verb: one sciences, i.e., deals with experience according to certain assumptions and with certain techniques. Science is one of two basic ways of dealing with experience. The other is art. And this word, too, may appropriately be used as a verb; one may art as well as science. The purpose of science and art is one: to render experience intelligible, i.e., to assist man to adjust himself to his environment in order that he may live. But although working toward the same goal, science and art approach it from opposite directions. Science deals with particulars in terms of universals: Uncle Tom disappears in the mass of Negro slaves. Art deals with universals in terms of particulars: the whole gamut of Negro slavery confronts us in the person of Uncle Tom. Art and science thus grasp a common experience, or reality, by opposite but inseparable poles.
