Metabolic dysfunction-associated steatotic liver disease (MASLD) is a major public health issue, with a recent estimated global prevalence of 30%(1). The pathogenesis of MASLD is complex, multifactorial, and incompletely understood(2). MicroRNAs (miRNAs) are small non-coding RNAs that typically inhibit gene expression as post-transcriptional regulators(3). MiRNAs are increasingly recognised for their potential as diagnostic biomarkers and therapeutic targets in liver diseases(3). The aim of this research was to consider the role of miRNAs in MASLD pathogenesis and their regulation by diet.

Liver RNA samples were extracted from male C57BL/6J mice (n = 6/group) fed for 8 weeks either a chow diet (CD) or 60% fat (high fat diet, HFD) and sent for RNA sequencing. The raw data obtained from next-generation sequencing (Novogene, Cambridge) were passed through a quality control pipeline. Differential expression analysis, Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses, and target predictions were performed in the R environment. Enriched GO terms were summarised and clustered by online tool Revigo. Rich factor (RF) was used to describe the intensity of dysregulated pathways, calculated as the ratio of differentially expressed genes (DEGs) to the total genes included in each pathway. MicroRNAs that targeted the DEGs were identified using two validated miRNA-mRNA interaction databases, Tarbase and miRTarbase.

A total of 1087 DEGs were found in HFD fed mice relative to CD (adjusted P<0.05) and their biological functions were examined using GO and KEGG enrichment analyses. In aggregate, significantly enriched components and functions included: small molecule metabolic processes, collagen-containing extracellular matrix and lipoprotein particles, and oxidoreductase activity. Circadian rhythm was the most enriched pathway (RF = 0.29) among 12 enriched KEGG pathways, which also included fatty acid degradation (RF = 0.21), PPAR signalling (RF = 0.16), retinol (RF = 0.17) and cholesterol metabolism (RF = 0.22) pathways. Indeed, out of the 34 genes that comprise the KEGG circadian rhythm pathway, 10 (29%) were found dysregulated by HFD, including the critical. Period genes (Per2, Per3). Further exploration in human found hsa-miR-133a-3p targets PER2 and PER3 via examination of validated miRNA-mRNA interaction databases. In addition to circadian rhythm, hsa-miR-133a-3p was found to target genes that function in macroautophagy, cold-induced thermogenesis, and insulin response pathways.

Peripheral clocks of circadian rhythm (liver, pancreas, adipose tissue and muscle) are regulated by miRNAs that contribute to the pathogenesis of MASLD through disruption of lipid metabolism and the insulin response. Future work includes the confirmation of miR-133a-3p expression and the related pathways in mouse liver and human primary hepatocytes.

The triple burden of malnutrition (TBM) is a growing public health issue worldwide. This study examined the prevalence and association between undernutrition, overnutrition and micronutrient deficiencies (MNDs), and the key demographic and socioeconomic determinants, among Vietnamese children, using the nationally representative General Nutrition Survey (GNS) 2020(1).

The GNS 2020 was reviewed and approved by the Ethical Committee of the National Institute of Nutrition, Vietnam. Written informed consent was obtained from each participant and/or their parents prior to data collection. Data on anthropometric parameters, micronutrients biomarkers, demographic and socioeconomic indicators for 7,289 children aged 6 months to 9 years old were analysed(1). All analyses were weighted and accounted for complex survey design using svyset commands in STATA. Age-specific and region-specific prevalence of malnutrition were examined. Determinants of malnutrition were assessed using bivariate and multivariate logistic regressions, crude or adjusted odds ratio (OR/AOR) and respective 95% confidence intervals (CI) were reported.

Overall, the prevalence of stunting, underweight and wasting/thinness of children was 12.7%, 10.5%, and 4.7%, respectively, in 2020. This finding underscores that child stunting reduction in Vietnam is on track towards the target of 40% reduction by 2025 set by the Global Nutrition Targets(2) (<20%). Meanwhile overweight and obesity among school aged children is off track the target(2), with the prevalence in urban areas increasing from 8.5% in 2010 to 31% in 2020. Low serum zinc, anaemia and iron deficiency (ID) were the common MNDs observed, affecting 53.1%, 15.2%, and 13.9% of children. Prevalence of low serum retinol was relatively low (<7%). In the bivariate regressions, older child (2-4 years old) [OR (95% CI): 1.43 (1.20, 1.72)], ethnic minorities [5.94 (3.78, 9.36)], and living in mountainous areas [5.06 (1.18, 14.42)] had increased odds of stunting, whereas reduced odds were found in children from the richest quintile [0.13 (0.05, 0.32)]. Similar determinants were found to be associated with underweight and MNDs. Males [1.43 (1.16, 1.76)], older children (5-9 years old) [10.02 (6.71, 14.97), and children from the richest quintile [2.91 (1.20, 7.05)] had increased odds of overweight. After adjusting for covariates including age, sex, region, wealth index and inflammation, children with anaemia, low serum retinol and low serum zinc had increased odds of stunting and underweight than non-micronutrient deficient children (AOR = 1.43-1.71). Compared to children without MNDs, those with ≥3 MNDs had almost double the odds of stunting and underweight, whereas those with ≤3 MNDs had reduced odds of overweight (AOR = 0.38−0.60).

TBM is becoming a major public health concern for Vietnamese children, and significant demographic variation and socioeconomic inequalities in malnutrition were observed. These findings suggest that national policies and programmes in Vietnam should address age-specific, sex-specific, geographical and socioeconomic disparities to accelerate progress in reducing child malnutrition.

Although in recent decades progress has been made in reducing the global burden of micronutrient deficiencies (MNDs), nonetheless MNDs prevalence remains high in some populations(1). Among the micronutrients, deficiencies in iron, zinc and vitamin A are particularly prevalent, and causally associated with adverse health outcomes for children and adolescents. The aim of this study was to examine the prevalence and associations between MNDs and components of the double burden of malnutrition in female adolescents utilizing the nationally representative Vietnam General Nutrition Survey 2020.

1,471 female adolescents were included and categorised into: pre-adolescent (10-12-year-old), middle-adolescent (13-15-year-old), and late-adolescent (16-18-year-old) groups. Biomarkers of micronutrients, anthropometry, and sociodemographic data were selected for analyses. WHO growth reference values were applied to define stunting (height-for-age z-score (HAZ) < −2 SD), thinness (BMI-for-age z-score (BAZ) < −2 SD) and overweight (BAZ > 1). Iron deficiency was defined as serum ferritin<15μg/L (<70 μg/L for individuals with inflammation), vitamin A insufficiency (VAI) as serum retinol<1.05 μmol/L, zinc deficiency was based on Zinc International Consultative Group Recommendation, and inflammation as C-reactive protein >5 mg/L or α1-acid glycoprotein >1 g/L. One or multiple MNDs were defined as individuals with ≥ 1 MNDs.

Data were analysed by STATA 17 (STATA Corporation, US). Descriptive and logistic regression analyses were performed based on the sampling weight adjustments for the estimation at population level. Multivariate logistic regressions between MNDs and growth indicators were presented as crude (COR) and 95% confidence intervals or adjusted odds ratio (AOR), after adjusting for sociodemographic variables (age, ecological area, demographic, ethnicity, and wealth index) and inflammation.

Compared to the pre-adolescent group, prevalence of stunting doubled in late adolescence (12.4% to 26.8%). Meanwhile, the prevalence of overweight and thinness decreased with age (29.7% to 22.0% and 9.4% to 2.7%, respectively). Overall, the prevalence of MNDs increased with age. Zinc deficiency occurred most often in the entire cohort (39.8% in 10-18) and contributed the most to multi-MNDs. Prevalence of one or multiple MNDs increased with age, from 58.1% in pre-adolescence to 80% in late adolescence. In logistic regression analyses adjusting for inflammation and sociodemographic factors, stunting increased the risk of having ≥1 MNDs (AOR: 1.94 (1.3, 2.88), p=0.002), but not individual MND alone. Thinness increased the risk of VAI (AOR: 2.38 (1.34, 4.22), p=0.005). No association was found between any MND and overweight, although overweight was protective against zinc deficiency (COR: 0.76 (0.59, 0.99), p=0.04) in the unadjusted model.

Female late adolescents in Vietnam had a higher prevalence of stunting and MNDs, but lower prevalence of thinness and overweight in comparison to pre-adolescents. Stunting was a risk factor associated with having one or multiple MNDs. Thinness was associated with higher risk of VAI. No association was found between overweight and MNDs in this cohort.

The status of the genera Euparagonimus Chen, 1963 and Pagumogonimus Chen, 1963 relative to Paragonimus Braun, 1899 was investigated using DNA sequences from the mitochondrial cytochrome c oxidase subunit I (CO1) gene (partial) and the nuclear ribosomal DNA second internal transcribed spacer (ITS2). In the phylogenetic trees constructed, the genus Pagumogonimus is clearly not monophyletic and therefore not a natural taxon. Indeed, the type species of Pagumogonimus,P. skrjabini from China, is very closely related to Paragonimus miyazakii from Japan. The status of Euparagonimus is less obvious. Euparagonimus cenocopiosus lies distant from other lungflukes included in the analysis. It can be placed as sister to Paragonimus in some analyses and falls within the genus in others. A recently published morphological study placed E. cenocopiosus within the genus Paragonimus and probably this is where it should remain.

Competition among the two-plasmon decay (TPD) of backscattered light of stimulated Raman scattering (SRS), filamentation of the electron-plasma wave (EPW) and forward side SRS is investigated by two-dimensional particle-in-cell simulations. Our previous work [K. Q. Pan et al., Nucl. Fusion 58, 096035 (2018)] showed that in a plasma with the density near 1/10 of the critical density, the backscattered light would excite the TPD, which results in suppression of the backward SRS. However, this work further shows that when the laser intensity is so high ($>{10}^{16}$ W/cm2) that the backward SRS cannot be totally suppressed, filamentation of the EPW and forward side SRS will be excited. Then the TPD of the backscattered light only occurs in the early stage and is suppressed in the latter stage. Electron distribution functions further show that trapped-particle-modulation instability should be responsible for filamentation of the EPW. This research can promote the understanding of hot-electron generation and SRS saturation in inertial confinement fusion experiments.

Two-dimensional particle-in-cell (PIC) simulations have been used to investigate the interaction between a laser pulse and a foil exposed to an external strong longitudinal magnetic field. Compared with that in the absence of the external magnetic field, the divergence of proton with the magnetic field in radiation pressure acceleration (RPA) regimes has improved remarkably due to the restriction of the electron transverse expansion. During the RPA process, the foil develops into a typical bubble-like shape resulting from the combined action of transversal ponderomotive force and instabilities. However, the foil prefers to be in a cone-like shape by using the magnetic field. The dependence of proton divergence on the strength of magnetic field has been studied, and an optimal magnetic field of nearly 60 kT is achieved in these simulations.

Keratin-associated proteins (KAPs) are constituents of wool and hair fibres and are believed to play an important role in determining the characteristics of the fibres. In the current study, a polymerase chain reaction-single stranded conformational polymorphism (PCR-SSCP) approach was used to screen for variation in the ovine KAP15-1 gene (KRTAP15-1). Four PCR-SSCP banding patterns, representing four different variants (named A to D), were detected. Four single nucleotide polymorphisms were found within the coding region and three of these were non-synonymous. The effect of this genetic variation on wool traits was investigated in 396 Merino × Southdown-cross sheep. Of the three variants found in these sheep (A, B and C), the presence of B was found to be associated with decreased wool yield, while C was associated with increased wool yield and decreased fibre diameter standard deviation. Sheep of genotype AC had a higher wool yield than those of genotype AA or AB.

Hepatitis E is an important zoonosis that is prevalent in China. Hepatitis E virus (HEV) is a pathogen that affects humans and animals and endangers public health in China. In this study, the detection of HEV epidemics in swine in Sichuan Province, China, was carried out by nested real-time PCR. A total of 174 stool samples and 160 bile samples from swine in Sichuan Province were examined. In addition, software was used to analyse the biological evolution of HEV. The results showed that within 2 years of swine HEV (SHEV) infection in China, SHEV was first detected in Sichuan Province. HEV was endemic in Sichuan; the positive rate for pig farms was 11.1%, and the total positive sample rate was 10.5%. The age of swine with the highest positive rate (17.9%) was 5–9 weeks. The examined swine species in order of highest to lowest HEV infection rates were Chenghua pig, Large White, Duroc, Pietrain, Landrace and Hampshire. Nucleotide and amino acid sequence analysis showed that the HEV epidemic in swine in Sichuan Province was related to genotype IV, which had the highest homology to HEV in Beijing. Sichuan strains have greater variation than Chinese representative strains, which may indicate the presence of new HEV strains.

Heat shock proteins (HSPs) participate in diverse physiological processes in insects, and HSP70 is one of the most highly conserved proteins in the HSP family. In this study, full-length cDNAs of three HSP70 genes (Lthsc70, Lthsp701, and Lthsp702) were cloned and characterized from Liriomyza trifolii, an important invasive pest of vegetable crops and horticultural crops worldwide. These three HSP70s exhibited signature sequences and motifs that are typical of the HSP70 family. The expression patterns of the three Lthsp70s during temperature stress and in different insect development stages were studied by real-time quantitative PCR. Lthsp701 was strongly induced by high- and low-temperature stress, but Lthsc70 and Lthsp702 were not very sensitive to temperature changes. All three Lthsp70s were expressed during insect development stages, but the expression patterns were quite different. The expression of Lthsc70 and Lthsp702 showed significant differences in expression during leafminer development; Lthsc70 was most highly expressed in female adults, whereas Lthsp702 was abundantly expressed in larvae and prepupae. Lthsp701 expression was not significantly different among leafminer stages. These results suggest that functional differentiation within the LtHSP70 subfamily has occurred in response to thermal stress and insect development.

The objectives of this study were to determine the effect and mode of action of Saccharomyces cerevisiae (YST2) on enteric methane (CH4) mitigation in pigs. A total of 12 Duroc×Landrace×Yorkshire male finisher pigs (60±1 kg), housed individually in open-circuit respiration chambers, were randomly assigned to two dietary groups: a basal diet (control); and a basal diet supplemented with 3 g/YST2 (1.8×1010 live cells/g) per kg diet. At the end of 32-day experiment, pigs were sacrificed and redox potential (Eh), pH, volatile fatty acid concentration, densities of methanogens and acetogens, and expression of methyl coenzyme-M reductase subunit A gene were determined in digesta contents from the cecum, colon and rectum. Results showed that S. cerevisiae YST2 decreased (P<0.05) the average daily enteric CH4 production by 25.3%, lowered the pH value from 6.99 to 6.69 in the rectum, and increased the Eh value in cecum and colon by up to −55 mV (P<0.05). Fermentation patterns were also altered by supplementation of YST2 as reflected by the lower acetate, and higher propionate molar proportion in the cecum and colon (P<0.05), resulting in lower acetate : propionate ratio (P<0.05). Moreover, there was a 61% decrease in Methanobrevibacter species in the upper colon (P<0.05) and a 19% increase in the acetogen community in the cecum (P<0.05) of treated pigs. Results of our study concluded that supplementation of S. cerevisiae YST2 at 3 g/kg substantially decreased enteric CH4 production in pigs.

Based on unstructured hybrid grid and dynamic overlapping grid technique, numerical simulations of Unsteady Reynolds Averaged Navier-Stokes equations were performed and investigation on isolated propeller aerodynamic characteristics and effects of propeller slipstream on turboprops were undertaken. The computational grid consisted of rotational subzone of propeller and stationary major-zone of aircraft, and walls criterion was used in the automatic hole-cutting procedure. Distance weight interpolation and tri-linear interpolation were developed to transfer information between the rotational and stationary subzones. The boundaries of overlapping grids were optimized for fixed axis rotation. The governing equations were solved by dual-time method and Lower Upper-Symmetric Gauss-Seidel method. The method and grid technique were verified by isolated propeller configuration and the computational results were in well agreement with the experimental data. The grid independence was studied to establish the numerical results. Finally, the flow around a turboprop case was simulated and the influence of propeller slipstream was presented by analyzing the surface pressure contours, profile pressure distribution, vorticity contours and profile streamline. It's indicated that the slipstream accelerates and rotates the free stream flow, changing the local angle of attack, enhancing the downwash effects, affecting the pressure distribution on wing and horizontal tail, as well as increasing the drag coefficient, pitching moment coefficient and the slope of lift coefficient.

A new trajectory tracking control approach for an under-actuated stratospheric airship is proposed. There is a two-level structure of the proposed controller. A low-level controller based on non-linear vectorial backstepping method, with the rigid-body dynamics expressed on vector form, stabilises the attitude and velocity of the airship, while a high-level controller performs guidance and trajectory tracking task in the three-dimensional (3D) space. Furthermore, a control allocation module based on the active set algorithm is incorporated into the low-level controller to optimise the practical control inputs under constraints of actuator saturation. The closed-loop trajectory tracking control plant is proved to be globally exponentially stable through the Lyapunov theory. Finally, simulations show that the vectorial backstepping trajectory tracking controller can achieve desired tracking performances even if the airship is affected by parametric uncertainties and exogenous disturbances.
