The purpose of the present study was to assess changes in the levels of cytochrome oxidase (CO) activity in the dorsal lateral geniculate nucleus (dLGN) of the adult cat following removal of primary visual cortical areas 17 and 18 on the day of birth (P1), P28, or in adulthood (≫6 months). Cytochrome oxidase activity was measured in histological sections 9 or more months after the cortical ablation. Control measures obtained from intact cats show that CO activity is normally highest in the A-laminae of dLGN, and slightly lower in the C-complex. Following visual cortex ablations incurred at any age, CO activity levels are reduced in the A-laminae. This reduction is most profound following ablations incurred on P28 or in adulthood. In contrast, CO activity in the C-complex of dLGN is at nearly normal levels following ablations on P1 or P28, but not in adulthood. These findings contribute to our understanding of the role played by the dLGN in the transfer of visual signals along retino-geniculo-extrastriate pathways that expand following early removal of areas 17 and 18. Moreover, they have implications for our understanding of spared behavioral functions attributed to the extrastriate cortex in cats which incurred early damage of areas 17 and 18.

Rod bipolar cells constitute the second-order neuron in the rod pathway. Previous investigations of the rabbit retina have evaluated the development of other components of the rod pathway, namely the dopaminergic and All amacrine cell populations. To gain further insights into the maturation of this retinal circuitry, we studied the development of rod bipolar cells, identified with antibodies directed to the α isoform of protein kinase C (PKC), in the rabbit retina. Lightly immunostained PKC-immunoreactive (IR) somata are first observed at postnatal day (PND) 6 in the distal inner nuclear layer (INL). Immunostaining is also observed in the outer plexiform layer (OPL), indicating the presence of PKC-IR dendrites. PKC-IR axons are present in the INL oriented toward the inner plexiform layer (IPL). Several of them terminate with enlarged structures resembling growth cones. At PND 8, some immunostained terminal bulbs, characteristic of rod bipolar cells, are detected in the proximal IPL. PKC-IR cells at PND 11 (eye opening) display stronger immunostaining and more mature characteristics than at earlier ages. The dendritic arborizations of these cells in the OPL and their axon terminals in the IPL attain mature morphology at later ages (PND 30 or older). The density of PKC-IR cells shows a peak at PND 11 followed by a drastic decrease up to adulthood. The total number of PKC-IR cells increases from PND 6 to PND 11 and then it remains almost unchanged until adulthood. The mosaic of PKC-IR cells is nonrandom in some retinal locations at PND 6, but the overall regularity index at PND 6 is lower than at older ages. The present data provide a comprehensive evaluation of the development of rod bipolar cells in the postnatal rabbit retina and are consistent with those previously reported for dopaminergic and All amacrine cell populations, indicating that different components of the rod pathway follow a similar pattern of maturation, presumably allowing the rod pathway to be functional at eye opening.

This study describes the connectivity between horizontal cells and short-wavelength-sensitive (SWS) cones in macaque monkey retina. H1 and H2 horizontal cells were either labelled with the carbocyanine dye, Dil, or injected intracellularly with Neurobiotin. The retinas were then processed with an antiserum against human SWS cone pigment, which usually stained the entire SWS cone. In these double-labelled retinas, the pattern of connectivity of H1 (n = 91) and H2 (n = 7) cells with SWS cones has been determined. About 85% of the H1 cells examined do not contact SWS cones. The dendritic terminal knobs of five H1 cells that do contact SWS cones were counted. They have, at most, 3% of their dendritic terminal knobs at SWS cones. All H2 cells examined make contact with SWS cones. The dendritic terminal knobs of one H2 cell were counted; about 11% of the dendritic terminal knobs are at the SWS cone. We conclude that horizontal cells in macaque monkey retina show specific patterns of connectivity to SWS cones.

Previous authors have hypothesized that retinotopic projections may be influenced by ‘preordering’ of the axons as they grow towards their targets. In some nonmammalian species, axons are reorganized at or near the optic nerve head to establish a retinotopic order. Data are ambiguous concerning the retinotopy of the mammalian retinal nerve fiber layer and whether fibers become reorganized at the optic nerve head. We have examined this question in fetal and newborn ferrets (Mustela putorius furo) by comparing the arrangement of axons in the retinal nerve fiber layer with that in the optic nerve. Dil or DiA crystals were implanted into fixed tissue in the innermost layers of the retinal periphery, or at a location midway between the periphery and the optic nerve head. Fluorescence labelling was examined in 100–200 μm Vibratome sections, or the eyecup and nerve were photooxidized and 1–2 μm longitudinal or transverse sections were examined. Regardless of fetal age, eccentricity or quadrant of the implant site, a segregation of labelled peripheral axons from unlabelled central ones was not detected within the nerve fiber layer. Axons coursed into the nerve head along the margin of their retinal quadrant of origin, often entering the optic nerve as a radial wedge, thus preserving a rough map of retinal circumference. However, peripheral axons were in no way restricted to the peripheral (nor central) portions of the nerve head or nerve, indicating that the optic axons do not establish a map of retinal eccentricity. Our results demonstrate that (1) the nerve fiber layer is retinotopic only with respect to circumferential position and (2) optic axons are not actively reorganized to establish a retinotopic ordering at the nerve head. The present results suggest that any degree of order present within the optic nerve is a passive consequence of combining the fascicles of the retinal nerve fiber layer; optic axons are not instructed to establish, nor constrained to maintain, a retinotopic order within the optic nerve.

NADPH diaphorase histochemistry is commonly used to identify cells containing nitric oxide synthase (NOS), the enzyme catalyzing the production of nitric oxide from L-arginine. NADPH diaphorase activity and NOS immunostaining was demonstrated in different cells of the vertebrate retina; photoreceptors, horizontal cells, amacrine cells, ganglion cells, and Müller cells. However, the physiological role of nitric oxide (NO) in the retina has yet to be elucidated. In this study, we tested the assumption that NADPH diaphorase activity in the retinas of rabbits and rats depended on the state of visual adaptation. In the rabbit, light adaptation enhanced NADPH diaphorase activity in amacrine cells and practically eliminated it in horizontal cells. Dark adaptation induced the opposite effects; the NADPH diaphorase activity was reduced in amacrine cells and enhanced in horizontal cells. Retinas from eyes that were injected intravitreally with L-glutamate exhibited a pattern of NADPH diaphorase activity that was similar to that seen in dark-adapted retinas. In rats, the NADPH diaphorase activity of amacrine and horizontal cells exhibited adaptation dependency similar to that of the rabbit retina. But, the most pronounced effect of dark adaptation in the rat's retina was an enhancement of NADPH diaphorase activity in Müller cells, especially of the endfoot region. Assuming that NADPH diaphorase activity is a marker for NOS, these findings suggest that NO production in the mammalian retina is modulated by the level of ambient illumination and support the notion that NO plays a physiological role in the retina.

A model of contrast discrimination performance in human observers is developed and then extended to cover effects on performance of anticholinergic drugs. It is shown that it is necessary to assume that neural noise increases at high spatial frequencies in order to provide a satisfactory model of variations in discrimination performance with spatial frequency. The model results are compared with the results of empirical studies in which the effects of the muscarinic antagonist scopolamine (hyoscine) on contrast discrimination performance in human observers are examined. The purpose of the pharmacological work is to test the hypothesis that the differential contrast gain found psychophysically at different spatial frequencies might reflect differential facilitation by extrinsic cholinergic neurons. Contrast discrimination and contrast increment detection are found to be impaired by scopolamine in a manner that depends on both spatial frequency and base contrast. By comparing the empirical data with the predictions of the model, it is concluded that contrast constancy may reflect differential cholinergic modulation.

The effects of dark adaptation on the response properties of ganglion cells have been documented extensively in the cat retina. To pinpoint the different retinal mechanisms that underlie these effects, we studied the response characteristics of cat horizontal (H) cells during prolonged dark adaptation. H-cell responses were recorded intracellularly in the optically intact, in vivo eye. To disentangle rod and cone contributions, sensitivity changes during dark adaptation were tracked with white light and with monochromatic lights that favored either rod or cone excitation. Stable, long-lasting recordings allowed us to measure changes of sensitivity for adaptation periods up to 45 min. Thresholds for white light and 503-nm monochromatic light decreased steadily and in parallel. The maximum increase of sensitivity, after extinguishing a photopic adaptation light, was 1.8 log units only, reached after about 35 min. Sensitivity for 581-nm lights also increased steadily, but at a shallower slope. The steady increase of sensitivity was concomitant with a linear shift in resting membrane potential and with an increase in relative rod contribution to the threshold responses. Even though small-amplitude responses were rod dominated after prolonged dark adaptation, sensitivity to rod signals remained relatively low, compared to sensitivity of cone responses or to the absolute sensitivity of ganglion cells. This suggests that the cone-H-cell pathway plays no role in the dark-adapted cat retina.

The Drosophila ninaC locus encodes two retinal specific proteins (p132 and p174) both consisting of a protein kinase joined to a myosin head domain and a C terminal with a calmodulin-binding domain. The role of p132 and p174 was studied via whole-cell recording and through measurements of the pupil mechanism, i.e. the pigment migration in the photoreceptor cells, in the ninaC mutants, P[ninaCΔ132] (p132 absent), P[ninaCΔ174] (p174 absent), and ninaCp235 (null mutant). Voltage-clamped flash responses in P[ninaCΔ174] and ninaCp235 showed delayed response termination. In response to steady light, plateau responses in both P[ninaCΔ174] and ninaCp235 were also large. In both cases the defect was significantly more severe in ninaCp235. Responses in P[ninaCΔ132] were apparently normal. P[ninaCΔ174] and ninaCP235 were also characterized by spontaneous quantum bump-like activity in the dark and by larger and longer light-induced quantum bumps. The turn-off of the pupil mechanism in P\ninaCΔ174] and ninaCp235 was also defective, although in this case the rate of return to baseline in both mutants was more or less the same. In all ninaC mutants, the amplitudes of the pupillary pigment migration were distinctly smaller than that in the wild type. The reduction of the amplitude was largest in P[ninaCΔ174]. The light sensitivity of the pupil mechanism of P[ninaCΔ174] compared to that of wild type was reduced by 1.3 log units. Remarkably, the light sensitivity of P[ninaCΔ132] and ninaCP235 was ca. 0.5 log units higher than that of the wild type. The results suggest that the p174 protein is required for normal termination of the transduction cascade. The diverse phenotypes observed may suggest multiple roles for calmodulin distribution for controlling response termination and regulating pigment migration in Drosophila photoreceptors.

We studied excitatory local circuits in the macaque primary visual cortex (V1) to investigate their relationships to the magnocellular (M) and parvocellular (P) streams. Sixty-two intracellularly labeled spiny neurons in layers 2–5 were analyzed. We made detailed observations of the laminar and columnar specificity of axonal arbors and noted correlations with dendritic arbors. We find evidence for considerable mixing of M and P streams by the local circuitry in V1. Such mixing is provided by neurons in the primary geniculate recipient layer 4C, as well as by neurons in both the supragranular and infragranular layers. We were also interested in possible differences in the axonal projections of neurons with different dendritic morphologies. We found that layer 4B spiny stellate and pyramidal neurons have similar axonal arbors. However, we identified two types of layer 5 pyramidal neuron. The majority have a conventional pyramidal dendritic morphology, a dense axonal arbor in layers 2–4B, and do not project to the white matter. Layer 5 projection neurons have an unusual “backbranching” dendritic morphology (apical dendritic branches arc downward rather than upward) and weak or no axonal arborization in layers 2–4B, but have long horizontal axonal projections in layer 5B. We find no strong projection from layer 5 pyramidal neurons to layer 6. In macaque V1 there appears to be no single source of strong local input to layer 6; only a minority of cells in layers 2–5 have axonal branches in layer 6 and these are sparse. Our results suggest that local circuits in V1 mediate interactions between M and P input that are complex and not easily incorporated into a simple framework.

In the present study, the early postnatal distribution and subsequent fate of visual callosal neurons were studied in neonatal rat pups. Previous studies had indicated that the adult pattern of visual callosal neurons was sculpted from an initially uniform distribution in the neonatal cortex. To reexamine this issue, we used a sensitive tracer, latex microspheres conjugated either to rhodamine or fluorescein, that was injected into the occipital cortex of one hemisphere in pups on the day of birth (PND 1), PND 6, or PND 12. Examination of the resulting retrograde labeling of cortical neurons in the opposite hemisphere indicates that features of the mature visual callosal pattern are present as early as PND 1. At all stages of postnatal development, the relative density of callosal projection cells varies consistently across the mediolateral extent of primary visual cortex —it is always highest in the region of the 17/18a border and lowest in the body of area 17. These data strongly suggest that, from the outset, visual cortical neurons in the region of the 17/18a border preferentially make connections with the opposite hemisphere. The results of experiments in which callosal neurons were labeled on the day of birth indicate that only those neurons that have migrated to their final cortical destinations have extended callosal axons into the vicinity of the visual cortex in the opposite hemisphere. The initial pattern of callosal neurons resembles a dense, compact version of the mature one, and the present study suggests that much of the remaining change in the appearance of this pathway may be accounted for by the decrease in the overall density of neurons that is due to expansion of the cortical gray matter during postnatal life. Taken together, these results suggest that the development of the visual callosal pathway in the rat may be more similar to that in the monkey than has been reported previously.

The generation of compensatory eye movements in response to rotational head movements involves the transformation of visual-optokinetic and vestibular signals into commands controlling the appropriate eye muscles. Previously, it has been shown that the three systems (optokinetic, vestibular, and eye muscle) share a similar three-dimensional reference frame. In this report, we suggest that a peculiarity in the structure of the horizontal recti in pigeons demonstrates that the optokinetic system is organized with respect to the eye muscles rather than the vestibular canals. Measurements of the orientation of the plane for each of the lateral and medial recti were obtained. These were compared with the direction preferences of optokinetic neurons responsive to horizontal motion, namely “back” units in the nucleus of the basal optic root (nBOR), “forward” units in the pretectal nucleus lentiformis mesencephali (LM), and “vertical axis” (VA) Purkinje cells in the flocculus. The average direction preference of LM neurons excited in response to forward (temporal to nasal) visual motion, and VA Purkinje cells in response to optokinetic motion in the ipsilateral visual field was approximately parallel to the visual horizontal. This corresponded to the orientation of the medial rectus, which was also approximately parallel to the visual horizontal. The average direction preference of nBOR neurons excited in response to backward (nasal to temporal) visual motion, and VA Purkinje cells in response to optokinetic motion in the contralateral visual field was approximately 20–30 deg down from the visual horizontal. The orientation of the lateral rectus was also approximately 20–30 deg down from the visual horizontal. These data suggest that the incoming optokinetic signals are organized with respect to the outgoing extraocular muscle commands.

Brn-3a, Brn-3b, and Brn-3c are highly homologous POU-domain transcription factors that are expressed in subsets of retinal ganglion cells. From each of the mouse Brn-3 genes, a DNA segment ranging in size from 4.6 to 13.4 kb and located immediately upstream of the start site of translation was joined to a human placental alkaline phosphatase (AP) reporter cDNA. Following the introduction of each construct into the mouse germline, a total of 19 transgenic lines were obtained, of which 16 expressed the AP reporter in the retina. Unexpectedly, at least 14 of the 16 expressing lines showed AP activity in subsets of amacrine cells, and these subsets typically differed among mouse lines injected with the same construct. Transgene expression was also found in ganglion cells in four lines and bipolar cells in seven lines. In all cases AP activity was confined to cells in the inner nuclear layer and the ganglion cell layer. The expression of Brn-3 transgenes in multiple cell types in the inner retina is reminiscent of earlier experiments in which visual pigment transgenes were found to be expressed in multiple cell types in the outer retina. Taken together, these observations suggest that anatomically and/or functionally related retinal neurons contain partially overlapping transcriptional regulatory specificities.

Fast, multiple-site optical recording of voltage-sensitive dye (VSD) signals and intracellular microelectrode recordings were combined to characterize visually evoked neuronal responses in the visual cortex of the pond turtle, Pseudemys scripta. By using an in vitro, eye-brain preparation stained with the merocyanine oxazolone voltage-sensitive dye, NK-2495 or a close analog, NK-2761, large VSD signals relatively free of vibrational noise could be recorded in single trials following a stroboscopic light flash to the contralateral eye. VSD signals recorded from the same cortical location in repeated trials exhibited considerable variability in the onset, duration, and amplitude of secondary depolarizations. Because of this variability, secondary depolarizations were largely absent in signal-averaged responses. Superposition of VSD signals with intracellular recordings obtained from cortical pyramidal cells revealed a close correspondence between their signal waveforms. The two signals were virtually identical in their onset, initial rate of rise, and time-to-peak. At later periods (>500 ms), the correspondence was less close, especially for large cortical depolarizations. Some of this disparity could be attributed to contamination of the VSD signal by a large intrinsic optical response. A second contribution was a failure of the VSD signal to register asynchronous regenerative effects occurring in single pyramidal cells. It is suggested that the close correspondence between the microelectrode and optical recordings in the early phase of the response may reflect the organization of pyramidal cells into clusters that receive virtually identical synaptic inputs.

The passive electrical properties of 17 ON-OFF retinal ganglion cells were derived from electrophysiological recordings. The parameters for each cells' equivalent model were obtained from the transient current responses to small step changes in clamp potential. Thirteen of the cells could be adequately approximated by a spherical soma connected to an equivalent dendritic cable. Estimates for the cell input conductance (GN), membrane time constant (τm), the dendritic-to-soma conductance ratio (ρ), and the normalized electrotonic length (L) were obtained (mean ± standard deviation, n = 13): GN = 580 ± 530 pS, τm = 97 ±72 ms, ρ = 2.8 ± 2.8, and L = 0.34 ± 0.13. Series resistance averaged 32 ± 11 MΩ The mean of the derived soma diameters was 18 ± 6 μm and the mean diameter and length of the equivalent cables were 1.4 ± 0.6 and 470 ± 90 μm, respectively. The average of the specific membrane conductances, 1.67 ± 1.08 S/cm2, corresponded to a membrane resistivity of 60 kΩ-cm2. Computer simulations of synaptic inputs were performed on a representative model, with an electrode at the soma and using the worst-case configuration, in which all synaptic inputs were confined to the tips of the dendrites. We draw three conclusions from the modeling: (1) Under voltage clamp, fast, spontaneous EPSCs would be significantly attenuated and slowed while the time course of the slower, light-evoked non-NMDA and NMDA EPSCs would be minimally distorted by dendritic filtering. (2) Excitatory synaptic reversal potentials can be accurately determined under voltage clamp. (3) In the absence of GABAergic and glycinergic inhibition, the efficacy at the soma of excitatory conductance changes is essentially independent of their dendritic location. The specific membrane resistivity appears to represent a good compromise between having a small membrane time constant and minimal EPSP attenuation.

Two synapse-rich layers are well recognized in the mammalian retina, the inner and outer plexiform layers. However, synapses occur also in other layers, particularly in the innermost nerve fiber layer. These synapses form a tenuous layer at times referred to as the superficial plexiform layer. We have found that staining for GABAA receptors in whole-mounted rabbit retina demonstrates this layer. It is most well developed in the region of the visual streak 2–4 mm below the center of the myelinated streak and is very sparse in other parts. Most or all of the processes in the plexus originate from cells in the ganglion cell layer.
