In May-June 1992 cases of infection with verocytotoxin-producing (VT+) Escherichia coli O157 in South Yorkshire could have been associated with prior consumption of beef from a local abattoir. During investigation of the abattoir, bovine rectal swabs and samples of meat and surface swabs from beef carcasses were examined for E. coli O157, isolates of which were tested for toxigenicity, plasmid content and phage type. E. coli O157 was isolated from 84 (4%) of 2103 bovine rectal swabs; of these 84, 78 (93%) were VT+, the most common phage types being 2 and 8, the types implicated in the cluster of human cases. Positive cattle were from diverse sources within England. E. coli O157 was isolated from 7 (30%) of 23 carcasses of rectal swab-positive cattle and from 2 (8%) of 25 carcasses of rectal swab-negative cattle. The study has shown that cattle may be a reservoir of VT+ E. coli O157, and that contamination of carcasses during slaughter and processing may be how beef and beef products become contaminated and thereby transmit the organism to man.

1. Experiments have been carried out to determine the relation between the moisture content of artificially dried grass and the relative humidity of the surrounding atmosphere. This relation is given by a smooth curve. At relative humidities which are typical of outdoor winter conditions in the British Isles (80–90%) the equilibrium moisture content lies between 18 and 30%.
2. The rate of moulding is directly related to the relative humidity and therefore to the moisture content of the dried grass. For reasonably safe storage a relative humidity of 67 % should not be exceeded. This corresponds to a moisture content of roughly 13 %. Even this low value does not confer absolute immunity from mould growth.
3. The importance of extending storage trials over long periods is stressed. In one instance storage for 300 days was required before mould growth occurred. Neglect of this fact may account for the widely held view that a relative humidity of 70–75 % provides safe storage conditions.
4. Attempts to detect mould growth at an early stage by plating methods proved unsuccessful. It is shown that the two earliest and most reliable indications of mould growth are (i) the appearance of visible mycelium when the product is examined under a low-power objective, and (ii) the detection of a musty smell.

1. Experiments were made on the respiration of bran at different moisture levels using a continuous absorption apparatus in which the humidity of the air-stream was adjusted to be at equilibrium with the respiring bran. The rate of respiration was accelerated with increasing moisture content and was very rapid deterioration due to the high respiration rate and microbiological damage.
2. Experiments were made on the storage of bran at known moisture contents in closed tins in the laboratory and also in sacks kept under farm conditions. The total nitrogen content of the bran stored in the tins appeared to increase during storage. From similar increases in total phosphorus, it is clear that the increase in total nitrogen was caused by a loss of dry matter from the stored feeding stuff, a fact which was further demonstrated by the loss in dry weight of the sacks of bran with attributed to two causes: (1) the respiration of the plant cells still active in the bran itself, and (2) the respiration of developing micro-organisms. It is evident from these experiments that fresh bran, free from insect infestation and stored at a moisture content of less than 13%, will have a very low respiration rate and will, therefore, not be liable to heating. Bran of 15–19% moisture content respires at a much faster rate and the time for which such material can be stored with freedom from heating or other types of deterioration will be limited.

1. A study has been made of the assimilation and retention of nitrogen, phosphorus and calcium by calves fed on raw and pasteurized milks. Sixteen calves were used in the experiments.
2. No significant differences were found between the two types of milk.
3. An examination of the percentage retentions of nitrogen, phosphorus and calcium indicates that milk is relatively deficient in calcium and to a less extent in phosphorus when used as the sole food for calves. The N/Ca balance can be improved by including pasture grass or hay in the ration.

1. As regards their nutritive value for milk production, the proteins of spring grass are markedly superior to those of autumn grass. This is shown not only from a study of milk yield, but from the increased excretion of N and S in the urine, the increased creatine excretion, and the lower biological value given by autumn grass.
2. A study of the N and S metabolism of cows receiving fresh and artificially dried grass fails to reveal any significant differences in the nutritive value as a result of artificial drying. There are no significant differences between the biological values of the proteins of fresh and of artificially dried autumn grass. A comparison of the mean biological values of the proteins of fresh and of artificially dried spring grass indicates that the latter has an inferior value for milk production, but the differences largely disappear when the biological values are calculated to a common standard.

The results of the present experiments may be summarised as follows:
(1) When minimal quantities of protein are fed in the maintenance rationof steers, a deficiency of lysine will lead to a marked increase in the urinary N output. A deficiency of cystine has no effect.
(2) The catabolism of body tissue is shown by the high creatine excretionand the S: N ratio of the excess S and N excreted. This is probably the resultof excessive deamination with the production of a mild acidosis.
(3) As regards urinary partition of N there is some indication that alysine-deficient diet causes a decreased percentage excretion of urea N and anincrease of ammonia N.
(4) The biological values have been calculated, the only marked difference occurring with wheat gluten, a protein very deficient in lysine
(5) An approximate value for the lysine requirements for maintenance has been calculated, i.e. 0·80 g. lysine N per 1000 lb. live weight.

1. The effect of increasing concentrations of milk solids on the degree of ionisation of electrolytes cannot be measured by determinations of electrical conductivity, owing to the influence of viscosity on ionic mobility.
2. An approximately linear relationship exists between concentration of milk solids and the freezing-point depression. Above a certain concentration (equivalent to 34 g. milk solids per 100 g. of water), however, the solutions are saturated with respect to lactose, and any increase in freezing-point depression beyond this point is limited to that due to the other osmotically active constituents of milk.
3. There is no evidence of the existence of “bound” water in solution of milk solids up to concentrations equivalent to 65 g. milk solids per 100 g. water.
4. An approximately linear relationship exists between concentration of milk solids and hydrogen-ion concentration, increase in the concentration of milk solids resulting in a markedly increased acidity.
5. The applications of these results to the heat coagulation of evaporated and dried milks are briefly discussed.

The results of the present experiments and of those reported in the previous paper(1) may be summarised as follows:
1. When minimal quantities of protein are fed in the production rations of milking cows, a deficiency of either lysine or tryptophane will lead to a marked reduction in milk yield. There is, however, some evidence of the storage of reserve N, which can be utilised when the food protein is inadequate.
2. The feeding of a lysine- or tryptophane-deficient ration causes a marked increase in urinary N, indicating a poor utilisation of food protein. On the other hand, the feeding of a ration containing adequate quantities of these essential amino acids reduces the urinary N, indicating efficient protein utilisation.
3. The utilisation of body tissue in an attempt to maintain normal milk production on a deficient protein ration is shown by the high creatine excretion. The fact that the S: N ratio of the excess sulphur and nitrogen excreted during the deficient protein periods approximates that of body tissue (circa 1: 15·7) confirms this conclusion.

The protein requirements for maintenance and milk production are based on the well-known investigations of Kellner, Armsby, Haecker, Savage and others, who recommend feeding standards varying from 0·5 to 0·;8 lb. digestible crude protein per 1000 lb. live weight for maintenance, and from 0·52 to 0·63 lb. digestible crude protein per 10 lb. milk for milk production. A detailed study of the work of these investigations shows that in many instances nitrogen equilibrium has been obtained on considerably lower planes of protein intake: but the figures have been arbitrarily raised, partly to provide a sufficient safeguard against underfeeding, and partly on the assumption that the excess food protein could exert a stimulating effect on mammary secretion. Recent investigations, particularly those of Hills and his associates (l), Perkins (2), Buschmann(3) and Savage and Harrison (4) indicate that the existing protein standards could, in fact, be considerably reduced. The above investigations have, however, been limited to a study of the quantity of protein required for maintenance and milk production, and no account has been taken of the possible effect of variations in the quality of proteins on their utilisation.

In the previous paper(1) it was shown that the heat treatment to which a milk powder is subjected during manufacture may result in the production of two distinct types of protein insolubility, namely insolubility due to denaturation of the protein while still in solution, and insolubility caused by over-heating the milk powder while in a dry state. The insolubility due to denaturation was shown to be independent of the temperature of reconstitution of the milk powder. On the other hand the insolubility due to dry heating was found to vary with the temperature of reconstitution, the protein being relatively insoluble when reconstituted in water held at 20° C, but completely soluble when mechanically stirred for 30 min. in water held at 90° C. It was suggested that this latter type of insolubility might be due to the removal of water of imbibition from the protein particles, since such an explanation would account for the effect of hot water in restoring the solubility of the protein. In the previous paper, however, observations on the effect of temperature of reconstitution were limited to the two extreme temperatures, i.e. 20 and 90° C.

No reliable information is at present available with regard to the effect of process of manufacture on the solubility of the protein of milk powders. Hunziker(1), it is true, gives a very well-balanced discussion of the subject, but it is apparent from his summary of the views of other workers that these are seldom based on adequate experimental evidence. In fact the only points on which there is general agreement among workers in this field are that the proteins of spray-dried powders are, on the whole, extremely soluble, while those of roller-dried powders are relatively insoluble. Moreover, even where experimental data are available, there remain many unexplained points. For example, Hunziker shows that the solubility of the proteins of a milk powder may vary according to the temperature at which the powder is reconstituted—a higher temperature giving a greater solubility. Such an observation cannot at present be brought into line with current theories regarding the properties of denatured proteins.
