The Millennium Ecosystem Assessment estimates that between 60,000 and 100,000 plant species are threatened with extinction–equivalent to around one-quarter of the total number of known plant species. Why should we care? There are a number of reasons. The first is that these plants may be useful to us in unknown ways. Secondly, ecology has taught us that resilience is found in diversity. Thirdly, we should be saving plant species from extinction because we can–there is no technological reason why any plant species should become extinct. Where we can't protect and manage plant diversity in situ, we should be employing ex situ conservation techniques, ranging from seed banks to habitat restoration. The Millennium Ecosystem Assessment describes such interventions as ‘techno-gardening’. This is not an abstract concept–it is already a reality in the majority of man-managed landscapes. In this context the perception of ex situ conservation as simply a back-up strategy for in situ conservation is mistaken. We are all involved in ex situ conservation to some degree, from cultivating our back gardens, to farming, to management of protected areas. Ex situ conservation should be seen as a complementary approach to in situ conservation and on the same spectrum. Kew's Millennium Seed Bank Partnership, comprising more than 120 plant science institutions in 50 countries, epitomizes this philosophy in action. We work actively on every seed collection we bank, finding out how useful it is and how we can grow it to enable human innovation, adaptation and resilience. Challenges remain at the policy level; for example, the need to factor-in the value of natural capital to development decision making, and better defining a role for public-sector science. At the technical level, also, there is much to do. Perhaps the greatest technical challenges relate to the restoration and management of complex, self-sustaining habitats or species assemblages. If we are to techno-garden effectively, in order to maintain ecosystem services and sustain biodiversity, then a multidisciplinary approach will be required. Many plant science institutions have recognized this and are becoming engaged increasingly in restoration activities and in situ management. Ultimately, humanity's ability to innovate and adapt is dependent on our having access to the full range of plant species and the alleles they contain.

Starch is globally important as a source of food and, in addition, has a wide range of industrial applications. Much of this agriculturally produced starch is synthesized in developing seeds, where its biological function is to provide energy for seedling establishment. Storage starch in developing seeds is synthesized in heterotrophic plastids called amyloplasts and is distinct from the transient synthesis of starch in chloroplasts. This article reviews our current understanding of storage starch biosynthesis occurring in these organelles and discusses recent advances in research in this field. The review discusses starch structure and granule initiation, emerging ideas on the evolution of the pathway, the enzymes of starch synthesis, and the post-translational modification and regulation of key enzymes of amylopectin biosynthesis.

Fritillaria tubiformis subsp. moggridgei (Liliaceae) is a rare, endemic species that inhabits open mountains and alpine grasslands of the Ligurian Alps. At the time of seed dispersal, the underdeveloped embryos were 27% the length of the seed. Here we report the results of laboratory experiments carried out to determine the temperature preferences for embryo growth and radicle emergence. Embryo growth commenced immediately after sowing at 4°C. Once the embryo had grown the length of the seed, the radicle emerged. The time required for embryo growth and radicle emergence was longer when seeds were placed through a seasonal sequence of temperatures, commencing with late summer (10/20°C), compared with seeds immediately placed at a temperature to simulate winter conditions (4°C). Prematurely transferring seeds from winter to spring temperatures (5/10°C) also slowed the progress of germination. Radicle emergence did not occur at 10 or − 5°C and less than 20% germination occurred in seeds placed at constant 0°C. Addition of gibberellic acid (GA3) did not promote embryo growth of seeds placed at 20°C. Overall, the temperature preferences for embryo growth and subsequent radicle emergence are such that, in situ, seed germination may occur during the winter under snow cover or at the end of winter to coincide with snow melt and warming temperatures.

The present study evaluated the size and germination characteristics of seeds of seven species of Syngonanthus (distributed among four sections) that occur in different microhabitats in campos rupestres vegetation in Minas Gerais State, to determine if (1) the germination responses of the various species characterize a single germination pattern for the genus; and (2) there is a correlation between the germination characteristics and the geographic distribution as well as the microhabitat of the different species. The experiments were undertaken with recently collected seeds exposed to a 12-h photoperiod and continuous darkness, at constant temperatures of 10–40°C (at intervals of 5°C) in germination chambers. The results indicated the existence of a pattern of reduced seed size and light requirements for germination within the genus. The geographic distribution of different species of the genus cannot be explained by the responses of seed germination to different temperatures, but suggests some relationship to their edaphic microhabitats. Therefore, germination characteristics of the populations studied may have been selected to colonize specific environments at different soil water conditions.

Whether and how seed mass and shape are related to seed persistence in soil is a controversial topic in plant ecology. There is little information on this relationship in sandy habitats. We tested whether or not this pattern is retained among 141 species within a range of growth forms, from a wide range of families, in the Horqin sandy land of northern China, which belongs to the inland sand ecosystem. We collected information on seed mass, shape and persistence of these species in this region from published articles. Seed mass and shape were found to be significantly and negatively correlated to persistence in the soil in the Chinese flora, in the same way as in most other floras examined, but not in the same way as the Israeli coastal sand dune flora. The value of seed mass and shape as good predictors of persistence in the soil could be applied to the Horqin sandy land. Although many factors affect the relationship between seed mass, shape and persistence, ease of burial appears to be the main potential interpretation in the Horqin sandy land.

Strigolactones are apocarotenoids regulating shoot branching. They are also known to be exuded by plant roots at very low concentrations, stimulating hyphal branching of arbuscular mycorrhizal fungi and germination of root parasitic weed seeds. We show that strigolactones play a major role in host specificity of Orobanche and Phelipanche (the broomrapes) seed germination. This observation confirms that host-derived germination stimulants are an important component determining the host specificity of these parasitic plants. Weedy broomrape species were less specialized in germination requirements than the non-weedy species except for O. cumana and O. foetida var. broteri. Similar results were obtained with the root exudates. Some species, such as P. aegyptiaca and O. minor, showed a broad spectrum of host specificity in terms of seed germination, which was stimulated by exudates from the majority of species tested, whereas others, such as O. cumana, O. hederae and O. densiflora, were highly specific. Some species, such as O. minor, P. aegyptiaca and P. nana, were responsive to the three strigolactones studied, whereas others were induced by only one of them, or did not respond to them at all. The synthetic strigolactone analogue GR24, generally used as a standard for germination tests, was not effective on some Orobanche and Phelipanche species. Seeds of some species that did not respond to GR24 were induced to germinate in the presence of fabacyl acetate or strigol, confirming the role of strigolactones in host specificity.

Five Euphorbiaceae species (Jatropha macrocarpa, J. hieronymi, J. excisa, Cnidoscolus tubulosus and Manihot guaranitica) were analysed for seed oil content and fatty acid profiles in order to evaluate their potential as sources of biodiesel. The five species are perennial shrubs adapted to arid and semi-arid environments of north-western Argentina. The seed oil content for all species ranged from 27.9 to 38.7% by dry weight. Fatty acid composition consisted mainly of linolenic, myristic, palmitic, stearic, arachidic, oleic and linoleic acids, with linoleic and oleic acid being the most abundant. Among the five species analysed, J. macrocarpa has the best potential to be used as a biodiesel source. The average total seed oil content (35.8%) and a high value of oleic acid (27.3%) together with the lowest values of linoleic acid (55.8%) make this species a promising crop as a source of biodiesel in arid and semi-arid regions.
