Two species of Proctoeces Odhner, 1911 have been described in marine organisms from Chile: P. humboldti George-Nascimento & Quiroga (1983), parasitizing the gonads of keyhole limpets (Fissurella spp.), and P. chilensis Oliva (1984), an intestinal parasite of Sicyases sanguineus (Teleostei); both species were subsequently considered as P. lintoni Siddiqi & Cable (1960). To assist in the resolution of the taxonomic identification of Proctoeces species in marine organisms from Chile, phylogenetic studies using DNA sequences from the V4 region of the SSU rRNA gene were performed. Several specimens of P. lintoni were isolated from keyhole limpets (Fissurella spp.) and clingfish (S. sanguineus) from Bahia San Jorge (23°40′S) and Bahia Concepción (36°50′S). Phylogenetic analyses were conducted using three different approaches: a neighbour-joining (NJ), a maximum likelihood (ML) and Bayesian inference (BI). The phylogenetic analysis confirms that specimens of Proctoeces obtained from keyhole limpets and those specimens from the clingfish are in fact the same species. We prefer to consider our specimens as Proctoeces cf. lintoni, as the morphology of Proctoeces appears to be of doubtful value and genetic information about P. lintoni Siddiqi & Cable (1960) is not available. In addition, our results strongly suggest that there are at least three species in this genus.

Fifteen years after our first investigation, a follow-up study was carried out with the purpose of assessing the evolution of schistosomiasis in the locality of Sabugo, Paracambi, state of Rio de Janeiro, Brazil, an area with low prevalence of the disease. The coprological techniques adopted were spontaneous sedimentation and Kato-Katz. Out of the 1356 individuals assessed, 13 (1%) were infected with Schistosoma mansoni. From those, 10 were males, 12 were over 15 years old, and at least 11 had been infected in Sabugo. All patients presented either the intestinal or the hepato-intestinal form of the disease, and 8 (61.5%) harboured light parasitic loads. In 1990, there were 27 (2.7%) infected individuals; less than half harboured light parasitic loads, with the predominance of moderate and heavy forms. Although our results indicate an improvement in the epidemiological situation of schistosomiasis in Sabugo, transmission of the disease in the locality is still active, especially among young males, and tends to be acquired during leisure activities.

The possibilities of disease prediction based on the environmental characteristics of geographical areas and specific requirements of the causative infectious agents are reviewed and, in the case of parasites whose life cycles involve more than one host, the needs of the intermediate hosts are also referred to. The geographical information systems framework includes epidemiological data, visualization (in the form of maps), modelling and exploratory analysis using spatial statistics. Examples include climate-based forecast systems, based on the concept of growing degree days, which now exist for several parasitic helminths such as fasciolosis, schistosomiasis, dirofilariasis and also for malaria. The paper discusses the limits of data collection by remote sensing in terms of resolution capabilities (spatial, temporal and spectral) of sensors on-board satellites. Although the data gained from the observation of oceans, land, elevations, land cover, land use, surface temperatures, rainfall, etc. are primarily for weather forecasting, military and commercial use, some of this information, particularly that from the climate research satellites, is of direct epidemiological utility. Disease surveillance systems and early-warning systems (EWS) are prime examples of academic approaches of practical importance. However, even commercial activities such as the construction of virtual globes, i.e. computer-based models of the Earth, have been used in this respect. Compared to conventional world maps, they do not only show geographical and man-made features, but can also be spatially annotated with data on disease distribution, demography, economy and other measures of particular interest.

The treatment of paramphistomiasis, a neglected tropical disease, has been carried out with different fasciolicidal compounds, all showing weak efficacy. Therefore, the search for alternative paramphistomicidal drugs is warranted. In the present study, the in vitro effects of artemether on adult Paramphistomum microbothrium were evaluated, for the first time, using scanning electron microscopy. After 24 h of incubation with 10 μg ml− 1 artemether, tegumental damage of both anterior and posterior ends of the fluke had occurred in the majority of the specimens examined. Sensory papillae surrounding the oral aperture were ruptured, while those at the acetabular region appeared to be sunken due to tegumental swelling. The tegumental disruption became more pronounced and both oral sucker and acetabulum were severely distorted, on increasing the concentration to 20 μg ml− 1. With higher concentration of 30 μg ml− 1, gross swellings of the body of the fluke, clearly visible to the naked eye, were observed, and damage to both oral sucker and acetabulum was so extreme that little recognizable structure remained.

Laboratory-bred goldfish were experimentally infected with Contracaecum rudolphii, either directly (with nematode larvae) or indirectly via infected zooplankton. Intensity and prevalence of infection were markedly higher in fish exposed to the infected zooplankton than in goldfish exposed to nematode larvae. When transmitted to fish via the zooplankton, larvae developed much faster and, after 8 weeks, became firmly encysted in the intestine wall. The digestive tract of larvae isolated from the intestine wall showed a well-developed ventriculus, ventricular appendix and intestinal caecum. The mouth was surrounded by three lips. When free-living larvae induced infection, they rapidly penetrated the intestine wall and migrated to internal organs (e.g. liver). The primordial lips of those larvae were poorly visible, the ventricular appendix was short, and the intestinal caecum was absent. During the 10-week experiment, no encysted larvae were found in fish infected with free-living larvae.

The parasites of 484 brown trout, Salmo trutta, were studied between 2004 and 2007. An indicator value (IndVal) method was used for analysis, which combines measures of fidelity and specificity. Because of its resilience to detect changes in abundance, IndVal is an effective ecological bio-indicator. The IndVal method demonstrated that altitude, hydrographic network and season could influence the occurrence of parasite species in brown trout. A randomization test identified three parasite species as having a significant indicator value for altitude (the trematode Nicolla wisniewskii, and the nematodes Spinitectus gordoni and Rhabdochona gnedini); five parasite species for hydrographic network (the trematodes Nicolla testiobliquum, N. wisniewskii, Plagioporus stefanskii, and the nematodes S. gordoni and R. gnedini) and two parasite species for season (the nematodes S. gordoni, and R. gnedini). Data for species composition and infection levels should help to improve the monitoring and management of parasitism in salmonid populations.

In vitro trials investigating the effects of albendazole and triclabendazole anthelmintics on the growth profiles of the egg-parasitic fungi Paecilomyces lilacinus and Verticillium chlamydosporium were undertaken. In addition, in vivo trials were conducted in goats fed on millet grain cultures of each fungus and administered albendazole and triclabendazole anthelmintics. In vitro growth revealed V. chlamydosporium to be more sensitive to albendazole compared to P. lilacinus. In contrast, triclabendazole had the least inhibitory effect on in vitro growth of both P. lilacinus and V. chlamydosporium. Similar to albendazole, growth of P. lilacinus was more vigorous at 0.5 ppm concentration of triclabendazole. Efforts to re-isolate these egg-parasitic fungi from faeces of goats fed on fungal millet grain cultures before and following single intraruminal administration of albendazole and triclabendazole showed that P. lilacinus was not able to be re-isolated from the faeces at any sampling period. In contrast, V. chlamydosporium was able to be re-isolated from the faeces at all of the sampling periods except for the samples taken at 8–18 h and 18–24 h after administration of albendazole and triclabendazole, respectively. Lack of fungal activity at these times coincided with peak plasma availability of anthelmintics and suggests faecal levels of drugs were also high at these times and impacted negatively on fungal viability.

The short-term effects of pentoxifylline (PTX) on granulomatous lesions during Schistosoma mansoni infection in Swiss mice were evaluated. Drug administration was initiated 42 and 140 days post-infection (DPI) for the acute and chronic infection groups, respectively. Treatment was carried out daily with 200 mg/kg (subcutaneous route) of the drug for five consecutive days. Recovery of parasites and tissues was performed at 49 DPI and 147 DPI, respectively. Liver histological analysis showed a decrease in the inflammatory reaction and fibrous content of the granulomas studied, and a significant reduction (P < 0.001) in their mean diameter was observed in the groups of rodents treated with PTX in acute and chronic infection, when compared to their respective control groups. However, no alteration in the number of S. mansoni recovered from the portal system was observed, and egg-laying kinetics was not notably modified by PTX treatment, and the immature stage distribution of S. mansoni eggs showed minor intrinsic variations with no statistical differences in the parameter second-stage/female/g among untreated mice and treated mice in acute and chronic infections, respectively, when evaluated by intestinal oograms. Data obtained indicate probable immunomodulatory effects of PTX in murine schistosomiasis both in acute and chronic infection.

Sixteen Tuli steers aged 1 year were subdivided into four equal groups (I–IV) and infected with Calicophoron microbothrium metacercariae. Group I received a low dose (LD) of 5000 metacercariae, group II a medium dose (MD) of 15,000 metacercariae, group III a high dose (HD) of 25,000 metacercariae while group IV was the non-infected control (C) group. The experimental animals were monitored daily for clinical signs while ethylene diamine tetraacetic acid (EDTA) blood and serum samples were collected every 7 days until day 28 post-infection, when sample collection was terminated. Samples were processed for full blood count, eosinophils and blood biochemical values for calcium, magnesium, phosphorus, total protein and albumin. Moderate to severe diarrhoea developed in the MD and HD groups at day 21 post-infection. The diarrhoea coincided with a significant decrease (P < 0.05) in total plasma protein, calcium and phosphorus levels, particularly in the MD group. Similarly, a significant decrease (P < 0.05) in the packed cell volume (PCV), the haemoglobin (Hb) and red blood cell (RBC) levels occurred in the MD and HD groups from day 21 post-infection, while a significant increase (P < 0.05) in the circulating eosinophils occurred between 7 and 21 days post-infection in the LD and the HD groups.

This article communicates the relative quantification of five isoforms of antigen B (AgB) of Echinococcus granulosus. Relative expression of the AgB was quantified in active and inactive cysts. Cysts with germinal membrane, clear cyst fluid and protoscoleces showed uniform expression of the five isoforms and were utilized as control. Relative expression of AgB1 was the highest in cysts, where calcification has initiated. AgB2 and AgB4 were expressed more in fertile cysts irrespective of the condition of germinal membrane. The lowest expression of AgB3 was seen in calcified cysts. The relative expression of AgB5 could not be correlated with respect to the condition of the cyst because AgB5 is typically expressed by the adult stage of the parasite.

A cross-sectional study was carried out to investigate the prevalence and risk factors for severe anaemia among schoolchildren in New Halfa, eastern Sudan. After taking age and gender, haemoglobin (Hb) levels were measured and all children were screened for malaria and intestinal parasitic infections, including schistosomiasis. Six hundred and forty (230 boys, 410 girls) schoolchildren aged 8–18 years were enrolled. The mean (SD) of the haemoglobin was 9.6 (1.7) mg/dl. One hundred and eleven (17.3%) and 33 (5.2%) children had Schistosoma mansoni infection and Hymenolepis nana infection, respectively. Five hundred and eighty (90.6%) of these children had anaemia (Hb < 12 g/dl), and 103 (16.0%) of them had severe anaemia (Hb < 8 g/dl). Girls (OR = 2.5, 95% CI = 1.1–5.3; P = 0.01) were at higher risk for severe anaemia in univariate and multivariate analyses. Schistosoma mansoni infections were associated with severe anaemia in univariate analyses only. Thus, there was a high prevalence of severe anaemia among these children. This needs to be investigated in more depth in the future, and more attention should be paid to the health of adolescent girls.

The action of four fungal isolates of the species Duddingtonia flagrans (AC001), Monacrosporium thaumasium (NF34a) and Pochonia chlamydosporia (VC1 and VC4) on eggs of Oxyuris equi and Austroxyuris finlaysoni was evaluated in two assays (A and B). Eggs of O. equi (Test A) and A. finlaysoni (Test B) were plated on Petri dishes with 2% water-agar with grown fungal isolates and control without fungus. After 5, 10 and 15 days, 100 eggs were collected and classified according to the following parameters: type 1 effect, physiological and biochemical effect without morphological damage to the eggshell; type 2 effect, lytic effect with morphological alteration of the eggshell and embryo; and type 3 effect, lytic effect with morphological alteration of the eggshell and embryo, hyphal penetration and internal egg colonization. Pochonia chlamydosporia isolates VC1 and VC4 showed ovicidal activity for type 1, 2 and 3 effects on eggs of O. equi and eggs of A. finlaysoni. In vitro assays A and B showed that P. chlamydosporia had a negative influence on eggs of O. equi and A. finlaysoni and can be considered as a potential biological control agent of nematodes.

Six species of digeneans (Proctoeces maculatus (Looss, 1901), Helicometra fasciata (Odhner, 1902), Helicometra pulchella (Odhner, 1902), Macvicaria alacris (Looss, 1901), Peracreadium genu (Nicoll, 1909) and Zoogonus rubellus (Olson, 1868)) were found for the first time in labrid fish (Symphodus tinca (Linnaeus, 1758), Labrus merula (Linnaeus, 1758) and Labrus viridis (Linnaeus, 1758)) from the Bay of Bizerte. Except for P. maculatus and Z. rubellus, which are limited to the rectum, these helminths colonize the entire digestive tract. The study of the seasonal prevalence, abundance and mean intensity shows that three parasites, P. maculatus, H. fasciata and P. genu are present throughout the year while H. pulchella, M. alacris and Z. rubellus are less frequent and appear only in spring (H. pulchella and M. alacris) or in spring and summer (Z. rubellus). The levels of infection and digenean faunal diversity in labrid fish from the Tunisian coasts are generally lower than those from localities within the western Mediterranean.

In order to study seasonal patterns of Meiogymnophallus minutus infections in its intermediate hosts, bivalve samples were collected monthly between April 2008 and March 2009 from a high intertidal flat at Courtmacsherry Bay, Ireland. Infection rates in the first intermediate host Scrobicularia plana did not fluctuate significantly with season. Completely developed M. minutus cercariae appeared in daughter sporocysts from June and prevailed from July to October, indicating that transmission of M. minutus from its first to its second intermediate host is confined to this period of the year. All analysed individuals of the second intermediate host Cerastoderma edule were found to be infected with metacercariae. Infection levels significantly increased in September, suggesting recent cercarial invasions. Throughout the year, the majority of metacercariae were hyperinfected by the pathogenic microsporidian Unikaryon legeri. Spreading of hyperinfections was confined to spring and summer. Newly settled metacercariae were not affected by hyperparasitism and presumably retained their infectivity for half a year. Our findings suggest that the spreading of hyperinfections is correlated with higher water temperatures and that the entire metacercarial population has to rebuild every year as a consequence of hyperparasite-induced mortality.

Two new species of spiruroid nematodes in birds from Kangean Island, Indonesia are reported: Diplotriaena anthreptis sp. nov. is found from the abdominal cavity of Anthreptes malacensis malacensis. The new species is similar to D. ozouxi, D. bargusinica, D. delta, D. isabellina and D. obtusa in the size of tridents and the length of spicules. However, it differs from the five similar species in the structure of the tridents, in the morphology of the right spicule, in the spicule ratio and in the size of the eggs. Acuaria irhami sp. nov. is described based on two male specimens from under the gizzard lining of Dicrurus hottentottus jentincki. The new species can be distinguished easily from all congeners except from A. microecae, in having equal rather than subequal or dissimilar spicules. However, the new species can be differentiated from A. microecae in the number of postanal papillae, in the median preanal papilla and in the length of the cordons.

The aim of the present study is to report morphological data from parasitic female, rhabditoid and filarioid larvae, free-living female worms and eggs of Strongyloides ophidiae (Nematoda, Strongyloididae). In addition, a molecular DNA analysis was carried out using a pool of eight S. ophidiae parasitic females. Samples were obtained from the small intestine of Oxyrhopus guibei (Serpentes, Colubridae) collected in the municipality of Lençóis Paulista, State of São Paulo, Brazil. DNA amplification by polymerase chain reaction (PCR) resulted in a 350 bp band for samples containing S. ophidiae and Strongyloides venezuelensis DNA. Strongyloides ophidiae nucleotide sequence analysis showed 98% similarity with Strongyloides procyonis and 97% with Strongyloides cebus, Strongyloides stercoralis, Strongyloides fuelleborni and Strongyloides sp. from snakes.

Echinostomes are common intestinal parasites causing zoonotic disease, which are endemic worldwide. Of the four species of medically important echinostomes reported in Thailand, two species, Echinostoma revolutum and Hypodereaum conoideum, have been detected in poultry. These two parasites are morphologically similar and are sometimes difficult to distinguish. In the present study, multilocus enzyme electrophoresis was used to differentiate E. revolutum from H. conoideum collected from domestic ducks in Thailand. The parasites were compared using 22 enzymes with 30 presumptive enzyme loci. The two species of echinostome could be distinguished at 17 of the 30 enzyme loci. Several loci were polymorphic within each species, suggesting that these can be used to examine their population genetics.

Trichinella zimbabwensis has been detected in wild and farmed Nile crocodiles (Crocodylus niloticus) and in wild monitor lizards (Varanus niloticus) of several African countries, but it has never been detected in mammals in nature, in spite of its infectivity to rodents, pigs, foxes and monkeys under laboratory conditions. The aim of this work was to describe the first detection of T. zimbabwensis in a naturally infected lion (Panthera leo) of the Kruger National Park (KNP) of South Africa. The sequence of the expansion segment V, a highly variable non-coding sequence of the large subunit ribosomal RNA of the genus Trichinella, of larvae from the lion was identical to that of larvae of T. zimbabwensis collected from a Nile crocodile originating from the same locality as the lion, suggesting a possible transmission of this parasite between mammals and reptiles. The KNP proves to be a very interesting area for parasites of the genus Trichinella since three taxa (Trichinella nelsoni, Trichinella T8 and T. zimbabwensis) circulate among the wildlife of this protected area.

Gongylonema ingluvicola and Spirocerca lupi are spirurid nematodes that require arthropod intermediate hosts in order to complete their life cycle. Beetles of the family Scarabaeidae are reported to serve as intermediate hosts for both these parasites. In this study selected species of beetles of the family Scarabaeidae as well as other groups of arthropods were screened for susceptibility to infection with S. lupi and G. ingluvicola. Arthropods were exposed to infective eggs of both parasites for a determined period of time and dissected/digested to determine the presence or absence of pre-infective and infective larvae. All the five species of dung beetles exposed to infection with S. lupi, namely, Pachylomerus femoralis, Scarabaeus rugosus, Gymnopleurus humanus, Kheper nigroaeneus and Anachalcos convexus were susceptible and, of the two species exposed to G. ingluvicola, only Gy. humanus was susceptible. Spirocerca lupi eggs developed in millipede species, Daratoagonus cristulatus, and remained as encysted larvae, while in Orthoporoides kyrhocephalus no development was observed. Spirocerca lupi larvae were not detected in the cricket species Gryllus assimilis, or the cockroach species Periplaneta americana, and, similarly, G. ingluvicola larvae were not detected in the millipede species O. kyrhocephalus. The difference in the susceptibility of the arthropods to the two parasite species may depend on their feeding biology.

FMRFamide-like peptides (FLPs) are a diverse group of neuropeptides that are expressed abundantly in nematodes. They exert potent physiological effects on locomotory, feeding and reproductive musculature and also act as neuromodulators. However, little is known about the specific expression patterns and functions of individual peptides. The current study employed rapid amplification of cDNA ends-polymerase chain reaction (RACE-PCR) to characterize flp genes from infective juveniles of the root knot nematodes, Meloidogyne incognita and Meloidogyne minor. The peptides identified from these transcripts are sequelogs of FLPs from the free-living nematode, Caenorhabditis elegans; the genes have therefore been designated as Mi-flp-1, Mi-flp-7, Mi-flp-12, Mm-flp-12 and Mi-flp-14. Mi-flp-1 encodes five FLPs with the common C-terminal moiety, NFLRFamide. Mi-flp-7 encodes two copies of APLDRSALVRFamide and APLDRAAMVRFamide and one copy of APFDRSSMVRFamide. Mi-flp-12 and Mm-flp-12 encode the novel peptide KNNKFEFIRFamide (a longer version of RNKFEFIRFamide found in C. elegans). Mi-flp-14 encodes a single copy of KHEYLRFamide (commonly known as AF2 and regarded as the most abundant nematode FLP), and a single copy of the novel peptide KHEFVRFamide. These FLPs share a high degree of conservation between Meloidogyne species and nematodes from other clades, including those of humans and animals, perhaps suggesting a common neurophysiological role which may be exploited by novel drugs. FLP immunoreactivity was observed for the first time in Meloidogyne, in the circumpharyngeal nerve ring, pharyngeal nerves and ventral nerve cord. Additionally, in situ hybridization revealed Mi-flp-12 expression in an RIR-like neuron and Mi-flp-14 expression in SMB-like neurons‡, respectively. These localizations imply physiological roles for FLP-12 and FLP-14 peptides, including locomotion and sensory perception.
