Spaceflight missions must limit biological contamination on both the outbound and return legs to comply with planetary protection requirements. Depending on the mission profile, contamination concerns may include the potential presence of bioactive molecules as defined by NASA’s Planetary Protection policies. Thus, the present study has examined the temperature and time requirements for sufficient inactivation/degradation of an infectious, heat-stable prion protein (Sup35NM), which serves as a model bioactive molecule. Bovine serum albumin was used to establish the method parameters and feasibility. Differential scanning calorimetry, Fourier transform infrared spectroscopy, analytical reversed-phase high-performance liquid chromatography, and mass spectrometry were utilized to analyze heat-treated samples, with non-treated samples serving as controls. Heat treatment at 400°C for 5 seconds was found to result in substantial decomposition of Sup35NM. In addition to the disruption of the protein backbone amide bonds, the side chain residues were also compromised. Fragments of molecular weight <4600 were observed by mass spectrometry but the impact of treatment on both the backbone and side chains of Sup35NM suggested that these fragments would not self-associate to create potentially pathogenic entities. The present methodology provided insight into the protein degradation process and can be applied to a variety of treatment strategies (e.g., any form of sterilization or inactivation) to ensure a lack of protein-based contamination of isolated extraterrestrial specimens.
Executives are important elites, and ideology is important to elite behavior, but measurement challenges and a focus on the presidency have kept scholars from fully exploring executive ideology. This article advocates studying US governors to learn more about executive ideology. It provides an overview of the data scholars can use to measure gubernatorial preferences, and highlights Bonica’s campaign finance-based ideology scores (CFscores) as offering the greatest coverage and allowing common-scale comparisons with other actors. As a validation exercise, I find that CFscores explain within-party variation in other measures and predict the decisions that governors make when in office. Then, I run a preliminary test of the substantive importance of executive ideology. Four models explain state policy liberalism as a function of executive, legislative, and citizen ideology. Gubernatorial preferences emerge as most predictive of the three. These results encourage greater investigation into the role of executive ideology in the policy process.
To evaluate the validity and reproducibility of a 152-item semi-quantitative FFQ (SFFQ) for estimating flavonoid intakes.
Design:
Over a 1-year period, participants completed two SFFQ and two weighed 7-d dietary records (7DDR). Flavonoid intakes from the SFFQ were estimated separately using Harvard (SFFQHarvard) and Phenol-Explorer (SFFQPE) food composition databases. 7DDR flavonoid intakes were derived using the Phenol-Explorer database (7DDRPE). Validity was assessed using Spearman’s rank correlation coefficients deattenuated for random measurement error (rs), and reproducibility was assessed using rank intraclass correlation coefficients.
Setting:
This validation study included primarily participants from two large observational cohort studies.

Six hundred forty-one men and 724 women.
Results:
When compared with two 7DDRPE, the validity of total flavonoid intake assessed by SFFQPE was high for both men and women (rs = 0·77 and rs = 0·74, respectively). The rs for flavonoid subclasses ranged from 0·47 for flavones to 0·78 for anthocyanins in men and from 0·46 for flavonols to 0·77 for anthocyanins in women. We observed similarly moderate (0·4–0·7) to high (≥0·7) validity when using SFFQHarvard estimates, except for flavonesHarvard (rs = 0·25 for men and rs = 0·19 for women). The SFFQ demonstrated high reproducibility for total flavonoid and flavonoid subclass intake estimates when using either food composition database. The intraclass correlation coefficients ranged from 0·69 (flavonolsPE) to 0·80 (proanthocyanidinsPE) in men and from 0·67 (flavonolsPE) to 0·77 (flavan-3-ol monomersHarvard) in women.
Conclusions:
SFFQ-derived intakes of total flavonoids and flavonoid subclasses (except for flavones) are valid and reproducible for both men and women.
Antimicrobial stewardship programs typically use days of therapy to assess antimicrobial use. However, this metric does not account for the antimicrobial spectrum of activity. We applied an antibiotic spectrum index to a population of very-low-birth-weight infants to assess its utility to evaluate the impact of antimicrobial stewardship interventions.
