Exploring cryptic diversity in publicly available strains of the model diatom Thalassiosira pseudonana (Bacillariophyceae)

The model diatom Thalassiosira pseudonana is believed to be a single species with a global distribution, but it has not been confirmed previously whether isolates from different environmental and geographic origins are genotypically and phenotypically identical. In the present study, a polyphasic approach was employed to characterize nine clonal isolates, plus an additional replicate of one of the isolates, of the diatom T. pseudonana from culture collections to investigate whether there was any cryptic speciation in the publicly available strains of this species. Morphological analysis using scanning electron microscopy concluded that the strains were indistinguishable. Furthermore, conventional DNA barcoding genes (SSU rDNA, ITS1 and ITS2 rDNA and rbcL), revealed no nucleotide variation among the strains tested. On employing a whole genome fingerprinting technique, Amplified Fragment Length Polymorphism (AFLP), three clusters were revealed, although the level of variation between the clusters was surprisingly low. These findings indicate a low level of diversity among these cultured T. pseudonana strains, despite their wide spatial and temporal distribution and the salinity range of their original habitats. Based on the limited number of available strains, this suggests that T. pseudonana is a highly conserved diatom that nevertheless has an ability to tolerate wide ranges of salinity and populate varied geographic locations.

Background: Several observational studies have suggested a link between health status and rate of decline among individuals with Alzheimer's disease (AD). We sought to quantify the relationship in a population-based study of incident AD, and to compare global comorbidity ratings to counts of comorbid conditions and medications as predictors of AD progression.
Methods: This was a case-only cohort study arising from a population-based longitudinal study of memory and aging, in Cache County, Utah. Participants comprised 335 individuals with incident AD followed for up to 11 years. Patient descriptors included sex, age, education, dementia duration at baseline, and APOE genotype. Measures of health status made at each visit included the General Medical Health Rating (GMHR), number of comorbid medical conditions, and number of non-psychiatric medications. Dementia outcomes included the Mini-Mental State Examination (MMSE), Clinical Dementia Rating – sum of boxes (CDR-sb), and the Neuropsychiatric Inventory (NPI).
Results: Health status tended to fluctuate over time within individuals. None of the baseline medical variables (GMHR, comorbidities, and non-psychiatric medications) was associated with differences in rates of decline in longitudinal linear mixed effects models. Over time, low GMHR ratings, but not comorbidities or medications, were associated with poorer outcomes (MMSE: β = –1.07 p = 0.01; CDR-sb: β = 1.79 p < 0.001; NPI: β = 4.57 p = 0.01).
Conclusions: Given that time-varying GMHR, but not baseline GMHR, was associated with the outcomes, it seems likely that there is a dynamic relationship between medical and cognitive health. GMHR is a more sensitive measure of health than simple counts of comorbidities or medications. Since health status is a potentially modifiable risk factor, further study is warranted.

The Diffusion of Radionuclides through Waste Encapsulation Grouts

United Kingdom Nirex Limited develops and advises on safe, environmentally sound and publicly acceptable options for the long-term management of radioactive waste. One option Nirex has developed is a phased geological repository concept for intermediate level waste and some low level wastes that makes use of a combination of engineered and natural barriers. Physical containment of radionuclides would be achieved by immobilisation and packaging of wastes (mostly) in stainless steel containers.
Existing models of the migration of dissolved radionuclides from packaged wastes suggest that radionuclide release is determined largely by the rate of diffusion through the encapsulation grout used to immobilise the waste. The use of such models requires diffusion coefficient data for radionuclides in waste encapsulation grouts. This paper describes a programme of through-diffusion experiments, and modelling interpretation, aimed at deriving diffusion coefficients for some radionuclides in two types of encapsulation grout.
An intrinsic diffusion coefficient of HTO of around 1×10−13 to 2×10−13 m2s−1 was determined for a 3:1 mix of blast furnace slag to ordinary Portland cement, compared to around 4×10−13 to 5×10−13 m2s−1 for a 3:1 mix of pulverised fuel ash to ordinary Portland cement. These values are lower than that assumed for a non-sorbing radionuclide in an earlier modelling exercise. Porosity values around 0.3 were obtained in each case. For 36Cl as chloride, the experiments showed no significant breakthrough over the experimental timescale of about one year, suggesting an intrinsic diffusion coefficient below 5×10−13 m2s−1. One possibility is that chlorine-containing solids are precipitating in the cement. An intrinsic diffusion coefficient for 137Cs in the 3:1 mix of pulverised fuel ash to ordinary Portland cement of 4×10−15 m2s−1 was estimated, significantly lower than that determined for HTO.
The results from three of the sixteen experiments could not be fitted with a simple diffusion model, and for a further five experiments there was some doubt as to whether simple diffusion behaviour had been observed. It is suggested that this may have been due to cracks in the grouts that were sufficiently large to affect the diffusion properties of the grouts, although none was visible to the naked eye. Cracking of the waste encapsulation grouts could provide a mechanism for enhanced migration of radionuclides from waste packages, compared with diffusion in a homogeneous porous medium alone.

The basis of an effective and satisfactory physician–patient relationship is found in the communication which occurs between these two individuals. By studying the interaction, we can learn much about the identities of the physician and patient, and how they view each other and the world. The interactional dynamics between physician and patient are unique. For example, even in initial medical encounters which involve the meeting of two strangers, patients and physicians deal with concerns as diverse as life and death as well as other intimate or personal issues. Researchers of physician–patient interaction seek to discover how communication evolves and how that communication reveals the multiple levels of meaning in the medical encounter.
