The extracellular matrices, such as the haemolymph, in insects are at the centre of most physiological processes and are protected from oxidative stress by the extracellular antioxidant enzymes. In this study, we identified two secreted superoxide dismutase genes (PxSOD3 and PxSOD5) and investigated the oxidative stress induced by chlorpyrifos (CPF) in the aquatic insect Protohermes xanthodes (Megaloptera: Corydalidae). PxSOD3 and PxSOD5 contain the signal peptides at the N-terminus. Structure analysis revealed that PxSOD3 and PxSOD5 contain the conserved CuZn-SOD domain, which is mainly composed of β-sheets and has conserved copper and zinc binding sites. Both PxSOD3 and PxSOD5 are predicted to be soluble proteins located in the extracellular space. After exposure to different concentrations of sublethal CPF, MDA content in P. xanthodes larvae were increased in a dose-dependent manner; SOD and CAT activities were also higher in CPF-treated groups than that in the no CPF control, indicating that sublethal CPF induces oxidative stress in P. xanthodes larvae. Furthermore, PxSOD3 and PxSOD5 expression levels and haemolymph SOD activity in the larvae were downregulated by sublethal CPF at different concentrations. Our results suggest that the PxSOD3 and PxSOD5 are putative extracellular antioxidant enzymes that may play a role in maintaining the oxidative balance in the extracellular space. Sublethal CPF may induce oxidative stress in the extracellular space of P. xanthodes by reducing the gene expression and catalytic activity of extracellular SODs.
The emotion regulation network (ERN) in the brain provides a framework for understanding the neuropathology of affective disorders. Although previous neuroimaging studies have investigated the neurobiological correlates of the ERN in major depressive disorder (MDD), whether patients with MDD exhibit abnormal functional connectivity (FC) patterns in the ERN and whether the abnormal FC in the ERN can serve as a therapeutic response signature remain unclear.
Methods
A large functional magnetic resonance imaging dataset comprising 709 patients with MDD and 725 healthy controls (HCs) recruited across five sites was analyzed. Using a seed-based FC approach, we first investigated the group differences in whole-brain resting-state FC of the 14 ERN seeds between participants with and without MDD. Furthermore, an independent sample (45 MDD patients) was used to evaluate the relationship between the aforementioned abnormal FC in the ERN and symptom improvement after 8 weeks of antidepressant monotherapy.
Results
Compared to the HCs, patients with MDD exhibited aberrant FC between 7 ERN seeds and several cortical and subcortical areas, including the bilateral middle temporal gyrus, bilateral occipital gyrus, right thalamus, calcarine cortex, middle frontal gyrus, and the bilateral superior temporal gyrus. In an independent sample, these aberrant FCs in the ERN were negatively correlated with the reduction rate of the HAMD17 score among MDD patients.
Conclusions
These results might extend our understanding of the neurobiological underpinnings underlying unadaptable or inflexible emotional processing in MDD patients and help to elucidate the mechanisms of therapeutic response.
Women with schizophrenia frequently discontinue antipsychotic medications during pregnancy. However, evidence on the risk of postpartum relapse associated with antipsychotic use during pregnancy is lacking.
Aims
To investigate the within-individual association between antipsychotic continuation during pregnancy and postpartum relapse in women with schizophrenia.
Method
This retrospective cohort study used data of women with schizophrenia who gave live birth between 2007 and 2018 identified from the National Health Information Database of South Korea. Women were classified according to antipsychotic use patterns during the 12 months before delivery as non-users, discontinuers and continuers. Relapse was defined as admission for psychosis (ICD-10, F20–29). The incidence rate ratio (IRR) for admission for psychosis in the 6-month postpartum period was estimated using conditional Poisson regression, with the reference period set between 2 and 1 years before delivery. Additionally, we calculated the relative risk ratios (RRRs) for the IRRs of different antipsychotic use patterns.
Results
Among the 3026 women included in the analysis (median age 34 years, interquartile range 31–37), the within-individual risk of admission for psychosis in the 6-month postpartum period was 0.56 times (RRR, 95% CI 0.36–0.87) lower in continuers (IRR = 1.31, 95% CI 0.89–1.72) than in discontinuers (IRR = 2.34, 95% CI 1.87–2.91). Among discontinuers, the IRRs of admission for psychosis in the 6-month postpartum period did not change significantly with the timing of discontinuation (trend P = 0.946).
Conclusions
Antipsychotic continuation during pregnancy was associated with a reduced risk of postpartum relapse in women with schizophrenia. Continuing antipsychotics during pregnancy would be recommended after a risk–benefit assessment.
The emerging era of big data in radio astronomy demands more efficient and higher-quality processing of observational data. While deep learning methods have been applied to tasks such as automatic radio frequency interference (RFI) detection, these methods often face limitations, including dependence on training data and poor generalisation, which are also common issues in other deep learning applications within astronomy. In this study, we investigate the use of the open-source image recognition and segmentation model, Segment Anything Model (SAM), and its optimised version, HQ-SAM, due to their impressive generalisation capabilities. We evaluate these models across various tasks, including RFI detection and solar radio burst (SRB) identification. For RFI detection, HQ-SAM (SAM) shows performance that is comparable to or even superior to the SumThreshold method, especially with large-area broadband RFI data. In the search for SRBs, HQ-SAM demonstrates strong recognition abilities for Type II and Type III bursts. Overall, with its impressive generalisation capability, SAM (HQ-SAM) can be a promising candidate for further optimisation and application in RFI and event detection tasks in radio astronomy.
Care of the dying is an essential part of holistic cancer nursing. Improving nurses’ attitudes and behaviors regarding care of the dying is one of the critical factors in increasing the quality of nursing service. This study aims to examine the impact of an educational program based on the CARES tool on nurses’ attitudes and behaviors toward care of the dying.
Methods
A quasi-experimental study with pre- and post-intervention measures was conducted. A total of 222 oncology nurses from 14 hospitals in Beijing, China, were enrolled using a convenient sampling method. This online educational course developed based on the CARES framework comprised 7 modules and 10 sessions. Each session was carried out twice a week over 30–60 min. Data were collected using a sociodemographic characteristics questionnaire, the Frommelt Attitude Towards Care of the Dying Scale (FATCOD) and the Nurses’ Practice Behavior Toward Care of the Dying Questionnaire (NPBTCOD). Reassessment of attitudes and behaviors was conducted when completed the learning and 6 months after the learning, respectively. The sociodemographic characteristics of the nurses were analyzed using descriptive statistics, and differences in attitudes and behaviors were reported and compared by the paired t-test.
Results
All the 222 oncology nurses completed educational courses, and 218 nurses (98.20%) completed the pre- and post-attitudes evaluation and 213 (95.9%) nurses completed the pre- and post-behaviors evaluation. The mean (SD) FATCOD score before and after the educational program was 108.83 (12.07) versus 115.09 (14.91), respectively (t = −8.546, p ≥ 0.001). The mean (SD) NPBTCOD score before and after the educational program was 69.14 (17.56) versus 73.40 (18.96), respectively (t = −3.231, p = 0.001).
Significance of results
This educational intervention was found to be an effective method for improving oncology nurses’ attitudes and behaviors toward caring for dying patients.
This paper proposes an air-filled substrate integrated waveguide (AFSIW) bandpass filter with a miniaturized non-resonant node (NRN). The NRN structure is introduced between the three resonators, and its size is smaller than the resonator size, which can realize the NRN structure’s miniaturization and reduce the model’s size. The NRN size of this filter is 41% of the NRN size of the existing AFSIW filter. This filter also introduces a transmission zero (TZ) above the passband. The measured results show that the filter’s center frequency is 20.73 GHz, and the bandwidth is 0.86 GHz. The insertion loss in the passband is 0.95 dB, and the return loss is better than 23 dB. Due to the TZ in the upper stopband, the AFSIW filter obtained good selectivity.
The Tang dynasty is the only period of Chinese history to which the word ‘cosmopolitan’ is now routinely applied in Western-language historical writing. This article traces the origins of this glamorous image of the Tang to the 1950s and 1960s, but also links its current popularity to a more recent increase in the appeal of the concept of cosmopolitanism, as well as the idea of a ‘cosmopolitan empire’ among Western intellectuals since the end of the Cold War. The article then proposes a less presentist and more critical and holistic reading of Tang ‘cosmopolitanism’ as part of a larger, interconnected, multi-centred, and changing medieval world of numerous coexisting cosmopolitanisms, and argues for recognizing the existence of a different but equally important mode of ‘cosmopolitanism’ in the Song.
The absorption and distribution of radiocarbon-labeled urea at the ultratrace level were investigated with a 14C-AMS biotracer method. The radiopharmaceutical concentrations in the plasma, heart, liver, spleen, lung, kidney, stomach, brain, bladder, muscle, testis, and fat of rats after oral administration of 14C urea at ultratrace doses were determined by AMS, and the concentration-time curves in plasma and tissues and pharmacokinetic distribution data were obtained. This study provides an analytical method for the pharmacokinetic parameters and tissue distribution of exogenous urea in rats at ultratrace doses and explores the feasibility of evaluation and long-term tracking of ultratrace doses of drugs with AMS.
Genes involved in melanin production directly impact insect pigmentation and can affect diverse physiology and behaviours. The role these genes have on sex behaviour, however, is unclear. In the present study, the crucial melanin pigment gene black was functionally characterised in an urban pest, the German cockroach, Blattella germanica. RNAi knockdown of B. germanica black (Bgblack) had no effect on survival, but did result in black pigmentation of the thoraxes, abdomens, heads, wings, legs, antennae, and cerci due to cuticular accumulation of melanin. Sex-specific variation in the pigmentation pattern was apparent, with females exhibiting darker coloration on the abdomen and thorax than males. Bgblack knockdown also resulted in wing deformation and negatively impacted the contact sex pheromone-based courtship behaviour of males. This study provides evidence for black function in multiple aspects of B. germanica biology and opens new avenues of exploration for novel pest control strategies.
The fall armyworm (FAW) Spodoptera frugiperda (J.E. Smith) is a highly damaging invasive omnivorous pest that has developed varying degrees of resistance to commonly used insecticides. To investigate the molecular mechanisms of tolerance to tetraniliprole, spinetoram, and emamectin benzoate, the enzyme activity, synergistic effect, and RNA interference were implemented in S. frugiperda. The functions of cytochrome P450 monooxygenase (P450) in the tolerance to tetraniliprole, spinetoram, and emamectin benzoate in S. frugiperda was determined by analysing changes in detoxification metabolic enzyme activity and the effects of enzyme inhibitors on susceptibility to the three insecticides. 102 P450 genes were screened via transcriptome and genome, of which 67 P450 genes were differentially expressed in response to tetraniliprole, spinetoram, and emamectin benzoate and validated by quantitative real-time PCR. The expression patterns of CYP9A75, CYP340AA4, CYP340AX8v2, CYP340L16, CYP341B15v2, and CYP341B17v2 were analysed in different tissues and at different developmental stages in S. frugiperda. Silencing CYP340L16 significantly increased the susceptibility of S. frugiperda to tetraniliprole, spinetoram, and emamectin benzoate. Furthermore, knockdown of CYP340AX8v2, CYP9A75, and CYP341B17v2 significantly increased the sensitivity of S. frugiperda to tetraniliprole. Knockdown of CYP340AX8v2 and CYP340AA4 significantly increased mortality of S. frugiperda to spinetoram. Knockdown of CYP9A75 and CYP341B15v2 significantly increased the susceptibility of S. frugiperda to emamectin benzoate. These results may help to elucidate the mechanisms of tolerance to tetraniliprole, spinetoram and emamectin benzoate in S. frugiperda.
The Righi–Leduc heat flux generated by the self-generated magnetic field in the ablative Rayleigh–Taylor instability driven by a laser irradiating thin targets is studied through two-dimensional extended-magnetohydrodynamic simulations. The perturbation structure gets into a low magnetization state though the peak strength of the self-generated magnetic field could reach hundreds of teslas. The Righi–Leduc effect plays an essential impact both in the linear and nonlinear stages, and it deflects the total heat flux towards the spike base. Compared to the case without the self-generated magnetic field included, less heat flux is concentrated at the spike tip, finally mitigating the ablative stabilization and leading to an increase in the velocity of the spike tip. It is shown that the linear growth rate is increased by about 10% and the amplitude during the nonlinear stage is increased by even more than 10% due to the feedback of the magnetic field, respectively. Our results reveal the importance of Righi–Leduc heat flux to the growth of the instability and promote deep understanding of the instability evolution together with the self-generated magnetic field, especially during the acceleration stage in inertial confinement fusion.
Mineral-microbe interactions are widespread in a number of environmental processes such as mineral weathering, decomposition, and transformation. Both clay minerals and silicate-weathering bacteria are widely distributed in nature, and the latter contribute to weathering, diagenesis, and mineralization of major rock-forming minerals. The purpose of this study was to observe changes in the chemical composition and structure, especially the phase transformation, of smectite after processing by a silicate-weathering bacterium. The interaction between Bacillus mucilaginosus and bentonite was studied using custom culture media. Results from Inductively Coupled Plasma-Atomic Emission Spectrometry revealed that the bacterium promoted release of Si and Al from solid bentonite to solution. Concomitantly, the Ka nd Fe contents of the mineral increased as shown by X-ray photoelectron spectroscopy results. After interaction with the bacterium, the montmorillonite underwent a possible structure transformation to smectite, as indicated by the emergence of a new weak peak (d = 9.08 Å) shown by X-ray diffraction patterns. The mineralogical changes were also demonstrated by the decrease in the specific surface area of the mineral from 33.0 to 24.0 m2/g (these lower values for SSA of bentonite are related to the particle size of the smectite examined (120-160 mesh) and the weakened absorption bands in Al-O-H and Si-O-Si vibrations by Micro Fourier-transform infrared spectroscopy. The morphology changes in the bacteria observed by environmental scanning electron microscopy and atomic force microscopy revealed an obvious growth of the flagella in the presence of bentonite.
Childhood is a crucial neurodevelopmental period. We investigated whether childhood reading for pleasure (RfP) was related to young adolescent assessments of cognition, mental health, and brain structure.
Methods
We conducted a cross-sectional and longitudinal study in a large-scale US national cohort (10 000 + young adolescents), using the well-established linear mixed model and structural equation methods for twin study, longitudinal and mediation analyses. A 2-sample Mendelian randomization (MR) analysis for potential causal inference was also performed. Important factors including socio-economic status were controlled.
Results
Early-initiated long-standing childhood RfP (early RfP) was highly positively correlated with performance on cognitive tests and significantly negatively correlated with mental health problem scores of young adolescents. These participants with higher early RfP scores exhibited moderately larger total brain cortical areas and volumes, with increased regions including the temporal, frontal, insula, supramarginal; left angular, para-hippocampal; right middle-occipital, anterior-cingulate, orbital areas; and subcortical ventral-diencephalon and thalamus. These brain structures were significantly related to their cognitive and mental health scores, and displayed significant mediation effects. Early RfP was longitudinally associated with higher crystallized cognition and lower attention symptoms at follow-up. Approximately 12 h/week of youth regular RfP was cognitively optimal. We further observed a moderately significant heritability of early RfP, with considerable contribution from environments. MR analysis revealed beneficial causal associations of early RfP with adult cognitive performance and left superior temporal structure.
Conclusions
These findings, for the first time, revealed the important relationships of early RfP with subsequent brain and cognitive development and mental well-being.
For about half a century, the Tang dynasty has held a reputation as the most 'cosmopolitan' period in Chinese history, marked by unsurpassed openness to foreign peoples and cultures and active promotion of international trade. Heavily influenced by Western liberal ideals and contemporary China's own self-fashioning efforts, this glamorous image of the Tang calls for some critical reexamination. This Element presents a broad and revisionist analysis of early Tang China's relations with the rest of the Eurasian world and argues that idealizing the Tang as exceptionally “cosmopolitan” limits our ability to think both critically and globally about its actions and policies as an empire.
In recent decades, the Tang dynasty (618-907) has acquired a reputation as the most 'cosmopolitan' period in Chinese history. The standard narrative also claims that this cosmopolitan openness faded after the An Lushan Rebellion of 755-763, to be replaced by xenophobic hostility toward all things foreign. This Element reassesses the cosmopolitanism-to-xenophobia narrative and presents a more empirically-grounded and nuanced interpretation of the Tang empire's foreign relations after 755.
IN 1120, THE Chinese Song dynasty negotiated an alliance with the Jurchen people of Manchuria, who had lately achieved remarkable military success in a revolt against their overlords, the Inner Mongolian Kitans (or Khitans).* The Song had enjoyed more than a century of peace with the Kitan Liao empire since 1005. But that peace rested on a treaty that required the Song to pay an annual indemnity of silk and silver to the Liao and renounce its claim to the strategic Yan-Yun region on its northern border, which a previous dynasty had ceded to the Liao in exchange for military aid in 938. A hawkish, irredentist segment of the Song literati elite had always resented the loss of Yan-Yun. Although a majority of ministers at the Song imperial court felt that war with the Liao was too risky, the chief minister Wang Fu (1079– 1126) and the eunuch general Tong Guan (1054– 1126) finally convinced emperor Zhao Ji (posthumously Huizong, r. 1100– 1126) that the Jurchen revolt was a heaven-sent opportunity to recover Yan-Yun.
Under the Song– Jurchen alliance's terms, the Song undertook to capture the Liao empire's Southern capital (Yan, modern Beijing) while the Jurchens overran the Kitan heartland in eastern Inner Mongolia and took the Western capital (Yun, modern Datong). Once the war was over, the Jurchens’ new Jin empire would recognize Song possession of the Yan region (northern Hebei) in return for receiving the same annual indemnity once made to the Liao: 300,000 bolts of silk and 200,000 ounces of silver. The question of who would get the Yun region (northern Shanxi) was left somewhat open, to be subject to further negotiation. It seemed like a relatively good deal for the Song, but things started going wrong almost immediately. A large rebellion broke out in southeast China in late 1120, delaying the planned Song invasion of Yan until late 1122. By that time, the Jurchens had already conquered all four of the Liao empire's other capitals, including Yun. Next, the Song army badly bungled its assault on the Southern capital, only to see its defenders surrender to a Jurchen army shortly afterwards.
The Song had clearly failed to fulfill its part of the alliance agreement. Nonetheless, under a new Song– Jin treaty concluded in 1123, the Jurchens grudgingly agreed to hand over the entire Yan-Yun region in exchange for financial compensation.
OBJECTIVES/GOALS: Clinical tissue specimens are primarily destined for formalin fixed, paraffin embedded processing to create a basis for diagnosis by microscopic examination. Innovations in specimen processing are required to expand its availability for inclusion as the substrate in assays that can contribute to the further development of Precision Medicine. METHODS/STUDY POPULATION: Transurethral resection of bladder tumors were selected for testing based on availability and tissue composition. A wash step was used to generate daughter aliquots composed of dislodged cells and a solution with prior contact to the parent tissue. This wash step served two purposes: 1) reduce the amount of contaminating material from spreading to other cases, a problem known to be associated with this type of specimen; and 2) create aliquots from which additional informative data could be generated. These daughter aliquots were then examined to determine their value as a source for exosome profiling, metabolomic studies, molecular characterization and organoid development. The parent tissue was not compromised, was able to undergo conventional processing and yielded results equivalent to unwashed specimens. RESULTS/ANTICIPATED RESULTS: Exosomes secreted by the tumor cells were identified to be present in the daughter aliquots by a combination of their isolation using CD31 and detection of miR-21 expression. These exosomes were confirmed to be not related to fragmented cells from testing for beta-tubulin. A global/discovery-based approach using mass spectrometry provided insights into early characterization of metabolomic profiles present in these tumor cells. Ample amounts of high quality DNA (226 ng/ul concentrations; 11.3 ug total) were recovered from the dislodged, excess cells in the wash for molecular studies. Finally, from viable cells recovered in one of the daughter wash aliquots, the ability to grow organoids was proven to be possible and reproducible. DISCUSSION/SIGNIFICANCE: Based on these results, the value of the clinical specimen can be markedly expanded for utilization in research and possible clinical use without detracting from the parent tissue. This non-destructive, easy to adopt wash procedure can potentially lead to an influx of data that may ultimately prove useful in improving patient care.
This study discusses why South Korea has not always succeeded in the ‘entrepreneurial state’ approach – defined as policy efforts to move away from the old developmental state model to a new industrial system of innovation in which small- and medium-sized enterprises (SMEs) are incorporated – by focusing on the limitation of new semiconductor industrial policies of the former Moon Jae-in government (2017–2022). Conventionally, many scholars have exclusively ascribed this limitation to large businesses' (chaebol's) practice of in-house production. Building upon historical institutionalism and its concept of increasing returns, alternatively, we shift attention to the way the Moon government played its entrepreneurial role. We argue that, as the government sought increasing returns from the developmental state idea and institution, the likelihood of wider SME incorporation decreased. Nationalism enabled the government to control the policymaking process but made it difficult to obtain new information through policy contestation. The government depended on developmental alliance to increase policy visibility through the chaebol's capabilities, but demands of small firms were downplayed. This study proposes to construct a more theoretical framework with which to explain how the old political economy model affects new entrepreneurial goals.
