Using a meta-analysis of literature data, this study aimed to quantify the dry matter (DM) intake response to changes in diet composition, and milk responses (yield, milk component yields and milk composition) to changes in dietary net energy for lactation (NEL) and metabolizable protein (MP) in dairy cows. From all studies included in the database, 282 experiments (825 treatments) with experimentally induced changes in either NEL or MP content were kept for this analysis. These treatments covered a wide range of diet characteristics and therefore a large part of the plausible NEL and MP contents and supplies that can be expected in practical situations. The average MP and NEL contents were, respectively (mean±SD), 97±12 g/kg DM and 6.71±0.42 MJ/kg DM. On a daily supply basis, there were high between-experiment correlations for MP and NEL above maintenance. Therefore, supplies of MP and NEL above maintenance were, respectively, centred on MP supply for which MP efficiency into milk protein is 0.67, and NEL above maintenance supply for which the ratio of NEL milk/NEL above maintenance is 1.00 (centred variables were called MP67 and NEL100). The majority of the selected studies used groups of multiparous Holstein-Friesian cows in mid lactation, milked twice a day. Using a mixed model, between- and within-experiment variation was split to estimate DM intake and milk responses. The use of NEL100 and MP67 supplies substantially improved the accuracy of the prediction of milk yield and milk component yields responses with, on average, a 27% lower root mean square error (RMSE) relative to using dietary NEL and MP contents as predictors. For milk composition (g/kg), the average RMSE was only 3% lower on a supply basis compared with a concentration basis. Effects of NEL and MP supplies on milk yield and milk component yields responses were additive. Increasing NEL supply increases energy partitioning towards body reserve, whereas increasing MP supply increases the partition of energy towards milk. On a nitrogen basis, the marginal efficiency decreases with increasing MP supply from 0.34 at MP67=−400 g/day to 0.07 at MP67=300 g/day. This difference in MP67 supply, assuming reference energy level of NEL100=0, equates to a global nitrogen efficiency decrease from 0.82 to 0.58. The equations accurately describe DM intake response to change in dietary contents and milk responses to change in dietary supply and content of NEL and MP across a wide range of dietary compositions.
Dynamics of myofiber differentiation/maturation in porcine skeletal muscle is associated with domestication, breeding and rearing conditions. This study was aimed to comparatively elucidate the age-dependent myosin heavy chain (MyHC) isoform expression and transition pattern in domestic and wild pig (WP) skeletal muscle from birth until adulthood. Domestic pigs (DPs) of Large White breed raised in conventional production system were compared with WPs reared in a large hunting enclosure. Muscle samples for immuno/enzyme histochemistry were taken from the longissimus dorsi muscle within 24 h postmortem at 24 to 48 h, 21 to 23 days, 7 months and ~2 years postpartum. Based on the antibody reactivity to MyHCs (NCL-MHCs, A4.74, BF-F3) and succinate dehydrogenase activity, myofibers were classified into I, I/IIa, IIa, IIx and IIb types. In addition, foetal MyHC expression was determined with the use of F158.4C10 antibody. Maturation of the longissimus dorsi muscle in the WP was characterized by an accelerated transformation of the fast to slow MyHC during the first hours postpartum, followed by differentiation towards oxidative myofibers in which type I, IIa and IIx MyHCs predominated. In the DP, the transformation shifted towards glycolytic myofibers that expressed MyHC-IIb. The expression of foetal MyHC was higher in the DP than in the WP at 1 day of age, and the decline in the foetal MyHC during the first 3 weeks was more rapid in the WP than in the DP denoting an accelerated early postnatal muscle maturation in WP than DP piglets. All foetal MyHC-positive myofibers co-expressed IIa isoform, but not vice versa. The intense myofiber hypertrophy was evident from 3 weeks until 7 months of age. In this period, the myofiber cross-sectional area increased up to 10- and 20-fold in the WP and the DP, respectively. In the DP, the hypertrophy of all myofiber types was more pronounced than in the WP, particularly the hypertrophy of IIx and IIb myofibers. To summarize, the comparison between growing DP with wild ancestors showed that genetic selection and rearing conditions lead to substantial changes in the direction and intensity of postnatal MyHC transformation as evidenced by different proportion of individual myofiber types and differences in their hypertrophic potential.
A total of 120 Duroc×(Large White×Landrace) pigs, 50% barrows and 50% gilts, with 54.1±0.14 kg BW and 103±3 days of age, were used to study the effect of advancing the shift to a standard finisher feed from 100 to 90 and 80 kg BW on production performances and carcass and pork quality. Each of the six treatments (two sexes×three BWs at time of feeding shift) was replicated four times and the experimental unit was the pen (with five pigs for growth performance and carcass variables and three pigs for pork and fat traits). The grower (163 g CP and 9.5 g total Lys/kg) and the finisher diets (152 g CP and 7.9 g total Lys/kg) were based on maize, barley and vegetal protein concentrates, contained 13.39 MJ metabolizable energy/kg and were offered ad libitum through the trial. Pigs intended for dry-cured product elaboration were slaughtered at 170±3 days of age as average (124 and 115 kg BW for barrows and gilts, respectively). For the overall period, barrows ate more feed (P<0.001) and grew faster (P=0.03) than gilts. No effect of feed shift was observed on growth performances, although the average daily CP intake (P=0.01) and feeding costs (P=0.04) were reduced by advancing the transition to the finisher feed. Carcasses from barrows were heavier (P<0.001) and had wider backfat depth (P<0.001) than those from gilts but no significant differences were observed in the meat chemical composition. The feed change schedule did not modify carcass or meat traits. It is concluded that an early shift to the finisher feed (at 80 kg BW instead of 100 kg BW) might be an interesting strategy in pigs intended for dry-cured products because, although it neither increased body fatness nor improved pork quality, CP intake and feeding costs were reduced without impairment of growth performances. Results were similar for barrows and gilts.
The primary objective of this study was to examine the effects of different inclusion rates of dietary triticale replacing corn grain in starter rations of dairy calves on feed intake, average daily gain (ADG), feed efficiency, skeletal growth, fecal score, and selected blood and ruminal parameters. In all, 30 4-day-old Holstein calves (15 female and 15 male) were blocked by gender and birth weight, and then randomly allotted to three treatment groups (n=10 calves/treatment) and received either a corn-based diet without triticale (T0), or a diet containing 16% (T16) or 32% (T32) triticale (25% and 50% corn replaced by triticale, respectively) for 10 weeks. Calves were weaned on day 43 of study and remained on the study until day 70. During post-weaning period, calves fed T32 had the lowest starter intake (1.78 kg/day; P=0.02). In contrast, ADG was not affected in pre-weaning and overall periods, but calves fed T32 had a lower ADG (P=0.04) as compared with calves fed T0 in post-weaning period. No dietary effect was detected for feed efficiency. Dietary treatment did not affect heart girth and body length; however, height at wither and hip at weaning increased in calves fed triticale compared with T0. No detectable effects were observed in serum glucose and β-hydroxybutyrate. No difference was detected in blood urea nitrogen on day 35 either but on day 50 and day 70 the greatest concentration was recorded for calves fed T16. Compared with the control, ruminal ammonia concentration was increased for calves fed T32 (4.34 v. 7.50 mmol/l) and T16 (4.01 v. 8.12 mmol/l) on day 35 and day 50. No difference was detected in ruminal pH on day 35 or day 50; however, calves fed T32 had the lowest pH (6.11) at 70 days. No significant effect was detected in days scoured, respiratory score and general appearance. Under our experimental conditions, it appears that triticale at 16% dry matter (i.e. replacing corn grain up to 25%) in the starter diet does not have adverse effects on the performance and intake of calves. Therefore, substituting corn partially with triticale in calf starter diets may prove beneficial in places where corn is less abundant or its price is prohibitive. The low number of calves per treatment however may have limited the statistical power to detect significant differences among treatments, possibly affecting the results, which should be interpreted with caution.
In order to expand with validated scientific data the limited knowledge regarding the potential application of insects as innovative feed ingredients for poultry, the present study tested a partial substitution of soya bean meal and soya bean oil with defatted black soldier fly (Hermetia illucens) larvae meal (H) in the diet for growing broiler quails (Coturnix coturnix japonica) on growth performance, mortality, nutrients apparent digestibility, microbiological composition of excreta, feed choice, carcass and meat traits. With this purpose, a total of 450 10-day-old birds were allocated to 15 cages (30 birds/cage) and received three dietary treatments: a Control diet (C) and two diets (H1 and H2) corresponding to 10% and 15% H inclusion levels, respectively (H substituted 28.4% soya bean oil and 16.1% soya bean meal for H1, and 100% soya bean oil and 24.8% soya bean meal for H2, respectively). At 28 days of age, quails were slaughtered, carcasses were weighed, breast muscles were then excised from 50 quails/treatment, weighed, and ultimate pH (pHu) and L*, a*, b* colour values were measured. Breast muscles were then cooked to assess cooking loss and meat toughness. For the digestibility trial, a total of 15 28-day-old quails were assigned to the three feeding groups. The excreta samples were subjected to chemical and microbiological analysis. The same 15 quails were then simultaneously provided with C and H2 diets for a 10-day feed choice trial. Productive performance, mortality and carcass traits were in line with commercial standards and similar in all experimental groups. With the exception of ether extract digestibility, which was lower in H1 group compared with C and H2 (P=0.0001), apparent digestibility of dry matter, CP, starch and energy did not differ among treatments. Microbial composition of excreta was also comparable among the three groups. Feed choice trial showed that quails did not express a preference toward C or H2 diets. Breast meat weight and yield did not differ among C, H1 and H2 quails. Differently, the inclusion of H meal reduced meat pHu compared with C. In conclusion, this study demonstrated that H. illucens larvae meal can partially replace conventional soya bean meal and soya bean oil in the diet for growing broiler quails, thus confirming to be a promising insect protein source for the feed industry. Further research to assess the impact of H meal on intestinal morphology as well as on meat quality and sensory profile would be of utmost importance.
The ability to accumulate intramuscular fat (IMF) is a highly variable characteristic in beef cattle. In breeds with a low tendency to accumulate IMF, this can lead to compromised meat quality because of the contribution of fat to such organoleptic attributes as juiciness and taste. This study considered adiposity and gene expression of some of the main markers involved in adipogenesis and lipid metabolism in the subcutaneous (SC) adipose tissue (AT) and the longissimus thoracis muscle (LM) and investigated differences in adipogenic regulation between the tissues during growth and fattening under different conditions. Pirenaica beef cattle were chosen for the study due to the breed’s low tendency to accumulate IMF and the breed’s regional importance. The young Pirenaica bulls used (n=16) were allocated to four groups and slaughtered at 6, 12 and 18 months. From 12 months onwards the bulls slaughtered at 18 months were fed diets having different energy densities. Backfat thickness increased from 6 to 12 months (P<0.05) but then was unchanged, while other fattening parameters such as percentage chemical fat and marbling did not vary. The adipose cell size distribution displayed a bimodal distribution for SC adipocytes and a unimodal distribution for IMF cells, suggestive of tissue-specific hyperplasia. Gene expression of peroxisome proliferator-activated receptor γ (PPARG), CCAAT/enhancer-binding protein α (CEBPA), sterol regulatory element-binding transcription factor 1 (SREBF1), wingless-type MMTV integration site family 10B (WNT10B), fatty acid-binding protein 4 (FABP4), acetyl Co-A carboxylase α, lipoprotein lipase and fatty acid synthase (FASN) were determined by real-time quantitative PCR. Expression did not differ between the experimental groups within the tissues but did differ between the tissues: PPARG, FABP4 and FASN were upregulated in the SC AT, while CEBPA, WNT10B and SREBF1 were upregulated in the LM. Although age and diet energy density did not have a significant effect on increasing the amount of IMF, these factors could have influenced adipocyte development in this tissue differently than in the SC AT. This was evidenced by the different size distributions of the cells in the two tissues, and the differing expression patterns of certain markers in the SC AT and the LM, which may indicate a differential role of PPARG and WNT10B in triggering adipocyte proliferation and fat accumulation capacity.
Survival during the first year after first calving was investigated over the last 25 years, 1989–2013, as well as how the association of survival with season of calving, age at first calving (AFC) and within-herd production level has changed over that period. The data set contained 1 108 745 Dutch black-and-white cows in 2185 herds. Linear models were used to estimate (1) effect of year and season and their interaction and (2) effect of AFC, within-herd production level, and 5-year intervals and their two-way interactions, and the genetic trend. All models contained AFC and percentage of Holstein Friesian as a fixed effect, and herd-year-season, sire and maternal grandsire as random effects. Survival and functional survival were analysed. Functional survival was defined as survival adjusted for within-herd production level. Survival rate increased by 8% up to 92% in the last 25 years. When accounting for pedigree, survival showed no improvement up to 1999, but improved since then. Genetically, survival increased 3% to 4% but functional survival did not increase over the 25 years. We found an interesting difference between the genetic trends for survival and functional survival for bulls born between 1985 and 1999, where the trend for survival was still increasing, but was negative for functional survival. Since 1999, genetic trend picked up again for both survival and functional survival. AFC, season of calving and within-herd production level affected survival. Survival rate decreased 0.6%/month for survival and 1.5% for functional survival between AFC of 24 and 32 months. Calving in summer resulted in 2.0% higher survival than calving in winter. Within herd, low-producing cows had a lower survival rate than high-producing cows. However, these effects became less important during the recent years. Based on survival optimum AFC is around 24 months, but based on functional survival it is better to have an AFC<24 months. Overall, survival rate of heifers has improved considerably in the past 25 years, initially due to the focus on a high milk production. More recently, the importance of a high milk production has been reduced towards attention for functional survival.
The number and size of free-range laying hen (Gallus gallus domesticus) production systems are increasing within Australia in response to consumer demand for perceived improvement in hen welfare. However, variation in outdoor stocking density has generated consumer dissatisfaction leading to the development of a national information standard on free-range egg labelling by the Australian Consumer Affairs Ministers. The current Australian Model Code of Practice for Domestic Poultry states a guideline of 1500 hens/ha, but no maximum density is set. Radio-frequency identification (RFID) tracking technology was used to measure daily range usage by individual ISA Brown hens housed in six small flocks (150 hens/flock – 50% of hens tagged), each with access to one of three outdoor stocking density treatments (two replicates per treatment: 2000, 10 000, 20 000 hens/ha), from 22 to 26, 27 to 31 and 32 to 36 weeks of age. There was some variation in range usage across the sampling periods and by weeks 32 to 36 individual hens from the lowest stocking density on average used the range for longer each day (P<0.001), with fewer visits and longer maximum durations per visit (P<0.001). Individual hens within all stocking densities varied in the percentage of days they accessed the range with 2% of tagged hens in each treatment never venturing outdoors and a large proportion that accessed the range daily (2000 hens/ha: 80.5%; 10 000 hens/ha: 66.5%; 20 000 hens/ha: 71.4%). On average, 38% to 48% of hens were seen on the range simultaneously and used all available areas of all ranges. These results of experimental-sized flocks have implications for determining optimal outdoor stocking densities for commercial free-range laying hens but further research would be needed to determine the effects of increased range usage on hen welfare.
Information on the genetic diversity and population structure of cattle breeds is useful when deciding the most optimal, for example, crossbreeding strategies to improve phenotypic performance by exploiting heterosis. The present study investigated the genetic diversity and population structure of the most prominent dairy and beef breeds used in Ireland. Illumina high-density genotypes (777 962 single nucleotide polymorphisms; SNPs) were available on 4623 purebred bulls from nine breeds; Angus (n=430), Belgian Blue (n=298), Charolais (n=893), Hereford (n=327), Holstein-Friesian (n=1261), Jersey (n=75), Limousin (n=943), Montbéliarde (n=33) and Simmental (n=363). Principal component analysis revealed that Angus, Hereford, and Jersey formed non-overlapping clusters, representing distinct populations. In contrast, overlapping clusters suggested geographical proximity of origin and genetic similarity between Limousin, Simmental and Montbéliarde and to a lesser extent between Holstein, Friesian and Belgian Blue. The observed SNP heterozygosity averaged across all loci was 0.379. The Belgian Blue had the greatest mean observed heterozygosity (HO=0.389) among individuals within breed while the Holstein-Friesian and Jersey populations had the lowest mean heterozygosity (HO=0.370 and 0.376, respectively). The correlation between the genomic-based and pedigree-based inbreeding coefficients was weak (r=0.171; P<0.001). Mean genomic inbreeding estimates were greatest for Jersey (0.173) and least for Hereford (0.051). The pair-wise breed fixation index (Fst) ranged from 0.049 (Limousin and Charolais) to 0.165 (Hereford and Jersey). In conclusion, substantial genetic variation exists among breeds commercially used in Ireland. Thus custom-mating strategies would be successful in maximising the exploitation of heterosis in crossbreeding strategies.
Sulla (Sulla coronarium L.) forage is valued for its positive impact on ruminant production, in part due to its moderate content of condensed tannin (CT). The duration of daily grazing is a factor affecting the feed intake and milk production of ewes. In this study, the effects of grazing sulla pasture compared with annual ryegrass, and the extension of grazing from 8 to 22 h/day, were evaluated with regard to ewe forage intake and milk production, as well as the physicochemical properties and fatty acid (FA) composition of cheese. During 42 days in the spring, 28 ewes of the Comisana breed were divided into four groups (S8, S22, R8 and R22) that grazed sulla (S) or ryegrass (R) for 8 (0800 to 1600 h) or 22 h/day, and received no feeding supplement. In six cheese-making sessions, cheeses were manufactured from the 48 h bulk milk of each group. Compared with ewes grazing ryegrass, those grazing sulla had higher dry matter (DM) intake, intake rate and milk yield, and produced milk that was lower in fat and higher in casein. Ewes grazing for 22 h spent more time eating, which reduced the intake rate, increased DM and nutrient intake and milk yield, and reduced milk fat. Due to the ability of CT to inhibit the complete ruminal biohydrogenation of polyunsaturated fatty acids (PUFA), the FA composition of sulla cheese was more beneficial for consumer health compared with ryegrass cheese, having lower levels of saturated fatty acids and higher levels of PUFA and n-3 FA. The FA profile of S8 cheese was better than that of S22 cheese, as it was higher in branched-chain FA, monounsaturated FA, PUFA, rumenic acid (c9,t11-C18:2), and had a greater health-promoting index. The effect of short grazing time on sulla was attributed to major inhibition of PUFA biohydrogenating ruminal bacteria, presumably stimulated by the higher accumulation of sulla CT in the rumen, which is related to a higher intake rate over a shorter eating time. Thus, grazing sulla improved the performance of ewes, thereby increasing, especially with short grazing time, the nutritional properties of cheese fat.
Tail lesions are important pig welfare indicators that could be recorded during meat inspection as they are more visible on the carcass than on the live animal. Tail biting is associated with reduced performance in the bitten pig, but it is not clear whether problems with tail biting are reflected in general farm performance figures. Farm advisory services aim to improve farm productivity which could be associated with improvements in pig welfare. Record keeping forms an integral part of such advisory services. The aim of this study was to examine the influence of record keeping in the Teagasc eProfit Monitor (ePM herds) on the prevalence of tail lesion severity scores in Irish slaughter pigs. In addition, we investigated associations between the prevalence of tail lesion scores and production parameters at farm level in ePM herds. Pigs were observed after scalding/dehairing and tail lesion score (0 to 4), sex and farm identification were recorded. Tail lesion scores were collapsed into none/mild lesions (score ⩽1), moderate lesions (score 2) and severe lesions (score ⩾3). The effect of record keeping (ePM herd) on the different tail lesion outcomes was analysed at batch level using the events/trials structure in generalized linear mixed models (PROC GLIMMIX). Spearman’s rank correlations were calculated between average tail lesion score of a batch and production parameters. A total of 13 133 pigs were assessed from 73 batches coming from 61 farms. In all, 23 farms were identified as ePM herds. The average prevalence of moderate tail lesions was 26.8% and of severe tail lesions was 3.4% in a batch. Batches coming from ePM herds had a lower prevalence of moderate tail lesions than non-ePM herds (P<0.001). Average tail lesion score was negatively associated with age (P<0.05) and weight (P<0.05) at sale/transfer of weaners, and tended to be positively associated with the number of finishing days (P=0.06). In addition, the prevalence of severe tail lesions was negatively associated with average daily gain in weaners (P<0.05) and tended to do so with average daily gain in finishers (P=0.08). This study provides the first indication that record keeping through an advisory service may help to lower the risk of tail biting, which is associated with improved farm performance.
The effects of feeding Cistus ladanifer (Cistus) and a blend of soybean and linseed oil (1 : 2 vol/vol) on fatty acid (FA) composition of lamb meat lipids and messenger RNA (mRNA) expression of desaturase enzymes was assessed. In total, 54 male lambs were randomly assigned to 18 pens and to nine diets, resulting from the combination of three inclusion levels of Cistus (50 v. 100 v. 200 g/kg of dry matter (DM)) and three inclusion levels of oil (0 v. 40 v. 80 g/kg of DM). The forage-to-concentrate ratio of the diets was 1 : 1. Longissimus muscle lipids were extracted, fractionated into neutral (NL) and polar lipid (PL) and FA methyl esters obtained and analyzed by GLC. The expression of genes encoding Δ5, Δ6 and Δ9 desaturases (fatty acid desaturase 1 (FADS1), fatty acid desaturase 2 (FADS2) and stearoyl CoA desaturase (SCD)) was determined. Intramuscular fat, NL and PL contents were not affected by oil or Cistus. Oil supplementation reduced (P<0.05) 16:0, c9-16:1, 17:0, c9-17:1 and c9-18:1 FA and increased (P<0.05) 18:2n-6, 18:3n-3 and the majority of biohydrogenation intermediates in NL. Cistus alone had few effects on FA of NL but interacted with oil (P<0.05) by increasing t10-18:1,t10,t12-18:2,t10,c12-18:2 and t7,c9-18:2. The t10-/t11-18:1 ratio increased with both Cistus and oil levels. The c9, t11-18:2 did not increase (P<0.05) with both oil and Cistus dietary inclusion. Oil reduced c9-16:1, 17:0, c9-17:1,c9-18:1, 20:4n-6, 22:4n-6 and 20:3n-9 proportions in PL, and increased 18:2n-6, 18:3n-3, 20:3n-3 and of most of the biohydrogenation intermediates. The Cistus had only minor effects on FA composition of PL. Cistus resulted in a reduction (P<0.05) of 20:5n-3 and 22:6n-3 in the meat PL. The expression level of SCD mRNA increased (P=0.015) with Cistus level, although a linear relationship with condensed tannins intake (P=0.11) could not be established. FADS1 mRNA expressed levels increased linearly (P=0.019) with condensed tannins intake. In summary, the inclusion of Cistus and oil in 1 : 1 forage-to-concentrate ratio diets resulted in a large increase in t10-18:1 and no increase in c9,t11-18:2 or n-3 long chain poor in polyunsaturated fatty acids in lamb meat.
Fertile eggs from Cobb 500 broiler breeder hens were incubated to provide low starting egg shell temperatures (EST; 36.9°C to 37.3°C) which were gradually increased to 37.8°C during the first 7 to 15 days of incubation compared with eggs incubated with a constant EST of 37.8°C (standard conditions) over the first 18 days of incubation. Time of individual chick hatching (measured at 6 h intervals from 468 h of incubation), chick weight, chick length and yolk weight were measured at take-off and BW was measured at 7, 14, 28, 34 and 42 days of age. Male birds at 34 and 42 days of age were assessed for their ability to remain standing in a latency-to-lie test. At 34 and 42 days, male birds were examined for leg symmetry, foot pad dermatitis, hock bruising and scored (scale 0 to 4, where 0=no lesion and 4=lesions extending completely across the tibial growth plate) for tibial dyschondroplasia (TD) lesions. The lower EST profiles caused chicks to hatch later than those incubated under the standard EST profile. Chicks which hatched at ⩽498 h incubation grew faster over the first 7 days than those that hatched later. There were significantly more birds (only males were studied) that hatched from the lower EST profiles with TD scores of 0 and 1 and fewer with score 4 at 34 days than those hatched under the standard profile. Male birds at 34 days with TD lesions ⩾3 stood for significantly shorter times than males with TD scores ⩽2. Moreover, male birds at 34 and 42 days with TD lesion scores of ⩾3 hatched significantly earlier and grew significantly faster over the first 2 weeks of age than did male birds with TD scores ⩽2. It appears possible to decrease the severity and prevalence of TD in the Cobb 500 broiler by ensuring that the birds do not hatch before 498 h of incubation.
Agroecology uses ecological processes and local resources rather than chemical inputs to develop productive and resilient livestock and crop production systems. In this context, breeding innovations are necessary to obtain animals that are both productive and adapted to a broad range of local contexts and diversity of systems. Breeding strategies to promote agroecological systems are similar for different animal species. However, current practices differ regarding the breeding of ruminants, pigs and poultry. Ruminant breeding is still an open system where farmers continue to choose their own breeds and strategies. Conversely, pig and poultry breeding is more or less the exclusive domain of international breeding companies which supply farmers with hybrid animals. Innovations in breeding strategies must therefore be adapted to the different species. In developed countries, reorienting current breeding programmes seems to be more effective than developing programmes dedicated to agroecological systems that will struggle to be really effective because of the small size of the populations currently concerned by such systems. Particular attention needs to be paid to determining the respective usefulness of cross-breeding v. straight breeding strategies of well-adapted local breeds. While cross-breeding may offer some immediate benefits in terms of improving certain traits that enable the animals to adapt well to local environmental conditions, it may be difficult to sustain these benefits in the longer term and could also induce an important loss of genetic diversity if the initial pure-bred populations are no longer produced. As well as supporting the value of within-breed diversity, we must preserve between-breed diversity in order to maintain numerous options for adaptation to a variety of production environments and contexts. This may involve specific public policies to maintain and characterize local breeds (in terms of both phenotypes and genotypes), which could be used more effectively if they benefited from the scientific and technical resources currently available for more common breeds. Last but not least, public policies need to enable improved information concerning the genetic resources and breeding tools available for the agroecological management of livestock production systems, and facilitate its assimilation by farmers and farm technicians.
Salers are a native French breed used for beef and dairy production that has expanded to all the continents. The Salers breed was introduced to the north of Spain in 1985 with only 15 individuals from France and has successfully increased to over 20 000 animals. Although over time new animals have been imported from France for breeding, it is possible that the limiting number of founder animals could have resulted in a reduction of the genetic diversity found in Spanish Salers. Thus, the purpose of the present study has been to characterize the genetic diversity of Salers breed in Spain and evaluate a possible founder effect due to reduced number of the first reproducers. A total of 403 individuals from 12 Salers herds were analyzed using 12 microsatellite markers and compared with phylogenetically and geographically close related Blonde d’Aquitaine, Limousin and Charolais French breeds but also other 16 European breeds. Microsatellites in Salers were polymorphic, with a mean allelic richness of 5.129 and an expected heterozygosity of 0.621 across loci (0.576 to 0.736 among all breeds). Average observed heterozygosity was 0.618. All the loci fit the Hardy–Weinberg (HW) equilibrium except TGLA227 locus due to a significant deficit of heterozygotes in only one of the herds, probably attributable to a sampling effect. When all loci were combined, Salers inbreeding coefficient did not differ statistically from 0 (FIS=0.005), indicating not significant excess or deficit of heterozygotes (P=0.309). Based in allelic distribution, Salers revealed a frequency of 0.488 in BM2113-131 and 0.064 in BM2113-143 diagnostic alleles, which are specific to the African zebu. These zebu alleles are also found in some French breeds, supported by STR data previously postulated hypothesis of a migration route through Mediterranean route by which North African cattle may have left a genetic signature in southern Europe. Phylogenetic tree and population structure analyses could unambiguously differentiate Salers cattle from the other populations and 10% of the total genetic variability could be attributed to differences among breeds (mean RST=0.105; P<0.01). Mutation-drift equilibrium tests (sign test and Wilcoxon’s sign rank test) were in correspondence to the absence of founder effect when Bonferroni was applied. Gene diversity previously reported in French Salers was comparable with the observed in our population. Thus, high genetic diversity in Spanish Salers highlights the resources of this population, which looks toward future breeding and selection programs.
Inherited optic neuropathies are a heterogeneous group of disorders characterized by mild to severe visual loss, colour vision deficit, central or paracentral visual field defects and optic disc pallor. Optic atrophies can be classified into isolated or non-syndromic and syndromic forms. While multiple modes of inheritance have been reported, autosomal dominant optic atrophy and mitochondrial inherited Leber's hereditary optic neuropathy are the most common forms. Optic atrophy type 1, caused by mutations in the OPA1 gene is believed to be the most common hereditary optic neuropathy, and most patients inherit a mutation from an affected parent. In this study we used whole-exome sequencing to investigate the genetic aetiology in a patient affected with isolated optic atrophy. Since the proband was the only affected individual in his extended family, and was a product of consanguineous marriage, homozygosity mapping followed by whole-exome sequencing were pursued. Exome results identified a novel de novo OPA1 mutation in the proband. We conclude, that though de novo OPA1 mutations are uncommon, testing of common optic atrophy-associated genes such as mitochondrial mutations and OPA1 gene sequencing should be performed first in single individuals presenting with optic neuropathy, even when dominant inheritance is not apparent.
The Danish plant scientist Wilhelm Johannsen (1857–1927) is remembered for his experimental work on plant heredity, and as a founding figure of modern genetics. The terms 'gene', 'genotype' and 'phenotype' were first used by him. The results of his studies on beans supported theories advanced during the 1890s by the Dutch botanist Hugo de Vries, who had unknowingly replicated the work of Gregor Mendel, published in English translation in 1902 (also reissued in this series) by William Bateson. Johannsen's proposal that changes in heredity resulted from sudden mutations rather than from slow processes of natural selection was seen at the time as a threat to Darwinian theory, though later research showed otherwise. This influential book, first published in 1909 (with later editions in 1913 and 1926), is a revised, expanded German translation of a 1905 Danish book by Johannsen, itself based on a journal article originally published in 1903.
Selection for high levels of prolificacy has allowed substantial improvements in the production efficiency of New Zealand (NZ) sheep farms, but the consequences on ewe lifetime performance are mostly unknown. In this study, the relationship between the level of prolificacy early in ewes’ productive lives and their probability to survive later (i.e. stayability) was evaluated in two contrasting NZ flock environments. Records were obtained from 6605 ewes from four ram breeder flocks representing either a moderate (n=2) or a highly variable (n=2) nutritional environment. All ewes lambed for the first time at 2 years of age and were mated the following year. The number of lambs born during the first 2 years of productive life (NLB2–3) was used as a measure of early prolificacy. Effects of NLB2–3 on stayability to 4, 5, 6, 7 and 8 years old were analysed using logistic regression. Curvilinear effects (logit-transformed) were detected (P<0.05) until stayability to 6 years and to 8 years old in the highly variable and the moderate environment, respectively. The NLB2–3 that resulted in maximum expected stayability to various ages was 3.9 to 4.2, and 4.5 to 4.7 lambs in the highly variable and in the moderate flock environments, respectively. In addition, ewe stayability was reduced when the proportion of the litter that survived from birth to weaning (i.e. ewe rearing ability) was submaximal during the early productive life. High prolific ewes had a low rearing ability whatever the environment whereas the rearing ability of lowly prolific ewes was apparently more sensitive to the nutritional environment. The poor maternal performance of ewes with low levels of NLB2–3 led to a premature culling by breeders whereas the high early reproductive effort associated with high levels of NLB2–3 seemed to be at the cost of ewes’ survival, even in the moderate flock environment. In conclusion, the flock environment influenced the level of early prolificacy beyond which ewe longevity was reduced. It is suggested that further selection for high and early prolificacy in NZ flocks is likely to impair ewes’ lifetime productivity.
