The evolutionary relationships of the nematode genus Loofilaimus are explored with an integrative approach, combining morphological and molecular (28S-rDNA) data. Never recorded since its original description in 1998, the finding of fresh specimens of its type and only species, L. phialistoma, allowed us to obtain SEM observations and sequencing, both for the first time, resulting in relevant aspects to elucidate its phylogeny. Morphologically, the genus is characterized by two autapomorphies affecting its lip region and pharynx. Molecular study revealed that it represents a very restricted evolutionary trend within Dorylaimida. The clade (Nygolaimina + (Loofilaimus + Dorylaimina)) is well supported. Loofilaimidae is accepted as a separate and valid family, which should also include Bertzuckermania.

Members of the genus Strigea Abildgaard, 1790 are endoparasites of birds distributed worldwide. Adults of an undescribed species of the genus Strigea were collected from the intestines of two hawk species (Rupornis magnirostris and Accipiter coperii). Other species identified as Parastrigea macrobursa that were described in Argentina were also recovered from two hawk species (Buteogallus urubitinga and Buteogallus anthracinus) in three localities along the coasts of Mexico. Specimens of the two species were sequenced for three molecular markers, the internal transcribed spacers locus (ITS1-5.8S rDNA- ITS2) and the domains D1–D3 from the large subunit from nuclear ribosomal DNA and the cytochrome c oxidase subunit 1 from mitochondrial DNA. The newly sequenced specimens were aligned with other strigeids sequences downloaded from GenBank. Maximum likelihood and Bayesian analyses inferred with each molecular marker revealed that our specimens of Strigea sp. formed an independent lineage, which is recognized herein as a new species, Strigea magnirostris n. sp., representing the first species in Mexico and the 16th in the Neotropical region. Morphologically, the new species is distinguished from other congeneric species from the Americas by having an oral sucker with several papillae around it, well-developed pseudosuckers (118–248 μm), a tegument covered with tiny spines, a larger cone genital (193–361 × 296–637) and a larger copulatory bursa (247–531 × 468–784). Our phylogenetic analyses revealed that P. macrobursa is not closely related to other members of the genus Parastrigea and is nested within Strigea, suggesting that P. macrobursa should be transferred to Strigea to form Strigea macrobursa n. comb., expanding its distribution range from Mexico to Argentina. Finally, the analyses also revealed that the taxonomy and systematics of Strigea should be re-evaluated, combining morphological and molecular characteristics.

Echinolaelaps echidninus is a gamasid mite that is of medical and veterinary significance as parasites and vectors of disease agents, which can carry pathogens of zoonosis such as Rickettsia tsutsugamushi, Rickettsia Q fever, Rickettsia mooseri, Rickettsia pox pathogens, Corynebacterium pseudotuberculosis and Leptospira. At present, only single mitochondrial genes have been analysed for E. echidninus in the world, and no complete mitochondrial genome has been reported. However, information carried by a single gene is limited. Therefore, the complete mitochondrial genome of E. echidninus was determined for the first time by Illumina Hiseq X-Ten platform in this study. The mitochondrial genome is 15 736 bp in length and contains 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes and a control region of 1561 bp in length. Codon analysis of 13 protein-coding genes revealed that UUU, UUA, AUU, AUA and AAU were the most frequently used, while cox2 had the fastest evolutionary rate and cob the slowest. Comparative analysis of genome structure and breakpoint distances of the mitochondrial genomes of 23 species in 17 genera from 10 families of Gamasida deposited in GenBank revealed a novel gene arrangement type of the E. echidninus mitochondrial genome, and different degrees of rearrangement among different taxa of Gamasida. Phylogenetic analyses of Gamasida were performed using the maximum likelihood and Bayesian inference methods. Echinolaelaps echidninus was clustered with Dermanyssoidea and formed a more supportive sister group with Varroa destructor. This study provides novel insights into rearrangement patterns and evolution of mitochondrial genomes of Gamasida.

The origin of carbon fixation is a fundamental question in astrobiology. While the Calvin cycle is the most active on the modern Earth, the reductive tricarboxylic acid (TCA) cycle (rTCA) pathway for carbon fixation has been proposed to have played an important role in early evolution. In this study, we examined the evolution of key enzymes in the rTCA, which are rare in extant organisms, occurring in a few groups of Bacteria and Archaea. We investigated one of the least common reactions of this pathway, cleavage of citrate into oxaloacetate and acetyl-CoA, which can be performed by either a two-enzyme system (CCS/CCL) or a single enzyme (ACL) that is assumed to be the result of fusion of the two active sites into a single polypeptide. For broader context, we also studied functionally diverged homologues of these enzymes, succinyl-CoA synthetase (SCS) and citrate synthase. Our phylogenetic analysis of these enzymes in Bacteria and Archaea shows that SCS, a homologue of CCS from distant bacterial taxa capable of citrate cleavage, are monophyletic, suggesting linked horizontal gene transfers of SCS and citrate cleavage enzymes. We also found evidence of the horizontal transfer of SCS from a clade of anaerobic Archaea (Archaeoglobi, Methanomicrobia or Crenarchaeota) to an ancestor of Cyanobacteria/Melainabacteria clade – both of which share a succinate semialdehyde shunt in their oxidative TCA cycles. We identified new bacterial and archaeal taxa for which complete rTCA cycles are theoretically possible, including Syntrophobacter, Desulfofundulus, Beggiatoa, Caldithrix, Ca. Acidulodesulfobacterales and Ca. Micrarchaeota. Finally, we propose a mechanism for syntrophically-regulated fluxes through oxidative and rTCA reactions in microbial communities particularly Haloarchaea-Nanohaloarchaea symbiosis and its implications for carbon fixation during retinal-based phototrophy and the Purple Earth hypothesis. We discuss how the inclusion of an ecological perspective in the studies of evolution of ancient metabolic pathways may be beneficial to understanding the origin of life.

The aquatic and terrestrial clades of species of Trypanosoma could provide insight into the evolutionary history of the genus, as well as complementary information for biomedical studies of medically and economically important species of Trypanosoma. The ecological interactions and phylogeny of aquatic trypanosomes are currently not well-understood, mostly due to their complex life cycles and a deficiency of data. The species of Trypanosoma from African anuran hosts are of the least understood taxa in the genus. Trypanosomes were collected from South African frogs and subjected to morphological and phylogenetic analyses. This study redescribes Trypanosoma (Trypanosoma) nelspruitense Laveran, 1904 and Trypanosoma (Haematomonas) grandicolor Pienaar, 1962, with morphological and molecular data. The present study aims to create a platform for further future research on African anuran trypanosomes.

Methodologies that analyse the colouration and external appearance of extant species are very useful tools when facing one of the greatest challenges in the palaeoartistic reconstructions of extinct fauna: inferring the colour patterns. Earlier works have applied this methodology, for example, in the reconstruction of the Miocene bovid Tethytragus, proving that the maximum likelihood (ML) analysis to infer ancestral states has promising potential. This study offers a proposal for the reconstruction of the external appearance of Heteroprox moralesi Azanza 1989, an early cervid of the Middle Miocene present in several fossil sites of Central Spain. For the reconstruction of the external appearance, the colour patterns of all the extant species of the family Cervidae were studied with the method of ML analysis, as well as recent works about their phylogeny. The results show the most probable basal colour pattern of the cervids: dark shades on the limbs, dorsal section, and head of the animal, and, in contrast, lighter colours on the neck and perianal region. This basal pattern can be used as a basis for reconstructing colouration and to hypothesise about the external appearance of extinct taxa. Furthermore, the inferred forest habitat of H. moralesi has been taken into consideration in order to adjust the colour pattern, comparing the final results of the analysis performed in this study with that of the pattern observed in extant forest deer as well as with previous works employing this methodology.

This study re-examined the status of species of Diorygma Eshw. known from the Western Ghats using an integrative taxonomy approach that includes morphological and chemical data, as well as multigene phylogenetic analyses. Prior to this work, the two species D. karnatakense and D. dandeliense were distinguished primarily on lirellae morphology (branching pattern) and the number of ascospores per ascus. Our study of the morphology, chemistry and molecular phylogeny (mtSSU, LSU and RPB2) of freshly collected samples and re-examination of type material suggests that both names should be synonymized. Consequently, D. karnatakense is accepted as the correct name, with D. dandeliense as a newly proposed synonym. Phylogenetically, D. karnatakense is allied to D. antillarum and D. hieroglyphicum.

Paravulvus zhongshanensis sp. nov., isolated from soil in a location at Jiangsu Province, China, is described and illustrated based on morphological, morphometric and molecular characterizations. The new species is characterized by its body 1.17–1.53 mm long, lip region offset by marked constriction and 12.1–13.8 μm broad, mural tooth deltoid and 9.6–11.7 μm long, neck 278–360 μm long, pharyngeal expansion 164–208 μm long or occupying more than one-half (54–62%) of total neck length, uterus 32.5–35.3 μm long or 1.0–1.1 times the corresponding body diameter, V = 47.8–53.4, paravulvae absent, female tail subcylindrical conoid (30.5–39.5 μm, c = 36.0–45.5, c′ = 1.7–2.2) with widely rounded end, and male unknown. The new species was compared with six known species of the genus including Paravulvus acuticaudatus, Paravulvus confusus, Paravulvus hartingii, Paravulvus iranicus, Paravulvus loofi and Paravulvus microdontus mainly by similarities in having conical tail and c′ value larger than 1.3. The rRNA and mitochondrial cytochrome oxidase subunit 1 genes of the new species were obtained and were used for reconstructing the phylogenetic relationships of the new species.

A new species of the genus Metaxonchium is described from a natural habitat in Iran. Metaxonchium magnum sp. n. is characterized by its 3.62–4.65 mm long body, lip region cap-like and offset by constriction and 13–16 μm wide, odontostyle fusiform and 14–17 μm long, neck 1016–1359 μm long, both parts of the pharynx separated by a short isthmus-like narrowing, pharyngeal expansion occupying 74.2 (73–77)% of total neck length in females and 70.4 (66–72)% in males, female genital system mono-opistho-ovarian, didelphic, anterior genital branch a large uterine sac with a small terminal mass occupying 7–14% of body length, posterior uterus long and tripartite with a Z-like differentiation, V = 50–52, caudal region short and rounded (24–41 μm, c = 99–161, c′ = 0.5–0.7), spicules 90–105 μm long and 10–13 spaced ventromedian supplements with hiatus. Analysis of D2–D3 28S rDNA sequences of the new species suggests that Metaxonchium might not be a monophyletic taxon, a matter that should be confirmed after future research.
