Along with increasing product complexity and quality requirements, the consistent consideration of inevitable production-induced variations within the product development process becomes a decisive factor for the market success of products. Consequently, various tolerancing approaches have emerged over the last few decades. However, tolerancing is considered complex in education, research, and industry, as it is a highly interdisciplinary task that takes place at different levels of detail, ranging from Robust Design to tolerance specification to manufacturing process design. This contribution proposes a novel approach that allows a holistic and structured description of tolerancing and fosters a common understanding among all involved stakeholders from design, manufacturing, and inspection. This is achieved by categorizing it into several distinct elements: activities, methods, tools, models and data, information, and knowledge. This ensures clarity and supports the utilization of existing approaches. While this contribution focuses on tolerancing in product design, the linkage to subsequent product realization stages and further engineering domains is also addressed in the proposed description scheme. An exemplary classification of research papers and a description of a practical development process of a technical system with its different tolerancing activities illustrate the benefits of the proposed description scheme.
Although the link between alcohol involvement and behavioral phenotypes (e.g. impulsivity, negative affect, executive function [EF]) is well-established, the directionality of these associations, specificity to stages of alcohol involvement, and extent of shared genetic liability remain unclear. We estimate longitudinal associations between transitions among alcohol milestones, behavioral phenotypes, and indices of genetic risk.
Methods
Data came from the Collaborative Study on the Genetics of Alcoholism (n = 3681; ages 11–36). Alcohol transitions (first: drink, intoxication, alcohol use disorder [AUD] symptom, AUD diagnosis), internalizing, and externalizing phenotypes came from the Semi-Structured Assessment for the Genetics of Alcoholism. EF was measured with the Tower of London and Visual Span Tasks. Polygenic scores (PGS) were computed for alcohol-related and behavioral phenotypes. Cox models estimated associations among PGS, behavior, and alcohol milestones.
Results
Externalizing phenotypes (e.g. conduct disorder symptoms) were associated with future initiation and drinking problems (hazard ratio (HR)⩾1.16). Internalizing (e.g. social anxiety) was associated with hazards for progression from first drink to severe AUD (HR⩾1.55). Initiation and AUD were associated with increased hazards for later depressive symptoms and suicidal ideation (HR⩾1.38), and initiation was associated with increased hazards for future conduct symptoms (HR = 1.60). EF was not associated with alcohol transitions. Drinks per week PGS was linked with increased hazards for alcohol transitions (HR⩾1.06). Problematic alcohol use PGS increased hazards for suicidal ideation (HR = 1.20).
Conclusions
Behavioral markers of addiction vulnerability precede and follow alcohol transitions, highlighting dynamic, bidirectional relationships between behavior and emerging addiction.
Electroencephalography is an accessible, portable, noninvasive and safe means of evaluating a patient’s brain activity. It can aid in diagnosis and management decisions for post-cardiac arrest patients with seizures, myoclonus and other non-epileptic movements. It also plays an important role in a multimodal approach to neuroprognostication predicting both poor and favorable outcomes. Individuals ordering, performing and interpreting these tests, regardless of the indication, should understand the supporting evidence, logistical considerations, limitations and impact the results may have on postarrest patients and their families as outlined herein.
Alcohol use is influenced by genetic and environmental factors. We examined the interactive effects between genome-wide polygenic risk scores for alcohol use (alc-PRS) and social support in relation to alcohol use among European American (EA) and African American (AA) adults across sex and developmental stages (emerging adulthood, young adulthood, and middle adulthood). Data were drawn from 4,011 EA and 1,274 AA adults from the Collaborative Study on the Genetics of Alcoholism who were between ages 18–65 and had ever used alcohol. Participants completed the Semi-Structured Assessment for the Genetics of Alcoholism and provided saliva or blood samples for genotyping. Results indicated that social support from friends, but not family, moderated the association between alc-PRS and alcohol use among EAs and AAs (only in middle adulthood for AAs); alc-PRS was associated with higher levels of alcohol use when friend support was low, but not when friend support was high. Associations were similar across sex but differed across developmental stages. Findings support the important role of social support from friends in buffering genetic risk for alcohol use among EA and AA adults and highlight the need to consider developmental changes in the role of social support in relation to alcohol use.
Researchers have identified genetic and neural risk factors for externalizing behaviors. However, it has not yet been determined if genetic liability is conferred in part through associations with more proximal neurophysiological risk markers.
Methods
Participants from the Collaborative Study on the Genetics of Alcoholism, a large, family-based study of alcohol use disorders were genotyped and polygenic scores for externalizing (EXT PGS) were calculated. Associations with target P3 amplitude from a visual oddball task (P3) and broad endorsement of externalizing behaviors (indexed via self-report of alcohol and cannabis use, and antisocial behavior) were assessed in participants of European (EA; N = 2851) and African ancestry (AA; N = 1402). Analyses were also stratified by age (adolescents, age 12–17 and young adults, age 18–32).
Results
The EXT PGS was significantly associated with higher levels of externalizing behaviors among EA adolescents and young adults as well as AA young adults. P3 was inversely associated with externalizing behaviors among EA young adults. EXT PGS was not significantly associated with P3 amplitude and therefore, there was no evidence that P3 amplitude indirectly accounted for the association between EXT PGS and externalizing behaviors.
Conclusions
Both the EXT PGS and P3 amplitude were significantly associated with externalizing behaviors among EA young adults. However, these associations with externalizing behaviors appear to be independent of each other, suggesting that they may index different facets of externalizing.
The modern Japanese nation-state that was established from 1868 onwards was marked by a strong tendency towards the separation of state and religion: religions were protected as a private matter, but the public sphere was resolutely kept free of them. This was mainly done so that competing religions would not get in the way of state-sanctioned emperor worship. The latter, although imbued with elements from Shinto, was carefully defined as non-religious, so that emperor worship could be prescribed without harm to the constitutionally guaranteed freedom of religion. This secularist approach to policing religions was broadly shared among Japanese elites—but it did not remain unopposed.
From around the turn of the twentieth century, dissatisfaction with the separation of the religious and the secular spheres began to be voiced, especially by pan-Asianist activists, who sought to combine the spiritual unity of Asia with the political liberation of Asian countries from Western colonialism and imperialism. Although Japanese pan-Asianism has conventionally been seen as a purely political movement, one cannot explain it fully without taking into account its spiritual dimension, which up to the 1920s drew its primary inspiration from India. This article will show how pan-Asianist activists in Japan opposed mainstream secularism and discuss what their vision for a unified Asia was. In doing so, it will focus on the Japanese reception of the Frenchman Paul Richard, an important political activist-cum-spiritual seeker who was a central node in the network of Indian and Japanese pan-Asianists in the early twentieth century.
The purpose of this study was to examine possible pathways by which genetic risk associated with externalizing is transmitted in families. We used molecular data to disentangle the genetic and environmental pathways contributing to adolescent externalizing behavior in a sample of 1,111 adolescents (50% female; 719 European and 392 African ancestry) and their parents from the Collaborative Study on the Genetics of Alcoholism. We found evidence for genetic nurture such that parental externalizing polygenic scores were associated with adolescent externalizing behavior, over and above the effect of adolescents’ own externalizing polygenic scores. Mediation analysis indicated that parental externalizing psychopathology partly explained the effect of parental genotype on children’s externalizing behavior. We also found evidence for evocative gene-environment correlation, whereby adolescent externalizing polygenic scores were associated with lower parent–child communication, less parent–child closeness, and lower parental knowledge, controlling for parental genotype. These effects were observed among participants of European ancestry but not African ancestry, likely due to the limited predictive power of polygenic scores across ancestral background. These results demonstrate that in addition to genetic transmission, genes influence offspring behavior through the influence of parental genotypes on their children’s environmental experiences, and the role of children’s genotypes in shaping parent–child relationships.
The coronavirus disease 2019 (COVID-19) pandemic challenged health care systems in an unprecedented way. Due to the enormous amount of hospital ward and intensive care unit (ICU) admissions, regular care came to a standstill, thereby overcrowding ICUs and endangering (regular and COVID-19-related) critical care. Acute care coordination centers were set up to safely manage the influx of COVID-19 patients. Furthermore, treatments requiring ICU surveillance were postponed leading to increased waiting lists.
Hypothesis:
A coordination center organizing patient transfers and admissions could reduce overcrowding and optimize in-hospital capacity.
Methods:
The acute lack of hospital capacity urged the region West-Netherlands to form a new regional system for patient triage and transfer: the Regional Capacity and Patient Transfer Service (RCPS). By combining hospital capacity data and a new method of triage and transfer, the RCPS was able to effectively select patients for transfer to other hospitals within the region or, in close collaboration with the National Capacity and Patient Transfer Service (LCPS), transfer patients to hospitals in other regions within the Netherlands.
Results:
From March 2020 through December 2021 (22 months), the RCPS West-Netherlands was requested to transfer 2,434 COVID-19 patients. After adequate triage, 1,720 patients with a mean age of 62 (SD = 13) years were transferred with the help of the RCPS West-Netherlands. This concerned 1,166 ward patients (68%) and 554 ICU patients (32%). Overcrowded hospitals were relieved by transferring these patients to hospitals with higher capacity.
Conclusion:
The health care system in the region West-Netherlands benefitted from the RCPS for both ward and ICU occupation. Due to the coordination by the RCPS, regional ICU occupation never exceeded the maximal ICU capacity, and therefore patients in need for acute direct care could always be admitted at the ICU. The presented method can be useful in reducing the waiting lists caused by the delayed care and for coordination and transfer of patients with new variants or other infectious diseases in the future.
Type I/II interferons (IFNα,β/IFNɣ) are cytokines that activate signal-transducer-and-activator-of-transcription-1 (STAT1). The STAT1 N-terminal domain (NTD) mediates dimerization and cooperative DNA-binding. The STAT1 DNA-binding domain (DBD) confers sequence-specific DNA-recognition. STAT1 has been connected to growth inhibition, replication stress and DNA-damage. We investigated how STAT1 and NTD/DBD mutants thereof affect fibrosarcoma cells. STAT1 and indicated mutants do not affect proliferation of resting and IFNα-treated cells as well as checkpoint kinase signaling, and phosphorylation of the tumor-suppressive transcription factor p53 ensuing ɣ-irradiation. Of the STAT1 reconstituted U3A cells those with STAT1 NTD mutants accumulate the highest levels of the replication stress/DNA-damage marker S139-phosphorylated histone H2AX (ɣH2AX). This is similarly seen with a STAT1 NTD/DBD double mutant, indicating transcription-independent effects. Furthermore, U3A cells with STAT1 NTD mutants are most susceptible to apoptotic DNA fragmentation and cleavage of the DNA repair protein PARP1. These data provide novel insights into the relevance of the STAT1 NTD.
We investigate the phonotactic behaviour of nasal consonants in a database of over 200 languages. Our findings challenge the common classification of nasals as intermediate between obstruents and liquids on the sonority hierarchy. Instead, we propose that there are two types of nasal consonants, one group with lower sonority than liquids and one with higher sonority. We propose that these two types of nasals differ in the presence or absence of a value for the feature [±continuant].
Studies suggest that alcohol consumption and alcohol use disorders have distinct genetic backgrounds.
Methods
We examined whether polygenic risk scores (PRS) for consumption and problem subscales of the Alcohol Use Disorders Identification Test (AUDIT-C, AUDIT-P) in the UK Biobank (UKB; N = 121 630) correlate with alcohol outcomes in four independent samples: an ascertained cohort, the Collaborative Study on the Genetics of Alcoholism (COGA; N = 6850), and population-based cohorts: Avon Longitudinal Study of Parents and Children (ALSPAC; N = 5911), Generation Scotland (GS; N = 17 461), and an independent subset of UKB (N = 245 947). Regression models and survival analyses tested whether the PRS were associated with the alcohol-related outcomes.
Results
In COGA, AUDIT-P PRS was associated with alcohol dependence, AUD symptom count, maximum drinks (R2 = 0.47–0.68%, p = 2.0 × 10−8–1.0 × 10−10), and increased likelihood of onset of alcohol dependence (hazard ratio = 1.15, p = 4.7 × 10−8); AUDIT-C PRS was not an independent predictor of any phenotype. In ALSPAC, the AUDIT-C PRS was associated with alcohol dependence (R2 = 0.96%, p = 4.8 × 10−6). In GS, AUDIT-C PRS was a better predictor of weekly alcohol use (R2 = 0.27%, p = 5.5 × 10−11), while AUDIT-P PRS was more associated with problem drinking (R2 = 0.40%, p = 9.0 × 10−7). Lastly, AUDIT-P PRS was associated with ICD-based alcohol-related disorders in the UKB subset (R2 = 0.18%, p < 2.0 × 10−16).
Conclusions
AUDIT-P PRS was associated with a range of alcohol-related phenotypes across population-based and ascertained cohorts, while AUDIT-C PRS showed less utility in the ascertained cohort. We show that AUDIT-P is genetically correlated with both use and misuse and demonstrate the influence of ascertainment schemes on PRS analyses.
Very Long Baseline Interferometry (VLBI) at sub-millimeter waves has the potential to image the shadow of the black hole in the Galactic Center, Sagittarius A* (Sgr A*), and thereby test basic predictions of the theory of general relativity. We investigate the imaging prospects of a new Space VLBI mission concept. The setup consists of two satellites in polar or equatorial circular Medium-Earth Orbits with slightly different radii, resulting in a dense spiral-shaped uv-coverage with long baselines, allowing for extremely high-resolution and high-fidelity imaging of radio sources. We simulate observations of a general relativistic magnetohydrodynamics model of Sgr A* for this configuration with noise calculated from model system parameters. After gridding the uv-plane and averaging visibilities accumulated over multiple months of integration, images of Sgr A* with a resolution of up to 4 μ as could be reconstructed, allowing for stronger tests of general relativity and accretion models than with ground-based VLBI.
All living things come in contact with radiation; most radiation exposure does not cause harmful complications. Certain types of radiation, specifically ionizing radiation, provide useful interventions in cancer treatment. This type of radiation provokes cellular injury from which normal tissue recovers more effectively than malignant tissue.
Radiation may be provided from isotopes such as radium, iridium, and cesium. Machines such as the linear accelerator or the betatron also create therapeutic forms of radiation which are important in cancer management. Terms used in the discussion of radiation are defined in Box 28.1. This chapter will describe treatment mechanisms and complications of radiation therapy in gynecologic malignancies. Cervical and endometrial cancers are the most commonly treated gynecologic cancers, but radiation also has a role in the management of leiomyosarcoma and vulvar cancers. There is no role for radiation therapy in the management of ovarian cancers due to the effects of radiation on the bowel.
Box 28.1 Terms Used in Discussion of Radiation
X-rays – a form of electromagnetic radiation. X-rays are emitted by electrons.
Gamma rays – these radiations originate from the decay of an atomic nucleus. The wavelengths of gamma rays are typically longer than the wavelengths of x-rays, but some types of gamma rays used in radiation therapy are also referred to as x-rays.
Photon – the elementary particle of ionizing radiation. Gamma rays and x-rays are both considered photons.
Ionization – this process occurs when an atom or proton gains or loses an electron; it thus forms an ion and becomes either negatively or positively charged.
Ionizing radiation – a form of radiation that is harmful to living tissue because it disrupts molecular bonds through the process of ionization.
Scope of the Problem
The most common gynecologic cancers treated by radiation therapy are cervical cancer and endometrial cancer. In early stages, surgery alone may be selected while in other stages the patient may be treated with a combination of surgery and radiation therapy, or with both chemotherapy and radiation. In the most advanced cases, radiation therapy may be used alone. The effects on normal surrounding tissues usually limit the dose of radiation. The sigmoid, the recto sigmoid, and the rectum are the most susceptible pelvic organs to radiation injury. The small bowel also limits radiation dose.
Vowel nasalisation in American English has been the subject of a long discussion as to whether it is a phonological process or whether it should better be analysed as phonetic coarticulation. As a predictable allophonic process, vowel nasalisation also provides a testing ground for theories of lexical representation, since if it turns out that language users store this predictable information in long-term memory, there is no reason to assume any kind of phonological underspecification. In this article, experimental studies on this phenomenon are reviewed with these two questions in mind: is the phenomenon a phonological process at all and is this predictable information, i.e. vowel nasalisation, stored in the mental lexicon. The majority of studies provide evidence supporting the phonological view and underspecified lexical representations.
Recent scholarship has highlighted the importance of Islam for prewar Japanese pan-Asianists. Yet, by considering Islam solely as a political factor, this strand of scholarship has largely overlooked the religious dimension of Japanese pan-Asianism. The existence of six different complete translations of the Qur'ān into Japanese, however, amply bespeaks a genuinely religious interest in Islam, an impression that is corroborated by a look at the sociopolitical contexts of the translations and the biographical backgrounds of the translators. While explicitly anti-modern, anti-Western, and anti-Christian notions were at work in these broadly pan-Asianist Japanese appropriations of Islam, an analysis of the terminology used in the translations shows that, ironically, Christian precedents were not easily overcome.
At the heart of generative phonology lies the assumption that the sounds of every language have abstract underlying representations, which undergo various changes in order to generate the 'surface' representations; that is, the sounds we actually pronounce. The existence, status and form of underlying representations have been hotly debated in phonological research since the introduction of the phoneme in the nineteenth century. This book provides a comprehensive overview of theories of the mental representation of the sounds of language. How does the mind store and process phonological representations? Krämer surveys the development of the concept of underlying representation over the last 100 years or so within the field of generative phonology. He considers phonological patterns, psycholinguistic experiments, statistical generalisations over data corpora and phenomena such as hypercorrection. The book offers a new understanding of contrastive features and proposes a modification of the optimality-theoretic approach to the generation of underlying representations.
In Chapter 3 we found that the omission of feature values in underlying representations became discredited by the end of the 1960s. Underspecification in the form of zeroes or blanks (rather than u and m markings) had a revival in the 1980s, which came in two flavours: Contrastive Underspecification (Steriade 1987, 1995, Clements 1988) and Radical Underspecification (Kiparsky 1982, Archangeli 1984, 1985, 1988, Pulleyblank 1986). The main point of disagreement between the two approaches lay in the question of how to determine feature values and whether this could be done on a language-specific basis or had to follow universal principles, i.e., make use of universally valid feature-filling rules and constraints. A third approach to underspecification, the deduction of feature specifications (and lack thereof) on the basis of a Contrastive Hierarchy, reappeared in the late 1990s and the following decade with the work of Dresher and his associates (Dresher 2003, 2008, 2009, 2010 and references there). We have seen such a hierarchy or division tree in the discussion of Trubetzkoy in Chapter 2 and it had been a common tool in generative phonology in the 1950s, e.g., in Halle (1959).
The 1980s also saw a renewed interest in unary or privative features, which we also have met in passing in the section on Trubetzkoy’s work (Section 2.3). A privative feature is either present or absent rather than taking a value. Thus, zero is one of two options rather than one of three and the adoption of privative features renders large parts of the discussion around underspecification obsolete. However, the issue does not vanish completely. If we take, for instance, the feature [voice], which has been argued to be privative by Lombardi (1991) and others, we see that underspecification is still an option. While in a language contrasting /p,t,k/ with /b,d,g/ the voiceless series is now necessarily underspecified, the sonorants in the same language, which are redundantly voiced at the surface, could be specified as [voice] underlyingly for the sake of completeness or lack this feature because it is redundant.
