OBJECTIVES/GOALS: The primary research goal was to identify brain alterations reliably associated with obesity using coordinate-based meta-analysis. A secondary goal was to compare brain alterations in metabolically healthy (MHO) and unhealthy (MUO) obesity. METHODS/STUDY POPULATION: Source data were peer-reviewed studies reporting locations of gray-matter alterations in group-average, case-control contrasts (obese vs. non-obese) cohorts, performed in a whole-brain, voxel-wise manner. Both voxel-based morphometry and voxel-based physiology studies were included. Three coordinate-based meta-analyses were performed: Pooled (MUO + MHO), MHO, and MUO. RESULTS/ANTICIPATED RESULTS: Thirty-two studies reporting a total of 50 case-control contrasts (MHO, 23; MUO, 27) met inclusion criteria, representing 3,368 participants (obese, 1,781; non-obese, 1587). The pooled analysis yielded 8 cerebral foci (3 nuclear, 5 cortical) in regions implicated in reward-seeking, cognitive, and interoceptive behaviors. MHO yielded 7 cerebral foci (4 nuclear, 3 cortical), partially overlapping Pooled results, with similar behavioral loadings. The MUO pattern was distinct, with 3 cerebellar and 1 occipital foci. DISCUSSION/SIGNIFICANCE: Brain alterations occurred reliably in obesity. The dominant pattern (Pooled & MHO) involved cerebral reward-system circuits, evident even in metabolically healthy obesity. Cerebellar alterations occurred exclusively in metabolically unhealthy obesity, a pattern previously reported in metabolic syndrome.
Information on pangolin life history and ecology is becoming increasingly available in African countries through community-based surveys and camera-trapping. However, there is a paucity of information on the largely arboreal black-bellied pangolin Phataginus tetradactyla, which is categorized as Vulnerable on the IUCN Red List. By targeting fallen trees or logs with camera traps we recorded the black-bellied pangolin opportunistically in Deng Deng National Park (East Region, Cameroon), a locality within the presumed distribution of the species within Cameroon. We obtained a low trapping rate of 0.063 events per 100 trap-days and the capture probability was one event recorded over 1,571 trap-days (0.0006 captures per day). Deploying camera traps that focus on fallen trees or logs could be explored as a method for detecting black-bellied pangolins.
Schizophrenia (SCZ) is one of the most severe mental disorders. Several elements involved in pathogenesis have been characterized recently. However, tools for diagnosis and risk prediction are limited. Elucidation of the underlying genomic and molecular mechanisms of SCA remains a challenge.
Objectives
In this study, we aimed to identify plasma biomarkers for SCZ using targeted metabolomics.
Methods
All enrolled patients were drug-free for at least 3 months prior to admission. Plasma from 31 SCZ patients and 70 matched controls were analyzed using the LC/MS- Api 4000 QTrap Sciex. A total of 188 targeted metabolites, including 21 amino acids, 21 biogenic amines and 145 lipids or lipid-related metabolites were analyzed. All data modeling and analysis is done using MetaboAnalyst 5.0
Results
There was no significant difference in the studied groups regarding BMI. Plasma Triglycerides, LDL-C, total proteins levels were significantly decreased in SCZ compared to controls. Heatmap identified 2 clusters with 25 significantly differentially expressed metabolites (FDR <0.05) between the drug-naïve group and the matched controls. The OPLS-DA score plot showed that the groups are clearly separated according to plasma phospholipids concentrations. Among these differential metabolites, the expression level of very long chain Phosphatidylcholines (PC 36 – PC p42) and acylcarnitines were significantly decreased in SCZ compared to controls, whereas sphingomyelin (SM) and lysoPC were significantly lower in drug-naive patients.
Conclusions
In this study, we found that plasma phospholipids were significantly dysregulated in the SCZ patients and could be a promising pathway to explore SCZ.
Pangolins have long been hunted for food and for their scales, but information on local hunting strategies in African range countries and the patterns of pangolin trafficking around sourcing areas is limited. Such knowledge is vital to inform monitoring approaches and conservation interventions. We administered questionnaires to 367 people from 20 villages around two national parks in Cameroon to gain insights into local hunting practices, and trading and trafficking patterns. We found that tracking was the preferred method of hunting pangolins. Trapping was also used, and setting traps on fallen logs and near burrows improved the likelihood of catching pangolins. The white-bellied pangolin Phataginus tricuspis was the most traded species, and the giant pangolin Smutsia gigantea was the most sought after. We found no evidence of trade in the black-bellied pangolin Phataginus tetradactyla. Most respondents stated that pangolin prices had increased over the last 5 years, and scarcity of pangolins was the primary reason given for this increase. We identified a pangolin scale trafficking network operating from rural communities to neighbouring cities. Although most respondents recognized that hunting pangolins is illegal, c. 30% indicated that they were engaged in pangolin hunting.
Much of our current understanding about novel coronavirus disease 2019 (COVID-19) comes from hospitalised patients. However, the spectrum of mild and subclinical disease has implications for population-level screening and control. Forty-nine participants were recruited from a group of 99 adults repatriated from a cruise ship with a high incidence of COVID-19. Respiratory and rectal swabs were tested by polymerase chain reaction (PCR) for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Sera were tested for anti-SARS-CoV-2 antibodies by enzyme-linked immunosorbent assay (ELISA) and microneutralisation assay. Symptoms, viral shedding and antibody response were examined. Forty-five participants (92%) were considered cases based on either positive PCR or positive ELISA for immunoglobulin G. Forty-two percent of cases were asymptomatic. Only 15% of symptomatic cases reported fever. Serial respiratory and rectal swabs were positive for 10% and 5% of participants respectively about 3 weeks after median symptom onset. Cycle threshold values were high (range 31–45). Attempts to isolate live virus were unsuccessful. The presence of symptoms was not associated with demographics, comorbidities or antibody response. In closed settings, incidence of COVID-19 could be almost double that suggested by symptom-based screening. Serology may be useful in diagnosis of mild disease and in aiding public health investigations.
By the end of their first year, infants can interpret many different types of complex dynamic visual events, such as caused-motion, chasing, and goal-directed action. Infants of this age are also in the early stages of vocabulary development, producing their first words at around 12 months. The present work examined whether there are meaningful individual differences in infants’ ability to represent dynamic causal events in visual scenes, and whether these differences influence vocabulary development. As part of the longitudinal Language 0–5 Project, 78 10-month-old infants were tested on their ability to interpret three dynamic motion events, involving (a) caused-motion, (b) chasing behaviour, and (c) goal-directed movement. Planned analyses found that infants showed evidence of understanding the first two event types, but not the third. Looking behaviour in each task was not meaningfully related to vocabulary development, nor were there any correlations between the tasks. The results of additional exploratory analyses and simulations suggested that the infants’ understanding of each event may not be predictive of their vocabulary development, and that looking times in these tasks may not be reliably capturing any meaningful individual differences in their knowledge. This raises questions about how to convert experimental group designs to individual differences measures, and how to interpret infant looking time behaviour.
There is lack of Cameroonian adult neuropsychological (NP) norms, limited knowledge concerning HIV-associated neurocognitive disorders in Sub-Saharan Africa, and evidence of differential inflammation and disease progression based on viral subtypes. In this study, we developed demographically corrected norms and assessed HIV and viral genotypes effects on attention/working memory (WM), learning, and memory.
Method:
We administered two tests of attention/WM [Paced Auditory Serial Addition Test (PASAT)-50, Wechsler Memory Scale (WMS)-III Spatial Span] and two tests of learning and memory [Brief Visuospatial Memory Test-Revised (BVMT-R), Hopkins Verbal Learning Test-Revised (HVLT-R)] to 347 HIV+ and 395 seronegative adult Cameroonians. We assessed the effects of viral factors on neurocognitive performance.
Results:
Compared to controls, people living with HIV (PLWH) had significantly lower T-scores on PASAT-50 and attention/WM summary scores, on HVLT-R total learning and learning summary scores, on HVLT-R delayed recall, BVMT-R delayed recall and memory summary scores. More PLWH had impairment in attention/WM, learning, and memory. Antiretroviral therapy (ART) and current immune status had no effect on T-scores. Compared to untreated cases with detectable viremia, untreated cases with undetectable viremia had significantly lower (worse) T-scores on BVMT-R total learning, BVMT-R delayed recall, and memory composite scores. Compared to PLWH infected with other subtypes (41.83%), those infected with HIV-1 CRF02_AG (58.17%) had higher (better) attention/WM T-scores.
Conclusions:
PLWH in Cameroon have impaired attention/WM, learning, and memory and those infected with CRF02_AG viruses showed reduced deficits in attention/WM. The first adult normative standards for assessing attention/WM, learning, and memory described, with equations for computing demographically adjusted T-scores, will facilitate future studies of diseases affecting cognitive function in Cameroonians.
In the current study, phage-exposed mimotopes as targets against tegumentary leishmaniasis (TL) were selected by means of bio-panning cycles employing sera of TL patients and healthy subjects, besides the immune stimulation of peripheral blood mononuclear cells (PBMCs) collected from untreated and treated TL patients and healthy subjects. The clones were evaluated regarding their specific interferon-γ (IFN-γ) and interleukin-4 (IL-4) production in the in vitro cultures, and selectivity and specificity values were calculated, and those presenting the best results were selected for the in vivo experiments. Two clones, namely A4 and A8, were identified and used in immunization protocols from BALB/c mice to protect against Leishmania amazonensis infection. Results showed a polarized Th1 response generated after vaccination, being based on significantly higher levels of IFN-γ, IL-2, IL-12, tumour necrosis factor-α (TNF-α) and granulocyte-macrophage colony-stimulating factor (GM-CSF); which were associated with lower production of specific IL-4, IL-10 and immunoglobulin G1 (IgG1) antibodies. Vaccinated mice presented significant reductions in the parasite load in the infected tissue and distinct organs, when compared with controls. In conclusion, we presented a strategy to identify new mimotopes able to induce Th1 response in PBMCs from TL patients and healthy subjects, and that were successfully used to protect against L. amazonensis infection.
Persistence of the embryonic “fifth aortic arch” in postnatal life is a rare, enigmatic – and at times controversial – condition, with variable anatomical forms and physiological consequences. First described in humans over 40 years ago by Van Praagh, the condition was labelled the “great pretender” by Gerlis 25 years later, because of its apparent propensity to mimic anatomically similar structures. Despite many subsequent case reports citing the condition, the true developmental origin of these structures remains unresolved, and has been the subject of debate among embryologists for more than a century. A persistent fifth aortic arch has been defined as an extrapericardial structure, arising from the ascending aorta opposite or proximal to the brachiocephalic artery, and terminating in the dorsal aorta or pulmonary arteries via a persistently patent arterial duct. This description may therefore encompass various anatomical forms, such as a unilateral double-lumen aortic arch, an unrestrictive aortopulmonary shunt, or a critical vascular channel for either the systemic or pulmonary circulation. The physiological properties of these vessels, such as their response to prostaglandins, may also be unpredictable. In this article, we demonstrate a number of cases that fulfil the contemporary definition of “persistent fifth aortic arch” while acknowledging the embryological controversies associated with this term. We also outline the key diagnostic features, particularly with respect to the use of new cross-sectional imaging techniques.
The University of Georgia (USA) is partnering with the University of Padova (Italy) for a dual Master’s degree program in sustainable agriculture, promoting collaboration on some of the biggest challenges facing agriculture today. This innovative program which was launched during 2016 provides students with outstanding training and a unique opportunity to learn about the challenges, opportunities, and leading edges of precision agriculture on another continent – an experience which will serve graduates well when they enter the job market in an increasingly global economy. This paper presents the goals of the program, the curriculum, and describes the opportunities available to prospective students. In addition it describes the process of developing the dual degree which can be used as guide by others wishing to develop similar programs.
At Sheep Mountain Bog, near Missoula, Montana, a late-glacial tephra, that probably fell in late summer, is preserved as an 8-mm-thick graded bed overlain by another 8 mm of redeposited ash mixed with lake deposits. Sediment surrounding the ash was 14C dated to about 11,200 yr B.P. Electron-microprobe analyses of the volcanic glass and hornblende phenocrysts from this ash layer indicate that they are similar in major-element chemistry to those of Glacier Peak layer G previously considered to be about 12,000 yr old or older.
The Glacier Peak tephra beds are among the most widespread and arguably some of the most important late Pleistocene chronostratigraphic markers in western North America. These beds represent a series of closely-spaced Plinian and sub-Plinian eruptions from Glacier Peak, Washington. The two most widespread beds, Glacier Peak ‘G’ and ‘B’, are reliably distinguished by their glass major and trace element abundances. These beds are also more broadly distributed than previously considered, covering at least 550,000 and 260,000 km2, respectively. A third bed, the Irvine bed, known only from southern Alberta, is similar in its major-element composition to the Glacier Peak G bed, but it shows considerable differences in trace element concentrations. The Irvine bed is likely considerably older than the G and B tephras and probably records an additional Plinian eruption, perhaps also from Glacier Peak but from a different magma than G through B. A review of the published radiocarbon ages, new ages in this study, and consideration in a Bayesian framework suggest that the widespread G and B beds are several hundred years older than widely assumed. Our revised age is about 11,600 14C yr BP or a calibrated age (at 2 sigma) of 13,710–13,410 cal yr BP.
The recent history of catastrophic disturbances in forests was reconstructed at Mount Rainier National Park. Basic data were ages of trees based on ring counts of early seral conifer species and maps of age-class boundaries from field work and aerial photographs. Maps illustrate age classes of the forests and show disturbances from fires, snow avalanches, and lahars (volcanic mudflows). Fires are by far the most important major disturbers, followed by snow avalanches and lahars. Fires over 250 ha in size are called fire events. Burns over 1000 ha, which may have been one fire or a series of fires within a short time, are called fire episodes. Important fire events or episodes occurred in the years 1230, 1303, 1403, 1503, 1628, 1688, 1703, 1803, 1825, 1856, 1858, 1872, 1886, 1894, 1930, and 1934 A.D. The largest fire episode was in 1230; it affected approximately 47% of the forests in the park. The majority of the forests are over 350 yr old, and several stands are over 1000 yr old. Stands 350 yr and 100 to 200 yr in age are the most extensive age classes in the park. Three fire frequency indices are compared. None describe fire frequency at Mount Rainier well. Natural fire rotation was estimated at about 434 yr. All but two episodes of major fires since 1300 A.D. correspond well with major droughts reconstructed for locations east of the Cascade Range crest. Impacts of humans on the disturbance regime may have increased the frequency of fire in the 1850–1900 period, followed by a decrease in frequency after 1900. Fuel build-up as a result of fire suppression should have no significant impact on fire frequency, since fires are relatively infrequent and fuels accumulate naturally.
The synthesis of paleoclimatic archives provided by loess and alluvial sequences of central Argentina has been hindered by the lack of a cohesive lithostratigraphic framework extending across the Chaco–Pampean plains and catchments of the Rios Desaguadero, Colorado, and Negro. This condition originates in part from the dearth of absolute chronological controls. The occurrence of discrete tephra layers across this region may provide an opportunity to address this deficiency if a tephrochronological framework can be established. The potential of such a project is assessed within the context of a pilot study constrained within alluvial sequences of central western Argentina proximal to potential source vents in the Southern Volcanic Zone. The intersite discrimination and correlation of tephra layers on a geochemical basis is examined, with indirect chronological control for the eruption of each generated by optical dating. Alluvial sediments on either side of each of five tephra units at a type site were dated using the optically stimulated luminescence of fine-silt-sized quartz, thus providing an age control on each tephra (ca. 24,000, 30,000, 32,000, 39,000, and 48,000 yr). The geochemical composition of each tephra was derived. Using these data, tephra layers at other sites in the study area were geochemically analyzed and in instances of statistical concordance in major oxide structure, correlated to the type site and therefore ascribed ages. This methodology identified a further sixth volcanic event between ca. 24,000 and 30,000 yr not registered by type-site tephras. The extension of this initial tephrochronological framework beyond the alluvial sequences of central western Argentina is encouraged by the occurrence of geochemically distinct tephra verified and dated in this study.
To better understand the regional tephra stratigraphy and chronology of northern Nevada and southern Oregon, tephras in archived cores, taken as part of the Steens Mountain Prehistory Project from four lakes, Diamond Pond, Fish and Wildhorse lakes in southeastern Oregon and Blue Lake in northwestern Nevada, were reexamined using more advanced electron microprobe analytical technology. The best preserved and most complete core from Fish Lake along with Wildhorse Lake hosted two tephras from Mt. Mazama (Llao Rock and the Climactic Mazama), a mid-Holocene basaltic tephra from Diamond Craters, Oregon, two Medicine Lake tephras and an unexpected late Holocene Chaos Crags (Mt. Lassen volcanic center) tephra which was also found in the other lakes. Blue Lake was the only lake that hosted a Devils Hill tephra from the Three Sisters volcano in west central Oregon. Another tephra from the Three Sisters Volcano previously reported in sediments of Twin Lakes in NE Oregon, has now been confirmed as Rock Mesa tephra. The Chaos Crags, Devils Hill and Rock Mesa tephras are important late Holocene stratigraphic markers for central and eastern Oregon and northwestern Nevada.
Tephra layers form important chronostratigraphic marker horizons within the Wisconsinan loess-paleosol stratigraphy of the Palouse, Washington State. This paper details the presence of disseminated glass shards of the Trego Hot Springs tephra that has not previously been recognized within the Palouse or environs. The tephra provides additional chronological control for the regional Late Wisconsinan stratigraphy and raises the possibility of correlating the paleoclimatic proxy record contained within the Palouse to those of lake sequences in Oregon, California, and Nevada.
A Glacier Peak tephra has been found in the mid-Holocene sediment records of two subalpine lakes, Frozen Lake in the southern Coast Mountains and Mount Barr Cirque Lake in the North Cascade Mountains of British Columbia, Canada. The age–depth relationship for each lake suggests an age of 5000–5080 14C yr B.P. (5500–5900 cal yr B.P.) for the eruption which closely approximates the estimated age (5100–5500 14C yr B.P.) of the Dusty Creek tephra assemblage found near Glacier Peak. The tephra layer, which has not been reported previously from distal sites and was not readily visible in the sediments, was located using contiguous sampling, magnetic susceptibility measurements, wet sieving, and light microscopy. The composition of the glass in pumice fragments was determined by electron microprobe analysis and used to confirm the probable source of this mid-Holocene tephra layer. Using the same methods, the A.D. 1481–1482 Mount St. Helens We tephra layer was identified in sediments from Dog Lake in southeastern British Columbia, suggesting the plume drifted further north than previously thought. This high-resolution method for identifying tephra layers in lake sediments, which has worldwide application in tephrachronologic/paleoenvironmental studies, has furthered our knowledge of the timing and airfall distribution of Holocene tephras from two important Cascade volcanoes.
Epidemiological studies suggest a higher prevalence of congenital malformations in children conceived through assisted reproductive technologies. There are a few studies that address CHD specifically and most have examined data from registries. We examined the relationship between CHD and assisted conception using data collected in a specialist paediatric cardiac service in the United Kingdom.
Between April, 2010 and July, 2011, the parents of children attending paediatric cardiology clinics at the Royal Brompton Hospital, London, were invited to complete a questionnaire that enquired about the nature of their child’s conception, the route for their original referral, and a number of potential confounding exposures. “Cases” were defined as children diagnosed with one or more carefully defined CHDs and “controls” as those with normal hearts.
Of 894 new attendees with complete data, half of them were cases (n=410, 45.9%). The overall prevalence of assisted conception was 5.4% (n=44). Logistic regression analysis demonstrated a non-significant increase in the crude odds for the use of assisted reproduction (odds ratio 1.21, 95% confidence interval 0.66–2.22) in this group. After adjustment for gestation, parity, year of birth, and maternal age, the odds ratio reduced (odds ratio 0.95, 95% confidence interval 0.48–1.88). Increased rates of assisted conception were observed in a number of CHD subgroups, although no significant differences were found.
These findings do not suggest an overall association between CHD and assisted reproduction in this population.
